Overexploitation is one of the main threats to biodiversity, but the intensity of this threat varies geographically. Project description:Species of Ganoderma, commonly called reishi (in Japan) or lingzhi (in China), have been used in traditional medicine for thousands of years, and their use has gained interest from pharmaceutical industries in recent years. Globally, the taxonomy of Ganoderma species is chaotic, and the taxon name Ganoderma lucidum has been used for most laccate (shiny) Ganoderma species. However, it is now known that G. lucidum sensu stricto has a limited native distribution in Europe and some parts of China. It is likely that differences in the quality and quantity of medicinally relevant chemicals occur among Ganoderma species. To determine what species are being sold in commercially available products, twenty manufactured products (e.g., pills, tablets, teas, etc.) and seventeen grow your own (GYO) kits labeled as containing G. lucidum were analyzed. DNA was extracted, and the internal transcribed spacer (ITS) region and translation elongation factor 1-alpha (tef1?) were sequenced with specific fungal primers. The majority (93%) of the manufactured reishi products and almost half of the GYO kits were identified as Ganoderma lingzhi. G. lingzhi is native to Asia and is the most widely cultivated and studied taxon for medicinal use. Illumina MiSeq sequencing of the ITS1 region was performed to determine if multiple Ganoderma species were present. None of the manufactured products tested contained G. lucidum sensu stricto, and it was detected in only one GYO kit. G. lingzhi was detected in most products, but other Ganoderma species were also present, including G. applanatum, G. australe, G. gibbosum, G. sessile, and G. sinense. Our results indicate that the content of these products vary and that better labeling is needed to inform consumers before these products are ingested or marketed as medicine. Of the 17 GYO kits tested, 11 kits contained Ganoderma taxa that are not native to the United States. If fruiting bodies of exotic Ganoderma taxa are cultivated, these GYO kits will likely end up in the environment. The effects of these exotic species to natural ecosystems needs investigation.

Project description:There is mounting evidence to suggest that the epigenetic reprogramming capacity of the oocyte is superior to that of the current factor-based reprogramming approaches and that some factor-reprogrammed induced pluripotent stem cells (iPSCs) retain a degree of epigenetic memory that can influence differentiation capacity and may be linked to the observed expression of immunogenicity genes in iPSC derivatives. One hypothesis for this differential reprogramming capacity is the "chromatin loosening/enhanced reprogramming" concept, as previously described by John Gurdon and Ian Wilmut, as well as others, which postulates that the oocyte possesses factors that loosen the somatic cell chromatin structure, providing the epigenetic and transcriptional regulatory factors more ready access to repressed genes and thereby significantly increasing epigenetic reprogramming. However, to empirically test this hypothesis a list of candidate oocyte reprogramming factors (CORFs) must be ascertained that are significantly expressed in metaphase II oocytes. Previous studies have focused on intraspecies or cross-species transcriptional analysis of up to two different species of oocytes. In this study, we have identified eight CORFs (ARID2, ASF1A, ASF1B, DPPA3, ING3, MSL3, H1FOO, and KDM6B) based on unbiased global transcriptional analysis of oocytes from three different species (human, rhesus monkey, and mouse) that both demonstrate significant (p<0.05, FC>3) expression in oocytes of all three species and have well-established roles in loosening/opening up chromatin structure. We also identified an additional 15 CORFs that fit within our proposed "chromatin opening/fate transformative" (COFT) model. These CORFs may be able to augment Shinya Yamanaka's previously identified reprogramming factors (OCT4, SOX2, KLF4, and cMYC) and potentially facilitate the removal of epigenetic memory in iPSCs and/or reduce the expression of immunogenicity genes in iPSC derivatives, and may have applications in future personalized pluripotent stem cell based therapeutics.

Project description:Unsustainable exploitation of wild species represents a serious threat to biodiversity and to the livelihoods of local communities and Indigenous peoples. However, managed, sustainable use has the potential to forestall extinctions, aid recovery, and meet human needs. We analyzed species-level data for 30,923 species from 13 taxonomic groups on the International Union for Conservation of Nature Red List of Threatened Species to investigate patterns of intentional biological resource use. Forty percent of species (10,098 of 25,009 species from 10 data-sufficient taxonomic groups) were used. The main purposes of use were pets, display animals, horticulture, and human consumption. Intentional use is currently contributing to elevated extinction risk for 28-29% of threatened or near threatened (NT) species (2752-2848 of 9753 species). Intentional use also affected 16% of all species used (1597-1631 of 10,098). However, 72% of used species (7291 of 10,098) were least concern, of which nearly half (3469) also had stable or improving population trends. The remainder were not documented as threatened by biological resource use, including at least 172 threatened or NT species with stable or improving populations. About one-third of species that had use documented as a threat had no targeted species management actions to directly address this threat. To improve use-related red-list data, we suggest small amendments to the relevant classification schemes and required supporting documentation. Our findings on the prevalence of sustainable and unsustainable use, and variation across taxa, can inform international policy making, including the Intergovernmental Science-Policy Platform on Biodiversity and Ecosystem Services, the Convention on Biological Diversity, and the Convention on International Trade in Endangered Species.

Project description:Oocysts of Cryptosporidium parvum, a zoonotic waterborne pathogen, can be removed by bivalve molluscs from contaminated water and retained on gills and in hemolymph. We identified oocysts of C. parvum in oysters from seven sites in the Chesapeake Bay area. These findings document the presence of C. parvum infectious for humans in oysters intended for human consumption.

Project description:A low-cost and scalable harvesting process was demonstrated for Chlorella sp. FC2 IITG, which offered an improved process economy for the production of a microalgal biomass feedstock via (i) the utilization of a cheaper commercial grade chemical flocculant; (ii) the recycling of post-harvested nutrient-rich spent water for the successive growth of the FC2 cells and (iii) the modulation of the flocculant dose, resulting in the non-requirement of a pH adjustment of the spent water and separate inoculum development step. Ferrous sulphate and ferric chloride were screened from a pool of four commercial grade flocculants, resulting in high harvesting efficiencies of 99.83% and 99.93% at the lower flocculant doses (g of flocculant g of dry biomass-1) of 2.5 and 3, respectively. The effect of the recycled nutrient-rich spent water and treated non-flocculated microalgal cells after harvesting was evaluated for the growth performance of the FC2 cells in six successive batches. It was found that ferrous sulphate was superior over ferric chloride in terms of the recyclability of the spent water for more number of batches, offering similar growth kinetics and nutrient recovery efficiency as compared with that of the control sample. The scale-up feasibility of the harvesting process was evaluated with a 5 L photobioreactor under indoor conditions and a 350 L open raceway pond under outdoor conditions with a modulated flocculant dose of 1.5 g ferrous sulphate. g dry biomass-1. The harvesting cost of 1 kg biomass using commercial grade ferrous sulphate was estimated to be in the range of 0.17-0.3 USD and was significantly lower as compared to that of analytical grade ferrous sulphate.

Project description:Global sustainability strategies require assessing whether countries' development trajectories are sustainable over time. However, sustainability assessments are limited because losses of natural capital and its ecosystem services through deforestation have not been comprehensively incorporated into national accounts. We update the national accounts of 80 nations that underwent tropical deforestation from 2000 to 2012 and evaluate their development trajectories using weak and strong sustainability criteria. Weak sustainability requires that countries do not decrease their aggregate capital over time. We adopt a strong sustainability criterion that countries do not decrease the value of their forest ecosystem services with respect to the year 2000. We identify several groups of countries: countries, such as Sri Lanka, Bangladesh, and India, that present sustainable development trajectories under both weak and strong sustainability criteria; countries, such as Brazil, Peru, and Indonesia, that present weak sustainable development but fail the strong sustainability criterion as a result of rapid losses of ecosystem services; countries, such as Madagascar, Laos, and Papua New Guinea, that present unsustainable development pathways as a result of deforestation; and countries, such as Democratic Republic of Congo and Sierra Leone, in which deforestation aggravates already unsustainable pathways. Our results reveal a large number of countries where tropical deforestation is both damaging to nature and not compensated by development in other sectors, thus compromising the well-being of their future generations.

Project description:The global epidemic of noncommunicable diseases (NCDs), such as cardiovascular disease and diabetes, is creating unsustainable burdens on health systems worldwide. NCDs are treatable but not curable. They are less amenable to top-down prevention and control than are the infectious diseases now in retreat. NCDs are mostly preventable, but only individuals themselves have the power to prevent and manage the diseases to which the enticements of modernity and rising prosperity have made them so susceptible (e.g., tobacco, fat-salt-carbohydrate laden food products). Rates of nonadherence to healthcare regimens for controlling NCDs are high, despite the predictable long-term ravages of not self-managing an NCD effectively. I use international data on adult functional literacy to show why the cognitive demands of today's NCD self-management (NCD-SM) regimens invite nonadherence, especially among individuals of below-average or declining cognitive capacity. I then describe ways to improve the cognitive accessibility of NCD-SM regimens, where required, so that more patients are better able and motivated to self-manage and less likely to err in life-threatening ways. For the healthcare professions, I list tools they can develop and deploy to increase patients' cognitive access to NCD-SM. Epidemiologists could identify more WHO "best buy" interventions to slow or reverse the world's "slow-motion disaster" of NCDs were they to add two neglected variables when modeling the rising burdens of disease. The neglected two are both cognitive: the distribution of cognitive capacity levels of people in a population and the cognitive complexity of their health environments.

Project description:Building upon previously published life cycle assessment (LCA) methodologies, we conducted an LCA of a commercial rainwater harvesting (RWH) system and compared it to a municipal water supply (MWS) system adapted to Washington, D.C. Eleven life cycle impact assessment (LCIA) indicators were assessed, with a functional unit of 1 m3 of rainwater and municipal water delivery system for toilets and urinals in a four-story commercial building with 1000 employees. Our assessment shows that the benchmark commercial RWH system outperforms the MWS system in all categories except Ozone Depletion. Sensitivity and performance analyses revealed pump and pumping energy to be key components for most categories, which further guides LCIA tradeoff analysis with respect to energy intensities. Tradeoff analysis revealed that commercial RWH performed better than MWS in Ozone Depletion if RWH's energy intensity was less than that of MWS by at least 0.86 kWh/m3 (249% of the benchmark MWS energy usage at 0.35 kWh/m3). RWH also outperformed MWS in Metal Depletion and Freshwater Withdrawal, regardless of energy intensities, up to 5.51 kWh/m3. An auxiliary commercial RWH system with 50% MWS reduced Ozone Depletion by 19% but showed an increase in all other impacts, which were still lower than benchmark MWS system impacts. Current models are transferrable to commercial RWH installations at other locations.

Project description:Changes in global (ocean and land) precipitation are among the most important and least well-understood consequences of climate change. Increasing greenhouse gas concentrations are thought to affect the zonal-mean distribution of precipitation through two basic mechanisms. First, increasing temperatures will lead to an intensification of the hydrological cycle ("thermodynamic" changes). Second, changes in atmospheric circulation patterns will lead to poleward displacement of the storm tracks and subtropical dry zones and to a widening of the tropical belt ("dynamic" changes). We demonstrate that both these changes are occurring simultaneously in global precipitation, that this behavior cannot be explained by internal variability alone, and that external influences are responsible for the observed precipitation changes. Whereas existing model experiments are not of sufficient length to differentiate between natural and anthropogenic forcing terms at the 95% confidence level, we present evidence that the observed trends result from human activities.

Project description:Wild edible mushrooms have been collected and consumed by human groups for centuries, and today they represent a relevant source of food and income for many rural families worldwide. Preserving these non-timber forest products is of great interest, and there is concern about the damage caused by intensive mushroom harvesting on macromycete communities. The aim of this study was to evaluate variation in diversity and composition of macromycete species between areas regularly used for mushroom harvesting and non-harvested areas in the Mixteca region of Oaxaca, Mexico, as well as to assess the influence of microclimatic and environmental factors on this variation. We selected two harvested and two non-harvested sites within the study area. In each one, we established 10 permanent plots of 10 m × 10 m where we sampled all the observed fruit bodies weekly from June to October 2017. We recorded a total of 856 individuals corresponding to 138 species, and 23 of these were identified as edible. Overall macromycete diversity, edible species diversity and composition were similar in Sites 1 (non-harvested) and 3 (harvested), and in Sites 2 (non-harvested) and 4 (harvested). Variation of diversity and species composition along the studied area was mainly related to microclimatic variables, while most environmental variables and variables related to vegetation structure similarly affected macromycete species in the four sites. Our results indicate that intensive harvesting of wild edible mushrooms is not affecting the diversity and distribution of macromycete species in our study area. Knowledge on the sustainability of mushroom harvesting practices can help improve current regulations regarding the management of these valuable non-timber forest products.
