Synthesis of 5-Methylene-2-pyrrolones.

ABSTRACT: A facile, one-pot synthetic method for the synthesis of 5-methylene-2-pyrrolones (5MPs) from inexpensive furfuryl acetate is described. Bromine oxidation and trapping of the in situ generated 1,4-dicarbonyl compound by a primary amine provided the corresponding 5MPs in 50-69% yield.

Thiol Specific and Tracelessly Removable Bioconjugation via Michael Addition to 5-Methylene Pyrrolones.

Project description:5-Methylene pyrrolones (5MPs) are highly thiol-specific and tracelessly removable bioconjugation tools. 5MPs are readily prepared from primary amines in one step. 5MPs exhibit significantly improved stability under physiologically relevant conditions and cysteine specificity compared to commonly used analogues, maleimides. Michael addition of thiol to 5MPs occurs rapidly, cleanly, and does not generate a stereocenter. The conjugates efficiently release thiols via retro-Michael reaction in alkaline buffer (pH 9.5) or via thiol exchange at pH 7.5. This unique property makes 5MPs valuable for the controlled release of conjugated cargo and temporary thiol protection. The utilization of 5MPs for protein immobilization and pull-down of active complexes is illustrated using E. coli. acetohydroxyacid synthase isozyme I.

Cysteine-specific protein multi-functionalization and disulfide bridging using 3-bromo-5-methylene pyrrolones.

Project description:Many reagents have been developed for cysteine-specific protein modification. However, few of them allow for multi-functionalization of a single Cys residue and disulfide bridging bioconjugation. Herein, we report 3-bromo-5-methylene pyrrolones (3Br-5MPs) as a simple, robust, and versatile class of reagents for cysteine-specific protein modification. These compounds can be facilely synthesized via a one-pot mild reaction and they show comparable tagging efficiency but higher cysteine specificity than the maleimide counterparts. The addition of cysteine to 3Br-5MPs generates conjugates that are amenable to secondary addition by another thiol or cysteine, making 3Br-5MPs valuable for multi-functionalization of a single cysteine and disulfide bridging bioconjugation. The labeling reaction and subsequent treatments are mild enough to produce stable and active protein conjugates for biological applications.

Organocatalytic Synthesis of Methylene-Bridged N-Heterobiaryls.

Project description:A one-step synthesis of 1,1'- and 2,2'-methylene-bridged N-heterobiaryls directly from the corresponding N-heterocycles in a reaction with methylmagnesium chloride in the presence of catalytic amounts of N,N,N',N'-tetramethylethylenediamine under thermal and microwave conditions is reported. The split-and-merge methylenation of 2,2'-N-heterobiaryls and the direct ortho-alkylation of quinoline and isoquinoline with Grignard reagents have also been developed. Mechanistic studies identified several intermediates and provided insight into the formation and roles of magnesium hydride species in the process.

Project description:Several heterocycles such as furanones, pyrrolones, and indolizines, which are of pharmacological importance, are easily accessed via the Pt(II)-catalyzed heterocyclization/1,2-migration of propargylic ketols or hydroxy imine derivatives. This method sidesteps the challenges of traditional heteroaromatic oxygenation strategies such as regioselectivity and functional group tolerance in the syntheses of these heterocycles.

Synthesis of TiO2NWS@AuNPS Composite Catalyst for Methylene Blue Removal.

Project description:In this article, HBP-NH₂-modified titania nanowire (TiO2NWS)-decorated Au nanoparticles (TiO2NWS@AuNPS) were synthesized by one-step method. The role of HBP-NH₂ concentration in the formation of TiO2NWS was investigated. The fineness and uniformity of pure TiO2NWS were enhanced by absorbed amino groups from amino-terminated hyperbranched polymer (HBP-NH₂). The morphology and crystal structure of TiO2NWS and TiO2NWS@AuNPS were examined by transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fournier transform infrared (FTIR) spectroscopy. The chemical states of gold, titanium and oxygen were analyzed by X-ray photoelectron spectroscopy (XPS). The results showed that at the concentration of HBP-NH₂ 100 g/L, the mean diameter of TiO2NWS was nearly 72 nm and Au nanoparticles were uniformly distributed on the surface of TiO2NWS. The presence of AuNPS improved the photocatalytic properties of TiO2NWS under UV light irradiation. The Au load was believed to improve the utilization rate of the photoelectron and activated the adsorbed oxygen. The obtained TiO2NWS@AuNPS decomposed 99.6% methylene blue (MB) after 300 min when subjected to UV light irradiation. After five cycles of the catalyzing process, the TiO2NWS@AuNPS still retained over 90% of its catalytic ability for MB. The Au deposition was found responsible for the high catalytic activity of TiO2NWS@AuNPS.

Bifurcated synthesis of methylene-lactone- and methylene-lactam-fused spirolactams via electrophilic amide allylation of γ-phenylthio-functionalized γ-lactams.

Project description:New synthetic methods for spirolactams bearing an α-methylene-γ-butyrolactone or its analogous methylene-lactam have been developed. The allylation of γ-phenylthio-functionalized γ-lactams with 2-(acetoxy)methyl acrylamides was accomplished by using 2.5 equivalents of NaH to give the corresponding adducts in excellent yields. The remaining phenylthio group was substituted with a hydroxy group by treatment with CuBr, and the resulting γ-hydroxyamides were cyclized under acidic conditions to afford the corresponding methylene-lactam-fused spirolactams in high yields. On the other hand, methylene-lactone-fused spirolactams could be delivered from the allyl adducts in high yields through a sequential N-Boc protection/desulfinative lactonization.

Synthesis and Antioxidant Activity of New Catechol Thioethers with the Methylene Linker.

Project description:Novel catechol thio-ethers with different heterocyclic substituents at sulfur atom were prepared by reacting 3,5-di-tert-butyl-6-methoxymethylcatechol with functionalized thiols under acidic conditions. A common feature of compounds is a methylene bridge between the catechol ring and thioether group. Two catechols with the thio-ether group, bound directly to the catechol ring, were also considered to assess the effect of the methylene linker on the antioxidant properties. The crystal structures of thio-ethers with benzo-thiazole moieties were established by single-crystal X-ray analysis. The radical scavenging and antioxidant activities were determined using 2,2'-diphenyl-1-picrylhydrazyl radical test, ABTS∙+, CUPRAC (TEAC) assays, the reaction with superoxide radical anion generated by xanthine oxidase (NBT assay), the oxidative damage of the DNA, and the process of lipid peroxidation of rat liver (Wistar) homogenates in vitro. Most catechol-thioethers exhibit the antioxidant effect, which varies from mild to moderate depending on the model system. The dual anti/prooxidant activity characterizes compounds with adamantyl or thio-phenol substituent at the sulfur atom. Catechol thio-ethers containing heterocyclic groups (thiazole, thiazoline, benzo-thiazole, benzo-xazole) can be considered effective antioxidants with cytoprotective properties. These compounds can protect molecules of DNA and lipids from the different radical species.

Synthesis of activated carbon from biowaste of fir bark for methylene blue removal.

Project description:Activated carbon (AC) was successfully prepared from low-cost forestry fir bark (FB) waste using KOH activation method. Morphology and texture properties of ACFB were studied by scanning and high-resolution transmission electron microscopies (SEM and HRTEM), respectively. The resulting fir bark-based activated carbon (ACFB) demonstrated high surface area (1552 m2 g-1) and pore volume (0.84 cm3 g-1), both of which reflect excellent potential adsorption properties of ACFB towards methylene blue (MB). The effect of various factors, such as pH, initial concentration, adsorbent content as well as adsorption duration, was studied individually. Adsorption isotherms of MB were fitted using all three nonlinear models (Freundlich, Langmuir and Tempkin). The best fitting of MB adsorption results was obtained using Freundlich and Temkin. Experimental results showed that kinetics of MB adsorption by our ACFB adsorbent followed pseudo-second-order model. The maximum adsorption capacity obtained was 330 mg g-1, which indicated that FB is an excellent raw material for low-cost production of AC suitable for cationic dye removal.
