The methanol sesquisolvate of sodium naproxen.

ABSTRACT: The asymmetric unit of the methanol solvate of sodium naproxen, systematic name: sodium (2S)-2-(6-meth-oxy-naphthalen-2-yl)propano-ate methanol sesquisolvate, Na⁺·C₁₄H₁₃O₃ ^-·1.5CH₃OH, comprises two formula units of the mol-ecular salt and three methanol mol-ecules. One of the sodium cations exhibits a coordination number of six and is bonded to three carboxyl-ate O atoms and three methanol OH groups whereas the second sodium cation has a coordination number of seven, defined by five carboxyl-ate O atoms and two methanol OH groups. Both coordination polyhedra around the sodium cations are considerably distorted. The two types of cations are bridged into polymeric chains extending parallel to [010]. This arrangement is stabilized by intrachain O-H⋯O hydrogen bonds between methanol ligands as donor and carboxyl-ate O atoms as acceptor groups. The hydro-phobic 6-meth-oxy-naphthyl moieties flank the hydro-philic sodium oxygen chains into ribbons parallel to [010]. There are no noticeable inter-molecular inter-actions between these ribbons. One of the 6-meth-oxy-naphthyl moieties is disordered over two sets of sites in a 0.723 (3):0.277 (3) ratio.

Project description:In the title compound, [RuCl(C(6)H(6))(C(10)H(8)N(2))]Cl·1.5CH(4)O, the Ru(II) atom is in a distorted octa-hedral environment coordinated by an ?(6)-benzene ring, a chelating 2,2'-bipyridine ligand and a chloride ion. The asymmetric unit is completed by a chloride anion and two methanol mol-ecules, one of which is disordered about a centre of inversion with an occupancy of 0.5. It is an example of a ruthenium complex with a less sterically congested environment than in similar derivatives. In the crystal structure, O-H?Cl hydrogen bonds, together with ?-? stacking inter-actions [centroid-centroid distances of 3.472Å(2)?Å], stabilize the structure.

Selective Adsorption of Aqueous Diclofenac Sodium, Naproxen Sodium, and Ibuprofen Using a Stable Fe3O4-FeBTC Metal-Organic Frameworka.

Project description:The FeBTC metal-organic framework (MOF) incorporated with magnetite is proposed as a novel material to solve water contamination with last generation pollutants. The material was synthesized by in situ solvothermal methods, and Fe3O4 nanoparticles were added during FeBTC MOF synthesis and used in drug adsorption. X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and Raman spectroscopy characterized the materials, with N2-physisorption at 77 K. Pseudo-second-order kinetic and Freundlich models were used to describe the adsorption process. The thermodynamic study revealed that the adsorption of three drugs was a feasible, spontaneous exothermic process. The incorporation of magnetite nanoparticles in the FeBTC increased the adsorption capacity of pristine FeBTC. The Fe3O4-FeBTC material showed a maximum adsorption capacity for diclofenac sodium (DCF), then by ibuprofen (IB), and to a lesser extent by naproxen sodium (NS). Additionally, hybridization of the FeBTC with magnetite nanoparticles reinforced the most vulnerable part of the MOF, increasing the stability of its thermal and aqueous media. The electrostatic interaction, H-bonding, and interactions in the open-metal sites played vital roles in the drug adsorption. The sites' competition in the multicomponent mixture's adsorption showed selective adsorption (DCF) and (NS). This work shows how superficial modification with a low-surface-area MOF can achieve significant adsorption results in water pollutants.

Structural basis for the transformation pathways of the sodium naproxen anhydrate-hydrate system.

Project description:Crystal structures are presented for two dihydrate polymorphs (DH-I and DH-II) of the non-steroidal anti-inflammatory drug sodium (S)-naproxen. The structure of DH-I is determined from twinned single crystals obtained by solution crystallization. DH-II is obtained by solid-state routes, and its structure is derived using powder X-ray diffraction, solid-state (13)C and (23)Na MAS NMR, and molecular modelling. The validity of both structures is supported by dispersion-corrected density functional theory (DFT-D) calculations. The structures of DH-I and DH-II, and in particular their relationships to the monohydrate (MH) and anhydrate (AH) structures, provide a basis to rationalize the observed transformation pathways in the sodium (S)-naproxen anhydrate-hydrate system. All structures contain Na(+)/carboxylate/H2O sections, alternating with sections containing the naproxen molecules. The structure of DH-I is essentially identical to MH in the naproxen region, containing face-to-face arrangements of the naphthalene rings, whereas the structure of DH-II is comparable to AH in the naproxen region, containing edge-to-face arrangements of the naphthalene rings. This structural similarity permits topotactic transformation between AH and DH-II, and between MH and DH-I, but requires re-organization of the naproxen molecules for transformation between any other pair of structures. The topotactic pathways dominate at room temperature or below, while the non-topotactic pathways become active at higher temperatures. Thermochemical data for the dehydration processes are rationalized in the light of this new structural information.

Anti-inflammatory effects of naproxen sodium on human osteoarthritis synovial fluid immune cells.

Project description:ObjectiveTo evaluate the anti-inflammatory effects of clinically relevant naproxen sodium (Nx) concentrations on human monocyte-derived macrophages in a controlled in vitro system and human primary synovial fluid (SF) cells.DesignUsing phorbol 12-myristate 13-acetate, THP-1 human monocytic cells were differentiated into mature monocyte-derived macrophages in vitro then treated with Nx pre- or post-activating an inflammatory response with lipopolysaccharide (LPS) and hyaluronan (HA) fragments (n = 8/group). Cell culture supernatants were assessed for NF-κB activity and prostaglandin E2 (PGE2), indicating cyclooxygenase enzyme activity. Under Duke IRB approval, primary human SF cells were collected at the time of knee joint replacement (n = 19 individuals) for osteoarthritis (OA), and cultured with LPS, HA and Nx; SF cells were characterized by polychromatic flow cytometry for cell surface markers and intracellular cytokines.ResultCompared to placebo treatment of THP-1 cells, low dose Nx (corresponding 27.5-440 mg/L orally) added both pre- and post-activation with LPS/HA, significantly reduced NF-κB activity and PGE2: mean reduction to 73%, 61%, 17% and 10% of placebo, respectively. LPS/HA treatment of primary OA SF cells significantly increased the number of IL-1β producing primary monocytes and macrophages, and by 24 h the overall production of secreted cytokines (IL-1β, IL-6, IL8, and TNF-α). Low dose Nx reduced the percentage of IL-1β producing primary monocytes and macrophages.ConclusionLPS/HA induced inflammation of THP-1 monocytic and primary human SF cells. Low dose Nx both prevented and reduced inflammatory responses of a human monocytic cell line and reduced IL-1β production by primary human SF monocytes and macrophages.

Evaluation of the migraine treatment sumatriptan/naproxen sodium on blood pressure following long-term administration.

Project description:Anti-inflammatory and pain therapies have been associated with blood pressure (BP) destabilization. Hence, the effects on BP of sumatriptan/naproxen sodium in fixed-dose combination, sumatriptan 85 mg, and naproxen sodium 500 mg administered intermittently for the acute treatment of migraine attacks were assessed. Patients with migraine with or without aura and no history of hypertension were randomized to sumatriptan/naproxen sodium (n=135), sumatriptan (n=136), or naproxen sodium (n=136) to treat migraine attacks for 6 months in a double-blind, parallel-group trial. Following a treated migraine attack, patients performed 2 consecutive days of self-measured BPs beginning ≥24 hours after the last dose of study medication and transmitted them by a transtelephonic modem. The primary end point was the change from baseline in self-measured BP at 6 months. Changes in self-measured BP from baseline to 6 months for sumatriptan/naproxen sodium were -2.1/-1.5 mm Hg (95% confidence intervals, -3.4 to -0.8 for systolic and -2.6 to -0.3 for diastolic). Mean changes from baseline in self-measured BP did not differ among the 3 treatment groups. Additional categorical analyses did not show increases from baseline with sumatriptan/naproxen sodium relative to either of the monotherapy groups. Intermittent acute migraine treatment with sumatriptan/naproxen sodium for up to 6 months was associated with clinically insignificant decreases in self-measured BP that were similar to those with sumatriptan or naproxen alone in normotensive patients with migraine.

Expression data from rat livers - Naproxen and NO-Naproxen treatment

Project description:To characterize naproxen and NO-naproxen, their effects on gene expression in livers of treated rats was examined. We have previously worked with liver and could therefore identify a moderate to strong antioxidant response element signature. There were two goals to this study: 1) to determine whether naproxen and NO-naproxen yield similar gene expression profiles, which would imply that their effects are mediated by the parent NSAID, and 2) to determine whether NO-naproxen, due to the release of NO, might cause increases in expression of genes associated with the antioxidant response element (ARE). In this dataset, we include the expression data obtained from liver of untreated rats and rats treated with naproxen (400mg/kg diet) or NO-naproxen (550mg/kg diet) for 7 days. Total 12 samples were analyzed. We generated the following comparisons: NO-Naproxen vs. Control and Naproxen vs. Control. Genes with a p-value < 0.05 and a fold-change M-bM-^IM-%1.5 were selected.

The In atom has a six-coordinate octa-hedral environment, being bonded to the N atoms of six thio-cyanate groups. The bond lengths and angles show normal values. The crystal packing exhibits no significantly short inter-molecular contacts.

Relief of menstrual symptoms and migraine with a single-tablet formulation of sumatriptan and naproxen sodium.

Project description:Dysmenorrhea and menstrual migraine may share a common pathogenic pathway. Both appear to be mediated, in part, by an excess of prostaglandin production that occurs during menstruation.Data were pooled from two replicate randomized controlled trials of 621 adult menstrual migraineurs with dysmenorrhea who treated migraine with sumatriptan-naproxen or placebo. Along with headache symptoms, nonpain menstrual symptoms (bloating, fatigue, and irritability) and menstrual pain symptoms (abdominal and back pain) were recorded at the time periods of 30 minutes and 1, 2, 4, and 4-24 hours. Relief of menstrual symptoms was compared using a Cochran-Mantel-Haenszel test. Logistic regression was used to determine the odds of a headache response with increasing numbers of moderate to severe dymenorrheic symptoms.Sumatriptan-naproxen was superior to placebo for relief of tiredness, irritability, and abdominal pain at the time periods of 2, 4, and 4-24 hours (p?0.023); back pain at the time periods of 4 and 4-24 hours (p?0.023); and bloating at 4-24 hours endpoint (p=0.01). The odds ratios (ORs) of attaining migraine pain freedom for 2 hours and for sustained 2-24 hours decreased as moderate to severe dysmenorrhea symptoms increased with sumatriptan-naproxen versus placebo.Treatment with sumatriptan-naproxen may provide relief of menstrual symptoms and migraine in female migraineurs with dysmenorrhea. The presence of moderate to severe dysmenorrhea symptoms is associated with decreased response rates for menstrual migraine, suggesting that the co-occurrence of these disorders may negatively impact the results of migraine-abortive therapy.

Project description:The asymmetric unit of the title compound, 2C(10)H(18)N(+)·HPO(4) (2-)·1.5C(4)H(4)O(4), contains two adamantan-1-aminium cations, one hydrogen phosphate anion, and one and a half mol-ecules of fumaric acid, one of which exhibits crystallographic inversion symmetry. Each HPO(4) (2-) anion is hydrogen bonded, via all of its O atoms, to four NH(3) (+) groups of the adamantan-1-aminium cations, forming chains along [100]. These chains are, in turn, inter-connected via a set of O-H⋯O hydrogen bonds involving the fumaric acid solvent mol-ecules, forming layers parallel to (001). Weak C-H⋯O inter-actions lead to a consolidation of the three-dimensional set-up.
