Ultrasound-Assisted Enantioselective Esterification of Ibuprofen Catalyzed by a Flower-Like Nanobioreactor.

ABSTRACT: A flower-like nanobioreactor was prepared for resolution of ibuprofen in organic solvents. Ultrasound irradiation has been used to improve the enzyme performance of APE1547 (a thermophilic esterase from the archaeon Aeropyrum pernix K1) in the enantioselective esterification. Under optimum reaction conditions (ultrasound power, 225 W; temperature, 45 °C; water activity, 0.21), the immobilized APE1547 showed an excellent catalytic performance (enzyme activity, 13.26 μmol/h/mg; E value, 147.1). After ten repeated reaction batches, the nanobioreactor retained almost 100% of its initial enzyme activity and enantioselectivity. These results indicated that the combination of the immobilization method and ultrasound irradiation can enhance the enzyme performance dramatically.

Dual-frequency ultrasound for ultrasonic-assisted esterification.

Project description:The optimization of wheat starch esterification (acetylation) with a high degree of substitution was performed through response surface methodology (RSM) via various concentrations of reagents (acetic anhydride), pHs, and temperatures under various ultrasonication frequencies (25, 40, and 25 + 40 kHz). According to RSM methodology, optimized samples were selected by achieving high degrees of substitution at various frequencies, temperatures, and pHs. Solubility, swelling, X-ray, RVA, DSC, freeze-thaw stability, texture, and SEM analysis of the optimized samples were performed at three frequencies. X-ray pattern exhibited a more significant reduction in the crystallinity percentage of esterified starch at frequency 25 + 40 kHz compared with 25 kHz, 40 kHz, and native starch. According to DSC analysis, To, Tp, Tc, and enthalpy of gelatinization (?H gel) were lower in AC at frequency 25 + 40 kHz compared with AC at frequency 25 and 40 kHz and N starches. According to morphology analysis, in acetylated starches at 25 and 40 kHz, the surfaces and small granules underwent more damage, whereas in 25 + 40 kHz, large granules were more affected than small granules. Upon acetylation, freeze-thaw stability and textural properties of the starch significantly increased and decreased, respectively. The peak and final viscosity of acetylated starch increased (25 + 40 kHz ? 25 kHz ? 40 kHz ? N starch).

Mild ultrasound-assisted alkali de-esterification modified pectins: Characterization and structure-activity relationships in immunomodulatory effects.

Project description:Apple pectin (AP), a well-established dietary fiber, offers significant health benefits, particularly in immunomodulation. However, the structure-activity relationship (SAR) in this context remains poorly understood. This study aimed to elucidate the impact of varying degrees of esterification (DE) on AP's SAR in immunomodulatory activity. AP-Es (AP-E1, AP-E2, AP-E3) with different DE were prepared using mild ultrasound-assisted alkali de-esterification, followed by SAR analysis. Results revealed that AP-E3, with the lowest DE (5.08 ± 0.22 %), demonstrated a significant reduction in homogalacturonan (HG) domains and a corresponding increase in rhamnogalacturonan-I (RG-I) domains, which coincided with enhanced immunomodulatory effects. The molecular weights of AP-E1, AP-E2, and AP-E3 were determined to be 30.94 ± 0.83 kDa, 27.61 ± 0.65 kDa, and 22.17 ± 0.57 kDa, respectively. To further explore the underlying mechanism, transgenic zebrafish with fluorescent macrophages were utilized. A positive correlation was observed between AP-E3 concentration and the number of fluorescent microspheres engulfed by macrophages. Additionally, AP-E3 significantly upregulated the expression of key immune response genes (tnf-α, il-1β, il-6, cox-2, inos, and nf-κb) and restored the gut microbiota composition and abundance in chloramphenicol-induced immunocompromised zebrafish. Metabolomics analysis revealed that AP-E3 effectively restored metabolic homeostasis by activating multiple signaling pathways associated with signal transduction, immune regulation, and metabolism. These findings highlight the potential of low-esterified AP enriched with RG-I domains as a promising candidate for applications in immune modulation and gut health management.

Nickel-Catalyzed Esterification of Aliphatic Amides.

Project description:Recent studies have demonstrated that amides can be used in nickel-catalyzed reactions that lead to cleavage of the amide C-N bond, with formation of a C-C or C-heteroatom bond. However, the general scope of these methodologies has been restricted to amides where the carbonyl is directly attached to an arene or heteroarene. We now report the nickel-catalyzed esterification of amides derived from aliphatic carboxylic acids. The transformation requires only a slight excess of the alcohol nucleophile and is tolerant of heterocycles, substrates with epimerizable stereocenters, and sterically congested coupling partners. Moreover, a series of amide competition experiments establish selectivity principles that will aid future synthetic design. These studies overcome a critical limitation of current Ni-catalyzed amide couplings and are expected to further stimulate the use of amides as synthetic building blocks in C-N bond cleavage processes.

Biocatalytic Approach for Direct Esterification of Ibuprofen with Sorbitol in Biphasic Media.

Project description:Ibuprofen is a nonsteroidal anti-inflammatory drug (NSAID) introduced in the 1960s and widely used as an analgesic, anti-inflammatory, and antipyretic. In its acid form, the solubility of 21 mg/L greatly limits its bioavailability. Since the bioavailability of a drug product plays a critical role in the design of oral administration dosage, this study investigated the enzymatic esterification of ibuprofen as a strategy for hydrophilization. This work proposes an enzymatic strategy for the covalent attack of highly hydrophilic molecules using acidic functions of commercially available bioactive compounds. The poorly water-soluble drug ibuprofen was esterified in a hexane/water biphasic system by direct esterification with sorbitol using the cheap biocatalyst porcine pancreas lipase (PPL), which demonstrated itself to be a suitable enzyme for the effective production of the IBU-sorbitol ester. This work reports the optimization of the esterification reaction.

Mechanistic elucidation of monoalkyltin(iv)-catalyzed esterification.

Project description:Monoalkyltin(iv) complexes are well-known catalysts for esterification reactions and polyester formation, yet the mode of operation of these Lewis acidic complexes is still unknown. Here, we report on mechanistic studies of n-butylstannoic acid in stoichiometric and catalytic reactions, analyzed by NMR, IR and MS techniques. While the chemistry of n-butyltin(iv) carboxylates is dominated by formation of multinuclear tin assemblies, we found that under catalytically relevant conditions only monomeric n-BuSn(OAc)3 and dimeric (n-BuSnOAc2OEt)2 are present. Density functional theory (DFT) calculations provide support for a mononuclear mechanism, where n-BuSn(OAc)3 and dimeric (n-BuSnOAc2OEt)2 are regarded as off-cycle species, and suggest that carbon-oxygen bond breaking is the rate-determining step.

Halloysite-Catalyzed Esterification of Bio-Mass Derived Acids.

Project description:Halloysite, a natural clay with a hollow tubular structure, was studied as a catalyst for the esterification of biomass-derived carboxylic acids (levulinic acid, fumaric acid, maleic acid, and succinic acid) with four different alcohols (MeOH, EtOH, n-PrOH, and n-BuOH). Reaction conditions were optimized (10 mol % halloysite, 170 °C, 24 h) and gave high yields of the corresponding esters and diesters (>90%). The halloysite was easily recovered and recycled after washing and drying.

In this protocol, a catalytic amount of additive, a halogenated salt, plays a crucial role in the success. Control experiments and theoretical studies disclosed that the four-membered ring transmetallation transition states which were stabilized by a halide anion are the key to yields and stereochemical outcomes.

Microwave-Assisted Enzymatic Esterification Production of Emollient Esters.

Project description:Currently, esterification production of isopropyl myristate (IPM) or isopropyl palmitate (IPP) uses a homogeneous or heterogeneous acid substance as a catalyst and is conducted at high temperatures and pressures. Utilization of this type of catalyst requires an additional postproduction process (neutralization and purification), which burdens the production cost. Microwave enzymatic esterification is a simple and fast method. The results showed that reaction time, ratio molar of fatty acids to isopropyl alcohol, catalyst weight, and microwave power have a significant effect on the IPM or IPP conversion. Further, the energy consumption of this process is less than other enzymatic esterification and is certainly more energy efficient, which could save 99 and 29% of processing time.

Titanium Tetrachloride-Assisted Direct Esterification of Carboxylic Acids.

Project description:Ester compounds, widely found in pharmaceutical and natural products, play a crucial role in organic synthesis, prompting the development of numerous methods for their synthesis. An important chemical approach in synthesizing esters from carboxylic acids involves the activation of the carboxyl function, requiring the conversion of the hydroxyl group into a suitable leaving group. This paper presents the findings of our investigations into an efficient method for producing esters from carboxylic acids and alcohols, using the Lewis acid titanium tetrachloride. Titanium tetrachloride has proven highly effective as a coupling reagent for the one-pot formation of esters from carboxylic acids and alcohols operating under mild and neutral conditions. Notably, the reaction eliminates the need for bases, yielding carboxylic esters in high purity and yields. The method is efficient, even with long-chain carboxylic acids, and operates well with primary alcohols in dichloromethane. Steric hindrance, potentially present in carboxylic acids, has a moderate effect on the reaction. Alcohol substrates that easily form stable carbocations require, instead, the use of non-polar solvents like hexane for the reaction.

Ultrasound-assisted extraction as an easy-to-perform analytical methodology for monitoring ibuprofen and its main metabolites in mussels.

Project description:Non-steroidal anti-inflammatory drugs (NSAIDs) have been reported to be the main pharmaceutical class accumulated in seafood. Among them, ibuprofen (IBU) is of special concern as it is used worldwide to treat common pain, does not require a medical prescription, it is often taken in a high daily dose, and has been reported to cause potential adverse effects on aquatic organisms. IBU is highly transformed into hydroxy- and carboxy-metabolites and/or degradation products generated not only after its administration but also during wastewater treatment or in the environment. These compounds can be present in the environment at higher concentrations than IBU and present higher toxicity. In this work, a low-cost and affordable routine analytical method was developed and validated for the first-time determination of IBU and its main metabolites in mussels. The method is based on ultrasound-assisted extraction (UAE), clean-up by dispersive solid-phase extraction (d-SPE) and analytical determination by liquid chromatography-tandem mass spectrometry. Box-Behnken experimental design was used for method optimisation to better evaluate the influence and interactions of UAE and d-SPE variables. Extraction recoveries were in the range from 81 to 115%. Precision, expressed as relative standard deviation, was lower than 7%. Method detection limits were in the range from 0.1 to 1.9 ng g-1 dry weight. The method was successfully applied to wild mussels.
