Spatial and spectral imaging of point-spread functions using a spatial light modulator.

ABSTRACT: We develop a point-spread function (PSF) engineering approach to imaging the spatial and spectral information of molecular emissions using a spatial light modulator (SLM). We show that a dispersive grating pattern imposed upon the emission reveals spectral information. We also propose a deconvolution model that allows the decoupling of the spectral and 3D spatial information in engineered PSFs. The work is readily applicable to single-molecule measurements and fluorescent microscopy.

Confocal imaging capacity on a widefield microscope using a spatial light modulator.

Project description:Confocal microscopes can reject out-of-focus and scattered light; however, widefield microscopes are far more common in biological laboratories due to their accessibility and lower cost. We report confocal imaging capacity on a widefield microscope by adding a spatial light modulator (SLM) and utilizing custom illumination and acquisition methods. We discuss our illumination strategy and compare several procedures for postprocessing the acquired image data. We assessed the performance of this system for rejecting out-of-focus light by comparing images taken at 1.4 NA using our widefield microscope, our SLM-enhanced setup, and a commercial confocal microscope. The optical sectioning capability, assessed on thin fluorescent film, was 0.85 ± 0.04 μm for our SLM-enhanced setup and 0.68 ± 0.04 μm for a confocal microscope, while a widefield microscope exhibited no sectioning capability. We demonstrate our setup by imaging the same set of neurons in C. elegans on widefield, SLM, and confocal microscopes. SLM enhancement greatly reduces background from the cell body, allowing visualization of dim fibers nearby. Our SLM-enhanced setup identified 96% of the dim neuronal fibers seen in confocal images while a widefield microscope only identified 50% of the same fibers. Our microscope add-on represents a very simple (2-component) and inexpensive (<$600) approach to enable widefield microscopes to optically section thick samples.

Generation of V-point polarization singularity using single phase encoding with a spatial light modulator.

Project description:A liquid crystal Spatial Light Modulator (SLM) can be used in various ways to produce vector-vortices. Superposition of scalar vortices with orthogonal polarization is a common approach, while a more recent technique is to use dual-phase modulation. These approaches require modulation of at least two phase patterns with a SLM or multiple SLMs. In this paper, we propose a novel technique to produce vector-vortices by modulating orthogonal light components through a single phase pattern with a SLM. It does not require interferometric setups, and simplifies the generation of light beams with V-point polarization singularities. Because of compact and robustness of our experimental setup, it can be easily integrated to any device for applications of vector-vortices.

Three-Dimensional Incoherent Imaging Using Spiral Rotating Point Spread Functions Created by Double-Helix Beams [Invited].

Project description:In recent years, there has been a significant transformation in the field of incoherent imaging with new possibilities of compressing three-dimensional (3D) information into a two-dimensional intensity distribution without two-beam interference (TBI). Most incoherent 3D imagers without TBI are based on scattering by a random phase mask exhibiting sharp autocorrelation and low cross-correlation along the depth axis. Consequently, during reconstruction, high lateral and axial resolutions are obtained. Scattering based-Imaging requires a wasteful photon budget and is therefore precluded in many power-sensitive applications. This study develops a proof-of-concept 3D incoherent imaging method using a rotating point spread function termed 3D Incoherent Imaging with Spiral Beams (3DI2SB). The rotation speed of the point spread function (PSF) with displacement and the orbital angular momentum has been theoretically analyzed. The imaging characteristics of 3DI2SB were compared with a direct imaging system using a diffractive lens, and the proposed system exhibited a higher focal depth than the direct imaging system. Different computational reconstruction methods such as the Lucy-Richardson algorithm (LRA), non-linear reconstruction (NLR), and the Lucy-Richardson-Rosen algorithm (LRRA) were compared. While LRRA performed better than both LRA and NLR for an ideal case, NLR performed better than both under real experimental conditions. Both single plane imaging, as well as synthetic 3D imaging, were demonstrated. We believe that the proposed approach might cause a paradigm shift in the current state-of-the-art incoherent imaging, fluorescence microscopy, and astronomical imaging.

Instantaneous three-dimensional sensing using spatial light modulator illumination with extended depth of field imaging.

Project description:Imaging three-dimensional structures represents a major challenge for conventional microscopies. Here we describe a Spatial Light Modulator (SLM) microscope that can simultaneously address and image multiple targets in three dimensions. A wavefront coding element and computational image processing enables extended depth-of-field imaging. High-resolution, multi-site three-dimensional targeting and sensing is demonstrated in both transparent and scattering media over a depth range of 300-1,000 microns.

Demonstration of polarization-insensitive spatial light modulation using a single polarization-sensitive spatial light modulator.

Project description:We present a simple configuration incorporating a single polarization-sensitive phase-only liquid crystal spatial light modulator (LC-SLM) to facilitate polarization-insensitive spatial light modulation. The polarization-insensitive configuration is formed by a polarization beam splitter (PBS), a polarization-sensitive phase-only LC-SLM, a half-wave plate (HWP), and a mirror in a loop structure. We experimentally demonstrate polarization-insensitive spatial light modulations for incident linearly polarized beams with different polarization states and polarization-multiplexed beams. Polarization-insensitive spatial light modulations generating orbital angular momentum (OAM) beams are demonstrated in the experiment. The designed polarization-insensitive configuration may find promising applications in spatial light modulations accommodating diverse incident polarizations.

Uniform intensity in multifocal microscopy using a spatial light modulator.

Project description:Multifocal microscopy (MFM) offers high-speed three-dimensional imaging through the simultaneous image capture from multiple focal planes. Conventional MFM systems use a fabricated grating in the emission path for a single emission wavelength band and one set of focal plane separations. While a Spatial Light Modulator (SLM) can add more flexibility as a replacement to the fabricated grating, the relatively small number of pixels in the SLM chip, cross-talk between the pixels, and aberrations in the imaging system can produce non-uniform intensity in the different axially separated image planes. We present an in situ iterative SLM calibration algorithm that overcomes these optical- and hardware-related limitations to deliver near-uniform intensity across all focal planes. Using immobilized gold nanoparticles under darkfield illumination, we demonstrate superior intensity evenness compared to current methods. We also demonstrate applicability across emission wavelengths, axial plane separations, imaging modalities, SLM settings, and different SLM manufacturers. Therefore, our microscope design and algorithms provide an alternative to the use of fabricated gratings in MFM, as they are relatively simple and could find broad applications in the wider research community.

Real-time 3D single-molecule localization using experimental point spread functions.

Project description:We present a real-time fitter for 3D single-molecule localization microscopy using experimental point spread functions (PSFs) that achieves minimal uncertainty in 3D on any microscope and is compatible with any PSF engineering approach. We used this method to image cellular structures and attained unprecedented image quality for astigmatic PSFs. The fitter compensates for most optical aberrations and makes accurate 3D super-resolution microscopy broadly accessible, even on standard microscopes without dedicated 3D optics.

Fourier light-field imaging of human organoids with a hybrid point-spread function.

Project description:Volumetric interrogation of the cellular morphology and dynamic processes of organoid systems with a high spatiotemporal resolution provides critical insights for understanding organogenesis, tissue homeostasis, and organ function. Fluorescence microscopy has emerged as one of the most vital and informative driving forces for probing the cellular complexity in organoid research. However, the underlying scanning mechanism of conventional imaging methods inevitably compromises the time resolution of volumetric acquisition, leading to increased photodamage and inability to capture fast cellular and tissue dynamic processes. Here, we report Fourier light-field microscopy using a hybrid point-spread function (hPSF-FLFM) for fast, volumetric, and high-resolution imaging of entire organoids. hPSF-FLFM transforms conventional 3D microscopy and enables exploration of less accessible spatiotemporally-challenging regimes for organoid research. To validate hPSF-FLFM, we demonstrate 3D imaging of rapid responses to extracellular physical cues such as osmotic and mechanical stresses on human induced pluripotent stem cells-derived colon organoids (hCOs). The system offers cellular (2-3 μm and 5-6 μm in x-y and z, respectively) and millisecond-scale spatiotemporal characterization of whole-organoid dynamic changes that span large imaging volumes (>900 μm × 900 μm × 200 μm in x, y, z, respectively). The hPSF-FLFM method provides a promising avenue to explore spatiotemporal-challenging cellular responses in a wide variety of organoid research.

Compact Holographic Projection Display Using Liquid-Crystal-on-Silicon Spatial Light Modulator.

Project description:This paper presents a holographic projection display in which a phase-only spatial light modulator (SLM) performs three functions: beam shaping, image display, and speckle reduction. The functions of beam shaping and image display are performed by dividing the SLM window into four sub-windows loaded with different diffractive phase elements (DPEs). The DPEs are calculated using a modified iterative Fourier transform algorithm (IFTA). The function of speckle reduction is performed using temporal integration of display images containing speckles. The speckle contrast ratio of the display image is 0.39 due to the integration of eight speckled images. The system can be extended to display full-color images also by using temporal addition of elementary color images. Because the system configuration needs only an SLM, a Fourier transform lens, and two mirrors, the system volume is very small, becoming a potential candidate for micro projectors.

Generalized recovery algorithm for 3D super-resolution microscopy using rotating point spread functions.

Project description:Super-resolution microscopy with phase masks is a promising technique for 3D imaging and tracking. Due to the complexity of the resultant point spread functions, generalized recovery algorithms are still missing. We introduce a 3D super-resolution recovery algorithm that works for a variety of phase masks generating 3D point spread functions. A fast deconvolution process generates initial guesses, which are further refined by least squares fitting. Overfitting is suppressed using a machine learning determined threshold. Preliminary results on experimental data show that our algorithm can be used to super-localize 3D adsorption events within a porous polymer film and is useful for evaluating potential phase masks. Finally, we demonstrate that parallel computation on graphics processing units can reduce the processing time required for 3D recovery. Simulations reveal that, through desktop parallelization, the ultimate limit of real-time processing is possible. Our program is the first open source recovery program for generalized 3D recovery using rotating point spread functions.
