ABSTRACT: This paper reports a case of Purpureocillium lilacinum infection in seven loggerhead sea turtle (Caretta caretta) hatchlings kept in an aquarium under inadequate condition. The fungus was isolated from skin and pulmonary lesions. Metilene blue and NaCl solutions, Schinus terebinthifolius and eucalyptus essential oils Minimum Inhibitory Concentrations were determined indicating new possibilities for treatment.

Long-term climate forcing in loggerhead sea turtle nesting.

Project description:The long-term variability of marine turtle populations remains poorly understood, limiting science and management. Here we use basin-scale climate indices and regional surface temperatures to estimate loggerhead sea turtle (Caretta caretta) nesting at a variety of spatial and temporal scales. Borrowing from fisheries research, our models investigate how oceanographic processes influence juvenile recruitment and regulate population dynamics. This novel approach finds local populations in the North Pacific and Northwest Atlantic are regionally synchronized and strongly correlated to ocean conditions--such that climate models alone explain up to 88% of the observed changes over the past several decades. In addition to its performance, climate-based modeling also provides mechanistic forecasts of historical and future population changes. Hindcasts in both regions indicate climatic conditions may have been a factor in recent declines, but future forecasts are mixed. Available climatic data suggests the Pacific population will be significantly reduced by 2040, but indicates the Atlantic population may increase substantially. These results do not exonerate anthropogenic impacts, but highlight the significance of bottom-up oceanographic processes to marine organisms. Future studies should consider environmental baselines in assessments of marine turtle population variability and persistence.

Anatomic Interactive Atlas of the Loggerhead Sea Turtle (Caretta caretta) Head.

Project description:The head of the sea turtle is susceptible to congenital, developmental, traumatic, and infectious disorders. An accurate interpretation and thorough understanding of the anatomy of this region could be useful for veterinary practice on sea turtles. The purpose of this study was to develop an interactive two-dimensional (2D) atlas viewing software of the head of the loggerhead sea turtle (Caretta caretta) using images obtained via osteology, gross dissections, and computed tomography (CT). The atlas is composed of 10 osteology, 13 gross dissection, 10 sagittal multiplanar reconstructed CT (bone and soft tissue kernels), and 22 transverse CT (bone and soft tissue windows) images. All images were segmented and colored using ITK-SNAP software. The visualization and image assessment were performed using the Unity 3D platform to facilitate the development of interactive content in 2D. This atlas can be useful as an interactive anatomic resource for assessment of the head of loggerhead sea turtles.

Loggerhead sea turtle (Caretta caretta) diving changes with productivity, behavioral mode, and sea surface temperature.

Project description:The relationship between dive behavior and oceanographic conditions is not well understood for marine predators, especially sea turtles. We tagged loggerhead turtles (Caretta caretta) with satellite-linked depth loggers in the Gulf of Mexico, where there is a minimal amount of dive data for this species. We tested for associations between four measurements of dive behavior (total daily dive frequency, frequency of dives to the bottom, frequency of long dives and time-at-depth) and both oceanographic conditions (sea surface temperature [SST], net primary productivity [NPP]) and behavioral mode (inter-nesting, migration, or foraging). From 2011-2013 we obtained 26 tracks from 25 adult female loggerheads tagged after nesting in the Gulf of Mexico. All turtles remained in the Gulf of Mexico and spent about 10% of their time at the surface (10% during inter-nesting, 14% during migration, 9% during foraging). Mean total dive frequency was 41.9 times per day. Most dives were ≤ 25 m and between 30-40 min. During inter-nesting and foraging, turtles dived to the bottom 95% of days. SST was an important explanatory variable for all dive patterns; higher SST was associated with more dives per day, more long dives and more dives to the seafloor. Increases in NPP were associated with more long dives and more dives to the bottom, while lower NPP resulted in an increased frequency of overall diving. Longer dives occurred more frequently during migration and a higher proportion of dives reached the seafloor during foraging when SST and NPP were higher. Our study stresses the importance of the interplay between SST and foraging resources for influencing dive behavior.

Hawksbill × loggerhead sea turtle hybrids at Bahia, Brazil: where do their offspring go?

Project description:Hybridization between hawksbill (Eretmochelys imbricata) and loggerhead (Caretta caretta) breeding groups is unusually common in Bahia state, Brazil. Such hybridization is possible because hawksbill and loggerhead nesting activities overlap temporally and spatially along the coast of this state. Nevertheless, the destinations of their offspring are not yet known. This study is the first to identify immature hawksbill × loggerhead hybrids (n = 4) from this rookery by analyzing the mitochondrial DNA (mtDNA) of 157 immature turtles morphologically identified as hawksbills. We also compare for the first time modeled dispersal patterns of hawksbill, loggerhead, and hybrid offspring considering hatching season and oceanic phase duration of turtles. Particle movements varied according to season, with a higher proportion of particles dispersing southwards throughout loggerhead and hybrid hatching seasons, and northwards during hawksbill season. Hybrids from Bahia were not present in important hawksbill feeding grounds of Brazil, being detected only at areas more common for loggerheads. The genetic and oceanographic findings of this work indicate that these immature hybrids, which are morphologically similar to hawksbills, could be adopting behavioral traits typical of loggerheads, such as feeding in temperate waters of the western South Atlantic. Understanding the distribution, ecology, and migrations of these hybrids is essential for the development of adequate conservation and management plans.

Transcriptional Analyses of Acute Exposure to Methylmercury on Erythrocytes of Loggerhead Sea Turtle.

Project description:To understand changes in enzyme activity and gene expression as biomarkers of exposure to methylmercury, we exposed loggerhead turtle erythrocytes (RBCs) to concentrations of 0, 1, and 5 mg L-1 of MeHg and de novo transcriptome were assembled using RNA-seq. The analysis of differentially expressed genes (DEGs) indicated that 79 unique genes were dysregulated (39 upregulated and 44 downregulated genes). The results showed that MeHg altered gene expression patterns as a response to the cellular stress produced, reflected in cell cycle regulation, lysosomal activity, autophagy, calcium regulation, mitochondrial regulation, apoptosis, and regulation of transcription and translation. The analysis of DEGs showed a low response of the antioxidant machinery to MeHg, evidenced by the fact that genes of early response to oxidative stress were not dysregulated. The RBCs maintained a constitutive expression of proteins that represented a good part of the defense against reactive oxygen species (ROS) induced by MeHg.

Correlation Between Microbial Community and Hatching Failure in Loggerhead Sea Turtle Caretta caretta.

Project description:Microbial communities provide essential information about host ecology and could be helpful as a tool to improve species conservation efforts. However, microbes can also infect and compromise the host development process and viability. Caretta caretta is the most widespread marine turtle species in the Mediterranean basin and is the only species of sea turtle nesting along the Italian coasts. Little is known about the microbiota composition of the nest of sea turtles and its correlation with hatching failures. In this study, the microbial composition of two nests of C. caretta featuring different rates of hatching success from a nesting beach in Lampedusa (Italy) was analyzed and compared. The bacterial community was determined using culture-dependent methods and next-generation sequencing based on 16S rRNA gene metabarcoding analysis. Our results showed five dominant bacterial phyla (Proteobacteria, Bacteroidetes, Actinobacteria, Verrucomicrobia, and Firmicutes) and indicated different bacterial families (Pseudomonadaceae and Brucellaceae) as likely causes of hatching failures. Besides, our findings demonstrated the nests' active role in modulating the sand's bacterial communities. This study suggests microbiological analysis could be a valuable tool in monitoring nests to take preventive actions and reduce hatching failures.

De novo transcriptome assembly of loggerhead sea turtle nesting of the Colombian Caribbean.

Project description:Loggerhead sea turtle Caretta caretta is widely distributed in the oceans of tropical and subtropical latitude. This turtle is an endangered species due to anthropic and natural factors that have decreased their population levels. In this study, RNA sequencing and de-novo assembly of genes expressed in blood were performed. The raw FASTQ files have been deposited on NCBI's SRA database with accession number SRX2629512. A total of 5.4 Gb raw sequence data were obtained, corresponding to 48,257,019 raw reads. Trinity pipeline was used to perform a de-novo assembly, we were able to identify 64,930 transcripts for female loggerhead turtle transcriptome with an N50 of 1131 bp. The obtained transcriptome data will be useful for further studies of the physiology, biochemistry and evolution in this species.

The Maternal Legacy: Female Identity Predicts Offspring Sex Ratio in the Loggerhead Sea Turtle.

Project description:In organisms with temperature-dependent sex determination, the incubation environment plays a key role in determining offspring sex ratios. Given that global temperatures have warmed approximately 0.6?°C in the last century, it is necessary to consider how organisms will adjust to climate change. To better understand the degree to which mothers influence the sex ratios of their offspring, we use 24 years of nesting data for individual female loggerhead sea turtles (Caretta caretta) observed on Bald Head Island, North Carolina. We find that maternal identity is the best predictor of nest sex ratio in univariate and multivariate predictive models. We find significant variability in estimated nest sex ratios among mothers, but a high degree of consistency within mothers, despite substantial spatial and temporal thermal variation. Our results suggest that individual differences in nesting preferences are the main driver behind divergences in nest sex ratios. As such, a female's ability to plastically adjust her nest sex ratios in response to environmental conditions is constrained, potentially limiting how individuals behaviorally mitigate the effects of environmental change. Given that many loggerhead populations already show female-biased offspring sex ratios, understanding maternal behavioral responses is critical for predicting the future of long-lived species vulnerable to extinction.

Sporadic nesting reveals long distance colonisation in the philopatric loggerhead sea turtle (Caretta caretta).

Project description:The colonisation of new suitable habitats is crucial for species survival at evolutionary scale under changing environmental conditions. However, colonisation potential may be limited by philopatry that facilitates exploiting successful habitats across generations. We examine the mechanisms of long distance dispersal of the philopatric loggerhead sea turtle (Caretta caretta) by analysing 40 sporadic nesting events in the western Mediterranean. The analysis of a fragment of the mitochondrial DNA and 7 microsatellites of 121 samples from 18 of these nesting events revealed that these nests were colonising events associated with juveniles from distant populations feeding in nearby foraging grounds. Considering the temperature-dependent sex determination of the species, we simulated the effect of the incubation temperature and propagule pressure on a potential colonisation scenario. Our results indicated that colonisation will succeed if warm temperature conditions, already existing in some of the beaches in the area, extend to the whole western Mediterranean. We hypothesize that the sporadic nesting events in developmental foraging grounds may be a mechanism to overcome philopatry limitations thus increasing the dispersal capabilities of the species and the adaptability to changing environments. Sporadic nesting in the western Mediterranean can be viewed as potential new populations in a scenario of rising temperatures.
