Direct Formation of Sub-Micron and Nanoparticles of a Bioinspired Coordination Polymer Based on Copper with Adenine.

ABSTRACT: We report on the use of different reaction conditions, e.g., temperature, time, and/or concentration of reactants, to gain control over the particle formation of a bioinspired coordination polymer based on copper(II) and adenine, allowing homogeneous particle production from micro- to submicro-, and up to nano-size. Additionally, studies on this reaction carried out in the presence of different surfactants gives rise to the control of the particle size due to the modulation of the electrostatic interactions. Stability of the water suspensions obtained within the time and pH has been evaluated. We have also studied that there is no significant effect of the size reduction in the magnetic properties of the Cu(II)-adenine coordination polymer.

Bioinspired Theranostic Coordination Polymer Nanoparticles for Intranasal Dopamine Replacement in Parkinson's Disease.

Project description:Dopamine (DA) is one of the main neurotransmitters found in the central nervous system and has a vital role in the function of dopaminergic (DArgic) neurons. A progressive loss of this specific subset of cells is one of the hallmarks of age-related neurodegenerative disorders such as Parkinson's disease (PD). Symptomatic therapy for PD has been centered in the precursor l-DOPA administration, an amino acid precursor of DA that crosses the blood-brain barrier (BBB) while DA does not, although this approach presents medium- to long-term side effects. To overcome this limitation, DA-nanoencapsulation therapies are actively being searched as an alternative for DA replacement. However, overcoming the low yield of encapsulation and/or poor biodistribution/bioavailability of DA is still a current challenge. Herein, we report the synthesis of a family of neuromelanin bioinspired polymeric nanoparticles. Our system is based on the encapsulation of DA within nanoparticles through its reversible coordination complexation to iron metal nodes polymerized with a bis-imidazol ligand. Our methodology, in addition to being simple and inexpensive, results in DA loading efficiencies of up to 60%. In vitro, DA nanoscale coordination polymers (DA-NCPs) exhibited lower toxicity, degradation kinetics, and enhanced uptake by BE(2)-M17 DArgic cells compared to free DA. Direct infusion of the particles in the ventricle of rats in vivo showed a rapid distribution within the brain of healthy rats, leading to an increase in striatal DA levels. More importantly, after 4 days of nasal administrations with DA-NCPs equivalent to 200 μg of the free drug per day, the number and duration of apomorphine-induced rotations was significantly lower from that in either vehicle or DA-treated rats performed for comparison purposes. Overall, this study demonstrates the advantages of using nanostructured DA for DA-replacement therapy.

Control of hierarchical polymer mechanics with bioinspired metal-coordination dynamics.

Project description:In conventional polymer materials, mechanical performance is traditionally engineered via material structure, using motifs such as polymer molecular weight, polymer branching, or block copolymer design. Here, by means of a model system of 4-arm poly(ethylene glycol) hydrogels crosslinked with multiple, kinetically distinct dynamic metal-ligand coordinate complexes, we show that polymer materials with decoupled spatial structure and mechanical performance can be designed. By tuning the relative concentration of two types of metal-ligand crosslinks, we demonstrate control over the material's mechanical hierarchy of energy-dissipating modes under dynamic mechanical loading, and therefore the ability to engineer a priori the viscoelastic properties of these materials by controlling the types of crosslinks rather than by modifying the polymer itself. This strategy to decouple material mechanics from structure is general and may inform the design of soft materials for use in complex mechanical environments. Three examples that demonstrate this are provided.

Multifunctional nanozyme-reinforced copper-coordination polymer nanoparticles for drug-resistance bacteria extinction and diabetic wound healing.

Project description:BackgroundDrug-resistant bacterial infections in chronic wounds are a persistent issue, as they are resistant to antibiotics and can cause excessive inflammation due to generation of reactive oxygen species (ROS). An effective solution would be to not only combat bacterial infections but also scavenge ROS to relieve inflammation at the wound site. Scaffolds with antioxidant properties are attractive for their ability to scavenge ROS, and there is medical demand in developing antioxidant enzyme-mimicking nanomaterials for wound healing.MethodsIn this study, we fabricated copper-coordination polymer nanoparticles (Cu-CPNs) through a self-assembly process. Furthermore, ε-polylysine (EPL), an antibacterial and cationic polymer, was integrated into the Cu-CPNs structure through a simple one-pot self-assembly process without sacrificing the glutathione peroxidase (GPx) and superoxide dismutase (SOD)-mimicking activity of Cu-CPNs.ResultsThe resulting Cu-CPNs exhibit excellent antioxidant propertiesin mimicking the activity of glutathione peroxidase and superoxide dismutase and allowing them to effectively scavenge harmful ROS produced in wound sites. The in vitro experiments showed that the resulting Cu-CPNs@EPL complex have superior antioxidant properties and antibacterial effects. Bacterial metabolic analysis revealed that the complex mainly affects the cell membrane integrity and nucleic acid synthesis that leads to bacterial death.ConclusionsThe Cu-CPNs@EPL complex has impressive antioxidant properties and antibacterial effects, making it a promising solution for treating drug-resistant bacterial infections in chronic wounds. The complex's ability to neutralize multiple ROS and reduce ROS-induced inflammation can help relieve inflammation at the wound site. Synthesis of [Fe(Leq)(Lax)] n coordination polymer nanoparticles using blockcopolymer micelles.

Project description:Spin-crossover compounds are a class of materials that can change their spin state from high spin (HS) to low spin (LS) by external stimuli such as light, pressure or temperature. Applications demand compounds with defined properties concerning the size and switchability that are maintained when the compound is integrated into composite materials. Here, we report the synthesis of [Fe(Leq)(Lax)]n coordination polymer (CP) nanoparticles using self-assembled polystyrene-block-poly(4-vinylpyridine) (PS-b-P4VP) block copolymer (BCP) micelles as template. Variation of the solvent (THF and toluene) and the rigidity of the axial ligand Lax (Lax = 1,2-di(pyridin-4-yl)ethane) (bpea), trans-1,2-di(pyridin-4-yl)ethene (bpee), and 1,2-di(pyridin-4-yl)ethyne) (bpey); Leq = 1,2-phenylenebis(iminomethylidyne)-bis(2,4-pentanedionato)(2-)) allowed the determination of the preconditions for the selective formation of nanoparticles. A low solubility of the CP in the used solvent and a high stability of the Fe-L bond with regard to ligand exchange are necessary for the formation of composite nanoparticles where the BCP micelle is filled with the CP, as in the case of the [FeLeq(bpey)] n @BCP. Otherwise, in the case of more flexible ligands or ligands that lead to high spin complexes, the formation of microcrystals next to the CP-BCP nanoparticles is observed above a certain concentration of [Fe(Leq)(Lax)] n . The core of the nanoparticles is about 45 nm in diameter due to the templating effect of the BCP micelle, independent of the used iron complex and [Fe(Leq)(Lax)] n concentration. The spin-crossover properties of the composite material are similar to those of the bulk for FeLeq(bpea)] n @BCP while pronounced differences are observed in the case of [FeLeq(bpey)] n @BCP nanoparticles.

A bioinspired molybdenum-copper molecular catalyst for CO2 electroreduction.

Project description:Non-noble metal molecular catalysts mediating the electrocatalytic reduction of carbon dioxide are still scarce. This work reports the electrochemical reduction of CO2 to formate catalyzed by the bimetallic complex [(bdt)MoVI(O)S2CuICN]2- (bdt = benzenedithiolate), a mimic of the active site of the Mo-Cu carbon monoxide dehydrogenase enzyme (CODH2). Infrared spectroelectrochemical (IR-SEC) studies coupled with density functional theory (DFT) computations revealed that the complex is only a pre-catalyst, the active catalyst being generated upon reduction in the presence of CO2. We found that the two-electron reduction of [(bdt)MoVI(O)S2CuICN]2- triggers the transfer of the oxo moiety to CO2 forming CO3 2- and the complex [(bdt)MoIVS2CuICN]2- and that a further one-electron reduction is needed to generate the active catalyst. Its protonation yields a reactive MoVH hydride intermediate which reacts with CO2 to produce formate. These findings are particularly relevant to the design of catalysts from metal oxo precursors.

Poly[di-μ-glycinato-copper(II)]: a two-dimensional coordination polymer.

Project description:The title coordination polymer, [Cu(C(2)H(4)NO(2))(2)](n), is two-dimensional and consists of a distorted octa-hedral copper coordination polyhedron with two bidentate glycine ligands chelating the metal through the O and N atoms in a trans-square-planar configuration. The two axial coordination sites are occupied by carbonyl O atoms of neighbouring glycine mol-ecules. The Cu-O distances for the axial O atoms [2.648 (2) and 2.837 (2) Å] are considerably longer than both the Cu-O [1.9475 (17) and 1.9483 (18) Å] and Cu-N [1.988 (2) and 1.948 (2) Å] distances in the equatorial plane, which indicates a strong Jahn-Teller effect. In the crystal, the two-dimensional networks are arranged parallel to (001) and are linked via N-H⋯O hydrogen bonds, forming a three-dimensional arrangement.

Photoactivable Ruthenium-Based Coordination Polymer Nanoparticles for Light-Induced Chemotherapy.

Project description:Green light photoactive Ru-based coordination polymer nanoparticles (CPNs), with chemical formula [[Ru(biqbpy)]1.5(bis)](PF6)3 (biqbpy = 6,6'-bis[N-(isoquinolyl)-1-amino]-2,2'-bipyridine; bis = bis(imidazol-1-yl)-hexane), were obtained through polymerization of the trans-[Ru(biqbpy)(dmso)Cl]Cl complex (Complex 1) and bis bridging ligands. The as-synthesized CPNs (50 ± 12 nm diameter) showed high colloidal and chemical stability in physiological solutions. The axial bis(imidazole) ligands coordinated to the ruthenium center were photosubstituted by water upon light irradiation in aqueous medium to generate the aqueous substituted and active ruthenium complexes. The UV-Vis spectral variations observed for the suspension upon irradiation corroborated the photoactivation of the CPNs, while High Performance Liquid Chromatography (HPLC) of irradiated particles in physiological media allowed for the first time precisely quantifying the amount of photoreleased complex from the polymeric material. In vitro studies with A431 and A549 cancer cell lines revealed an 11-fold increased uptake for the nanoparticles compared to the monomeric complex [Ru(biqbpy)(N-methylimidazole)2](PF6)2 (Complex 2). After irradiation (520 nm, 39.3 J/cm2), the CPNs yielded up to a two-fold increase in cytotoxicity compared to the same CPNs kept in the dark, indicating a selective effect by light irradiation. Meanwhile, the absence of 1O2 production from both nanostructured and monomeric prodrugs concluded that light-induced cell death is not caused by a photodynamic effect but rather by photoactivated chemotherapy.

Mesosphere of Carbon-Shelled Copper Nanoparticles with High Conductivity and Thermal Stability via Direct Carbonization of Polymer Soft Templates.

Project description:Copper nanoparticle (Cu NP) is a promising replacement for noble metal nanoparticles due to its high electrical conductivity and low cost. However, Cu NPs are relatively active compared to noble metals, and current ways of protecting Cu NPs from oxidation by encapsulation have severe drawbacks, such as a long reaction time and complicated processes. Here, a facial and effective method to prepare the mesosphere of carbon-shelled copper nanoparticles (Cu@MC) was demonstrated, and the resulting Cu@MC was both highly electrically conductive and thermally stable. Cu@organic (100 nm) was first synthesized by the reduction of Cu ions with poly (vinyl pyrrolidone) (PVP) and sodium poly ((naphthalene-formaldehyde) sulfonate) (Na-PNFS) as soft templates. Then, the carbon shells were obtained by in situ carbonization of the polymer soft templates. The Cu@organic and Cu@MC showed an anti-oxidation ability up to 175 and 250 °C in the air atmosphere, respectively. Furthermore, the Cu@MC exhibited excellent volume resistivity of 7.2 × 10-3 Ω·cm under 20 MPa, and showed promising application potential in electric sensors and devices.

Multiple Roles of Alkanethiolate-Ligands in Direct Formation of H2 O2 over Pd Nanoparticles.

Project description:Coadsorbed organic species including thiolates can promote direct synthesis of hydrogen peroxide from H2 and O2 over Pd particles. Here, density functional theory based kinetic modeling, augmented with activity measurements and vibrational spectroscopy are used to provide atomistic understanding of direct H2 O2 formation over alkylthiolate(RS) Pd. We find that the RS species are oxidized during reaction conditions yielding RSO2 as the effective ligand. The RSO2 ligand shows superior ability for proton transfer to the intermediate surface species OOH, which accelerates the formation of H2 O2 . The ligands promote the selectivity also by blocking sites for unselective water formation and by modifying the electronic structure of Pd. The work rationalizes observations of enhanced selectivity of direct H2 O2 formation over ligand-funtionalized Pd nanoparticles and shows that engineering of organic surface modifiers can be used to promote desired hydrogen transfer routes.

Site Communication in Direct Formation of H2O2 over Single-Atom Pd@Au Nanoparticles.

Project description:Single atom alloy catalysts offer possibilities to obtain turnover frequencies and selectivities unattainable by their monometallic counterparts. One example is direct formation of H2O2 from O2 and H2 over Pd embedded in Au hosts. Here, a first-principles-based kinetic Monte Carlo approach is developed to investigate the catalytic performance of Pd embedded in Au nanoparticles in an aqueous solution. The simulations reveal an efficient site separation where Pd monomers act as active centers for H2 dissociation, whereas H2O2 is formed over undercoordinated Au sites. After dissociation, atomic H may undergo an exothermic redox reaction, forming a hydronium ion in the solution and a negative charge on the surface. H2O2 is preferably formed from reactions between dissolved H+ and oxygen species on the Au surface. The simulations show that tuning the nanoparticle composition and reaction conditions can enhance the selectivity toward H2O2. The outlined approach is general and applicable for a range of different hydrogenation reactions over single atom alloy nanoparticles.
