Description and molecular analysis of Tylencholaimushelanensis sp. n. from China (Dorylaimida, Tylencholaimidea).

ABSTRACT: A new species, Tylencholaimushelanensis sp. n., extracted from the rhizosphere soil of unidentified grasses from Helan Mountain, Inner Mongolia, China was identified. The new species is characterized by having a body length of 0.93-1.07 mm with the lip region approximately one-quarter of the body diameter at the posterior end of the neck region wide; female didelphic-amphidelphic; pars proximalis vaginae violin-shaped. Males were not found. SEM observations of the new species were made and a phylogenetic analysis of both the 18S rDNA and the D2-D3 region of 28S rDNA is presented.

Description of Longidorus azarbaijanensis n. sp. (Dorylaimida: Longidoridae) from Iran.

Project description:Longidorus azarbaijanensis n. sp. is described and illustrated using morphological and molecular data. It was recovered in West Azarbaijan province, northwestern Iran, from the rhizospheric soil of foxtail weed. The new species is characterized by having 5.4 to 6.8 mm long females, offset, anteriorly flat lip region and separated from the rest of the body by a shallow constriction, funnel-shaped amphidial pouches, the guiding ring at 23 to 27 µm from the anterior end, 73 to 81 and 44 to 50 µm long odontostyle and odontophore, respectively, 95.0 to 113.5 µm long pharyngeal bulb, didelphic-amphidelphic reproductive system with long tubular uteri lacking sperm cells, vulva located at 52.0% to 56.4%, conical tail dorsally convex, ventrally flat, with bluntly rounded wide tip, for juvenile developmental stages and absence of males. The general body shape of the new species is similar to that of five known species of the genus namely L. euonymus, L. perangustus, L. persicus, L. protae and L. sturhani . The morphological differences of the new species with the aforementioned species are discussed. For all the aforementioned species (except L. protae, currently lacking molecular data) the differences of the new species was also confirmed with differences in molecular sequences of D2-D3 expansion domains of 28S rDNA and the corresponding phylogenetic analyses. The partial sequence of the internal transcribed spacer 1 (ITS1) of the new species was also used in phylogenetic analyses. In partial 28S tree, the clade including the new species and six other species ( L. attenuatus, Longidorus sp. and four above-mentioned species having molecular data for this fragment) was well supported in Bayesian inference. In the ITS1 tree, the new species formed a clade with L. euonymus , L. perangustus and L. persicus , as in 28S tree. This is one of the cases from which the morphologically similar species are separated using molecular sequences.

Description of Longidorella (Saevadorella) caspica n. sp. (Dorylaimida: Nordiidae) from north Iran.

Project description:This contribution provides the morphological and molecular identification of a new species of the genus Longidorella (Saevadorella). L. (S.) caspica n. sp., was recovered from the rhizospheric soil of grasses in Mazandaran province in the seashore of the Caspian Sea. It is characterized by females with a length of 788 to 874 μm and a cephalic region with prominent papillae; and separated from the rest of the body by a remarkable constriction, an odontostyle of 32 to 33 μm, vulva at 52.5 to 59.0%, and a tail of 33 to 38 μm with a rounded tip. Males have 32 to 35 μm long spicules of dorylaimoid form and five to seven ventral supplements ending at 48 to 55 μm distance from cloacal pair. The new species was morphologically compared with seven nominal species viz. L. (S.) arenicola, L. (S.) cuspidata, L. (S.) magna, L. (S.) perplexa, L. (S.) saadi, L. (S.) saevae, and L. (S.) tharensis. It was further compared with similar species under the subgenus Longidorella (Enchodorella) viz. L. (E.) deliblatica and L. (E.) murithi. Besides morphological studies, molecular phylogenetic studies using partial sequences of D2 to D3 expansion fragments of 28S rDNA were performed for the new species and its phylogenetic relationships with other species and genera were discussed.

Description of Enchodorus yeatsi n. sp. (Dorylaimida, Nordiidae) from Southern Iran and Its Molecular Phylogenetic Study.

Project description:Enchodorus yeatsi n. sp., a new species of the rare genus Enchodorus is described and illustrated based on its morphological and molecular characters. It was recovered from southern Iran. Females of the new species are characterized by having 1,511.3- to 1,792.5-µm long slender body, angular lip region having prominent papillae, 12- to 13-µm long odontostyle, double guiding ring, simple rod-like odontophore, didelphic-amphidelphic reproductive system, and 102- to 160-µm long elongate-conoid tail with rounded tip. Males of the new species are abundant and functional, characterized by 1,484- to 1,576-µm long body, 40- to 43-µm long spicules, 5 to 6 ventromedian supplements, and ventrally bent elongate conical tail. Compared to the type species, Enchodorus dolichurus, the new species has differences in its tail morphology and V value. These morphological differences and the separation of two species was further supported with basic differences in sequences of 28S rDNA D2/D3 and internal transcribed spacer 1 (ITS1) fragments. Compared to Enchodorus neodolichurus, it has basic differences in tail characters and spicule lengths. Molecular phylogenetic studies using partial sequences of 28S rDNA D2/D3 fragment of the new species and available sequences of Nordiidae members and several other dorylaim species/genera, revealed E. yeatsi n. sp. and E. dolichurus forming a clade with 0.81 Bayesian posterior probability (BPP). This clade forms a sister clade to the clade of Heterodorus sp. and Rhyssocolpus vinciguerrae, again with 0.81 BPP. In ITS1 tree, reconstructed using few available sequences, the new species and E. dolichurus formed a clade with 0.98 BPP.

Description of Crassolabium persicum sp. n. (Nematoda, Dorylaimida, Qudsianematidae), an interesting species from Iran.

Project description:A new species of the genus Crassolabium, Crassolabium persicumsp. n., collected from Arasbaran rangelands of Iran, is described and illustrated. It is characterized by its body 1.92-2.40 mm long, lip region offset by constriction and 17-19 ?m wide, odontostyle 16-19 ?m long with aperture occupying less than one-third (27-30%) its length, neck 428-690 ?m long, pharyngeal expansion 369-390 ?m long or occupying 54-56% of total neck length, female genital system amphidelphic, uterus bipartite and 162-218 ?m long or 2.3-3.5 times as long as body diameter, pars refringens vaginae well developed, V = 54-57.5, vulva longitudinal, prerectum bearing a blind sac, tail conical with rounded tip to conoid (25-36 ?m, c=60-69, c'=0.5-0.9), spicules 68-72 ?m long, precloacal pair of supplements far (22-27 ?m) from cloacal aperture, and 13-17 shortly spaced ventromedian supplements with hiatus. The new taxon is compared in depth to its relatives in Crassolabium as well as other similar species of Aporcelaimellus and Amblydorylaimus.

Oostenbrinkia pedrami n. sp. (Dorylaimida: Aulolaimoididae) from Iran, with molecular phylogenetic relationships to some other Dorylaimida Pearse, 1942.

Project description:A population of the rarely collected aulolaimoidid genus, Oostenbrinkia, was recovered from the rhizosphere soil of Fagus orientalis in Mazandaran province, north Iran. Oostenbrinkia pedrami n. sp. is mainly characterized by long females, 920 µm with a 6 to 8 µm long odontostyle, 18 to 21 µm long odontophore with well-developed basal ﬂanges, 124 to 187 µm long neck, vulva post equatorial (V = 58.5-61.0), and tail short (10-18 µm long, c = 42.5-65.2, c' = 0.6-0.9) and rounded. Compared to the generotype species, O. oostenbrinki, the new species has a longer odontostyle, total stylet, a posteriorly located vulva, and shorter rounded tail, and compared to the only one other representative, O. parva, longer females with longer odontostyle and total stylet. In molecular phylogenetic analyses using a near-full-length sequence of the small subunit ribosomal DNA (SSU rDNA), the new species appeared as an independent basal lineage to the included ingroup species. This is the first molecular data of this family.

Description of Longidorus cholevae sp. n. (Nematoda, Dorylaimida) from a riparian habitat in the Rila Mountains, Bulgaria.

Project description:A description is provided of Longidorus cholevae sp. n., a bisexual species associated with wild cherry (Prunus avium L.) from the Rila Mountains, Bulgaria. The position of L. cholevae sp. n. among other species of the genus was elucidated by using morphological and molecular data. Phylogenetic analyses were performed of D2-D3 expansion domains of the 28S rRNA and the partial ITS1 containing regions by Neighbor-Joining, Maximum Likelihood and Bayesian Inference methods. The species is characterised by a female body length of 6.1-8.1 mm; long odontostyle (106-129 ?m); lip region wide (21.5-24 ?m) rounded and continuous with the body profile; amphidial pouches short and wide, funnel-shaped; a posteriorly situated guide ring (30-37 ?m); normal arrangement of pharyngeal glands, and short bluntly rounded to hemispherical tail. Four juvenile stages indentified, first stage with elongate conoid tail. Males with 2-4 adanal pairs and a row of 11-13 single ventromedian supplements, spicules 96-120 ?m long. Based both on morphological and molecular data the new species appearred to be the most similar witha group of species distributed in Europe sharing common charcters such as amphidial fovea, lip region and tail shapes, and having similar odontostyle and body length: L. poessneckensis, L. caespiticola, L. macrososma, L. helveticus, L. carniolensis and L. pius. An updated list of Longidorus species and a partial polytomous keys to the Longidorus species with long odontostyle (code A45) and short tail (code H1) are provided.

Morphological and molecular characterization of Epidorylaimus procerus sp. n. (Dorylaimida: Qudsianematidae) from Vietnam.

Project description:Epidorylaimus procerus sp. n., collected from a natural habitat in Vietnam, is described and illustrated. It is distinguishable by its 2.16 to 2.46-mm-long body, lip region offset by depression and 15 to 17-µm broad, odontostyle 32 to 35-µm long, neck 415 to 461-µm long, pharyngeal expansion occupying 47 to 52% of the total neck length, uterus 76 to 130-µm long or 1.0 to 1.5-body diameters, vulva transverse (V = 40-43), caudal region conical elongate (157-186 µm, c =12.1-14.4, c' = 4.4-5.5) with blister-like bodies, and hyaline portion occupying one-fourth its length, and males absent. Molecular analysis shows a close relationship of the new species and E. lugdunensis, supporting monophyly of the genus Epidorylaimus.

Project description:Sectonema caobangense sp. n. from evergreen forest soil in Vietnam is described, including scanning electron micrograph (SEM) observations and D2-D3 LSU rDNA analysis. The new species is characterized by its 3.12 to 5.80 mm long body, lip region offset by deep constriction and 21 to 23 ?m broad, mural tooth 13 to 14 ?m long at its ventral side, 940 to 1,112 ?m long neck, pharyngeal expansion occupying 61% to 69% of total neck length, uterus a long simple tube-like structure 292 to 363 ?m long or 2.7 to 2.9 times the corresponding body diameter, pars refringens vaginae well developed, V = 48 to 56, short (36-51 ?m, c = 77-132, c' = 0.5-0.8) and rounded tail, 87 to 99 ?m long spicules, and four or five irregularly spaced ventromedian supplements bearing hiatus. Sectonema caobangense sp. n. differs from the typical pattern of Sectonema in the nature of the stomatal protrusible structure, bearing a mural tooth attached to the ventral side of the stoma. Molecular data obtained and the derived evolutionary trees support a close phylogenetic relationship with other Sectonema species.
