DNA-Based Assembly of Quantum Dots into Dimers and Helices.

ABSTRACT: Owing to their unique optical properties, colloidal quantum dots (QDs) have attracted much attention as versatile fluorescent markers with broad biological and physical applications. On the other hand, DNA-based assembly has proven to be a powerful bottom-up approach to create designer nanoscale objects and to use these objects for the site-directed arrangement of guest components. To achieve good colloidal stability and accurate positioning of QDs on DNA templates, robust QD surface functionalization is crucial. Here, we present a simple and reliable conjugation method for the direct attachment of DNA molecules to QDs. Phosphorothiolated regions of chimera oligonucleotides are attached and incorporated into a ZnS layer freshly growing in situ on QDs that were rendered water soluble with hydrophilic ligands in a prior step. The reaction can be completed in a 2 mL plastic tube without any special equipment. The utility of these DNA-labeled QDs is demonstrated via prototypical assemblies such as QDs dimers with various spacings and chiral helical architectures.

Construction of Photoelectrochemical DNA Biosensors Based on TiO2@Carbon Dots@Black Phosphorous Quantum Dots.

Project description:In this work, carbon dots (CDs) and black phosphorus quantum dots (BPQDs) were used to decorate titanium dioxide to enhance the photoelectrochemical (PEC) properties of the nanocomposites (TiO2@CDs@BPQDs), and the modified nanocomposites were used to sensitively detect DNA. We used the hydrothermal method and citric acid as a raw material to prepare CDs with good dispersion and strong fluorescence properties. BPQDs with a uniform particle size were prepared from black phosphorus crystals. The nanocomposites were characterized by fluorescence spectroscopy, UV-Vis absorption spectroscopy, Fourier transform infrared spectroscopy (FTIR) and transmission electron microscopy (TEM). The preparation method of the working electrode was explored, the detection conditions were optimized, and the sensitive detection of target DNA was achieved. The results demonstrate that CDs and BPQDs with good optical properties were successfully prepared, and they were successfully combined with TiO2 to improve the PEC performance of TiO2@CDs@BPQDs. The TiO2-based PEC DNA detection method was constructed with a detection limit of 8.39 nM. The constructed detection method has many advantages, including good sensitivity, a wide detection range, and good specificity. This work provides a promising PEC strategy for the detection of other biomolecules.

Evaporation-induced assembly of quantum dots into nanorings.

Project description:Herein, we demonstrate the controlled formation of two-dimensional periodic arrays of ring-shaped nanostructures assembled from CdSe semiconductor quantum dots (QDs). The patterns were fabricated by using an evaporative templating method. This involves the introduction of an aqueous solution containing both quantum dots and polystyrene microspheres onto the surface of a planar hydrophilic glass substrate. The quantum dots became confined to the meniscus of the microspheres during evaporation, which drove ring assembly via capillary forces at the polystyrene sphere/glass substrate interface. The geometric parameters for nanoring formation could be controlled by tuning the size of the microspheres and the concentration of the QDs employed. This allowed hexagonal arrays of nanorings to be formed with thicknesses ranging from single dot necklaces to thick multilayer structures over surface areas of many square millimeters. Moreover, the diameter of the ring structures could be simultaneously controlled. A simple model was employed to explain the forces involved in the formation of nanoparticle nanorings.

Reconfigurable quantum photonic circuits based on quantum dots.

Project description:Quantum photonic integrated circuits, composed of linear-optical elements, offer an efficient way for encoding and processing quantum information on-chip. At their core, these circuits rely on reconfigurable phase shifters, typically constructed from classical components such as thermo- or electro-optical materials, while quantum solid-state emitters such as quantum dots are limited to acting as single-photon sources. Here, we demonstrate the potential of quantum dots as reconfigurable phase shifters. We use numerical models based on established literature parameters to show that circuits utilizing these emitters enable high-fidelity operation and are scalable. Despite the inherent imperfections associated with quantum dots, such as imperfect coupling, dephasing, or spectral diffusion, we show that circuits based on these emitters may be optimized such that these do not significantly impact the unitary infidelity. Specifically, they do not increase the infidelity by more than 0.001 in circuits with up to 10 modes, compared to those affected only by standard nanophotonic losses and routing errors. For example, we achieve fidelities of 0.9998 in quantum-dot-based circuits enacting controlled-phase and - not gates without any redundancies. These findings demonstrate the feasibility of quantum emitter-driven quantum information processing and pave the way for cryogenically-compatible, fast, and low-loss reconfigurable quantum photonic circuits.

Assembly and targeting of liposomal nanoparticles encapsulating quantum dots.

Project description:Quantum dots (QDs) are attracting intense interest as fluorescence labeling agents for biomedical imaging because biocompatible coatings and relatively nontoxic rare earth metal QDs have emerged as possible options. QD photoemissions are bright, of narrow wavelength range, and very stable. We sought to encapsulate QDs within targeted PEGylated liposomes to reduce their propensity for liver uptake and to amplify the already strong QD emission signal. A novel lipid-QD conjugate initialized a process by which lipids in solution coalesced around the QDs. The liposomal structure was confirmed with size measurements, SEM, and IR spectroscopy. PEGylated QD liposomes injected into a xenograft tumor model largely cleared from the body within 24 h. Residual liver labeling was low. Targeted QD liposomes exhibited robust tumor labeling compared with controls. This study highlights the potential of these near IR emitting QD liposomes for preclinical/clinical applications.

Discrete Stacked Dimers of Aromatic Oligoamide Helices.

Project description:Tight binding was observed between the C-terminal cross section of aromatic oligoamide helices in aqueous solution, leading to the formation of discrete head-to-head dimers in slow exchange on the NMR timescale with the corresponding monomers. The nature and structure of the dimers was evidenced by 2D NOESY and DOSY spectroscopy, mass spectrometry and X-ray crystallography. The binding interface involves a large hydrophobic aromatic surface and hydrogen bonding. Dimerization requires that helices have the same handedness and the presence of a C-terminal carboxy function. The protonation state of the carboxy group plays a crucial role, resulting in pH dependence of the association. Dimerization is also influenced by neighboring side chains and can be programmed to selectively produce heteromeric aggregates.

Controlling Superstructure-Property Relationships via Critical Casimir Assembly of Quantum Dots.

Project description:The assembly of colloidal quantum dots (QDs) into dense superstructures holds great promise for the development of novel optoelectronic devices. Several assembly techniques have been explored; however, achieving direct and precise control over the interparticle potential that controls the assembly has proven to be challenging. Here, we exploit the application of critical Casimir forces to drive the growth of QDs into superstructures. We show that the exquisite temperature-dependence of the critical Casimir potential offers new opportunities to control the assembly process and morphology of the resulting QD superstructures. The direct assembly control allows us to elucidate the relation between structural, optical, and conductive properties of the critical Casimir-grown QD superstructures. We find that the choice of the temperature setting the interparticle potential plays a central role in maximizing charge percolation across QD thin-films. These results open up new directions for controlling the assembly of nanostructures and their optoelectronic properties.

Supramolecular Assembly of Edge Functionalized Top-Down Chiral Graphene Quantum Dots.

Project description:The construction of supramolecular assemblies of heterogeneous materials at the nanoscale is an open challenge in science. Herein, new chiral graphene quantum dots (GQDs) prepared by amidation reaction introducing chiral amide groups and pyrene moieties into the periphery of GQDs are described. The analytical and spectroscopic data show an efficient chemical functionalization and the morphological study of the supramolecular ensembles using SEM and AFM microscopies reveals the presence of highly ordered fibers of several micrometers length. Fluorescence studies, using emission spectroscopy and confocal microscopy, reveal that the fibers stem from the π-π stacking of both pyrenes and GQDs, together with the hydrogen bonding interactions of the amide groups. Circular dichroism analysis supports the chiral nature of the supramolecular aggregates.

Antibody Self-Assembly Maximizes Cytoplasmic Immunostaining Accuracy of Compact Quantum Dots.

Project description:Antibody conjugates of quantum dots (QDs) are expected to transform immunofluorescence staining by expanding multiplexed analysis and improving target quantification. Recently, a new generation of small QDs coated with multidentate polymers has improved QD labeling density in diverse biospecimens, but new challenges prevent their routine use. In particular, these QDs exhibit nonspecific binding to fixed cell nuclei and their antibody conjugates have random attachment orientations. This report describes four high-efficiency chemical approaches to conjugate antibodies to compact QDs. Methods include click chemistry and self-assembly through polyhistidine coordination, both with and without adaptor proteins that directionally orient antibodies. Specific and nonspecific labeling are independently analyzed after application of diverse blocking agent classes, and a new assay is developed to quantitatively measure intracellular labeling density based on microtubule stain connectivity. Results show that protein conjugation to the QD surface is required to simultaneously eliminate nonspecific binding and maintain antigen specificity. Of the four conjugation schemes, polyhistidine-based coordination of adaptor proteins with antibody self-assembly yields the highest intracellular staining density and the simplest conjugation procedure. Therefore, antibody and adaptor protein orientation, in addition to blocking optimization, are important determinants of labeling outcomes, insights that can inform translational development of these more compact nanomaterials.
