ABSTRACT: A mechanically strong composite hydrogel was produced based on an interpenetrating network (IPN) between gelatin and silk fibroin. When two layers of the IPN were created, the resulting hydrogel exhibited much improved mechanical properties. This hydrogel is biodegradable and non-cytotoxic and allows for cell adhesion and proliferation on the surface.

Synthesis and characterization of photocrosslinkable gelatin and silk fibroin interpenetrating polymer network hydrogels.

Project description:To effectively repair or replace damaged tissues, it is necessary to design scaffolds with tunable structural and biomechanical properties that closely mimic the host tissue. In this paper, we describe a newly synthesized photocrosslinkable interpenetrating polymer network (IPN) hydrogel based on gelatin methacrylate (GelMA) and silk fibroin (SF) formed by sequential polymerization, which possesses tunable structural and biological properties. Experimental results revealed that IPNs, where both the GelMA and SF were independently crosslinked in interpenetrating networks, demonstrated a lower swelling ratio, higher compressive modulus and lower degradation rate as compared to the GelMA and semi-IPN hydrogels, where only GelMA was crosslinked. These differences were likely caused by a higher degree of overall crosslinking due to the presence of crystallized SF in the IPN hydrogels. NIH-3T3 fibroblasts readily attached to, spread and proliferated on the surface of IPN hydrogels, as demonstrated by F-actin staining and analysis of mitochondrial activity (MTT). In addition, photolithography combined with lyophilization techniques was used to fabricate three-dimensional micropatterned and porous microscaffolds from GelMA-SF IPN hydrogels, furthering their versatility for use in various microscale tissue engineering applications. Overall, this study introduces a class of photocrosslinkable, mechanically robust and tunable IPN hydrogels that could be useful for various tissue engineering and regenerative medicine applications.

CuAAC-methacrylate interpenetrating polymer network (IPN) properties modulated by visible-light photoinitiation.

Project description:Interpenetrating polymer networks (IPNs) are a class of materials with interwoven polymers that exhibit unique blended or enhanced properties useful to a variety of applications, ranging from restorative protective materials to conductive membranes and hydrophobic adhesives. The IPN formation kinetics can play a critical role in the development of the underlying morphology and in turn the properties of the material. Dual photoinitiation of copper-catalyzed azide-alkyne (CuAAC) and radical mediated methacrylate polymerization chemistries enable the manipulation of IPN microstructure and properties by controlling the kinetics of IPN formation via the intensity of the initiating light. Specifically, azide and alkyne-based polyethylene glycol monomers and tetraethylene glycol dimethacrylate (TEGDMA) were polymerized in a single pot to form IPNs and the properties were evaluated as a function of the photoinitiating light intensity. Morphological differences as a function of intensity were observed in the IPNs as determined by thermomechanical properties and phase-contrast imaging in tapping mode atomic force microscopy (AFM). At moderate intensities (20 mW/cm2) of visible light (470 nm), the TEGDMA polymerization gels first and therefore forms the underlying network scaffold. At low intensities (0.2 mW/cm2), the CuAAC polymerization can gel first. The ability to switch sequence of gelation and IPN trajectory (simultaneous vs. sequential), affords control over phase separation behavior. Thus, light not only allows for spatial and temporal control over the IPN formation but also provides control over their thermomechanical properties, representing a route for facile IPNs design, synthesis, and application.

Processing and characterisation of a novel electropolymerized silk fibroin hydrogel membrane.

Project description:Silk fibroin can be made into various forms of biocompatible medical materials, including hydrogel due to its excellent properties. Here, we report a novel method for the preparation of electropolymerized silk fibroin hydrogel membrane (ESFHM), which is formed on a nanoporous film as a barrier using a homemade device at a higher DC voltage. Regenerated silk fibroin solution in Tris buffer (pH 6.55-7.55) was added into a reservoir with a negative charge, and the silk molecules migrated toward the positive charge at 80VDC, resulting in the formation of the ESFHM on the barrier film. Barrier film with a MWCO of 10 kDa is favourable to the formation of the ESFHM. Semi-transparent ESFHM with a swelling ratio of 1056.4% predominantly consisted of a mixture of β-sheets and α-helix crystalline structures. SEM studies revealed that the ESFHM consisted of a 3D mesh structure woven by a chain of silk fibroin nanoparticles with a size of approximately 30 nanometres, similar to a pearl necklace. In vitro studies indicated that the ESFHM was degradable and was sufficient for cell adhesion and growth. Thus, ESFHM is a promising candidate for loading bioactive protein and appropriate cells, as artificial skin or for use in transplantation.

One-pot blue-light triggered tough interpenetrating polymeric network (IPN) using CuAAC and methacrylate reactions.

Project description:An interpenetrating polymeric network (IPN) is formed in a one-pot blue-light activated scheme, where the step- and chain- growth polymerizations of the CuAAC and methacrylate reactions, respectively, are simultaneously triggered but proceed sequentially. The glassy IPN is polymerized under ambient conditions and is able to withstand high strain before failure owing to its significantly enhanced toughness. Additionally, this material exhibits shape memory attributes with readily tunable mechanical properties at high temperature.

Silk fibroin hydrogel adhesive enables sealed-tight reconstruction of meniscus tears.

Project description:Despite orientationally variant tears of the meniscus, suture repair is the current clinical gold treatment. However, inaccessible tears in company with re-tears susceptibility remain unresolved. To extend meniscal repair tools from the perspective of adhesion and regeneration, we design a dual functional biologic-released bioadhesive (S-PIL10) comprised of methacrylated silk fibroin crosslinked with phenylboronic acid-ionic liquid loading with growth factor TGF-β1, which integrates chemo-mechanical restoration with inner meniscal regeneration. Supramolecular interactions of β-sheets and hydrogen bonds richened by phenylboronic acid-ionic liquid (PIL) result in enhanced wet adhesion, swelling resistance, and anti-fatigue capabilities, compared to neat silk fibroin gel. Besides, elimination of reactive oxygen species (ROS) by S-PIL10 further fortifies localized meniscus tear repair by affecting inflammatory microenvironment with dynamic borate ester bonds, and S-PIL10 continuously releases TGF-β1 for cell recruitment and bridging of defect edge. In vivo rabbit models functionally evidence the seamless and dense reconstruction of torn meniscus, verifying that the concept of meniscus adhesive is feasible and providing a promising revolutionary strategy for preclinical research to repair meniscus tears.

Fabrication of a Double-Cross-Linked Interpenetrating Polymeric Network (IPN) Hydrogel Surface Modified with Polydopamine to Modulate the Osteogenic Differentiation of Adipose-Derived Stem Cells.

Project description:Hydrogel surface properties can be modified to form bioactive interfaces to modulate the osteogenic differentiation of stem cells. In this work, a hydrogel made of gelatin methacrylamide (GelMA) and alginate was designed and tested as a scaffold to control stem-cell osteogenic differentiation. The hydrogel's surface was treated with polydopamine (pDA) to create an adhesive layer for the adsorption of the osteoinductive drug dexamethasone (Dex). The presence of the pDA coating enhanced Dex adsorption and retention over 21 days. This effect resulted in a delay in the osteogenic differentiation of hASCs cultured on the hydrogel treated with a pDA layer.

Enzyme-Mediated Conjugation of Peptides to Silk Fibroin for Facile Hydrogel Functionalization.

Project description:Enzymatic crosslinking of tyrosine is a simple and modular method for adding functional peptides to silk fibroin (SF) hydrogels. Silk fibroin is a naturally derived polymer notable for its robust mechanical properties, biological compatibility, and versatility. Hydrogels fabricated from SF are elastic, optically clear, and have tunable moduli, however, they do not contain native biological epitopes for cell interactions. In this work we demonstrate the attachment of peptides to SF hydrogels through crosslinking of tyrosine with horseradish peroxidase (HRP) and hydrogen peroxide (H2O2). The goal was to understand the utility of this approach and to study how the addition of peptides affects the SF material properties. SF hydrogels conjugated to model peptides with different molecular weights and hydrophobic properties were studied by liquid chromatography/tandem mass spectroscopy (LC-MS/MS) (bond formation), fluorescent imaging (spatial distribution), Fourier transform infrared spectroscopy (FTIR) (protein secondary structure), and rheology (gelation time, modulus). As a proof of concept using a biologically relevant peptide, a peptide containing the cell binding domain Arg-Gly-Asp (RGD) was conjugated to SF, and the density and morphology of primary human fibroblasts were assessed. This work demonstrates a facile method for adding peptides to silk fibroin that can be adopted for a variety of biomaterials applications.

Boosting cartilage repair with silk fibroin-DNA hydrogel-based cartilage organoid precursor.

Project description:Osteoarthritis (OA), a common degenerative disease, is characterized by high disability and imposes substantial economic impacts on individuals and society. Current clinical treatments remain inadequate for effectively managing OA. Organoids, miniature 3D tissue structures from directed differentiation of stem or progenitor cells, mimic native organ structures and functions. They are useful for drug testing and serve as active grafts for organ repair. However, organoid construction requires extracellular matrix-like 3D scaffolds for cellular growth. Hydrogel microspheres, with tunable physical and chemical properties, show promise in cartilage tissue engineering by replicating the natural microenvironment. Building on prior work on SF-DNA dual-network hydrogels for cartilage regeneration, we developed a novel RGD-SF-DNA hydrogel microsphere (RSD-MS) via a microfluidic system by integrating photopolymerization with self-assembly techniques and then modified with Pep-RGDfKA. The RSD-MSs exhibited uniform size, porous surface, and optimal swelling and degradation properties. In vitro studies demonstrated that RSD-MSs enhanced bone marrow mesenchymal stem cells (BMSCs) proliferation, adhesion, and chondrogenic differentiation. Transcriptomic analysis showed RSD-MSs induced chondrogenesis mainly through integrin-mediated adhesion pathways and glycosaminoglycan biosynthesis. Moreover, in vivo studies showed that seeding BMSCs onto RSD-MSs to create cartilage organoid precursors (COPs) significantly enhanced cartilage regeneration. In conclusion, RSD-MS was an ideal candidate for the construction and long-term cultivation of cartilage organoids, offering an innovative strategy and material choice for cartilage regeneration and tissue engineering.

Bioinspired biliverdin/silk fibroin hydrogel for antiglioma photothermal therapy and wound healing.

Project description:Rationale: Photothermal therapy employs the photoabsorbers to generate heat under the near-infrared (NIR) irradiation for thermal tumor ablation. However, NIR irradiation might damage the adjacent tissue due to the leakage of the photoabsorbers and the residual materials after treatment might hinder the local healing process. A bifunctional hydrogel that holds both photothermal property and potent pro-healing ability provides a viable option to resolve this issue. Methods: In this study, we developed a bioinspired green hydrogel (BVSF) with the integration of bioproduct biliverdin into natural derived silk fibroin matrix for antiglioma photothermal therapy and wound healing. Results: The BVSF hydrogel possessed excellent and controllable photothermal activity under NIR irradiation and resulted in effective tumor ablation both in vitro and in vivo. Additionally, the BVSF hydrogel exerted anti-inflammatory effects both in vitro and in vivo, and stimulated angiogenesis and wound healing in a full-thickness defect rat model. Conclusion: Overall, this proof-of-concept study was aimed to determine the feasibility and reliability of using an all-natural green formulation for photothermal therapy and post-treatment care.
