Project description:Rationale: Maternal immune responses can promote allergy development in offspring. Pilot data show that neonates of mother mice exposed during pregnancy to air pollution particles have increased allergic susceptibility. Objective: We investigated whether inflammatory response to titanium dioxide (TiO2) particles earlier considered immunologically ‘inert’ is enhanced during pregnancy. Methods: Pregnant BALB/c mice (or non-pregnant controls) received particle suspensions intranasally at day 14 of pregnancy. Lung inflammatory responses were evaluated 19 and 48 h after exposure. Results: Pregnant mice showed robust and persistent acute inflammatory responses after exposure to TiO2, while non-pregnant females had the expected minimal responses. Genomic profiling identified genes differentially expressed in pregnant lungs exposed to TiO2. Neonates of mothers exposed to TiO2 (but not PBS) developed increased susceptibility to allergens. We exposed adult male fathead minnows for 96 hr to three doses (0, 1, and 10 mg/L nominal) of TiO2 under static renewal conditions. TiO2 is stearate coated, 15 nm particle size.

Keywords: Toxicology, disease state analysis, biomarkers of health effects Female C57BL/6 mice were exposed to 40 mg nanoTiO2/m3 for one hour/day for 11 consecutive days and were sacrificed 5 days following the last exposure. Left lung lobes and liver were removed and flash frozen. Total RNA was isolated from a small random part of the frozen lung and liver and was hybridized against universal mouse reference RNA to Agilent Oligo DNA microarrays (Agilent Technologies) containing 44,000 transcripts. Microarrays were normalized using a global LOWESS approach and analyzed by MAANOVA 2.0 and SAM. Microarray results were validated by real time RT-PCR. Impact of alteration in expression of select genes was further validated by analysing their total protein levels in lung tissue homogenates.

Project description:The growing application of materials containing TiO2 particles has led to an increased risk of human exposure, while a gap in knowledge about the possible adverse effects of TiO2 still exists. In this work, TiO2 particles of rutile, anatase, and their commercial mixture were exposed to various environments, including simulated gastric fluids and human blood plasma (both representing in vivo conditions), and media used in in vitro experiments. Simulated body fluids of different compositions, ionic strengths, and pH were used, and the impact of the absence or presence of chosen enzymes was investigated. The physicochemical properties and agglomeration of TiO2 in these media were determined. The time dependent agglomeration of TiO2 related to the type of TiO2, and mainly to the type and composition of the environment that was observed. The presence of enzymes either prevented or promoted TiO2 agglomeration. TiO2 was also observed to exhibit concentration-dependent cytotoxicity. This knowledge about TiO2 behavior in all the abovementioned environments is critical when TiO2 safety is considered, especially with respect to the significant impact of the presence of proteins and size-related cytotoxicity.

Experiment Overall Design: To test whether pregnancy alters the normally minimal inflammatory response to âinertâ particles, we have administered TiO2 and DEP suspensions (50 ug/mouse) or PBS solution by intranasal insufflation to normal or pregnant E14 mice (see Figure 1B). The mice were subjected to pathologic analysis 19 or 48 hrs later. Experiment Overall Design: Respirable-size TiO2 particles were generously provided by Dr. Ian Gilmour (US EPA). Particle samples were baked at 165 0C for 3 h to eliminate endotoxin, aliquoted and stored frozen at â 80 0C. Particle suspensions (50 ug in 50 uL) or PBS solution (vehicle) were administered by single intranasal insufflation of pregnant or normal Balb/c mice under light halothane anesthesia Experiment Overall Design: Total lung RNA extraction and isolation was performed 19 hrs after exposure using a Qiagen RNAeasy Mini kit according to manufacturerâs instructions (Qiagen, Valencia, CA). RNA purity and quality were analyzed by Agilent Bioanalyzer 2100 scan. The hybridization was carried out at the Harvard Partners Genomic Center Microarray facility (Cambridge, MA) using the Affymetrix GeneChip ® platform and Affymetrix mouse 430 2.0 chips (Affymetrix, Santa Clara, CA). Experiment Overall Design: 4 samples were analyzed in each of 4 groups: Normal PBS, Normal TiO2, Pregnant PBS and Pregnant TiO2, for a total of 16 samples.

Project description:The search for new antimicrobial substances has increased in recent years. Antimicrobial nanostructures are one of the most promising alternatives. In this work, titanium dioxide nanotubes were obtained by an atomic layer deposition (ALD) process over electrospun polyvinyl alcohol nanofibers (PVN) at different temperatures with the purpose of obtaining antimicrobial nanostructures with a high specific area. Electrospinning and ALD parameters were studied in order to obtain PVN with smallest diameter and highest deposition rate, respectively. Chamber temperature was a key factor during ALD process and an appropriate titanium dioxide deposition performance was achieved at 200 °C. Subsequently, thermal and morphological analysis by SEM and TEM microscopies revealed hollow nanotubes were obtained after calcination process at 600 °C. This temperature allowed complete polymer removal and influenced the resulting anatase crystallographic structure of titanium dioxide that positively affected their antimicrobial activities. X-ray analysis confirmed the change of titanium dioxide crystallographic structure from amorphous phase of deposited PVN to anatase crystalline structure of nanotubes. These new nanostructures with very large surface areas resulted in interesting antimicrobial properties against Gram-positive and Gram-negative bacteria. Titanium dioxide nanotubes presented the highest activity against Escherichia coli with 5 log cycles reduction at 200 μg/mL concentration.

Project description:Sanitary sewer overflows (SSOs) are a common problem across the United States. An estimated 23,000-75,000 SSOs occurred annually in 2004 discharging between 11 and 38 billion liters of untreated wastewater to receiving waters. SSOs release many contaminants, including engineered nanomaterials (ENMs), to receiving water bodies. Measuring ENM concentrations in environmental samples remains a key challenge in environmental nanotechnology and requires the distinction between natural and engineered particles. This distinction between natural and engineered particles is often hampered by the similarities in the intrinsic properties of natural and engineered particles such as particle size, composition, density, surface chemistry, and by the limitations of the available nanometrology tools. To overcome these challenges, we applied a multi-method approach to measure the concentrations and properties of TiO2 engineered particles (e.g., ENMs and pigments) including 1) multi-element single particle-inductively coupled plasma-mass spectrometry (ME-SP-ICP-MS) to identify elemental associations and to determine elemental ratios in natural particles, 2) total elemental concentrations and ratios calculated from total metal concentrations measured following total sample digestion to estimate engineered particle concentrations, and 3) transmission electron microscopy (TEM) to characterize engineered particle size and morphology. ME-SP-ICP-MS analysis revealed that natural TiO2 particles are often associated with at least one of the following elements Al, Fe, Ce, Si, La, Zr, Nb, Pb, Ba, Th, Ta, W and U, and that elemental ratios of Ti to these elements is typical of riverine particulates and the average crustal ratios, except for Pb likely due to anthropogenic Pb contamination. High TiO2 engineered particle concentrations up to 100 ?g L-1 were found in SSOs-impacted surface waters. TEM analysis demonstrated the presence of regular-shape TiO2 particles in SSOs-impacted surface waters. This study provides a comprehensive approach for measuring TiO2 engineered particle concentrations in surface waters. The quantitative data produced in this work can be used as input for modeling studies and pave the road toward routine monitoring of ENMs in environmental systems, validation of ENM fate models, and more accurate ENM exposure and risk assessment.

Project description:Despite previous studies on exploring the environmental effects of titanium dioxide nanoparticles particle (nTiO2) on plants, the detailed impacts of nTiO2 on the antioxidant system and photosynthesis of plants is still not well understood. This study was aimed at investigating the physiological and biochemical responses to nTiO2 by oxidative damage, Ti bioaccumulation, cell death, and photosynthesis in wheat. The results showed that 5.0 g nTiO2 L-1 resulted in a significant decrease in plant growth, chlorophyll contents, and photosynthetic activity. However, the obvious accumulation of reactive oxygen species (ROS) and cell death were observed under nTiO2 treatments in wheat roots and leaves. In addition, the concentrations of Ti in the roots were significantly higher than that in leaves with increased nTiO2 concentrations. Significant increase in enzyme activities and the levels of ascorbate were found in leaves exposed to 1.0 and 5.0 g nTiO2 L-1. Furthermore, the level of D1 and PsbS remarkably decreased in wheat leaves at 5.0 g nTiO2 L-1. However, the strong phosphorylation of photosystem II (PSII) reaction center protein D1 and D2 was observed at 5.0 g nTiO2 L-1. Altogether, these findings demonstrated that the roots suffered from more severe toxic damage from nTiO2 than the leaves and wheat plants respond to nTiO2 through the different physiological and biochemical mechanisms in the roots and leaves.
