Data on a real-time tripodal colorimetric/fluorescence sensor for multiple target metal ions.

ABSTRACT: Currently considerable research both in life and in environmental sciences is dedicated to chemosensors able to detect metals of biological interest such as zinc and iron or other toxic and carcinogenic, as cadmium, mercury, chromium, lead. Recently, a new chemosensor strategy of "single chemosensor for multiple metals" has emerged. For this scope, many fluorescent sensors for Cd(II) and Zn(II) have been designed and synthetized, as ligand systems or in polymeric matrices [1], [2], [3]. The data presented in this article include experimental data on the of a pyridyl/phenolic/benzothiazole functionalized colorimetric receptor (BPAP) and its selectively recognise Fe(III) and Fe(II) ions with visible, naked eye colour changes and fluorometric selectivity towards Zn²⁺ and Cd²⁺ ions in aqueous medium. This article is submitted as a companion paper to Caruso et al. (2018) [4].

Development of a Colorimetric Sensor for Autonomous, Networked, Real-Time Application.

Project description:This review describes an ongoing effort intended to develop wireless sensor networks for real-time monitoring of airborne targets across a broad area. The goal is to apply the spectrophotometric characteristics of porphyrins and metalloporphyrins in a colorimetric array for detection and discrimination of changes in the chemical composition of environmental air samples. The work includes hardware, software, and firmware design as well as development of algorithms for identification of event occurrence and discrimination of targets. Here, we describe the prototype devices and algorithms related to this effort as well as work directed at selection of indicator arrays for use with the system. Finally, we review the field trials completed with the prototype devices and discuss the outlook for further development.

Towards A Wireless Image Sensor for Real-Time Fluorescence Microscopy in Cancer Therapy.

Project description:We present a mm-sized, ultrasonically powered lensless CMOS image sensor as a progress towards wireless fluorescence microscopy. Access to biological information within the tissue has the potential to provide insights guiding diagnosis and treatment across numerous medical conditions including cancer therapy. This information, in conjunction with current clinical imaging techniques that have limitations in obtaining images continuously and lack wireless compatibility, can improve continual detection of multicell clusters deep within tissue. The proposed platform incorporates a 2.4×4.7 mm2 integrated circuit (IC) fabricated in TSMC 0.18 μm, a micro laser diode (μLD), a single piezoceramic and off-chip storage capacitors. The IC consists of a 36×40 array of capacitive trans-impedance amplifier-based pixels, wireless power management and communication via ultrasound and a laser driver all controlled by a Finite State Machine. The piezoceramic harvests energy from the acoustic waves at a depth of 2 cm to power up the IC and transfer 11.5 kbits/frame via backscattering. During Charge-Up, the off-chip capacitor stores charge to later supply a high-power 78 mW μLD during Imaging. Proof of concept of the imaging front end is shown by imaging distributions of CD8 T-cells, an indicator of the immune response to cancer, ex vivo, in the lymph nodes of a functional immune system (BL6 mice) against colorectal cancer consistent with the results of a fluorescence microscope. The overall system performance is verified by detecting 140 μm features on a USAF resolution target with 32 ms exposure time and 389 ms ultrasound backscattering.

A Fluorescence Sensor Capable of Real-Time Herbicide Effect Monitoring in Greenhouses and the Field.

Project description:Herbicide resistant weeds need to be identified early so that yield loss can be avoided by applying proper field management strategies. A novel chlorophyll-fluorescence-imaging sensor has been developed to conduct real-time herbicide effect evaluation. In this research, greenhouse and field experiments were conducted to calibrate the capability of the sensor in monitoring herbicide effects on different biotypes of two grass weeds (Alopecurus myosuroides, Apera spica-venti) in southwestern Germany. Herbicides with different modes of action were applied for the effect monitoring. Chlorophyll fluorescence yield of the plants was measured 3?15 days after treatment (DAT) using the new fluorescence sensor. Visual assessment of the weeds was carried out on 21 DAT. The results showed that the maximal PS II quantum yield (Fv/Fm) of herbicide sensitive weeds was significantly lower than the values of resistant populations in 5 DAT. The new technology was capable of quickly identifying the herbicide's effect on plants. It can be used to optimize management strategies to control herbicide resistant weeds.

Device-independent, real-time identification of bacterial pathogens with a metal oxide-based olfactory sensor.

Project description:A novel olfactory method for bacterial species identification using an electronic nose device called the MonoNose was developed. Differential speciation of micro-organisms present in primary cultures of clinical samples could be performed by real-time identification of volatile organic compounds (VOCs) produced during microbial replication. Kinetic measurements show that the dynamic changes in headspace gas composition are orders of magnitude larger than the static differences at the end of fermentation. Eleven different, clinically relevant bacterial species were included in this study. For each of the species, two to eight different strains were used to take intra-species biodiversity into account. A total of 52 different strains were measured in an incubator at 37 degrees C. The results show that the diagnostic specificities varied from 100% for Clostridium difficile to 67% for Enterobacter cloacae with an overall average of 87%. Pathogen identification with a MonoNose can be achieved within 6-8 h of inoculation of the culture broths. The diagnostic specificity can be improved by broth modification to improve the VOC production of the pathogens involved.

Steady-state and time-resolved fluorescence quenching with transition metal ions as short-distance probes for protein conformation.

Project description:A series of model dye-labeled histidine-containing peptides was used to investigate the nature of the quenching mechanism with Cu(2+) and Ni(2+). The strong reduction in steady-state fluorescence was found to be unaccompanied by any noticeable changes in lifetime kinetics. This static nature of quenching is not consistent with the dynamic Förster resonance energy transfer (FRET) phenomenon, which was assumed to dominate the quenching mechanism, and is likely caused by shorter range orbital coupling. Our results indicate that the FRET-like sixth power of distance dependence of quenching cannot be automatically assumed for transition metal ions and that time-resolved measurements should be used to distinguish various quenching mechanisms.

Functionalized Silver Nano-Sensor for Colorimetric Detection of Hg2+ Ions: Facile Synthesis and Docking Studies.

Project description:In the present study, we describe the facile synthesis of silver nanoparticles (AgNPs) and their nanostructures functionalized with 2-aminopyrimidine-4,6-diol (APD-AgNPs) for Hg2+ ion detection. The promising colorimetric response of APD-AgNPs to detect Hg2+ ions was visible with naked eyes and spectroscopic changes were examined by using a UV-Visible spectrophotometer. The aggregation of APD-AgNPs upon addition of Hg2+ ions was due to the chelation effect of the functionalized nanostructures and results in a color change from pale brown to deep yellow color. The probing sensitivity was observed within five minutes with a detection limit of about 0.35 µM/L. The TEM images of APD-AgNPs showed polydispersed morphologies with hexagonal, heptagonal and spherical nanostructures with an average size between 10 to 40 nm. Furthermore, the sensing behavior of APD-AgNPs towards Hg2+ ions detection was investigated using docking and interaction studies.

Benzothiazole appended 2,2'-(1,4-phenylene)diacetonitrile for the colorimetric and fluorescence detection of cyanide ions.

Project description:A benzothiazole appended 2,2'-(1,4-phenylene)diacetonitrile derivative (2Z,2'Z)-2,2'-(1,4-phenylene)bis(3-(3-(benzo[d]thiazol-2-yl)-4-hydroxyphenyl)acrylonitrile) (PDBT) has been synthesized and investigated as a novel sensor, capable of showing high selectivity and sensitivity towards CN- over a wide range of other interfering anions. After reaction with CN-, PDBT shows a new absorption peak at 451 nm with a color transformation from colorless to reddish-brown. When yellow fluorescent PDBT is exposed to CN-, it displays a significant increase in fluorescence at 445 nm, resulting in strong sky-blue fluorescence emission. The nucleophilic addition reaction of CN- plays a role in the sensing mechanism of PDBT to CN-. PDBT can distinguish between a broad variety of interfering anions and CN- with remarkable selectivity and sensitivity. Furthermore, the detection limit of the PDBT probe for CN- is 0.62 μM, which is significantly lower than the WHO standard of 1.9 μM for drinking water. Density functional theory simulations corroborated the observed fluorescence changes and the internal charge transfer process that occurs after cyanide ion addition. In addition, real-time applications of PDBT, such as cell imaging investigations and the detection of CN- in water samples, were successfully carried out.

Fluorescence spectroscopy approaches for the development of a real-time organophosphate detection system using an enzymatic sensor.

Project description:Organophosphates are organic substances that contain a phosphoryl or a thiophosphoryl bond. They are mainly used around the world as pesticides, but can also be used as chemical warfare agents. Their detection is normally entrusted to techniques like GC- and LC-MS that, although sensitive, do not allow their identification on site and in real time. We have approached their identification by exploiting the high-affinity binding of these compounds with the esterase 2 from Alicyclobacillus acidocaldarius. Using an in silico analysis to evaluate the binding affinities of the enzyme with organophosphate inhibitors, like paraoxon, and other organophosphate compounds, like parathion, chlorpyriphos, and other organophosphate thio-derivatives, we have designed fluorescence spectroscopy experiments to study the quenching of the tryptophan residues after esterase 2 binding with the organophosphate pesticides. The changes in the fluorescence signals permitted an immediate and quantitative identification of these compounds from nano- to picomolar concentrations. A fluorescence based polarity-sensitive probe (ANS) was also employed as a means to understand the extent of the interactions involved, as well as to explore other ways to detect organophosphate pesticides. Finally, we designed a framework for the development of a biosensor that exploits fluorescence technology in combination with a sensitive and very stable bio-receptor.

Colorimetric-Luminance Readout for Quantitative Analysis of Fluorescence Signals with a Smartphone CMOS Sensor.

Project description:Smartphones have shown promise as an enabling technology for portable and distributed point-of-care diagnostic tests. The CMOS camera sensor can be used for detecting optical signals, including fluorescence for applications such as isothermal nucleic acid amplification tests. However, such analysis is typically limited mostly to end point detection of single targets. Here we present a smartphone-based image analysis pipeline that utilizes the CIE xyY (chromaticity-luminance) color space to measure the luminance (in lieu of RGB intensities) of fluorescent signals arising from nucleic acid amplification targets, with a discrimination sensitivity (ratio between the positive to negative signals), which is an order of magnitude more than traditional RGB intensity based analysis. Furthermore, the chromaticity part of the analysis enables reliable multiplexed detection of different targets labeled with spectrally separated fluorophores. We apply this chromaticity-luminance formulation to simultaneously detect Zika and chikungunya viral RNA via end point RT-LAMP (Reverse transcription Loop-Mediated isothermal amplification). We also show real time LAMP detection of Neisseria gonorrhoeae samples down to a copy number of 3.5 copies per 10 ?L of reaction volume in our smartphone-operated portable LAMP box. Our chromaticity-luminance analysis is readily adaptable to other types of multiplexed fluorescence measurements using a smartphone camera.

Real-time observation of interfacial ions during electrocrystallization.

Project description:Understanding the electrocrystallization mechanisms of metal cations is of importance for many industrial and scientific fields. We have determined the transitional structures during underpotential deposition (upd) of various metal cations on Au(111) electrode using time-resolved surface X-ray diffraction and step-scan IR spectroscopy. At the initial stage of upd, a characteristic intensity transient appears in the time-resolved crystal truncation rod depending on metal cations. Metal cations with relatively high coordination energies of hydration water are deposited in two steps: first, the hydrated metal cations approached the surface and are metastably located at the outer Helmholtz plane, then they are deposited via the destruction of the hydration shell. However, Tl+ and Ag+, which have low hydration energy, are rapidly adsorbed on Au(111) electrode without any metastable states of dehydration. Therefore, the deposition rate is strongly related to the coordination energy of the hydration water. Metal cations strongly interacting with the counter coadsorbed anions such as Cu2+ in sulfuric acid causes the deposition rate to be slower because of the formation of complexes.
