Zebra finches identify individuals using vocal signatures unique to each call type.

ABSTRACT: Individual recognition is critical in social animal communication, but it has not been demonstrated for a complete vocal repertoire. Deciphering the nature of individual signatures across call types is necessary to understand how animals solve the problem of combining, in the same signal, information about identity and behavioral state. We show that distinct signatures differentiate zebra finch individuals for each call type. The distinctiveness of these signatures varies: contact calls bear strong individual signatures while calls used during aggressive encounters are less individualized. We propose that the costly solution of using multiple signatures evolved because of the limitations of the passive filtering properties of the birds' vocal organ for generating sufficiently individualized features. Thus, individual recognition requires the memorization of multiple signatures for the entire repertoire of conspecifics of interests. We show that zebra finches excel at these tasks.

Vocal tract articulation in zebra finches.

Project description:BACKGROUND: Birdsong and human vocal communication are both complex behaviours which show striking similarities mainly thought to be present in the area of development and learning. Recent studies, however, suggest that there are also parallels in vocal production mechanisms. While it has been long thought that vocal tract filtering, as it occurs in human speech, only plays a minor role in birdsong there is an increasing number of studies indicating the presence of sound filtering mechanisms in bird vocalizations as well. METHODOLOGY/PRINCIPAL FINDINGS: Correlating high-speed X-ray cinematographic imaging of singing zebra finches (Taeniopygia guttata) to song structures we identified beak gape and the expansion of the oropharyngeal-esophageal cavity (OEC) as potential articulators. We subsequently manipulated both structures in an experiment in which we played sound through the vocal tract of dead birds. Comparing acoustic input with acoustic output showed that OEC expansion causes an energy shift towards lower frequencies and an amplitude increase whereas a wide beak gape emphasizes frequencies around 5 kilohertz and above. CONCLUSION: These findings confirm that birds can modulate their song by using vocal tract filtering and demonstrate how OEC and beak gape contribute to this modulation.

Parental influence on begging call structure in zebra finches (Taeniopygia guttata): evidence of early vocal plasticity.

Project description:Begging calls are signals of need used by young birds to elicit care from adults. Different theoretical frameworks have been proposed to understand this parent-offspring communication. But relationships between parental response and begging intensity, or between begging characteristics and proxies of a young's need remain puzzling. Few studies have considered the adjustment of nestling begging features to previous experience as a possible explanation of these discrepancies. In this study, we tested the effect of a heterospecific rearing environment on individual developmental trajectories of the acoustic structure of nestling begging calls. Fifty-two zebra finch chicks were fostered either to Bengalese finch or to zebra finch parents, and begging calls were recorded at several stages of nestling development. Acoustic analyses revealed that the development of the spectral features of the begging calls differed between experimental conditions: chicks reared by Bengalese finches produced higher pitched and less broadband begging calls than chicks reared by conspecific parents. Differences were stronger in males than females and were not explained by differences in growth rate. We conclude that nestling begging calls can be plastic in response to social interactions with parents.

A juvenile locomotor program promotes vocal learning in zebra finches.

Project description:The evolution and development of complex, learned motor skills are thought to be closely associated with other locomotor movement and cognitive functions. However, it remains largely unknown how different neuromuscular programs may interconnect during the protracted developmental process. Here we use a songbird to examine the behavioral and neural substrates between the development of locomotor movement and vocal-motor learning. Juvenile songbirds escalate their locomotor activity during the sensitive period for vocal learning, followed by a surge of vocal practice. Individual variability of locomotor production is positively correlated with precision of tutor imitation and duration of multi-syllable sequences. Manipulation of juvenile locomotion significantly impacts the precision of vocal imitation and neural plasticity. The locomotor program developed during the sensitive period of vocal learning may enrich the neural substrates that promote the subsequent development of vocal learning.

A feedforward inhibitory premotor circuit for auditory-vocal interactions in zebra finches.

Project description:Significance During conversations, we frequently alternate between listening and speaking. This involves withholding responses while the other person is vocalizing and rapidly initiating a reply once they stop. Similar exchanges also occur in other animals, such as songbirds, yet little is known about how brain areas responsible for vocal production are influenced by areas dedicated to listening. Here, we combined neural recordings and mathematical modeling of a sensorimotor circuit to show that input-dependent inhibition can both suppress vocal responses and regulate the onset latencies of vocalizations. Our resulting model provides a simple generalizable circuit mechanism by which inhibition precisely times vocal output and integrates auditory input within a premotor nucleus.

The Forebrain Song System Mediates Predictive Call Timing in Female and Male Zebra Finches.

Project description:The dichotomy between vocal learners and non-learners is a fundamental distinction in the study of animal communication. Male zebra finches (Taeniopygia guttata) are vocal learners that acquire a song resembling their tutors', whereas females can only produce innate calls. The acoustic structure of short calls, produced by both males and females, is not learned. However, these calls can be precisely coordinated across individuals. To examine how birds learn to synchronize their calls, we developed a vocal robot that exchanges calls with a partner bird. Because birds answer the robot with stereotyped latencies, we could program it to disrupt each bird's responses by producing calls that are likely to coincide with the bird's. Within minutes, the birds learned to avoid this disruptive masking (jamming) by adjusting the timing of their responses. Notably, females exhibited greater adaptive timing plasticity than males. Further, when challenged with complex rhythms containing jamming elements, birds dynamically adjusted the timing of their calls in anticipation of jamming. Blocking the song system cortical output dramatically reduced the precision of birds' response timing and abolished their ability to avoid jamming. Surprisingly, we observed this effect in both males and females, indicating that the female song system is functional rather than vestigial. We suggest that descending forebrain projections, including the song-production pathway, function as a general-purpose sensorimotor communication system. In the case of calls, it enables plasticity in vocal timing to facilitate social interactions, whereas in the case of songs, plasticity extends to developmental changes in vocal structure.

Patterns of call communication between group-housed zebra finches change during the breeding cycle.

Project description:Vocal signals such as calls play a crucial role for survival and successful reproduction, especially in group-living animals. However, call interactions and call dynamics within groups remain largely unexplored because their relation to relevant contexts or life-history stages could not be studied with individual-level resolution. Using on-bird microphone transmitters, we recorded the vocalisations of individual zebra finches (Taeniopygia guttata) behaving freely in social groups, while females and males previously unknown to each other passed through different stages of the breeding cycle. As birds formed pairs and shifted their reproductive status, their call repertoire composition changed. The recordings revealed that calls occurred non-randomly in fine-tuned vocal interactions and decreased within groups while pair-specific patterns emerged. Call-type combinations of vocal interactions changed within pairs and were associated with successful egg-laying, highlighting a potential fitness relevance of calling dynamics in communication systems.

A memory-driven auditory program ensures selective and precise vocal imitation in zebra finches.

Project description:In the vocal learning model, the juvenile first memorizes a model sound, and the imprinted memory gradually converts into vocal-motor output during the sensorimotor integration. However, early acquired memory may not precisely represent the fine structures of a model sound. How do juveniles ensure precise model imitation? Here we show that juvenile songbirds develop an auditory learning program by actively and attentively engaging with tutor's singing during the sensorimotor phase. The listening/approaching behavior requires previously acquired model memory and the individual variability of approaching behavior correlates with the precision of tutor song imitation. Moreover, it is modulated by dopamine and associated with forebrain regions for sensory processing. Overall, precise vocal learning may involve two steps of auditory processing: a passive imprinting of model memory occurs during the early sensory period; the previously acquired memory then guides an active and selective engagement of the re-exposed model to fine tune model imitation.

Neural circuits. Inhibition protects acquired song segments during vocal learning in zebra finches.

Project description:Vocal imitation involves incorporating instructive auditory information into relevant motor circuits through processes that are poorly understood. In zebra finches, we found that exposure to a tutor's song drives spiking activity within premotor neurons in the juvenile, whereas inhibition suppresses such responses upon learning in adulthood. We measured inhibitory currents evoked by the tutor song throughout development while simultaneously quantifying each bird's learning trajectory. Surprisingly, we found that the maturation of synaptic inhibition onto premotor neurons is correlated with learning but not age. We used synthetic tutoring to demonstrate that inhibition is selective for specific song elements that have already been learned and not those still in refinement. Our results suggest that structured inhibition plays a crucial role during song acquisition, enabling a piece-by-piece mastery of complex tasks.

Studying the Mechanisms of Developmental Vocal Learning and Adult Vocal Performance in Zebra Finches through Lentiviral Injection.

Project description:Here we provide a detailed step-by-step protocol for using lentivirus to manipulate miRNA expression in Area X of juvenile zebra finches and for analyzing the consequences on song learning and song performance. This protocol has four parts: 1) making the lentiviral construct to overexpress miRNA miR-9; 2) packaging the lentiviral vector; 3) stereotaxic injection of the lentivirus into Area X of juvenile zebra finches; 4) analysis of song learning and song performance in juvenile and adult zebra finches. These methods complement the methods employed in recent works that showed changing FoxP2 gene expression in Area X with lentivirus or adeno-associated virus leads to impairments in song behavior.

Inhibition within a premotor circuit controls the timing of vocal turn-taking in zebra finches.

Project description:Vocal turn-taking is a fundamental organizing principle of human conversation but the neural circuit mechanisms that structure coordinated vocal interactions are unknown. The ability to exchange vocalizations in an alternating fashion is also exhibited by other species, including zebra finches. With a combination of behavioral testing, electrophysiological recordings, and pharmacological manipulations we demonstrate that activity within a cortical premotor nucleus orchestrates the timing of calls in socially interacting zebra finches. Within this circuit, local inhibition precedes premotor neuron activation associated with calling. Blocking inhibition results in faster vocal responses as well as an impaired ability to flexibly avoid overlapping with a partner. These results support a working model in which premotor inhibition regulates context-dependent timing of vocalizations and enables the precise interleaving of vocal signals during turn-taking.
