Hydnophanerochaete and Odontoefibula, two new genera of phanerochaetoid fungi (Polyporales, Basidiomycota) from East Asia.

ABSTRACT: Two new genera with phylogenetic affinities to Phanerochaete s.l. are presented, namely Hydnophanerochaete and Odontoefibula. The generic type of Hydnophanerochaete is Phanerochaeteodontoidea. Odontoefibula is established based on a new species: O.orientalis (generic type). Both genera have effused basidiocarps with odontioid hymenial surface, simple-septate generative hyphae, cystidia lacking, clavate basidia and ellipsoid basidiospores that are smooth, thin-walled, inamyloid, non-dextrinoid and acyanophilous. Hydnophanerochaete is additionally characterised by a compact texture in the subiculum with thick-walled generative hyphae and quasi-binding hyphae. Odontoefibula has a dense texture of subiculum with thin- to slightly thick-walled hyphae and further a dark reddish reaction of basidiocarps when treated with KOH. Multi-marker phylogenetic analyses based on sequences, inferred from the ITS+nuc 28S+rpb1+rpb2+tef1 dataset, indicate that Hydnophanerochaete and Odontoefibula are placed in the Meruliaceae and Donkia clades of Phanerochaetaceae, respectively. Phanerochaetesubodontoidea is a synonym of P.odontoidea, according to morphological and molecular evidence.

Project description:Two new species of Perenniporia, P. pseudotephropora sp. nov. and P. subcorticola sp. nov., are introduced respectively from Brazil and China based on morphological characteristics and molecular data. Perenniporia pseudotephropora is characterised by perennial, pileate basidiocarps with distinctly stratified tubes, grey pores, tissues becoming dark in KOH, a dimitic hyphal system with slightly dextrinoid arboriform skeletal hyphae and broadly ellipsoid to subglobose, truncate, weakly dextrinoid, cyanophilous basidiospores, measuring 4.9-5.2 × 4-4.8 ?m. Perenniporia subcorticola is characterised by resupinate basidiocarps, yellow pores with thick dissepiments, tissues becoming dark in KOH, flexuous skeletal hyphae, ellipsoid, truncate and slightly dextrinoid basidiospores, measuring 4.2-5 × 3.5-4.2 µm. The morphologically-similar species and phylogenetically closely-related species to the two new species are discussed.

Multiple-marker phylogeny and morphological evidence reveal two new species in Steccherinaceae (Polyporales, Basidiomycota) from Asia.

Project description:Two new wood-inhabiting fungi, Mycorrhaphium subadustum sp. nov. and Trullella conifericola sp. nov., are proposed and described from Asia based on ITS, nrLSU and tef1 molecular phylogeny and morphological characteristics. Mycorrhaphium subadustum is characterized by a stipitate basidiocarp, velutinate pileal surface concentrically zoned, hydnoid hymenophore, a dimitic hyphal system in spine trama and monomitic in context, absence of gloeocystidia, presence of cystidioles and the non-amyloid, cylindrical to ellipsoid basidiospores. Trullella conifericola is characterized by a laterally stipitate basidiocarp with flabelliform to semicircular pileus, hirtellous pileal surface with appressed coarse hair and concentrically zoned and sulcate, tiny pores (10-12 per mm), a dimitic hyphal system, absence of any type of cystidia, short clavate basidia and thin-walled, smooth, cylindrical to allantoid basidiospores. Phylogenetic analyses based on a three-marker dataset were performed using maximum likelihood and Bayesian inference methods. The two new species formed isolated lineages with full support in Steccherinaceae. The distinguishing characters of the two new species as well as allied species are discussed, and a key to species of Mycorrhaphium is provided.

Taxonomy and phylogeny of Postia. Multi-gene phylogeny and taxonomy of the brown-rot fungi: Postia (Polyporales, Basidiomycota) and related genera.

Project description:Phylogenetic and taxonomic studies on the brown-rot fungi Postia and related genera, are carried out. Phylogenies of these fungi are reconstructed with multiple loci DNA sequences including the internal transcribed spacer regions (ITS), the large subunit (nLSU) and the small subunit (nSSU) of nuclear ribosomal RNA gene, the small subunit of mitochondrial rRNA gene (mtSSU), the translation elongation factor 1-α gene (TEF1), the largest subunit of RNA polymerase II (RPB1) and the second subunit of RNA polymerase II (RPB2). Ten distinct clades of Postia s.lat. are recognized. Four new genera, Amaropostia, Calcipostia, Cystidiopostia and Fuscopostia, are established, and nine new species, Amaropostia hainanensis, Cyanosporus fusiformis, C. microporus, C. mongolicus, C. piceicola, C. subhirsutus, C. tricolor, C. ungulatus and Postia sublowei, are identified. Illustrated descriptions of the new genera and species are presented. Identification keys to Postia and related genera, as well as keys to the species of each genus, are provided.

Phylogeny and diversity of Bjerkandera (Polyporales, Basidiomycota), including four new species from South America and Asia.

Project description:Four new species of Bjerkandera, viz. B. ecuadorensis, B. fulgida, B. minispora, and B. resupinata spp. nov., are described from tropical America and Asia. B. ecuadorensis is characterised by dark grey to black pore surface, a monomitic hyphal system, hyaline to yellowish-brown generative hyphae, and ellipsoid basidiospores measuring 3.9-4.5 × 2.7-3 μm. B. fulgida is distinguished from the other species in the genus by clay buff to pale brown and shiny pore surface. B. minispora is characterised by white tomentose pore mouth and small basidiospores measuring 3.1-4.2 × 2-2.8 μm. B. resupinata is characterised by resupinate basidiomata, pinkish buff to pale brownish pore surface, and ellipsoid to broadly ellipsoid basidiospores measuring 4.5-6 × 3.2-4.1 µm. All these new species grow on angiosperm trunks or rotten wood, and cause a white rot. The closely related taxa to four new species are discussed. An identification key to the ten accepted species of Bjerkandera is provided, and a phylogeny comprising all known Bjerkandera species is provided.

Three new species of Phanerochaete (Polyporales, Basidiomycota).

Project description:Phanerochaetecanobrunnea, P.cystidiata and P.fusca are presented as new species, supported by morphological studies and two sets of phylogenetic analyses. The 5.8S+nuc 28S+rpb1 dataset shows the generic placement of the three species within the phlebioid clade of Polyporales. The ITS+nuc 28S dataset displays relationships for the new taxa within Phanerochaete s.s. Phanerochaetecanobrunnea grew on angiosperm branches in subtropical Taiwan and is characterised by greyish brown hymenial surface, brown generative hyphae and skeletal hyphae and absence of cystidia. Phanerochaetecystidiata grew on angiosperm branches above 1000 m in montane Taiwan and SW Yunnan Province of China and is characterised by cream to yellowish hymenial surface and more or less encrusted leptocystidia. Phanerochaetefusca grew on angiosperm branches at 1700 m in Hubei Province of China and is characterised by dark brown hymenial surface, leptocystidia, brown subicular hyphae and colourless to brownish basidiospores.

Phylogeny and taxonomy of Laetiporus (Basidiomycota, Polyporales) with descriptions of two new species from western China.

Project description:Laetiporus is a cosmopolitan genus of brown rot fungi. In this study, L.medogensis and L.xinjiangensis are described as new species from western China, based on morphological and molecular evidence. L.medogensis has only been found on gymnosperms so far and is distinguished by pinkish-buff to clay-buff pileal surface and buff-yellow pore surface, azonate to faintly zonate pileus and ellipsoid to ovoid basidiospores (5-6.2 × 4.2-5.2 ?m). L.xinjiangensis is found on angiosperms and is characterised by pale-buff to clay-pink pileal surface, cream to light yellow pore surface, azonate to faintly zonate pileus, large pores (2-3 per mm) and small basidiospores (4.5-5 × 3-4.2 ?m). The phylogeny of Laetiporus is reconstructed with multi-gene sequences including the internal transcribed spacer regions (ITS), the large subunit (nrLSU) and small subunit (nrSSU) of the nuclear ribosomal RNA gene, the small subunit of the mitochondrial rRNA gene (mtSSU), the translation elongation factor 1-? gene (EF-1?) and the second subunit of RNA polymerase II (RPB2). The results show that L.medogensis and L.xinjiangensis formed two distinct lineages belonging to Laetiporus. Illustrated descriptions of the two new species are presented. An identification key to species of L.sulphureus complex is provided.

Three new species of Junghuhnia (Polyporales, Basidiomycota) from China.

Project description:In this study, taxonomic and phylogenetic analyses of Junghuhnia were performed. Three new species were characterised according to morphological characteristics and molecular phylogenetic analysis using ITS and nLSU sequences. They are J. austrosinensis sp. nov., J. nandinae sp. nov. and J. subcollabens sp. nov. Junghuhnia austrosinensis is characterised by resupinate, thin basidiomata with white to buff-yellow hymenophore, small pores (9-11 per mm), clamped generative hyphae possessing hymenial cystidia, ellipsoid basidiospores (2.5-3 × 1.7-2 µm) and growth on fallen bamboo or angiosperm branch. Junghuhnia nandinae is characterised by resupinate basidiomata with pink to salmon pores and a distinct white margin, clamp generative hyphae, interwoven tramal hyphae, ellipsoid basidiospores measuring 2.6-3.2 × 1.8-2 µm and growth on Nandina domestica. Junghuhnia subcollabens is characterised by resupinate basidiomata with pale salmon to brownish vinaceous hymenophore, small pores (10-12 per mm), generative hyphae with simple septa and clamp connections, interwoven tramal hyphae, lunate basidiospores measuring 2.9-3.4 × 1.6-1.8 µm and thriving on rotten wood of angiosperms.

Global Taxonomy and Phylogeny of Irpicaceae (Polyporales, Basidiomycota) With Descriptions of Seven New Species and Proposals of Two New Combinations.

Project description:The phylogenetic analyses of the family Irpicaceae were carried out based on a complete global sampling. The dataset that included concatenated ITS1-5.8S-ITS2 and nrLSU sequences of 67 taxa of Irpicaceae from around the world was subjected to the maximum likelihood analyses and Bayesian inference. In the phylogenetic tree, species from 14 genera were distributed in nine clades, among which five genera-Irpex, Phanerochaetella, Byssomerulius, Cytidiella, and Meruliopsis, received high support values. The genus Efibula was shown to be paraphyletic and four subclades could be recognized, while Phanerochaete allantospora, Leptoporus mollis, and several species from Ceriporia and Candelabrochaete formed a large clade with relatively strong support. Based on the molecular and morphological evidence, seven new corticioid species- Candelabrochaete guangdongensis, Efibula grandinosa, E. hainanensis, E. shenghuae, E. taiwanensis, Irpex alboflavescens, and Phanerochaetella sinensis, were revealed from the materials mostly from East Asia. The monotypic genus Flavodontia, newly described from southwestern China, is regarded as a later synonym of Irpex, and the new combination I. rosea is proposed. In addition, Phanerochaetella queletii is proposed for a taxon first described from Italy and newly recorded from China; Phanerochaete jose-ferreirae from Portugal is determined to be a later synonym. Descriptions and illustrations of the new species and the newly combined taxa are presented, and morphological comparisons for the known species of Efibula and Phanerochaetella are provided.

Steccherinum tenuissimum and S. xanthum spp. nov. (Polyporales, Basidiomycota): New species from China.

Project description:Two new wood-inhabiting fungal species, Steccherinum tenuissimum and S. xanthum spp. nov. are described based on a combination of morphological features and molecular evidence. Steccherinum tenuissimum is characterized by an annual growth habit, resupinate basidiomata with an odontioid hymenial surface, a dimitic hyphal system with clamped generative hyphae, strongly encrusted cystidia and basidiospores measuring 3-5 × 2-3.5 ?m. Steccherinum xanthum is characterized by odontioid basidiomata and a monomitic hyphal system with generative hyphae bearing clamp connections and covering by crystals, colourless, thin-walled, smooth, IKI-, CB-and has basidiospores measuring 2.7-5.5 × 1.8-4.0 ?m. Sequences of the ITS and nLSU nrRNA gene regions of the studied samples were generated, and phylogenetic analyses were performed with maximum likelihood, maximum parsimony and Bayesian inference methods. The phylogenetic analyses based on molecular data of ITS + nLSU sequences showed that two new Steccherinum species felled into the residual polyporoid clade. Further investigation was obtained for more representative taxa in Steccherinum based on ITS + nLSU sequences, which demonstrated that S. tenuissimum and S. xanthum were sister to S. robustius with high support (100% BP, 100% BS and 1.00 BPP).

Four new corticioid species in Trechisporales (Basidiomycota) from East Asia and notes on phylogeny of the order.

Project description:Four new species in Trechisporales from East Asia, Dextrinocystiscalamicola, Subulicystidiumacerosum, S.tropicum and Tubuliciumbambusicola, are described and illustrated, based on morphological and molecular evidence. The phylogeny of Trechisporales was inferred from a combined dataset of ITS-nrLSU sequences. In the phylogenetic tree, Sistotremastrum formed a family-level clade of its own, sister to the Hydnodontaceae clade formed by all other genera. Dextrinocystis, is for the first time, confirmed as a member of Hydnodontaceae. A key to all the accepted genera in Trechisporales is given.
