ABSTRACT: Data on temperature-time curves measured at chimney-roof penetration are reported here. The tests were performed in different configurations in order to reproduce all the possible conditions in which a chimney may operate. To do this, a chimney was installed in a corner test structure and in an axi-simmetric test structure, and in three roofs of different thickness, thermal resistance, and layers position. The space between chimney and roof was left open, sealed with metal sheets, sealed with insulating panels, and filled of insulating material respectively.

Magnetization curves of haemoproteins measured by low-temperature magnetic-circular-dichroism spectroscopy.

Project description:The magnetic-circular-dichroism (m.c.d.) spectra of methymyoglobin cyanide and oxidized horse heart cytochrome c were measured in the region of the Soret band over a range of temperatures from 1.5 to 50 K and in fields from 0 to 5T. A similar study has been made with reduced bovine heart cytochrome c oxidase, which contains one high-spin ferrous haem, namely a3. M.c.d. magnetization curves characteristic of an isolated Kramer's ground state with spin S = 1/2. These curves contrast with the magnetization curve of the high-spin ferrous haem with spin S = 2. The electronic ground state of the latter compound contains zero-field components that are thermally accessible over the temperature range of the experiment. Hence the magnetization curves are a complex nested set. The magnetization curves of the S = 1/2 proteins were analysed and it is shown that it is possible to make estimates of the ground-state g-factors even in the presence of rhombic anisotropy, provided that some knowledge of the polarizations of the electronic transitions is available. The striking difference between the m.c.d. magnetization curves of a simple S = 1/2 paramagnet and magnetically complex ground state should prove extremely useful when m.c.d. spectroscopy is sued to probe the magentic properties of metal centres in proteins, and should have wide application beyond the field of haemoproteins.

Reconstructing time-to-event data from published Kaplan-Meier curves.

Project description:Hazard ratios can be approximated by data extracted from published Kaplan-Meier curves. Recently, this curve approach has been extended beyond hazard-ratio approximation with the capability of constructing time-to-event data at the individual level. In this article, we introduce a command, ipdfc, to implement the reconstruction method to convert Kaplan-Meier curves to time-to-event data. We give examples to illustrate how to use the command.

Nonstationary Temperature-Duration-Frequency curves.

Project description:Persistent extreme heat events are of growing concern in a climate change context. An increase in the intensity, frequency and duration of heat waves is observed in several regions. Temperature extremes are also influenced by global-scale modes of climate variability. Temperature-Duration-Frequency (TDF) curves, which relate the intensity of heat events of different durations to their frequencies, can be useful tools for the analysis of heat extremes. To account for climate external forcings, we develop a nonstationary approach to the TDF curves by introducing indices that account for the temporal trend and teleconnections. Nonstationary TDF modeling can find applications in adaptive management in the fields of health care, public safety and energy production. We present a one-step method, based on the maximization of the composite likelihood of observed heat extremes, to build the nonstationary TDF curves. We show the importance of integrating the information concerning climate change and climate oscillations. In an application to the province of Quebec, Canada, the influence of Atlantic Multidecadal Oscillations (AMO) on heat events is shown to be more important than the temporal trend.

Orbital roof fracture and orbital cellulitis secondary to halo pin penetration: case report.

Project description:Study Design Case report. Objective To report and discuss a rare complication after a patient was treated conservatively with a halo vest. Methods A 51-year-old man sustained a hangman's injury of the C2 vertebra following a motor vehicle collision. He was treated conservatively in a halo vest appliance and following mobilization was discharged from the hospital. Two weeks after discharge, the patient presented to the emergency department complaining of proptosis, ptosis, diplopia, and pin loosening. He was readmitted to the hospital, the halo vest was removed, and urgent imaging studies including computed tomography scan and magnetic resonance imaging were performed. They revealed that one of the halo pins had penetrated the orbital roof with active infection of the extraocular soft tissues. In consultation with the ophthalmologist, he was treated conservatively with antibiotics for 10 days. Results His ophthalmologic complaints resolved gradually and his eye returned to normal appearance and function. In the meantime, he was immobilized in a sterno-occipital mandibular immobilizer brace. Conclusion Though rare, penetrating injuries after cranial pin insertion can occur. Halo devices must be applied by, or under close supervision of, experienced personnel to avoid such complications, and halo vests should be reviewed frequently to detect such incidents early.

Particle Diffusivity and Free-Energy Profiles in Hydrogels from Time-Resolved Penetration Data.

Project description:A combined experimental and theoretical method to simultaneously determine diffusivity and free-energy profiles of particles that penetrate into inhomogeneous hydrogel systems is presented. As the only input, arbitrarily normalized concentration profiles from fluorescence intensity data of labeled tracer particles for different penetration times are needed. The method is applied to dextran molecules of varying size that penetrate into hydrogels of polyethylene-glycol chains with different lengths that are covalently cross-linked by hyperbranched polyglycerol hubs. Extracted dextran bulk diffusivities agree well with fluorescence correlation spectroscopy data obtained separately. Empirical scaling laws for dextran diffusivities and free energies inside the hydrogel are identified as a function of the dextran mass. An elastic free-volume model that includes dextran as well as polyethylene-glycol linker flexibility quantitively describes the repulsive dextran-hydrogel interaction free energy, which is of steric origin, and furthermore suggests that the hydrogel mesh-size distribution is rather broad and particle penetration is dominated by large hydrogel pores. Particle penetration into hydrogels for steric particle-hydrogel interactions is thus suggested to be governed by an elastic size-filtering mechanism that involves the tail of the hydrogel pore-size distribution.

Handling temperature bursts reaching 464°C: different microbial strategies in the sisters peak hydrothermal chimney.

Project description:The active venting Sisters Peak (SP) chimney on the Mid-Atlantic Ridge holds the current temperature record for the hottest ever measured hydrothermal fluids (400°C, accompanied by sudden temperature bursts reaching 464°C). Given the unprecedented temperature regime, we investigated the biome of this chimney with a focus on special microbial adaptations for thermal tolerance. The SP metagenome reveals considerable differences in the taxonomic composition from those of other hydrothermal vent and subsurface samples; these could be better explained by temperature than by other available abiotic parameters. The most common species to which SP genes were assigned were thermophilic Aciduliprofundum sp. strain MAR08-339 (11.8%), Hippea maritima (3.8%), Caldisericum exile (1.5%), and Caminibacter mediatlanticus (1.4%) as well as to the mesophilic Niastella koreensis (2.8%). A statistical analysis of associations between taxonomic and functional gene assignments revealed specific overrepresented functional categories: for Aciduliprofundum, protein biosynthesis, nucleotide metabolism, and energy metabolism genes; for Hippea and Caminibacter, cell motility and/or DNA replication and repair system genes; and for Niastella, cell wall and membrane biogenesis genes. Cultured representatives of these organisms inhabit different thermal niches; i.e., Aciduliprofundum has an optimal growth temperature of 70°C, Hippea and Caminibacter have optimal growth temperatures around 55°C, and Niastella grows between 10 and 37°C. Therefore, we posit that the different enrichment profiles of functional categories reflect distinct microbial strategies to deal with the different impacts of the local sudden temperature bursts in disparate regions of the chimney.

Deep Learning-Based Real-Time Auto Classification of Smartphone Measured Bridge Vibration Data.

Project description:In this study, a simple and customizable convolution neural network framework was used to train a vibration classification model that can be integrated into the measurement application in order to realize accurate and real-time bridge vibration status on mobile platforms. The inputs for the network model are basically the multichannel time-series signals acquired from the built-in accelerometer sensor of smartphones, while the outputs are the predefined vibration categories. To verify the effectiveness of the proposed framework, data collected from long-term monitoring of bridge were used for training a model, and its classification performance was evaluated on the test set constituting the data collected from the same bridge but not used previously for training. An iOS application program was developed on the smartphone for incorporating the trained model with predefined classification labels so that it can classify vibration datasets measured on any other bridges in real-time. The results justify the practical feasibility of using a low-latency, high-accuracy smartphone-based system amid which bottlenecks of processing large amounts of data will be eliminated, and stable observation of structural conditions can be promoted.

Time-to-event data with time-varying biomarkers measured only at study entry, with applications to Alzheimer's disease.

Project description:Relating time-varying biomarkers of Alzheimer's disease to time-to-event using a Cox model is complicated by the fact that Alzheimer's disease biomarkers are sparsely collected, typically only at study entry; this is problematic since Cox regression with time-varying covariates requires observation of the covariate process at all failure times. The analysis might be simplified by using study entry as the time origin and treating the time-varying covariate measured at study entry as a fixed baseline covariate. In this paper, we first derive conditions under which using an incorrect time origin of study entry results in consistent estimation of regression parameters when the time-varying covariate is continuous and fully observed. We then derive conditions under which treating the time-varying covariate as fixed at study entry results in consistent estimation. We provide methods for estimating the regression parameter when a functional form can be assumed for the time-varying biomarker, which is measured only at study entry. We demonstrate our analytical results in a simulation study and apply our methods to data from the Rush Religious Orders Study and Memory and Aging Project and data from the Alzheimer's Disease Neuroimaging Initiative.

DAG expression: high-throughput gene expression analysis of real-time PCR data using standard curves for relative quantification.

Project description:BackgroundReal-time quantitative PCR (qPCR) is still the gold-standard technique for gene-expression quantification. Recent technological advances of this method allow for the high-throughput gene-expression analysis, without the limitations of sample space and reagent used. However, non-commercial and user-friendly software for the management and analysis of these data is not available.ResultsThe recently developed commercial microarrays allow for the drawing of standard curves of multiple assays using the same n-fold diluted samples. Data Analysis Gene (DAG) Expression software has been developed to perform high-throughput gene-expression data analysis using standard curves for relative quantification and one or multiple reference genes for sample normalization. We discuss the application of DAG Expression in the analysis of data from an experiment performed with Fluidigm technology, in which 48 genes and 115 samples were measured. Furthermore, the quality of our analysis was tested and compared with other available methods.ConclusionsDAG Expression is a freely available software that permits the automated analysis and visualization of high-throughput qPCR. Voltage-induced penetration effect in liquid metals at room temperature.

Project description:Room-temperature liquid metal is discovered to be capable of penetrating through macro- and microporous materials by applying a voltage. The liquid metal penetration effects are demonstrated in various porous materials such as tissue paper, thick and fine sponges, fabrics, and meshes. The underlying mechanism is that the high surface tension of liquid metal can be significantly reduced to near-zero due to the voltage-induced oxidation of the liquid metal surface in a solution. It is the extremely low surface tension and gravity that cause the liquid metal to superwet the solid surface, leading to the penetration phenomena. These findings offer new opportunities for novel microfluidic applications and could promote further discovery of more exotic fluid states of liquid metals.
