Attraction effect of different colored cards on thrips Frankliniella intonsa in cowpea greenhouses in China.

ABSTRACT: The flower thrips Frankliniella intonsa (Trybom) is one of the most economically important pests in cowpea greenhouses in China. Widespread pesticide resistance of thrips and the negative environmental effects limit the application of pesticides for thrips control. Two commercial cowpea greenhouse experiments were designed to determine the color preference of F. intonsa to colored cards, including white, pink, pale green, light yellow, powder blue, crimson, yellow green, deep sky blue, dark slate blue, dark orange, medium orchid, gold, and black. Clear pieces of plastic were used as the control cards. Additionally, the effects of placement height and orientation (cardinal direction) of the cards were also studied. Both greenhouse trials showed that white cards were significantly more attractive to F. intonsa than the other 13 card colors, followed by deep sky blue cards. White or deep sky blue cards placed low to the ground were found to be most attractive to F. intonsa. Orientation of the colored cards also affected the attractiveness to F. intonsa. The results indicate that white sticky cards were significantly more attractive to F. intonsa than blue cards and therefore can be recommended to monitor F. intonsa population densities and to control them in cowpea greenhouses as part of integrated pest management programs.

Onion thrips, Thrips tabaci, have gut bacteria that are closely related to the symbionts of the western flower thrips, Frankliniella occidentalis.

Project description:It has been shown that many insects have Enterobacteriaceae bacteria in their gut system. The western flower thrips, Frankliniella occidentalis Pergande [Thysanoptera: Thripidae], has a symbiotic relation with Erwinia species gut bacteria. To determine if other Thripidae species have similar bacterial symbionts, the onion thrips, Thrips tabaci, was studied because, like F. occidentalis, it is phytophagous. Contrary to F. occidentalis, T. tabaci is endemic in Europe and biotypes have been described. Bacteria were isolated from the majority of populations and biotypes of T. tabaci examined. Bacteria were present in high numbers in most individuals of the populations studied. Like F. occidentalis, T. tabaci contained one type of bacterium that clearly outnumbered all other types present in the gut. This bacterium was identified as an Erwinia species, as was also the case for F. occidentalis. However, its biochemical characteristics and 16S rDNA sequence differed from the bacteria present in F. occidentalis.

Characterization of E93 in neometabolous thrips Frankliniella occidentalis and Haplothrips brevitubus.

Project description:Insect metamorphosis into an adult occurs after the juvenile hormone (JH) titer decreases at the end of the juvenile stage. This generally coincides with decreased transcript levels of JH-response transcription factors Krüppel homolog 1 (Kr-h1) and broad (br), and increased transcript levels of the adult specifier E93. Thrips (Thysanoptera) develop through inactive and non-feeding stages referred to as "propupa" and "pupa", and this type of distinctive metamorphosis is called neometaboly. To understand the mechanisms of hormonal regulation in thrips metamorphosis, we previously analyzed the transcript levels of Kr-h1 and br in two thrips species, Frankliniella occidentalis (Thripidae) and Haplothrips brevitubus (Phlaeothripidae). In both species, the transcript levels of Kr-h1 and br decreased in the "propupal" and "pupal" stages, and their transcription was upregulated by exogenous JH mimic treatment. Here we analyzed the developmental profiles of E93 in these two thrips species. Quantitative RT-PCR revealed that E93 expression started to increase at the end of the larval stage in F. occidentalis and in the "propupal" stage of H. brevitubus, as Kr-h1 and br mRNA levels decreased. Treatment with an exogenous JH mimic at the onset of metamorphosis prevented pupal-adult transition and caused repression of E93. These results indicated that E93 is involved in adult differentiation after JH titer decreases at the end of the larval stage of thrips. By comparing the expression profiles of Kr-h1, br, and E93 among insect species, we propose that the "propupal" and "pupal" stages of thrips have some similarities with the holometabolous prepupal and pupal stages, respectively.

Developing a Phototactic Electrostatic Insect Trap Targeting Whiteflies, Leafminers, and Thrips in Greenhouses.

Project description:Our aim was to develop an electrostatic apparatus to lure and capture silverleaf whiteflies (Bemisia tabaci), vegetable leafminers (Liriomyza sativae), and western flower thrips (Frankliniella occidentalis) that invade tomato greenhouses. A double-charged dipolar electric field producer (DD-EFP) was constructed by filling water in two identical transparent soft polyvinyl chloride tubes arrayed in parallel with fixed separation, and then, inserting the probes of grounded negative and positive voltage generators into the water of the two tubes to generate negatively and positively charged waters, respectively. These charged waters electrified the outer surfaces of the opposite tubes via dielectric polarization. An electric field formed between the oppositely charged tubes. To lure these phototactic insects, the water was colored yellow using watercolor paste, then introduced into the transparent insulator tubes to construct the yellow-colored DD-EFP. This apparatus lured insects in a manner similar to commercially available yellow sticky traps. The yellow-colored DD-EFP was easily placed as a movable upright screen along the plants, such that invading pests were preferentially attracted to the trap before reaching the plants. Furthermore, pests settling on the plants were attracted to the apparatus, which used a plant-tapping method to drive them off the plants. Our study provided an experimental basis for developing an electrostatic device to attract and capture insects that enter greenhouses.

A new brood-pollination mutualism between Stellera chamaejasme and flower thrips Frankliniella intonsa.

Project description:BackgroundBrood pollination mutualism is a special type of plant-pollinator interaction in which adult insects pollinate plants, and the plants provide breeding sites for the insects as a reward. To manifest such a mutualism between Stellera chamaejasme and flower thrips of Frankliniella intonsa, the study tested the mutualistic association of the thrips life cycle with the plant flowering phenology and determined the pollination effectiveness of adult thrips and their relative contribution to the host's fitness by experimental pollinator manipulation.ResultsThe adult thrips of F. intonsa, along with some long-tongue Lepidoptera, could serve as efficient pollinators of the host S. chamaejasme. The thrips preferentially foraged half-flowering inflorescences of the plants and oviposited in floral tubes. The floral longevity was 11.8 ± 0.55 (mean ± se) days, which might precisely accommodate the thrips life cycle from spawning to prepupation. The exclusion of adult thrips from foraging flowers led to a significant decrease in the fitness (i.e., seed set) of host plants, with a corresponding reduction in thrips fecundity (i.e., larva no.) in the flowers.ConclusionsThe thrips of F. intonsa and the host S. chamaejasme mutualistically interact to contribute to each other's fitness such that the thrips pollinate host plants and, as a reward, the plants provide the insects with brooding sites and food, indicating the coevolution of the thrips life cycle and the reproductive traits (e.g., floral longevity and morphology) of S. chamaejasme.

Bulk development and stringent selection of microsatellite markers in the western flower thrips Frankliniella occidentalis.

Project description:Recent improvements in next-generation sequencing technologies have enabled investigation of microsatellites on a genome-wide scale. Faced with a huge amount of candidates, the use of appropriate marker selection criteria is crucial. Here, we used the western flower thrips Frankliniella occidentalis for an empirical microsatellite survey and validation; 132,251 candidate microsatellites were identified, 92,102 of which were perfect. Dinucleotides were the most abundant category, while (AG)n was the most abundant motif. Sixty primer pairs were designed and validated in two natural populations, of which 30 loci were polymorphic, stable, and repeatable, but not all in Hardy-Weinberg equilibrium (HWE) and linkage equilibrium. Four marker panels were constructed to understand effect of marker selection on population genetic analyses: (i) only accept loci with single nucleotide insertions (SNI); (ii) only accept the most polymorphic loci (MP); (iii) only accept loci that did not deviate from HWE, did not show SNIs, and had unambiguous peaks (SS) and (iv) all developed markers (ALL). Although the MP panel resulted in microsatellites of highest genetic diversity followed by the SNI, the SS performed best in individual assignment. Our study proposes stringent criteria for selection of microsatellites from a large-scale number of genomic candidates for population genetic studies.

Development of Near-Isogenic Lines in a Parthenogenetically Reproduced Thrips Species, <i>Frankliniella occidentalis</i>.

Project description:Although near-isogenic lines (NILs) can standardize genetic backgrounds among individuals, it has never been applied in parthenogenetically reproduced animals. Here, through multiple rounds of backcrossing and spinosad screening, we generated spinosad resistant NILs in the western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), with a haplo-diploid reproduction system. The resultant F. occidentalis NIL-R strain maintained a resistance ratio over 30,000-fold, which was comparable to its parental resistant strain, Spin-R. More importantly, F. occidentalis NIL-R shared 98.90% genetic similarity with its susceptible parental strain Ivf03. By developing this toolset, we are able to segregate individual resistance and facilitate the mechanistic study of insecticide resistances in phloem-feeding arthropods, a group of devastating pest species reproducing sexually as well as asexually.

Development and characterization of 18 novel EST-SSRs from the western flower Thrips, Frankliniella occidentalis (Pergande).

Project description:The western flower thrips, Frankliniella occidentalis (Pergande), is an invasive species and the most economically important pest within the insect order Thysanoptera. For a better understanding of the genetic makeup and migration patterns of F. occidentalis throughout the world, we characterized 18 novel polymorphic EST-derived microsatellites. The mutational mechanism of these EST-SSRs was also investigated to facilitate the selection of appropriate combinations of markers for population genetic studies. Genetic diversity of these novel markers was assessed in 96 individuals from three populations in China (Harbin, Dali, and Guiyang). The results showed that all these 18 loci were highly polymorphic; the number of alleles ranged from 2 to 15, with an average of 5.50 alleles per locus. The observed (H(O)) and expected (H(E)) heterozygosities ranged from 0.072 to 0.707 and 0.089 to 0.851, respectively. Furthermore, only two locus/population combinations (WFT144 in Dali and WFT50 in Guiyang) significantly deviated from Hardy-Weinberg equilibrium (HWE). Pairwise F(ST) analysis showed a low but significant differentiation (0.026 < F(ST) < 0.032) among all three pairwise population comparisons. Sequence analysis of alleles per locus revealed a complex mutational pattern of these EST-SSRs. Thus, these EST-SSRs are useful markers but greater attention should be paid to the mutational characteristics of these microsatellites when they are used in population genetic studies.

Wavelength-Specific Behavior of the Western Flower Thrips (Frankliniella occidentalis): Evidence for a Blue-Green Chromatic Mechanism.

Project description:The western flower thrips (Frankliniella occidentalis) is a serious pest in horticulture, feeding on leaf tissue and floral resources. Blue and yellow sticky traps are commonly used for monitoring and control in greenhouses. The mechanisms underlying the color preferences are largely unknown. The use of light-emitting diodes (LEDs) is a promising approach to increase the attractiveness of visual traps and to study the color choice behavior in insects. The color preferences of F. occidentalis were systematically investigated in a series of choice experiments with several LEDs from the ultraviolet (UV) and visible spectral range. Blue LEDs were most attractive, followed by green, while only a moderate attractiveness of UV was observed. Blue and green were identified as two separate attractive ranges. When light from blue and green LEDs was mixed, the attractiveness decreased compared to its single components. In conclusion, F. occidentalis exhibits two different wavelength specific behaviors towards blue and green. Compelling indications are provided that these behaviors are controlled by two photoreceptors maximally sensitive in the blue and green range with an inhibitory chromatic interaction between both. Since the known UV sensitive photoreceptor could be confirmed, a trichromatic photoreceptor setup is suggested for F. occidentalis. For advanced plant protection strategies, the results offer several opportunities to optimize monitoring or even develop mass trapping devices.

Population analysis reveals genetic structure of an invasive agricultural thrips pest related to invasion of greenhouses and suitable climatic space.

Project description:Biological invasions of pests into climatically unsuitable areas can be facilitated by human-regulated environments, in which case there may be an impact on genetic structure through population processes and/or adaptation. Here, we investigated the population genetic structure of an invasive agricultural pest, Thrips palmi, in China, which has expanded its distribution range through using greenhouses. Early invaded populations showed a relatively higher level of genetic diversity than recently expanded greenhouse populations. Strong population genetic structure corresponded to a pattern of isolation by distance, with no recent gene flow and low historical gene flow among populations, reflecting limited ongoing dispersal. A genetic signature of population expansion was detected in early invaded populations and three northern populations from greenhouses, suggesting that the greenhouse environments facilitated expansion of this species. Redundancy analysis showed that the independent effects of environment and geography could explain 51.68% and 32.06% of the genetic variance, respectively. These findings point to climate- and greenhouse-related spatial expansion, with the potential for adaptation by T. palmi. They emphasize the contribution of human-regulated environments on the successes of this invasive species, a situation likely to apply to other invasive species that use greenhouse environments.
