The fourth instar larva and the pupa of <i>Bezziachilensis</i> Spinelli & Ronderos, 2001 are described for the first time. The immature stages were collected from macrophytes and filamentous algae in streams of the Patagonian steppe, in the provinces of Neuquén and Chubut, Argentina. The described stages were photographed and illustrated with a phase-contrast microscope and scanning electron microscope. Project description:Background:This study is one in a series of planned studies on different Egyptian dipteran taxa aiming to catalogue the whole order in Egypt. New information:All known Egyptian taxa of the family Ceratopogonidae (biting midges) are systematically catalogued. A total number of 64 species belonging to 11 genera, four tribes and four subfamilies has been treated. Data for this study have been compiled from both available literature and specimens collected from different Egyptian localities by the authors. An updated classification, synonymies, type localities, world distributions by biogeographic realm(s) and country, Egyptian localities and dates of collection are provided comprising some new locality records. The study treats all previous inaccuracies in the classification of the family in Egypt.

Project description:To evaluate the host preferences of Culicoides species (Diptera: Ceratopogonidae) in Central Tunisia, we identified the source of blood meals of field collected specimens by sequencing of the cytochrome b (cyt b) mitochondrial locus and Prepronociceptine single copy nuclear gene. The study includes the most common and abundant livestock associated species of biting midges in Tunisia: C. imicola, C. jumineri, C. newsteadi, C. paolae, C. cataneii, C. circumscriptus, C. kingi, C. pseudojumineri, C. submaritimus, C. langeroni, C. jumineri var and some unidentified C. species. Analysis of cyt b PCR products from 182 field collected blood-engorged females' midges revealed that 92% of them fed solely on mammalian species, 1.6% on birds, 2.4% on insects and 0.8% on reptiles. The blast results identified the blood origin of biting midges to the species level with exact or nearly exact matches (?98%). The results confirm the presence of several Culicoides species, including proven vectors in Central Tunisia. Blood meal analyses show that these species will indeed feed on bigger mammals, thereby highlighting the risk that these viruses will be able to spread in Tunisia.

Project description:BackgroundTibet Orbivirus (TIBOV) is a recently discovered Orbivirus known to infect cattle, Asian buffalo and goats in south-western China. It was first isolated from mosquitoes and subsequently from biting midges (Culicoides spp.) in Yunnan, China, indicating that it is an arbovirus. Little is known of its potential to cause disease, but the economic importance of related viruses promoted an investigation of potential Culicoides spp. vectors of TIBOV.MethodsBiting midges were collected approximately once per week between May and December 2020, at a cattle farm in Wulong village, Shizong County, Yunnan Province, China. Approximately 3000 specimens of nine species were subsequently used in attempts to isolate virus, and a further 2000 specimens of six species were tested for the presence of bluetongue virus (BTV) and TIBOV using a RT-qPCR test.ResultsVirus isolation attempts resulted in the isolation of three viruses. One isolate from a pool of Culicoides jacobsoni was identified as TIBOV, while the other two viruses from C. orientalis and C. tainanus remain unidentified but are not BTV or TIBOV. RT-qPCR analysis did not detect BTV in any specimens, but a single pool containing five specimens of C. jacobsoni and another containing five specimens of C. tainanus produced PCR quantification cycle (Cq) values of around 28 that may indicate infection with TIBOV.ConclusionsThe isolation of TIBOV from C. jacobsoni satisfies one criterion required to prove its status as a vector of this virus. This isolation is supported by a low Cq value produced from a different pool of this species in the RT-qPCR test. The low Cq value obtained from a pool of C. tainanus suggests that this species may also be able to satisfy this criterion. Both of these species are widespread throughout Asia, with C. jacobsoni extending into the Pacific region, which raises the possibility that TIBOV may be more widespread than is currently known.

Project description:Leishmania parasites, causative agents of leishmaniasis, are currently divided into four subgenera: Leishmania, Viannia, Sauroleishmania and Mundinia. The recently established subgenus Mundinia has a wide geographical distribution and contains five species, three of which have the potential to infect and cause disease in humans. While the other Leishmania subgenera are transmitted exclusively by phlebotomine sand flies (Diptera: Psychodidae), natural vectors of Mundinia remain uncertain. This study investigates the potential of sand flies and biting midges of the genus Culicoides (Diptera: Ceratopogonidae) to transmit Leishmania parasites of the subgenus Mundinia. Sand flies (Phlebotomus argentipes, P. duboscqi and Lutzomyia migonei) and Culicoides biting midges (Culicoides sonorensis) were exposed to five Mundinia species through a chicken skin membrane and dissected at specific time intervals post bloodmeal. Potentially infected insects were also allowed to feed on ear pinnae of anaesthetized BALB/c mice and the presence of Leishmania DNA was subsequently confirmed in the mice using polymerase chain reaction analyses. In C. sonorensis, all Mundinia species tested were able to establish infection at a high rate, successfully colonize the stomodeal valve and produce a higher proportion of metacyclic forms than in sand flies. Subsequently, three parasite species, L. martiniquensis, L. orientalis and L. sp. from Ghana, were transmitted to the host mouse ear by C. sonorensis bite. In contrast, transmission experiments entirely failed with P. argentipes, although colonisation of the stomodeal valve was observed for L. orientalis and L. martiniquensis and metacyclic forms of L. orientalis were recorded. This laboratory-based transmission of Mundinia species highlights that Culicoides are potential vectors of members of this ancestral subgenus of Leishmania and we suggest further studies in endemic areas to confirm their role in the lifecycles of neglected pathogens.

Project description:Biting midges of the genus Culicoides (Diptera, Ceratopognidae) are demonstrably or putatively involved in the transmission of both bluetongue (BTV) and Schmallenberg viruses (SBV) in Central Europe. Although these insects are ubiquitous in Europe, relatively little is known about their requirements in terms of breeding habitats and substrates. Culicoides species composition and relative abundance in potential breeding habitats were therefore studied at various locations in Northeastern Germany and one location in Western Germany by emergence trap collections. Forty-three potential breeding sites were analyzed in ten landscape structures, with 28,091 adult biting midges emerging from them. Among these, 2116 specimens belonged to the genus Culicoides. Species of the culicoid subgenus Avaritia were most abundant (70.6% of all specimens) and widespread (91.6% of all sites), while the subgenus Culicoides accounted for 15.6% of the specimens registered but emerged from 70.8% of all sites sampled. Culicoides species of other subgenera were collected in 75.0% of all studied sites, with a relative abundance of 8.7%. The results indicate that various types of dung, but probably also some landscape habitats, offer suitable substrates for the development of potential Culicoides vector species. Adaptations in dung management on farms and landscape design and use might therefore be appropriate approaches to reduce the risk of BTV or SBV transmission.

Project description:BackgroundBiting midges, Culicoides, of the Obsoletus group and the Pulicaris group have been involved in recent outbreaks of bluetongue virus and the former was also involved in the Schmallenberg virus outbreak in northern Europe.MethodsFor the first time, here we investigate the local abundance pattern of these two species groups in the field by intensive sampling with a grid of light traps on 16 catch nights. Neighboring trap catches can be spatially dependent on each other, hence we developed a conditional autoregressive (CAR) model framework to test a number of spatial and non-spatial covariates expected to affect Culicoides abundance.ResultsThe distance to sheep penned in the corner of the study field significantly increased the abundance level up to 200 meters away from the sheep. Spatial clustering was found to be significant but could not be explained by any known factors, and cluster locations shifted between catch nights. No significant temporal autocorrelation was detected. CAR models for both species groups identified a significant positive impact of humidity and significant negative impacts of precipitation and wind turbulence. Temperature was also found to be significant with a peak at just below 16 degrees Celcius. Surprisingly, there was a significant positive impact of wind speed. The CAR model for the Pulicaris group also identified a significant attraction to the smaller groups of sheep placed in the field. Furthermore, a large number of spatial covariates which were incorrectly found to be significant in ordinary regression models were not significant in the CAR models. The 95% C.I. on the prediction estimates ranged from 20.4% to 304.8%, underlining the difficulties of predicting the abundance of Culicoides.ConclusionsWe found that significant spatial clusters of Culicoides moved around in a dynamic pattern varying between catch nights. This conforms with the modeling but was not explained by any of the tested covariates. The mean abundance within these clusters was up to 11 times higher for the Obsoletus group and 4 times higher for the Pulicaris group compared to the rest of the field.

Project description:The species of the Culicoides genus are hematophagous, and some of them are vectors of important human and animal diseases. This group of insects is distributed worldwide, varying according to local species. Knowledge of the geographic distribution of specific species is crucial for the development and implementation of control strategies. The aim of this work was to investigate the occurrence of Culicoides in the state of Alagoas in northeast Brazil. Midges were captured with CDC light traps, and their identification and morphological analyses were performed by the Ceratopogonidae Collection of the Oswaldo Cruz Foundation (FIOCRUZ/CCER) in Rio de Janeiro, Brazil. Morphological analyses were performed using the key to Culicoides from the guttatus group and comparison with other deposited specimens. DNA sequencing, genetic analysis and comparison with sequences in the Genbank database, confirmed the identification of the flies as Culicoides insignis. This was the first formal report of C. insignis being found in Alagoas.

Project description:The genus Culicoides includes vectors of important animal diseases such as bluetongue and Schmallenberg virus (BTV and SBV). This genus includes 1300 species classified in 32 subgenera and 38 unclassified species. However, the phylogenetic relationships between different subgenera of Culicoides have never been studied. Phylogenetic analyses of 42 species belonging to 12 subgenera and 8 ungrouped species of genus Culicoides from Ecuador, France, Gabon, Madagascar and Tunisia were carried out using two molecular markers (28S rDNA D1 and D2 domains and COI mtDNA). Sequences were subjected to non-probabilistic (maximum parsimony) and probabilistic (Bayesian inference (BI)) approaches. The subgenera Monoculicoides, Culicoides, Haematomyidium, Hoffmania, Remmia and Avaritia (including the main vectors of bluetongue disease) were monophyletic, whereas the subgenus Oecacta was paraphyletic. Our study validates the subgenus Remmia (= Schultzei group) as a valid subgenus, outside of the subgenus Oecacta. In Europe, Culicoides obsoletus, Culicoides scoticus and Culicoides chiopterus should be part of the Obsoletus complex whereas Culicoides dewulfi should be excluded from this complex. Our study suggests that the current Culicoides classification needs to be revisited with modern tools.

Project description:Climate change is affecting the distribution of pathogens and their arthropod vectors worldwide, particularly at northern latitudes. The distribution of Culicoides sonorensis (Diptera: Ceratopogonidae) plays a key role in affecting the emergence and spread of significant vector borne diseases such as Bluetongue (BT) and Epizootic Hemorrhagic Disease (EHD) at the border between USA and Canada. We used 50 presence points for C. sonorensis collected in Montana (USA) and south-central Alberta (Canada) between 2002 and 2012, together with monthly climatic and environmental predictors to develop a series of alternative maximum entropy distribution models. The best distribution model under current climatic conditions was selected through the Akaike Information Criterion, and included four predictors: Vapour Pressure Deficit of July, standard deviation of Elevation, Land Cover and mean Precipitation of May. This model was then projected into three climate change scenarios adopted by the IPCC in its 5th assessment report and defined as Representative Concentration Pathways (RCP) 2.6, 4.5 and 8.5. Climate change data for each predictor and each RCP were calculated for two time points pooling decadal data around each one of them: 2030 (2021-2040) and 2050 (2041-2060). Our projections showed that the areas predicted to be at moderate-high probability of C. sonorensis occurrence would increase from the baseline scenario to 2030 and from 2030 to 2050 for each RCP. The projection also indicated that the current northern limit of C. sonorensis distribution is expected to move northwards to above 53°N. This may indicate an increased risk of Culicoides-borne diseases occurrence over the next decades, particularly at the USA-Canada border, as a result of changes which favor C. sonorensis presence when associated to other factors (i.e. host and pathogen factors). Recent observations of EHD outbreaks in northern Montana and southern Alberta supported our projections and considerations. The results of this study can inform the development of cost effective surveillance programs, targeting areas within the predicted limits of C. sonorensis geographical occurrence under current and future climatic conditions.

Project description:BACKGROUND:Stable isotope labeling is a promising method for use in insect mark-capture and dispersal studies. Culicoides biting midges, which transmit several important animal pathogens, including bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV), are small flies that develop in various semi-aquatic habitats. Previous Culicoides dispersal studies have suffered from the limitations of other labeling techniques, and an inability to definitively connect collected adult midges to specific immature development sites. RESULTS:Adult C. sonorensis were successfully labeled with 13C and 15N stable isotopes as larvae developing in a semi-aquatic mud substrate in the laboratory. High and low-dose isotope treatments for both elements significantly enriched midges above the background isotope levels of unenriched controls. Enrichment had no effect on C. sonorensis survival, though a slight (~ 5 day) delay in emergence was observed, and there was no significant effect of pool size on 13C or 15N enrichment levels. CONCLUSIONS:Stable isotope labeling is life-long, and does not interfere with natural insect behaviors. Stable isotope enrichment using 13C or 15N shows promise for Culicoides dispersal studies in the field. This method can be used to identify adult dispersal from larval source habitat where a midge developed. It may be possible to detect a single enriched midge in a pool of unenriched individuals, though further testing is needed to confirm the sensitivity of this method.
