Project description:Ionic-liquid-based aqueous biphasic systems (IL-based ABS) have been largely investigated as promising extraction and purification routes. In this context, the determination of their phase diagrams and the physical properties of the coexisting phases are of high relevance when envisaging their large-scale applications. Low viscosities improve the mass transfer and reduce energy consumptions, while the knowledge on their densities is important for the equipment design. In this work, novel phase diagrams for aqueous solutions of imidazolium-based ILs combined with acetate-based salts, namely KCH3CO2 or NaCH3CO2, are reported and discussed. The ability of the acetate-based salts to induce the phase separation not only depends on the ions hydration energy, but also on the concentration of "free" ions in solution. The tie-lines, tie-line lengths and critical points are also addressed. Experimental measurements of density and viscosity of the coexisting phases, for the different systems and at several compositions and temperatures, are additionally presented. The Othmer-Tobias and Bancroft equations are also applied to ascertain on the tie-lines coherence. It is here shown that low-viscous IL-based ABS, with a high difference in the densities of the coexisting phases, can be formed with organic and biodegradable salts thus offering enhanced features over conventional polymer-based systems.

Odd-even effect on the formation of aqueous biphasic systems formed by 1-alkyl-3-methylimidazolium chloride ionic liquids and salts.

Project description:This work provides a comprehensive evaluation of the effect of the cation alkyl side chain length of the 1-alkyl-3-methylimidazolium chloride series ([C n C1im]Cl, n = 2-14) of ionic liquids (ILs) on their capability to form aqueous biphasic systems (ABSs) with salts and self-aggregation derived properties. The liquid-liquid phase behavior of ternary systems composed of [C n C1im]Cl, water, and K3PO4 or K2CO3 and the respective Setschenow salting-out coefficients (ks ), a quantitative measure of the two-phase formation ability, were determined. An odd-even effect in the ks values along the number of methylene groups of the longest IL cation alkyl side chain was identified for the ABS formed by K2CO3, a weaker salting-out agent where the phenomenon is clearly identified. In general, cations with even alkyl side chains, being likely to display higher molar volumes, are more easily salted-out and thus more prone to undergo phase separation. The odd-even effect in the ks values is, however, more significant in ILs up to n = 6, where the nanostructuration/nanosegregation of ILs plays a less relevant role. Still, with the [C n C1im]Cl (n = 7-14) series of ILs, an odd-even effect was also identified in the ILs' ionization degree, molar conductivity, and conductivity at infinite dilution. In summary, it is shown here that the ILs' odd-even effect occurs in IL aqueous solutions and not just in neat ILs, an already well-established phenomenon occurring in a series of ILs' properties described as a result of the orientation of the terminal methyl groups to the imidazolium ring cation and consequent effect in the ILs' cohesive energy.

Hydrogen bond basicity of ionic liquids and molar entropy of hydration of salts as major descriptors in the formation of aqueous biphasic systems.

Project description:Aqueous biphasic systems (ABS) composed of ionic liquids (ILs) and conventional salts have been largely investigated and successfully used in separation processes, for which the determination of the corresponding ternary phase diagrams is a prerequisite. However, due the large number of ILs that can be prepared and their high structural versatility, it is impossible to experimentally cover and characterize all possible combinations of ILs and salts that may form ABS. The development of tools for the prediction and design of IL-based ABS is thus a crucial requirement. Based on a large compilation of experimental data, a correlation describing the formation of IL-based ABS is shown here, based on the hydrogen-bonding interaction energies of ILs (EHB) obtained by the COnductor-like Screening MOdel for Real Solvents (COSMO-RS) and the molar entropy of hydration of the salt ions. The ability of the proposed model to predict the formation of novel IL-based ABS is further ascertained.

Enhanced tunability afforded by aqueous biphasic systems formed by fluorinated ionic liquids and carbohydrates.

Project description:This work unveils the formation of novel aqueous biphasic systems (ABS) formed by perfluoroalkylsulfonate-based ionic liquids (ILs) and a large number of carbohydrates (monosaccharides, disaccharides and polyols) aiming at establishing more benign alternatives to the salts commonly used. The respective ternary phase diagrams were determined at 298 K. The aptitude of the carbohydrates to induce phase separation closely follows their hydration capability, while the length of the IL cation/anion fluorinated chain also plays a crucial role. Finally, these systems were investigated as liquid-liquid extraction strategies for four food dyes. Single-step extraction efficiencies for the carbohydrate-rich phase up to 94% were obtained. Remarkably and contrarily to the most investigated IL-salt ABS, most dyes preferentially migrate for the most hydrophilic and biocompatible carbohydrate-rich phase - an outstanding advantage when envisaging the products recovery and further use. On the other hand, more hydrophobic dyes preferentially partition to the IL-rich phase, disclosing therefore these novel systems as highly amenable to be tuned by the proper choice of the phase-forming components.

Good's Buffer Ionic Liquids as Relevant Phase-Forming Components of Self-Buffered Aqueous Biphasic Systems.

Project description:A series of new self-buffering ionic liquids (ILs) based on Good's buffers (GBs) anions and the tetrabutylphosphonium cation ([P4444]+) was here synthesized and characterized. The self-buffering behaviour of the GB-ILs was confirmed by measuring their protonation constants by potentiometry. Further, their ability to form aqueous biphasic systems with the biodegradable potassium citrate salt was evaluated, and further investigated for the extraction of proteins, using bovine serum albumin (BSA) as a model protein. If these ionic structures display self-buffering characteristics as well as a low toxicity towards the luminescent bacteria Vibrio fischeri, they were additionally found to be highly effective in the formation of ABS and in the extraction of BSA - extraction efficiencies of 100% to the IL-rich phase obtained in a single-step. The BSA secondary structure in the aqueous IL-rich solutions was evaluated through infrared spectroscopic studies revealing the protein-friendly nature of the synthesized ILs. Dynamic light scattering (DLS), "COnductor-like Screening MOdel for Real Solvents" (COSMO-RS), and molecular docking studies were finally carried out to better understand the main driving forces of the extraction process. The results suggest that van der Waals and hydrogen-bonding interactions are important driving forces of the protein migration towards the GB-IL-rich phase, while the molecular docking investigations demonstrated a stabilizing effect of the studied ILs over the protein.

Mechanisms ruling the partition of solutes in ionic-liquid-based aqueous biphasic systems - the multiple effects of ionic liquids.

Project description:In the past decade, the remarkable potential of ionic-liquid-based aqueous biphasic systems (IL-based ABSs) to extract and purify a large range of valued-added biocompounds has been demonstrated. However, the translation of lab-scale experiments to an industrial scale has been precluded by a poor understanding of the molecular-level mechanisms ruling the separation or partition of target compounds between the coexisting phases. To overcome this limitation, we carried out a systematic evaluation of specific interactions, induced by ILs and several salts used as phase-forming components, and their impact on the partition of several solutes in IL-based ABSs. To this end, the physicochemical characterization of ABSs composed of imidazolium-based ILs, three salts (Na2SO4, K2CO3 and K3C6H5O7) and water was performed. The ability of the coexisting phases to participate in different solute-solvent interactions (where "solvent" corresponds to each ABS phase) was estimated based on the Gibbs free energy of transfer of a methylene group between the phases in equilibrium, ΔG(CH2), and on the Kamlet-Taft parameters - dipolarity/polarizability (π*), hydrogen-bonding donor acidity (α) and hydrogen-bonding acceptor basicity (β) - of the coexisting phases. Relationships between the partition coefficients, the phase properties expressed as Kamlet-Taft parameters and COSMO-RS descriptors were established, highlighting the ability of ILs to establish specific interactions with given solutes. The assembled results clearly support the idea that the partition of solutes in IL-based ABSs is due to multiple effects resulting from both global solute-solvent and specific solute-IL interactions. Solute-IL specific interactions are often dominant in IL-based ABSs, explaining the higher partition coefficients, extraction efficiencies and selectivities observed with these systems when compared to more traditional ones majorly composed of polymers.

Aqueous biphasic systems composed of ionic liquids and polypropylene glycol: insights into their liquid-liquid demixing mechanisms.

Project description:Novel ternary phase diagrams of aqueous biphasic systems (ABSs) composed of polypropylene glycol with an average molecular weight of 400 g mol(-1) (PPG-400) and a vast number of ionic liquids (ILs) were determined. The large array of selected ILs allowed us to evaluate their tuneable structural features, namely the effect of the anion nature, cation core and cation alkyl side chain length on the phase behaviour. Additional evidence on the molecular-level mechanisms which rule the phase splitting was obtained by (1)H NMR (Nuclear Magnetic Resonance) spectroscopy and by COSMO-RS (Conductor-like Screening Model for Real Solvents). Some systems, for which the IL-PPG-400 pairs are completely miscible, revealed to be of type "0". All data collected suggest that the formation of PPG-IL-based ABSs is controlled by the interactions established between the IL and PPG, contrarily to previous reports where a "salting-out" phenomenon exerted by the IL over the polymer in aqueous media was proposed as the dominant effect in ABS formation. The influence of temperature on the liquid-liquid demixing was also evaluated. In general, an increase in temperature favours the formation of an ABS in agreement with the lower critical solution temperature (LCST) phase behaviour usually observed in polymer-IL binary mixtures. Partition results of a dye (chloroanilic acid, in its neutral form) further confirm the possibility of tailoring the phases' polarities of IL-PPG-based ABSs.

Enhanced extraction of bovine serum albumin with aqueous biphasic systems of phosphonium- and ammonium-based ionic liquids.

Project description:Novel aqueous biphasic systems (ABS) composed of phosphonium- or ammonium-based ionic liquids (ILs), combined with a buffered aqueous solution of potassium citrate/citric acid (pH=7.0), were investigated for the extraction of proteins. For that purpose, the phase diagrams, tie-lines and tie-line lengths were determined at 25 °C, and the performance of these ABS for the extraction of bovine serum albumin (BSA) was then evaluated. The obtained results reveal that, with the exception of the more hydrophobic ILs, most of the systems investigated allow the complete extraction of BSA for the IL-rich phase in a single-step. These remarkable extraction efficiencies are far superior to those afforded by more conventional extraction systems previously reported. The composition of the biphasic systems, i.e., the amount of phase-forming components, was also investigated aiming at reducing the overall costs of the process without losing efficiency on the protein extraction. It is shown that the extraction efficiencies of BSA are maintained at 100% up to high protein concentrations (at least up to 10 g L(-1)). The recovery of the BSA from the IL-rich phase by dialysis is also shown in addition to the demonstration of the IL recyclability and reusability, at least for 3 times. In the sequential three-step extractions (BSA recovery/IL reusability), the extraction efficiencies of BSA for the IL-rich phase were maintained at 100%. For the improved ABS, the preservation of the protein native conformation was confirmed by Size Exclusion High-Performance Liquid Chromatography (used also as the quantification method) and by Fourier Transform Infra-Red spectroscopy. According to the results herein reported, ABS composed of phosphonium- or ammonium-based ILs and a biodegradable organic salt represent an alternative and remarkable platform for the extraction of BSA and may be extended to other proteins of interest.

Project description:Aiming at establishing more effective and sustainable separation processes, herein we propose the use of carbohydrates combined with tetralkylphosphonium- and tetralkylammonium-based ionic liquids (ILs) to form aqueous biphasic systems (ABS). The formation of ABS composed of non-aromatic and non-fluorinated ILs with carbohydrates is here shown for the first time. These novel systems are competive extraction platforms when compared against more conventional ABS formed by ILs and salts or fluorinated ILs and carbohydrates. Finally, it is shown that these systems can be efficiently recovered and reused.
