The dependences of the concentrations of the non-ligated, uni-ligated and bi-ligated forms of a molecule that binds two molecules of ligand are expressed as functions of the logarithm of free ligand concentration by means of hyperbolic functions. Expressions are also given for the saturation both of an individual site and of the molecule as a whole. This form of expression allows derivation of the following points. 2. The sharpness of bell-shaped curves of concentration of the uni-ligated form is analysed in terms of the heights of their points of inflexion; these can rise to 1/ radical2 of the curve. 3. A single group can exhibit a doubly sigmoid saturation curve if this group and another have comparable affinities for a ligand, and if ligand binding at one of them diminishes the affinity at the other. If the molecular pK values pK(1) and pK(2) for the first and second molecules of ligand are called pK*+/-logm, so that K*(2)=K(1)K(2) and m(2)=K(1)/K(2), then the doubly sigmoid curve can be represented by the sum of two independent one-site saturation curves, in general of unequal height, of pK values pK*+/-log(1/2)[m+ radical(m(2)-4)]. The error in such representation is small either if the mutual interaction between the groups (i.e. m) is large, or if the groups have very similar affinities for the ligand. 4. The sum of two one-site saturation curves, again of pK values of pK*+/-log(1/2)[m+ radical(m(2)-4)] but of equal heights, gives a precise value for the total saturation, provided that the binding of one molecule does not promote the binding of a second, i.e. providing that m>/=2. Hence determinations of saturation cannot distinguish interacting and possibly identical sites from independent and different ones.

Project description:Age-related white matter (WM) microstructure maturation and decline occur throughout the human lifespan, complementing the process of gray matter development and degeneration. Here, we create normative lifespan reference curves for global and regional WM microstructure by harmonizing diffusion MRI (dMRI)-derived data from ten public datasets (N = 40,898 subjects; age: 3-95 years; 47.6% male). We tested three harmonization methods on regional diffusion tensor imaging (DTI) based fractional anisotropy (FA), a metric of WM microstructure, extracted using the ENIGMA-DTI pipeline. ComBat-GAM harmonization provided multi-study trajectories most consistent with known WM maturation peaks. Lifespan FA reference curves were validated with test-retest data and used to assess the effect of the ApoE4 risk factor for dementia in WM across the lifespan. We found significant associations between ApoE4 and FA in WM regions associated with neurodegenerative disease even in healthy individuals across the lifespan, with regional age-by-genotype interactions. Our lifespan reference curves and tools to harmonize new dMRI data to the curves are publicly available as eHarmonize (https://github.com/ahzhu/eharmonize).

Project description:1. A theoretical appraisal of the alternative pathway mechanism for a two-substrate enzyme shows that this mechanism is capable of giving rise to apparent substrate inhibition or substrate activation (Dalziel, 1958). It has now been shown that these phenomena may occur simultaneously in the following ways. With certain relationships between the kinetic parameters and the constant concentration of one substrate, A, the plot of initial rate, v, against the concentration of the other substrate, B, may show substrate ;activation' at low concentrations of B and substrate ;inhibition' at high concentrations of B. In other circumstances the plot of v against [B], with [A] constant, may be sigmoid (substrate activation), whereas the plot of v against [A], with [B] constant, may pass through a maximum (substrate inhibition). 2. Kinetic data for phosphofructokinase are of the latter type and it is suggested that the mechanism of this enzyme may involve a kinetically preferred pathway. It is emphasized that the phenomena of substrate inhibition and activation need not necessarily involve more than one binding site for each substrate on the enzyme molecule, nor more than one monomer per molecule.

Project description:Objectives: Post Mortem Human Surrogate (PMHS) experiments are used for describing tolerance and improve safety. For nearside impacts, the United States Standard Federal Motor Vehicle Safety Standards (FMVSS-214) used PMHS tests and binary regression methods to achieve these goals. Since this promulgation, Parametric Statistical Survival Modeling (PSSM) has become a de facto standard for developing injury risk curves (IRCs). This study is focused on pelvic injuries from side impacts. The objectives are as follows. Analyze impactor-based intact PMHS tests and develop IRCs at different AIS levels using the force metric and examine the effectiveness of other force-related variables on IRCs.Methods: Impactor-driven pelvic tests conducted using whole body PMHS were selected from published studies. The dataset had 63 tests. Peak force, 3-ms clip force, and impulse were used to develop IRCs for Abbreviated Injury Scores (AIS) AIS2+ and AIS3+, i.e., groups A and B. Brier Score Metric (BSM) was used for ranking metrics. 95% confidence intervals were computed, Normalized Confidence Interval Sizes (NCIS) were determined, and quality of the IRCs were obtained.Results: Impulse best described the underlying response of the pelvis. BSMs were the lowest for the impulse for both groups. At 10% and 50% probabilities, impulses were 71 Ns and 125 Ns for group A and 79 Ns and 160 Ns for group B; peak forces were 3.8 kN and 7.1 kN and 4 kN and 10 kN for groups A and B; and clip forces were 2.7 kN and 6.5 kN and 3.6 kN and 8.6 kN, for groups A and B. NCIS at discrete probability levels, qualities of risk curves, and individual IRCs are given.Conclusion: This study underscores the importance of using impulse to describe pelvis injury criteria in lateral impacts. These findings are applicable to anthropomorphic test devices, as matched pair tests are done to determine dummy-based injury criteria/injury assessment risk curves (IARCs). Although IRCs have been developed for WorldSID, it may be appropriate to use impulse-based IARCs. Because THOR is a potential device for automated vehicle environments, it may be appropriate to develop THOR-based IARCS. The present IRCs act as fundamental human-based injury criteria. These responses can also be used in human body and subsystem computational models.

Project description:Hyperbolic neural networks have been popular in the recent past due to their ability to represent hierarchical data sets effectively and efficiently. The challenge in developing these networks lies in the nonlinearity of the embedding space namely, the Hyperbolic space. Hyperbolic space is a homogeneous Riemannian manifold of the Lorentz group which is a semi-Riemannian manifold, i.e. a manifold equipped with an indefinite metric. Most existing methods (with some exceptions) use local linearization to define a variety of operations paralleling those used in traditional deep neural networks in Euclidean spaces. In this paper, we present a novel fully hyperbolic neural network which uses the concept of projections (embeddings) followed by an intrinsic aggregation and a nonlinearity all within the hyperbolic space. The novelty here lies in the projection which is designed to project data on to a lower-dimensional embedded hyperbolic space and hence leads to a nested hyperbolic space representation independently useful for dimensionality reduction. The main theoretical contribution is that the proposed embedding is proved to be isometric and equivariant under the Lorentz transformations, which are the natural isometric transformations in hyperbolic spaces. This projection is computationally efficient since it can be expressed by simple linear operations, and, due to the aforementioned equivariance property, it allows for weight sharing. The nested hyperbolic space representation is the core component of our network and therefore, we first compare this representation - independent of the network - with other dimensionality reduction methods such as tangent PCA, principal geodesic analysis (PGA) and HoroPCA. Based on this equivariant embedding, we develop a novel fully hyperbolic graph convolutional neural network architecture to learn the parameters of the projection. Finally, we present experiments demonstrating comparative performance of our network on several publicly available data sets.

Project description:When engineered on scales much smaller than the operating wavelength, metal-semiconductor nanostructures exhibit properties unobtainable in nature. Namely, a uniaxial optical metamaterial described by a hyperbolic dispersion relation can simultaneously behave as a reflective metal and an absorptive or emissive semiconductor for electromagnetic waves with orthogonal linear polarization states. Using an unconventional multilayer architecture, we demonstrate luminescent hyperbolic metasurfaces, wherein distributed semiconducting quantum wells display extreme absorption and emission polarization anisotropy. Through normally incident micro-photoluminescence measurements, we observe absorption anisotropies greater than a factor of 10 and degree-of-linear polarization of emission >0.9. We observe the modification of emission spectra and, by incorporating wavelength-scale gratings, show a controlled reduction of polarization anisotropy. We verify hyperbolic dispersion with numerical simulations that model the metasurface as a composite nanoscale structure and according to the effective medium approximation. Finally, we experimentally demonstrate >350% emission intensity enhancement relative to the bare semiconducting quantum wells.

Project description:The notions of Bloch wave, crystal momentum, and energy bands are commonly regarded as unique features of crystalline materials with commutative translation symmetries. Motivated by the recent realization of hyperbolic lattices in circuit quantum electrodynamics, we exploit ideas from algebraic geometry to construct a hyperbolic generalization of Bloch theory, despite the absence of commutative translation symmetries. For a quantum particle propagating in a hyperbolic lattice potential, we construct a continuous family of eigenstates that acquire Bloch-like phase factors under a discrete but noncommutative group of hyperbolic translations, the Fuchsian group of the lattice. A hyperbolic analog of crystal momentum arises as the set of Aharonov-Bohm phases threading the cycles of a higher-genus Riemann surface associated with this group. This crystal momentum lives in a higher-dimensional Brillouin zone torus, the Jacobian of the Riemann surface, over which a discrete set of continuous energy bands can be computed.

Project description:The friction coefficient between the tire and the road is one of the key parameters affecting road traffic safety. The purpose of this paper is to quantify the risk of skidding for the vehicles due to the friction evolution caused by the traffic polishing in the horizontal curve. Based on the reliability theory, an innovative dynamic risk assessment model is developed in the present study for passenger cars and trucks. The influence of two traffic characteristics for pavement friction is quantified: cumulative traffic volume (CTV) and annual average daily traffic of trucks (AADTT). The speed distribution on the horizontal curve of the motorway is obtained through field experiments as the basic parameter of the model. The Hasofer-Lind Method is adopted to solve the reliability and the risk probability of vehicle skidding. The results show that in the traffic characteristics, the AADTT has a significant impact on the pavement friction; When the AADTT on the road exceeds 2000 veh/d, the increasing CTV leads to friction decrease rapidly and therefore has a significant impact on the risk of horizontal curve. Especially for roads with more than 50 million vehicles of the CTV, the risk of the horizontal curve shows a sharp increase with CTV rising. The research results can provide reference for the road maintenance department to determine the timing of road maintenance.

Project description:Patterns of gene expressions play a key role in determining cell state. Although correlations in gene expressions have been well documented, most of the current methods treat them as independent variables. One way to take into account gene correlations is to find a low-dimensional curved geometry that describes variation in the data. Here we develop such a method and find that gene expression across multiple cell types exhibits a low-dimensional hyperbolic structure. When more genes are taken into account, hyperbolic effects become stronger but representation remains low dimensional. The size of the hyperbolic map, which indicates the hierarchical depth of the data, was the largest for human cells, the smallest for mouse embryonic cells, and intermediate in differentiated cells from different mouse organs. We also describe how hyperbolic metric can be incorporated into the t-SNE method to improve visualizations compared with leading methods.

Project description:Models of evolution of simple languages have typically assumed full alignment of the speaker and listeners interests, with perfect understanding representing the optimal outcome for both parties. In more realistic settings, communicating individuals will often desire different outcomes from one another. Previous work has shown that misalignment of speaker-listener interests reduces the maximum informativeness among Nash-equilibrium languages, and that multiple equilibrium languages (with different degrees of informativeness) are supported. We study the stochastic evolutionary dynamics of signaling games in which the alignment of speaker-listener interests can vary. We find that increased misalignment of speaker-listener interests is associated with a decrease in information transmission. Moreover, the most common languages to evolve are typically the most informative languages supportable as static Nash equilibria, suggesting a solution to the 'equilibrium selection problem'. In addition, our dynamics reveal the mechanism by which less informative languages evolve: words that previously signaled intense states come to be used hyperbolically for less intense states, with listeners' interpretation of these newly-ambiguous words evolving downward in response. We ground our results in linguistic data on intensifiers such as so and very, words which have unique dynamics-with constant recycling and innovation that match our theoretical results well.
