The second century CE Roman watermills of Barbegal: Unraveling the enigma of one of the oldest industrial complexes.

ABSTRACT: The second century CE Roman watermill complex of Barbegal, France, is regarded as one of the first industrial complexes in human history. The 16 water wheels are no longer extant as all woodwork has decayed. However, carbonate deposits precipitated from water during operation of the mills forming casts on the woodwork. These casts are preserved in fragments and provide unique insights into the frequency of use and maintenance of the mills, and even into the structure of the water wheel chambers. Stable isotope time series of carbonate deposits reveal that the mill activity was regularly interrupted for several months. This strongly suggests that the mill complex was not used for a steady supply of flour to a major population center, as previously thought, but likely served to produce nonperishable hardtack for the nearby harbors.

Reconstructing the hydraulics of the world's first industrial complex, the second century CE Barbegal watermills, France.

Project description:The Barbegal watermill complex, a unique cluster of 16 waterwheels in southern France, was the first known attempt in Europe to set up an industrial-scale complex of machines during the culmination of Roman Civilization in the second century CE. Little is known about the state of technological advance in this period, especially in hydraulics and the contemporary diffusion of knowledge. Since the upper part of the Barbegal mill complex has been destroyed and no traces of the wooden machinery survived, the mode of operation of these mills has long remained elusive. Carbonate incrustations that formed on the woodwork of the mills were used to reconstruct its structure and function, revealing a sophisticated hydraulic setup unique in the history of water mills. The lower mills used an elbow shaped flume to bring water onto overshot millwheels. This flume was specially adapted to the small water basins and serial arrangement of the mills on the slope. Carbonate deposits from ancient water systems are therefore a powerful tool in archaeological reconstructions and provide tantalizing insights into the skills of Roman engineers during a period of history that is the direct predecessor of our modern civilization.

The parisite-(Ce) enigma: challenges in the identification of fluorcarbonate minerals.

Project description:A multi-methodological study was conducted in order to provide further insight into the structural and compositional complexity of rare earth element (REE) fluorcarbonates, with particular attention to their correct assignment to a mineral species. Polycrystals from La Pita Mine, Municipality de Maripí, Boyacá Department, Colombia, show syntaxic intergrowth of parisite-(Ce) with röntgenite-(Ce) and a phase which is assigned to B 3 S 4 (i.e., bastnäsite-3-synchisite-4; still unnamed) fluorcarbonate. Transmission electron microscope (TEM) images reveal well-ordered stacking patterns of two monoclinic polytypes of parisite-(Ce) as well as heavily disordered layer sequences with varying lattice fringe spacings. The crystal structure refinement from single crystal X-ray diffraction data - impeded by twinning, complex stacking patterns, sequential and compositional faults - indicates that the dominant parisite-(Ce) polytype M 1 has space group Cc. Parisite-(Ce), the B 3 S 4 phase and röntgenite-(Ce) show different BSE intensities from high to low. Raman spectroscopic analyses of parisite-(Ce), the B 3 S 4 phase and röntgenite-(Ce) reveal different intensity ratios of the three symmetric CO3 stretching bands at around 1100 cm-1. We propose to non-destructively differentiate parisite-(Ce) and röntgenite-(Ce) by their 1092 cm-1 / 1081 cm-1 ν1(CO3) band height ratio.

Evolutionary history of carbon monoxide dehydrogenase/acetyl-CoA synthase, one of the oldest enzymatic complexes.

Project description:Carbon monoxide dehydrogenase/acetyl-CoA synthase (CODH/ACS) is a five-subunit enzyme complex responsible for the carbonyl branch of the Wood-Ljungdahl (WL) pathway, considered one of the most ancient metabolisms for anaerobic carbon fixation, but its origin and evolutionary history have been unclear. While traditionally associated with methanogens and acetogens, the presence of CODH/ACS homologs has been reported in a large number of uncultured anaerobic lineages. Here, we have carried out an exhaustive phylogenomic study of CODH/ACS in over 6,400 archaeal and bacterial genomes. The identification of complete and likely functional CODH/ACS complexes in these genomes significantly expands its distribution in microbial lineages. The CODH/ACS complex displays astounding conservation and vertical inheritance over geological times. Rare intradomain and interdomain transfer events might tie into important functional transitions, including the acquisition of CODH/ACS in some archaeal methanogens not known to fix carbon, the tinkering of the complex in a clade of model bacterial acetogens, or emergence of archaeal-bacterial hybrid complexes. Once these transfers were clearly identified, our results allowed us to infer the presence of a CODH/ACS complex with at least four subunits in the last universal common ancestor (LUCA). Different scenarios on the possible role of ancestral CODH/ACS are discussed. Despite common assumptions, all are equally compatible with an autotrophic, mixotrophic, or heterotrophic LUCA. Functional characterization of CODH/ACS from a larger spectrum of bacterial and archaeal lineages and detailed evolutionary analysis of the WL methyl branch will help resolve this issue.

Climate change, society, and pandemic disease in Roman Italy between 200 BCE and 600 CE.

Project description:Records of past societies confronted with natural climate change can illuminate social responses to environmental stress and environment-disease connections, especially when locally constrained high-temporal resolution paleoclimate reconstructions are available. We present a temperature and precipitation reconstruction for ~200 BCE to ~600 CE, from a southern Italian marine sedimentary archive-the first high-resolution (~3 years) climate record from the heartland of the Roman Empire, stretching from the so-called Roman Climate Optimum to the Late Antique Little Ice Age. We document phases of instability and cooling from ~100 CE onward but more notably after ~130 CE. Pronounced cold phases between ~160 to 180 CE, ~245 to 275 CE, and after ~530 CE associate with pandemic disease, suggesting that climate stress interacted with social and biological variables. The importance of environment-disease dynamics in past civilizations underscores the need to incorporate health in risk assessments of climate change.

Unraveling the enigma: progress towards understanding the coronin family of actin regulators.

Project description:Coronins are a conserved family of actin cytoskeleton regulators that promote cell motility and modulate other actin-dependent processes. Although these proteins have been known for 20 years, substantial progress has been made in the past 5 years towards their understanding. In this review, we examine this progress, place it into the context of what was already known, and pose several questions that remain to be addressed. In particular, we cover the emerging consensus about the role of Type I coronins in coordinating the function of Arp2/3 complex and ADF/cofilin proteins. This coordination plays an important role in leading-edge actin dynamics and overall cell motility. Finally, we discuss the roles played by the more exotic coronins of the Type II and III classes in cellular processes away from the leading edge.

A multidisciplinary study unveils the nature of a Roman ink of the I century AD.

Project description:A multi-instrumental approach combining highly sensitive Synchrotron Radiation-based techniques was used to provide information on the real composition of a dry black ink powder found in a bronze inkwell of the first century AD. The presence of Pb, Cu and Fe in the powder, revealed by XRF and ICP-OES data, leads to raise several hypotheses on their origin. The inkpot and its lid were also investigated by Hand-Held XRF, revealing a bronze alloy (Cu-Sn) with a certain amount of Fe and Pb. The lid was found to be particularly enriched in lead. XRPD, XAS and FTIR measurements showed a substantial presence of silicates and common clay minerals in the ink along with cerussite and malachite, Pb and Cu bearing-carbonates, respectively. These evidences support the hypothesis of an important contamination of the ink sample by the burial environment (soil) and the presence of degradation products of the bronze inkpot. The combined use of IR, Raman, and GC-MS evidenced that the black ink is mainly composed of amorphous carbon deriving from the combustion of organic material mixed with a natural binding agent, Arabic gum.

Liquid-Liquid Phase Separation: Unraveling the Enigma of Biomolecular Condensates in Microbial Cells.

Project description:Numerous examples of microbial phase-separated biomolecular condensates have now been identified following advances in fluorescence imaging and single molecule microscopy technologies. The structure, function, and potential applications of these microbial condensates are currently receiving a great deal of attention. By neatly compartmentalizing proteins and their interactors in membrane-less organizations while maintaining free communication between these macromolecules and the external environment, microbial cells are able to achieve enhanced metabolic efficiency. Typically, these condensates also possess the ability to rapidly adapt to internal and external changes. The biological functions of several phase-separated condensates in small bacterial cells show evolutionary convergence with the biological functions of their eukaryotic paralogs. Artificial microbial membrane-less organelles are being constructed with application prospects in biocatalysis, biosynthesis, and biomedicine. In this review, we provide an overview of currently known biomolecular condensates driven by liquid-liquid phase separation (LLPS) in microbial cells, and we elaborate on their biogenesis mechanisms and biological functions. Additionally, we highlight the major challenges and future research prospects in studying microbial LLPS.

Non-conventional Ce:YAG nanostructures via urea complexes.

Project description:Ce:YAG nanostructures (Ce:YAG = Cerium in Yttrium Aluminium Garnet), easy to control and shape, have been prepared via templating approach using natural and synthetic materials (i.e. paper, cotton wool and glass wool) previously soaked with a gel-like metals precursor and then thermally treated to achieve the wished morphology. The final material, otherwise difficult to process, can be easily moulded, it is lightweight, portable and forms, at the nanoscale, homogeneous layers of interconnected but not agglomerated nanoparticles (15 ± 5 nm). Using the same synthetic route, called Urea-Glass-Route, but in absence of a template, extremely pure Ce:YAG nanoparticle (45 ± 5 nm) can be also prepared, highly crystalline and well-defined in size and shape. Both structural and optical properties of the final materials were investigated, showing high optical quality. The support allows the production of a multifunctional material with mouldable shape and potential lighting application for large structures combining the strength, chemical durability, fire resistance, and translucency of glass fibres. Last, but not least, the synthetic path also allows an easy scaling up of the process: the first, key step for practical application of nanosized rare-earth doped YAG on large scale.

Revisiting Lebedev's one-century old experiment.

Project description:One hundred years ago, world-famous scientist A. A. Lebedev performed a set of classical measurements on annealed optic crown glasses. He found that these glasses exhibited characteristic endothermic effects in a particular temperature range. To explain these phenomena, Lebedev proposed a hypothesis that the glasses contain tiny quartz crystals. This initial hypothesis was quickly disapproved, and the origin of the endothermic effect of glasses remains an unsolved puzzle. This work uses recently proposed nanoflake model of silica glass structure to explain the endothermic effect of various glasses. The new model differs from the popular continuous random network theory in that it emphasizes the medium-range ordering structure of glasses. According to the nanoflake based theory, the endothermic effect of glasses is caused by the transition from ordered one-dimensional structures into disordered structure in glasses. The new theory also predicts that the temperature range of the endothermic effect is dependent on both glass composition and cooling rates during glass formation.

Alzheimer's disease: the challenge of the second century.

Project description:Alzheimer's disease (AD) was first described a little more than 100 years ago. It is the most common cause of dementia with an estimated prevalence of 30 million people worldwide, a number that is expected to quadruple in 40 years. There currently is no effective treatment that delays the onset or slows the progression of AD. However, major scientific advances in the areas of genetics, biochemistry, cell biology, and neuroscience over the past 25 years have changed the way we think about AD. This review discusses some of the challenges to translating these basic molecular and cellular discoveries into clinical therapies. Current information suggests that if the disease is detected before the onset of overt symptoms, it is possible that treatments based on knowledge of underlying pathogenesis can and will be effective.
