Characterization of furnace oil bioremediation potential of hydrocarbonoclastic bacteria isolated from petroleum contaminated sites of the Sundarbans, Bangladesh.

ABSTRACT: Spillage of furnace oil is a more frequent event in recent times. In this study, environmental samples from furnace oil spillage sites of the Shela River, the Sundarbans, Bangladesh, were collected after three weeks of spillage. Serial dilution was applied and total seven bacterial isolates were separated as pure cultures. The oil-degrading potentiality of all seven isolates was further assessed, confirmed and compared with the growth pattern in furnace oil supplemented media, 2, 6-dichlorophenolindophenol test, and gravimetric analysis. After 7 days of incubation, isolates SS3, RW2, and SB degraded 56%, 43%, and 52% of supplemented furnace oil, respectively. The top three hydrocarbonoclastic bacterial isolates were selected as potential and identified as Pseudomonas aeruginosa (SS3), Bacillus sp. (RW2), and Serratia sp. (SB). All three isolates showed significant oil-degrading capacity compared to negative control, when incubated in sterile pond water supplemented with 2% furnace oil, suggesting them as potential bioremediation agents.

Project description:In this study, we compared the culturability of aerobic bacteria isolated from long-term oil-contaminated soils via enrichment and direct-plating methods; bacteria were cultured at 30°C or ambient temperatures. Two soil samples were collected from two sites in the Shengli oilfield located in Dongying, China. One sample (S0) was close to the outlet of an oil-production water treatment plant, and the other sample (S1) was located 500 m downstream of the outlet. In total, 595 bacterial isolates belonging to 56 genera were isolated, distributed in Actinobacteria, Firmicutes, Bacterioidetes, and Proteobacteria. It was interesting that Actinobacteria and Firmicutes were not detected from the 16S rRNA gene clone library. The results suggested the activation of rare species during culture. Using the enrichment method, 239 isolates (31 genera) and 96 (22 genera) isolates were obtained at ambient temperatures and 30°C, respectively, from S0 soil. Using the direct-plating method, 97 isolates (15 genera) and 163 isolates (20 genera) were obtained at ambient temperatures and 30°C, respectively, from two soils. Of the 595 isolates, 244 isolates (41.7% of total isolates) could degrade n-hexadecane. A greater number of alkane-degraders was isolated at ambient temperatures using the enrichment method, suggesting that this method could significantly improve bacterial culturability. Interestingly, the proportion of alkane degrading isolates was lower in the isolates obtained using enrichment method than that obtained using direct-plating methods. Considering the greater species diversity of isolates obtained via the enrichment method, this technique could be used to increase the diversity of the microbial consortia. Furthermore, phenol hydroxylase genes (pheN), medium-chain alkane monooxygenases genes (alkB and CYP153A), and long-chain alkane monooxygenase gene (almA) were detected in 60 isolates (11 genotypes), 91 isolates (27 genotypes) and 93 isolates (24 genotypes), and 34 isolates (14 genotypes), respectively. This study could provide new insights into microbial resources from oil fields or other environments, and this information will be beneficial for bioremediation of petroleum contamination and for other industrial applications.

Project description:Petroleum is, up to this date, an inimitable nonrenewable energy resource. Petroleum leakage, which arises during transport, storage, and refining, is the most important contaminant in the environment, as it produces harm to the surrounding ecosystem. Bioremediation is an efficient method used to treat petroleum hydrocarbon-contaminated soil using indigenous microorganisms. The degradation characteristics for a variety of hydrocarbons (hexane, benzene, gasoline, and diesel) were qualitatively and quantitatively investigated using Bacillus isolates. Microbiological and biochemical methods have been used including isolation of oil-degrading bacteria, enzymatic activities, the determination of physicochemical parameters, biosurfactant production and extraction assay, oil displacement assay, antimicrobial assay of the biosurfactants, and bioremediation kinetics. Consequently, of the 60 isolates capable of degrading different hydrocarbons at fast rates, 34 were suspected to be Bacillus isolates capable of growing in 24 h or 48 h on BH medium supplemented with 2% of hexane, benzene, gasoline, diesel, and olive oil, respectively. Among the 34 isolates, 61% (21/34) are capable of producing biosurfactant-like molecules by using gasoline, 70% (24/34) with diesel oil, 85% (29/34) with hexane, and 82% (28/34) with benzene. It was found that biosurfactant-producing isolates are extractable with HCl (100%), ammonium sulphate (95%), chloroform (95%), and ethanol (100%). Biosurfactants showed stability at 20°C, 37°C, 40°C, and 60°C. Biosurfactant secreted by Bacillus strains has shown an antagonistic effect in Escherichia coli, Shigella flexneri 5a M90T, and Bacillus cereus. The selected isolates could therefore be safely used for biodegradation. Substrate biodegradation patterns by individual isolates were found to significantly differ. The study shows that benzene was degraded faster, followed by hexane, gasoline, and finally diesel. The Bacillus consortium used can decrease hydrocarbon content from 195 to 112 (g/kg) in 15 days.

Project description:Soil remediation industries continue to seek technologies to speed-up treatment and reduce operating costs. Some processes are energy intensive and, in some cases, transport can be the main source of carbon emissions. Residual fertilizing materials (RFM), such as organic residues, have the potential to be beneficial bioremediation agents. Following a circular economy framework, we investigated the feasibility of sourcing RFMs locally to reduce transport and assess possible bioremediation efficiency gains. RFMs were recruited within 100 km of the treatment site: ramial chipped wood (RCW), horse manure (MANR) and brewer spent grain (BSG). They were added to the land treatment unit's baseline fertilizer treatment (FERT, "F") to measure if they improved the remediation efficiency of an engine oil-contaminated soil (7,500 ± 100 mg kg-1). Results indicate that MANR-F was the only amendment more effective than FERT for petroleum hydrocarbons (PHC) reduction, while emitting the least CO2 overall. RCW-F was equivalent to FERT but retained more moisture. Although BSG contributed the most nitrogen to the soil, BSG-F retained excessive moisture, emitted more volatile organic compounds, contained less soil O2, and was less effective than the baseline treatment. Significantly more of the C16-C22 fraction was removed (63% ± 22%) than all other fractions (C22-C28, C28-C34, C34-C40), which were equally removed. Microbial community-level physiological profiling was conducted with Biolog Ecoplates™, and catabolic diversity differed between treatments (utilization rates of 31 carbon sources). MANR-F has the potential to increase PHC-remediation speed and efficiency compared to inorganic fertilizer alone. Other RFM promote moisture retention and diverse microbial catabolic activity. A variety of RFM are present across the globe and some can offer low-cost amendments to boost remediation efficiency, while reducing treatment time compared to traditional fertilizer-only methods.
