Coryneumheveanum sp. nov. (Coryneaceae, Diaporthales) on twigs of Para rubber in Thailand.

ABSTRACT: During studies of microfungi on para rubber in Thailand, we collected a new Coryneum species on twigs which we introduce herein as C.heveanum with support from phylogenetic analyses of LSU, ITS and TEF1 sequence data and morphological characters. Coryneumheveanum is distinct from other known taxa by its conidial measurements, number of pseudosepta and lack of a hyaline tip to the apical cell.

New polyketides from the liquid culture of Diaporthebreyniae sp. nov. (Diaporthales, Diaporthaceae).

Project description:During the course of a study on the biodiversity of endophytes from Cameroon, a fungal strain was isolated. A multigene phylogenetic inference using five DNA loci revealed that this strain represents an undescribed species of Diaporthe, which is introduced here as D.breyniae. Investigation into the chemistry of this fungus led to the isolation of two previously undescribed secondary metabolites for which the trivial names fusaristatins G (7) and H (8) are proposed, together with eleven known compounds. The structures of all of the metabolites were established by using one-dimensional (1D) and two-dimensional (2D) Nuclear Magnetic Resonance (NMR) spectroscopic data in combination with High-Resolution ElectroSpray Ionization Mass Spectrometry (HR-ESIMS) data. The absolute configuration of phomopchalasin N (4), which was reported for the first time concurrently to the present publication, was determined by analysis of its Rotating frame Overhauser Effect SpectroscopY (ROESY) spectrum and by comparison of its Electronic Circular Dichroism (ECD) spectrum with that of related compounds. A selection of the isolated secondary metabolites were tested for antimicrobial and cytotoxic activities, and compounds 4 and 7 showed weak antifungal and antibacterial activity. On the other hand, compound 4 showed moderate cytotoxic activity against all tested cancer cell lines with IC50 values in the range of 5.8-45.9 µM. The latter was found to be less toxic than the other isolated cytochalasins (1-3) and gave hints in regards to the structure-activity relationship (SAR) of the studied cytochalasins. Fusaristatin H (8) also exhibited weak cytotoxicity against KB3.1 cell lines with an IC50 value of 30.3 µM. Graphical abstract.

Phaeotubakialithocarpicola gen. et sp. nov. (Tubakiaceae, Diaporthales) from leaf spots in China.

Project description:Tubakiaceae represents a distinct lineage of Diaporthales, including its type genus Tubakia and nine additional known genera. Tubakiaceous species are commonly known as endophytes in leaves and twigs of many tree species, but can also be plant pathogens causing conspicuous leaf symptoms. In the present study, isolates were obtained from diseased leaves of Lithocarpusglaber collected in Guangdong Province, China. The identification was conducted based on morphology and phylogeny of combined loci of 28S nrRNA gene (LSU), internal transcribed spacer regions and intervening 5.8S nrRNA gene (ITS) of the nrDNA operon, translation elongation factor 1-alpha (tef1) and beta tubulin (tub2). As a result, a distinct clade in Tubakiaceae was revealed named Phaeotubakialithocarpicola gen. et sp. nov., which was distinguished from the other tubakiaceous taxa by its dark brown conidiogenous cells and conidia.

Dwiroopa punicae sp. nov. (Dwiroopaceae fam. nov., Diaporthales), associated with leaf spot and fruit rot of pomegranate (Punica granatum).

Project description:During a survey of diseases affecting pomegranate in the southeastern USA we identified a unique species of Diaporthales causing leaf spotting and fruit rot. Objectives of this study were to provide a morphological description of the putative new species, use DNA sequence data of three gene loci (LSU, ITS and rpb2) to accurately place the fungus within the Diaporthales, and to prove Koch's postulates. Morphological and phylogenetic comparisons confirmed the fungus to represent a new species and family, for which the names Dwiroopa punicae sp. nov. and Dwiroopaceae fam. nov. are proposed. This is the first report of a species of Dwiroopa being pathogenic to Punica granatum.

Lignicolous freshwater ascomycota from Thailand: Phylogenetic and morphological characterisation of two new freshwater fungi: Tingoldiago hydei sp. nov. and T. clavata sp. nov. from Eastern Thailand.

Project description:Lignicolous freshwater fungi represent one of the largest groups of Ascomycota. This taxonomically highly diverse group plays an important role in nutrient and carbon cycling, biological diversity and ecosystem functioning. The diversity of lignicolous freshwater fungi along a north-south latitudinal gradient is currently being studied in Asia. In this paper, we introduce two novel freshwater taxa viz. Tingoldiago hydei sp. nov. and T. clavata sp. nov. which were collected from freshwater substrates in Eastern Thailand. Morphological comparison based on the size of ascomata, asci and ascospores, as well as multi-gene phylogenetic analyses based on LSU, SSU, ITS and TEF1-α DNA sequences, supports their placement in Tingoldiago (Lentitheciaceae). Descriptions and illustrations of these two new species are provided.

A Novel Yeast Genus and Two Novel Species Isolated from Pineapple Leaves in Thailand: Savitreella phatthalungensis gen. nov., sp. nov. and Goffeauzyma siamensis sp. nov.

Project description:Four yeast strains, representing one genus and two novel anamorphic yeast species, were isolated from pineapple leaves collected in Thailand. Analysis of the sequences of the D1/D2 domains of the large subunit (LSU) rRNA gene and the internal transcribed spacer (ITS) regions showed the two strains (DMKU-PAL186 and DMKU-PAL178) were closely related to the type strains of the Protomyces and Taphrina species, but with high nucleotide divergence. Two strains (DMKU-PAL39 and DMKU-PAL18) were found to be closely related to the type strains of Goffeauzyma iberica, but with eight nucleotide substitutions in the D1/D2 domains and 26 nucleotide substitutions in the ITS regions. In phylogenetic analyses, the strains DMKU-PAL186 and DMKU-PAL178 formed a well-separated lineage from Protomyces and Taphrina genera, confirming that they represented a distinct genus, while the strains DMKU-PAL39 and DMKU-PAL18 represented a species in the genus Goffeauzyma, which was phylogenetically distinct from other recognized species of the genus. Based on molecular analyses and phenotypic characteristics, the names Savitreella gen. nov. (Taphrinomycetes, Ascomycota) and Savitreella phatthalungensis sp. nov. are proposed to accommodate the strains DMKU-PAL186 and DMKU-PAL178, and the name Goffeauzyma siamensis sp. nov. (Tremellomycetes, Basidiomycota) is proposed to accommodate the strains DMKU-PAL39 and DMKU-PAL18.

Pseudocryphonectriaelaeocarpicola gen. et sp. nov. (Cryphonectriaceae, Diaporthales) causing stem blight of Elaeocarpus spp. in China.

Project description:Cryphonectriaceae is a diaporthalean family containing important plant pathogens of which Cryphonectriaparasitica is the most notorious one. An emerging stem blight disease on Elaeocarpusapiculatus (Elaeocarpaceae) and E.hainanensis was observed in Guangdong Province of China recently. Typical Cryphonectria blight-like symptoms including cankers on tree barks with obvious orange conidial tendrils were observed. Forty-eight isolates were obtained from diseased tissues and conidiomata formed on the hosts E.apiculatus and E.hainanensis. These isolates were further identified based on both morphology and molecular methods using the combined sequence data of the internal transcribed spacer (ITS) region, large subunit of the nrDNA (LSU), the translation elongation factor 1-alpha (tef1) and DNA-directed RNA polymerase II second largest subunit (rpb2) genes. As a result, the fungus represents an undescribed genus and species within the family Cryphonectriaceae. Hence, Pseudocryphonectriaelaeocarpicola gen. et sp. nov. is proposed herein to represent these isolates from diseased barks of E.apiculatus and E.hainanensis. Pseudocryphonectria differs from the other genera of Cryphonectriaceae in having dimorphic conidia. Further inoculation results showed that P.elaeocarpicola is the causal agent of this emerging blight disease in China, which can quickly infect and kill the hosts E.apiculatus and E.hainanensis.

Gelatinomyces siamensis gen. sp. nov. (Ascomycota, Leotiomycetes, incertae sedis) on bamboo in Thailand.

Project description:Gelatinomyces siamensis gen. sp. nov., incertae sedis within Leotiomycetes, the Siamese jelly-ball, is described. The fungus was collected from bamboo culms and branches in Nam Nao National Park, Phetchabun, Thailand. It presents as a ping-pong ball-sized and golf ball-like gelatinous ascostroma. The asci have numerous ascospores, are thick-walled, and arise on discoid apothecia which are aggregated and clustered to form the spherical gelatinous structures. An hyphomycete asexual morph is morphologically somewhat phialophora-like, and produces red pigments. On the basis of phylogenetic analysis based on rRNA, SSU, and LSU gene sequences, the lineage is closest to Collophora rubra. However, ITS sequences place the fungus on a well-separated branch from that fungus, and the morphological and ecological differences exclude it from Collophora.

Distoseptispora bambusae sp. nov. (Distoseptisporaceae) on bamboo from China and Thailand.

During our taxonomic study on medicinal plants, three collections of Distoseptispora were made from China and Thailand. Phylogenetic analyses of combined LSU, ITS and RPB2 sequence data showed that two collections represented a new species, phylogenetically distinct from other described species in Distoseptispora.New informationThis new species has macronematous, mononematous conidiophores, polyblastic or monoblastic conidiogenous cells and acrogenous, solitary, straight, obclavate, multi-septate, thick-walled conidia. Distoseptispora bambusae sp. nov. is introduced with illustrations and a comprehensive description. The third collection on dead wood from Thailand was identified as D. tectona with newly-generated molecular data for this taxon.

Construction of Para rubber tree genome and multi-transcriptome database accelerates rubber researches.

Project description:BackgroundNatural rubber is an economically important material. Currently the Pará rubber tree, Hevea brasiliensis is the main commercial source. Little is known about rubber biosynthesis at the molecular level. Next-generation sequencing (NGS) technologies brought draft genomes of three rubber cultivars and a variety of RNA sequencing (RNA-seq) data. However, no current genome or transcriptome databases (DB) are organized by gene.ResultsA gene-oriented database is a valuable support for rubber research. Based on our original draft genome sequence of H. brasiliensis RRIM600, we constructed a rubber tree genome and transcriptome DB. Our DB provides genome information including gene functional annotations and multi-transcriptome data of RNA-seq, full-length cDNAs including PacBio Isoform sequencing (Iso-Seq), ESTs and genome wide transcription start sites (TSSs) derived from CAGE technology. Using our original and publically available RNA-seq data, we calculated co-expressed genes for identifying functionally related gene sets and/or genes regulated by the same transcription factor (TF). Users can access multi-transcriptome data through both a gene-oriented web page and a genome browser. For the gene searching system, we provide keyword search, sequence homology search and gene expression search; users can also select their expression threshold easily.ConclusionThe rubber genome and transcriptome DB provides rubber tree genome sequence and multi-transcriptomics data. This DB is useful for comprehensive understanding of the rubber transcriptome. This will assist both industrial and academic researchers for rubber and economically important close relatives such as R. communis, M. esculenta and J. curcas. First Report of Colletotrichum fructicola, C. rhizophorae sp. nov. and C. thailandica sp. nov. on Mangrove in Thailand.

Project description:Colletotrichum, a genus within the phylum Ascomycota (Fungi) and family Glomerellaceae are important plant pathogens globally. In this paper, we detail four Colletotrichum species found in mangrove ecosystems. Two new species, Colletotrichum rhizophorae and C. thailandica, and a new host record for Colletotrichum fructicola were identified in Thailand. Colletotrichum tropicale was collected from Taiwan's mangroves and is a new record for Rhizophora mucronata. These identifications were established through a combination of molecular analysis and morphological characteristics. This expanded dataset for Colletotrichum enhances our understanding of the genetic diversity within this genus and its associations with mangrove ecosystems. The findings outlined herein provide data on our exploration of mangrove pathogens in Asia.
