Complete Genome Sequence of an Alphabaculovirus from the Southern Armyworm, Spodoptera eridania.

ABSTRACT: We report the complete genome sequence of a baculovirus from the moth Spodoptera eridania, the southern armyworm. The genome sequence is 149,090?bp and exhibits the greatest degree of sequence similarity with genomes from alphabaculoviruses isolated from other moths of the genus Spodoptera.

Purification and characterization of NADPH--cytochrome c reductase from the midgut of the southern armyworm (Spodoptera eridania).

Project description:1. NADPH-cytochrome c reductase was solubilized with bromelain and purified about 400-fold from sucrose/pyrophosphate-washed microsomal fractions from southern armyworm (Spodoptera eridania) larval midguts. 2. The enzyme has a mol.wt. of 70 035 +/- 1300 and contained 2 mol of flavin/mol of enzyme consisting of almost equimolar amounts of FMN and FAD. 3. Aerobic titration of the enzyme with NADPH caused the formation of a stable half-reduced state at 0.5 mol of NADPH/mol of flavin. 4. Kinetic analysis showed that the reduction of cytochrome c proceeded by a Bi Bi Ping Pong mechanism. 5. Apparent Km values for NADPH and cytochrome c and Ki values for NADP+ and 2'-AMP were considerably higher for the insect reductase than for the mammalian liver enzyme. 6. These are discussed in relation to possible differences in the active sites of the enzymes.

Properties of 5-aminolaevulinate synthetase and its relationship to microsomal mixed-function oxidation in the southern armyworm (Spodoptera eridania).

Project description:1. Activity of 5-aminolaevulinate synthetase was measured in the midgut and other tissues of the last larval instar of the southern armyworm (Spodoptera eridania Cramer, formerly Prodenia eridania Cramer). 2. Optimum conditions for measuring the activity were established with respect to all variables involved and considerable differences from those reported for mammalian enzyme preparations were found. 3. Maximum activity (20 nmol/h per mg of protein) occurs 18-24 h after the fifth moult and thereafter decreases to trace amounts as the larvae age and approach pupation. 4. Synthetase activity was rapidly induced by oral administration (in the diet) of pentamethylbenzene, phenobarbital, diethyl 1,4-dihydro-2,4,6-trimethylpyridine-3, 5-dicarboxylate, and 2-allyl-2-isopropylacetamide. 5. Puromycin inhibited the induction of synthetase by pentamethylbenzene. 6. Induction of 5-aminolaevulinate synthetase correlated well with the induction of microsomal N-demethylation of p-chloro-N-methylaniline, except for phenobarbital, which induced the microsomal oxidase relatively more than the synthetase.

The complete genome sequence of an alphabaculovirus from Spodoptera exempta, an agricultural pest of major economic significance in Africa.

Project description:Spodoptera exempta nucleopolyhedrovirus (SpexNPV) is a viral pathogen of the African armyworm, Spodoptera exempta (Lepidoptera: Noctuidae), a significant agricultural pest of cereal crops in Africa. SpexNPV has been evaluated as a potential insecticide for control of this pest and has served as the subject of research on baculovirus pathology and transmission. Occlusion bodies (OBs) of SpexNPV isolate 244.1 were examined, and the nucleotide sequence of the genome was determined and characterized. SpexNPV-244.1 OBs consisted of irregular polyhedra with a size and appearance typical for alphabaculoviruses. Virions within the polyhedra contained 1-8 nucleocapsids per unit envelope. The SpexNPV-244.1 genome was comprised of a 129,528 bp circular sequence, in which 139 ORFs were annotated. Five homologous regions (hrs) consisting of a variable number of 28-bp imperfect palindromes were identified in the genome. The genome sequence contained the 38 core genes of family Baculoviridae, as well as three ORFs unique to the SpexNPV sequence and one ORF that was apparently acquired by horizontal gene transfer with a betabaculovirus ancestor. Phylogenetic inference with core gene amino acid sequence alignments placed SpexNPV-244.1 in a lineage containing alphabaculoviruses of Spodoptera frugiperda and Spodopotera exigua which in turn is part of a larger group of alphabaculoviruses from the subfamily Noctuinae in the lepidopteran family Noctuidae. Kimura-2-parameter pairwise nucleotide distances indicated that SpexNPV-244.1 represented a different and previously unlisted species in the genus Alphabaculovirus. Gene parity plots indicated that the gene order of SpexNPV-244.l was extensively collinear with that of Spodoptera exigua NPV (SeMNPV). These plots also revealed a group of 17 core genes whose order was conserved in other alpha- and betabaculoviruses.

Pest categorisation of Spodoptera eridania.

Project description:The EFSA Panel on Plant Health performed a pest categorisation of Spodoptera eridania (Lepidoptera: Noctuidae) for the European Union (EU). S. eridania (southern armyworm) is a highly polyphagous pest native to the Americas which has spread to Africa being first reported there in 2016. There are multiple generations per year. Although it can endure short freezing periods, prolonged frosts are lethal. Eggs are laid in batches on the leaves of host plants. Five to seven larval instars follow. Like other armyworms, early instars are gregarious and cause leaf skeletonisation. Older instars disperse and become more solitary and nocturnal. Larvae feed on field vegetables and can bore into tomato fruit. They can eat apical portions of branches and can bore into stems and tubers if preferred foods are scarce. Pupation takes place in the soil. S. eridania is regulated in the EU by Directive 2000/29/EC (Annex IAI). Within this Directive, a prohibition of soil imported from countries where S. eridania occurs, prevents the entry of S. eridania pupae. However, immature stages on plants (excluding seeds), fruit and flowers provide potential pathways for entry into the EU. S. eridania adults have been intercepted in the EU as hitchhikers. Climatic conditions and the wide availability of host plants provide conditions to support establishment in frost-free regions of the EU. It could spread more widely forming transient populations during summer months. Impacts on field vegetables and ornamentals would be possible. Phytosanitary measures are available to reduce the likelihood of entry. S. eridania satisfies the criteria that are within the remit of EFSA to assess for it to be regarded as a potential Union quarantine pest. S. eridania does not meet the criteria of (a) occurring in the EU, and (b) plants for planting being the principal means of spread for it to be regarded as a potential Union regulated non-quarantine pest.

Project description:1. A potent inhibitor of microsomal mixed-function oxidation has been isolated from the gut contents of larvae of the southern armyworm (Prodenia eridania). 2. Progressive inhibition is associated with a soluble proteinase with a molecular weight of approx. 26000. 3. Although the proteolytic activity of the inhibitor can be reduced by phenylmethanesulphonyl fluoride and other reagents, these materials are only partially effective in protecting the microsomal enzymes. 4. Inhibitory activity is decreased in the presence of bovine serum albumin.

Mechanism of inhibition of microsomal mixed-function oxidation by the gut-contents inhibitor of the southern armyworm (Prodenia eridania).

Project description:A potent inhibitor of microsomal mixed-function oxidation reactions in insects had previously been isolated and partially purified from the gut contents of Prodenia eridania and shown to be associated with proteinase activity. Incubation of rat liver microsomal fraction with low concentrations of this inhibitor led to solubilization of NADPH-cytochrome c reductase, which was paralleled by the inactivation of reduction of cytochrome P-450 by NADPH and by the inhibition of NADPH-linked benzo[3,4]pyrene hydroxylation and aminopyrine demethylation. There was little or no effect on cytochromes b(5) and P-450, nor was the capacity of the latter catalyst to combine with exogenous substrates decreased. Contrary to the findings with NADPH, preincubation of microsomal fraction with the inhibitor did not cause a significant decrease in the rate of cytochrome P-450 reduction by NADH, supporting the assumption that different catalysts are involved in the electron transfer from NADH and NADPH to cytochrome P-450. The findings indicate the importance of taking the possible presence of endogenous inhibitors into consideration when evaluating low or absent mixed-function oxidation activities found in insect systems in vitro.

We have determined a 15,388?bp mitogenome of S. frugiperda which includes 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNAs. The base composition was AT-biased (81.3%). Phylogenetic trees present that Korean S. frugiperda placed in basal position of S. frugiperda clade. S. frugiperda mitochondrial genome can be used for understanding recent active migration of S. frugiperda.
