ABSTRACT: The caprylic, lauric, palmitic and stearic esters of solketal and glycerol formal were synthesized with high selectivity and in good yields by a solvent-free acid catalyzed procedure. No acetal hydrolysis was observed, notwithstanding the acidic reaction conditions.

Selective Ammonolysis of Bioderived Esters for Biobased Amide Synthesis.

Project description:Amidation is an important reaction for bioderived platform molecules, which can be upgraded for use in applications such as polymers. However, fundamental understanding of the reaction especially in the presence of multiple groups is still lacking. In this study, the amidation of dimethyl fumarate, maleate, and succinate through ester ammonolysis was examined. The reaction networks and significant side reactions, such as conjugate addition and ring closing, were determined. A preliminary kinetic comparison among additional C4 and C6 esters showed a significant correlation between molecular structure and ammonolysis reactivity. Esters with a C=C double bond in the molecule backbone were found to have higher ammonolysis reactivity. To improve the selectivity to unsaturated amides rather than byproducts, the effects of thermal conditions and additives in dimethyl fumarate ammonolysis were examined. Lower temperature and decreasing methoxide ion concentration in the solution relative to the base case conditions increased the fumaramide selectivity from 67.1 to 90.6%.

Iodine-mediated formal [3 + 2] annulation for synthesis of furocoumarin from oxime esters.

Project description:A novel synthesis of furocoumarins was developed by a reaction between oxime esters and 4-hydroxycoumarins. The reaction was proposed to undergo radical mechanism mediated by iodine, a cheap and common laboratory reagent. Mechanistic studies showed the key for the successful transformation was the presence of α-iodoimine intermediate which facilitated the ring-closing step. The developed conditions produced good functional group tolerance with a wide range of high-profile furocoumarin product. The potential for this strategy to be applied in other syntheses of heterocyclic compounds is highly achievable.

Project description:To handle huge amount of glycerol produced in biodiesel industry, glycerol is transformed to value-added products. In this regard, glycerol acetalization to solketal is industrially attractive. As in this process various by-products can be formed, designing highly selective catalysts is of great importance. In this line, we wish to report a novel catalyst that benefits from strong acidity, high specific surface area and thermal stability, which can selectively form solketal in glycerol acetalization. To prepare the catalyst, hierarchical zeolite was prepared via a novel method, in which partially dealuminated NaY was treated with PluronicF-127 and then reacted with NH4NO3 to furnish the H-form zeolite. Hierarchical faujasite was then achieved through calcination and template removal. Subsequently, it was functionalized with ionic liquid and used for the immobilization of heteropolyacid. The results indicated the importance of the mesoprosity of zeolite and the presense of ionic liquid functionality for achiveing high solketal yield. Moreover, among three investigated heteropolyacids, phosphomolybdic acid exhibited the highest catalytic activity. In fact, using 10 wt% catalyst at 55 °C and glycerol to acetone molar ratio of 1:20, solketal with yield of 98% was furnished under solvent-less condition. Besides, the catalyst was recyclable with low leaching of heteropolyacid.

A Facile Solid-Phase Route to Renewable Aromatic Chemicals from Biobased Furanics.

Project description:Renewable aromatics can be conveniently synthesized from furanics by introducing an intermediate hydrogenation step in the Diels-Alder (DA) aromatization route, to effectively block retro-DA activity. Aromatization of the hydrogenated DA adducts requires tandem catalysis, using a metal-based dehydrogenation catalyst and solid acid dehydration catalyst in toluene. Herein it is demonstrated that the hydrogenated DA adducts can instead be conveniently converted into renewable aromatics with up to 80% selectivity in a solid-phase reaction with shorter reaction times using only an acidic zeolite, that is, without solvent or dehydrogenation catalyst. Hydrogenated adducts from diene/dienophile combinations of (methylated) furans with maleic anhydride are efficiently converted into renewable aromatics with this new route. The zeolite H-Y was found to perform the best and can be easily reused after calcination.

Biobased Chemicals: 1,2,4-Benzenetriol, Selective Deuteration and Dimerization to Bifunctional Aromatic Compounds.

Project description:1,2,4-Benzenetriol (BTO), sourced from the carbohydrate-derived platform chemical 5-hydroxylmethylfurfural (HMF), is an interesting starting point for the synthesis of various biobased aromatic products. However, BTO readily undergoes dimerization and other reactions under mild conditions, making analysis and isolation challenging. To both control and utilize the reactivity of BTO to produce biobased building blocks, its reactivity needs to be better understood. Here it was found that specific BTO aromatic C-H bonds are reactive toward deuterium exchange with D2O, which appears pronounced under acidic conditions at room temperature and can lead to the selective formation of BTO with an aromatic ring that contains one or two deuterium atoms, the first at the five and the second at the three position. By exposure to air, it was shown that BTO forms a 5,5'-linked BTO dimer [1,1'-biphenyl]-2,2',4,4',5,5'-hexaol (1) and subsequently a hydroxyquinone containing dimeric structure 2',4,4',5'-tetrahydroxy-[1,1'-biphenyl]-2,5-dione (2). Additionally, condensed dimer dibenzo[b,d]furan-2,3,7,8-tetraol (3) can be relatively easily accessed. The controlled formation of these symmetric and asymmetric multifunctional dimers illustrates diverse possibilities for BTO to be converted to valuable biobased aromatic compounds. Deuterium exchange was attributed to electrophilic aromatic substitution because this reactivity was found to be independent of oxygen and acid mediated. On the contrary, the dimerization was dependent on the presence of oxygen and thus likely involves radical intermediates. Thus this report overall displays different accessible reaction pathways for BTO that can be exploited for the production of BTO-derived compounds.

Opportunities and Challenges for Microbial Synthesis of Fatty Acid-Derived Chemicals (FACs).

Project description:Global warming and uneven distribution of fossil fuels worldwide concerns have spurred the development of alternative, renewable, sustainable, and environmentally friendly resources. From an engineering perspective, biosynthesis of fatty acid-derived chemicals (FACs) is an attractive and promising solution to produce chemicals from abundant renewable feedstocks and carbon dioxide in microbial chassis. However, several factors limit the viability of this process. This review first summarizes the types of FACs and their widely applications. Next, we take a deep look into the microbial platform to produce FACs, give an outlook for the platform development. Then we discuss the bottlenecks in metabolic pathways and supply possible solutions correspondingly. Finally, we highlight the most recent advances in the fast-growing model-based strain design for FACs biosynthesis.

Ultrasound-assisted enzymatic synthesis of xylitol fatty acid esters in solvent-free conditions.

Project description:A commercial immobilized lipase was successfully used for the synthesis of five xylityl acyl esters by means of the esterification of free fatty acids (caprylic, capric, lauric and myristic, respectively) with xylitol under solvent-free conditions. Ultrasound-assistance was shown to be a key tool to overcome the handicap imposed by both the mutual immiscibility of fatty acids and xylitol substrates, and the semisolid character of the initial reaction mixtures. In such semisolid systems, ultrasonic irradiation may enable the transport of substrate molecules to the enzyme catalytic-site, leading to the efficient synthesis of xylityl fatty ester (e.g. up to 95% yield after 90 min at 40 °C), with xylityl monoacyl ester and xylitol diacyl ester appearing as the main products (greater than 96%), assessed by HPLC and NMR analyses. The separation of products was carried out by heating and simple centrifugation of the reaction medium, which was possible due to different densities of the resulting fractions.

Upgrading of Biobased Glycerol to Glycerol Carbonate as a Tool to Reduce the CO2 Emissions of the Biodiesel Fuel Life Cycle.

Project description:With regards to oil-based diesel fuel, the adoption of bio-derived diesel fuel was estimated to reduce CO2 emissions by approximately 75%, considering the whole life cycle. In this paper, we present a novel continuous-flow process able to transfer an equimolar amount of CO2 (through urea) to glycerol, producing glycerol carbonate. This represents a convenient tool, able to both improve the efficiency of the biodiesel production through the conversion of waste streams into added-value chemicals and to beneficially contribute to the whole carbon cycle. By means of a Design of Experiments approach, the influence of key operating variables on the product yield was studied and statistically modeled.

Formal synthesis of (+/-)-platensimycin.

Project description:[reaction: see text] Reductive alkylation of 5-methoxy-1-tetralone (6) with 2,3-dibromopropene gave an equilibrium mixture of bicyclic diones 7 (51%) and 8 (35%). Radical cyclization of 7 afforded tricyclic dione 5 (84%), which was reduced, cyclized, and dehydrated to give tetracyclic alkene 13 in 63% yield. Allylic oxidation of 13 with SeO2 and activated MnO2 afforded enone 2 in 85% yield, thereby completing a short formal synthesis of (+/-)-platensimycin.
