Project description:During a survey of entomopathogenic nematodes (EPNs) in the eastern Black Sea region of Turkey in 2009-2012, a steinernematid species was recorded and isolated using the Galleria-baiting method. The isolate was identified as Steinernema kraussei based on its morphological and molecular properties. The analysis of the ITS rDNA sequence placed the Turkish population of S. kraussei in the "feltiae-kraussei" group in the clade that contains different isolates of the species. This is the first record of S. kraussei from Turkey. The efficacy of S. kraussei was tested on Agrotis segetum (Lepidoptera: Noctuidea) larvae at different densities (100, 300, and 500 infective juveniles (IJs) g(-1) dry sand ) in laboratory conditions at 25 °C. The highest mortality (98%) was obtained with 500 IJs g(-1) dry sand within 7 d after inoculation. Our results indicate that the new isolate is a highly promising biological control agent against A. segetum, one of the most serious soil pests of agricultural area and fruits worldwide.

Project description:The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), spread rapidly in Africa and Asia recently, causing huge economic losses in crop production. Fall armyworm caterpillars were first detected in South Korea and Japan in June 2019. Here, the migration timing and path for FAW into the countries were estimated by a trajectory simulation approach implementing the insect's flight behavior. The result showed that FAWs found in both South Korea and Japan were estimated to have come from eastern China by crossing the Yellow Sea or the East China Sea in 10-36 h in three series of migrations. In the first series, FAW moths that arrived on Jeju Island during 22-24 May were estimated to be from Zhejiang, Anhui and Fujian Provinces after 1-2 nights' flights. In the second series, it was estimated that FAW moths landed in southern Korea and Kyushu region of Japan simultaneously or successively during 5-9 June, and these moths mostly came from Guangdong and Fujian Provinces. The FAW moths in the third series were estimated to have immigrated from Taiwan Province onto Okinawa Islands during 19-24 June. During these migrations, southwesterly low-level jets extending from eastern China to southern Korea and/or Japan were observed in the northwestern periphery of the western Pacific Subtropical High. These results, for the first time, suggested that the overseas FAW immigrants invading Korea and Japan came from eastern and southern China. This study is helpful for future monitoring, early warning and the source control of this pest in the two countries.

Project description:Fall armyworm is recognized as one of most highly destructive global agricultural pests. In January 2020, it had first invaded Australia, posing a significant risk to its biosecurity, food security, and agricultural productivity. In this study, the migration paths and wind systems for the case of fall armyworm invading Australia were analyzed using a three-dimensional trajectory simulation approach, combined with its flight behavior and NCEP meteorological reanalysis data. The analysis showed that fall armyworm in Torres Strait most likely came from surrounding islands of central Indonesia on two occasions via wind migration. Specifically, fall armyworm moths detected on Saibai and Erub Islands might have arrived from southern Sulawesi Island, Indonesia, between January 15 and 16. The fall armyworm in Bamaga most likely arrived from the islands around Arafura Sea and Sulawesi Island of Indonesia, between January 26 and 27. The high risk period for the invasion of fall armyworm is only likely to have occurred in January-February due to monsoon winds, which were conducive to flight across the Timor Sea towards Australia. This case study is the first to confirm the immigration paths and timing of fall armyworm from Indonesia to Australia via its surrounding islands.

Project description:The alien invasive insect pest Spodoptera frugiperda Smith (Lepidoptera: Noctuidae), commonly referred to as fall armyworm (FAW), is causing significant losses to maize production in Africa since its detection in 2016. As an emergency response, governments in several countries distributed and/or promoted massive use of synthetic insecticides among smallholder farmers to fight FAW. The inappropriate use of synthetic insecticides by non-trained and ill-equipped farmers raises environmental and health concerns. This study aimed to assess the occurrence of native parasitoids of FAW, their parasitism rates, and relative abundance in the central province of Manica, Mozambique. A field collection of FAW egg masses and larvae was conducted from May to August 2019 (dry season of the 2018/2019 cropping season) and in December 2019 and January 2020 (rainy season of 2019/2020 cropping season). A total of 101 egg masses and 1444 larvae of FAW were collected from infested fields. Five larval parasitoids were recorded, but no egg parasitism was observed. Coccygidium luteum Brullé (Hymenoptera: Braconidae) and Drino quadrizonula Thomson (Diptera: Tachinidae) were the primary parasitoids. Maximum parasitism of 23.68% and 8.86% and relative abundance of 100 and 96.3 were recorded for C. luteum and D. quadrizonula, respectively. Total parasitism by different parasitoid species was at 9.49%. Cultural practices favoring the action of these parasitoids should be advocated.

Additionally, coding sequences for the 18 S and 28 S rDNA were amplified from the nematode genome, and phylogenetic analysis revealed that the nematode belongs to Ovomermis sinensis, a known entomoparasitic nematode. Our finding is the first record that S. frugiperda was naturally parasitized by O. sinensis. Project description:The aim of this experiment was to compare the transciptome of the fall armyworm (Spodoptera frugiperda) strain SUS (a laboratory insecticide-susceptible standard) with organophosphate (OP) and pyrethroid (PYR) resistant strains were collected in cornfields located in Minas Gerais and Mato Grosso States, Brazil, in 2008 and maintained in the laboratory under selection with chlorpyrifos (at increasing discriminating doses from 100 up to 400 µg of insecticide/g of insect in a topical bioassay) or lambda-cyhalothrin (from 8.4 up to 27 µg of insecticide/g of insect) respectively The custom microarray used in this study was designed using the Agilent eArray platform (Agilent Technologies). A SurePrint HD (8×15k) expression array was designed using the base composition and the best probe methodologies to design sense orientation 60-mer probes with a 3′ bias. The FAW EST database (SPODOBASE) was used as the reference transcriptome (Negre, Hotelier et al. 2006). These sequences are derived from 8 cDNA libraries as follows: Sf1F: Fat body, Sf1H: Hemocyte, Sf1M: Midgut, Sf1P: Pool of various tissues, Sf2H: Immune Challenged hemocytes, Sf2L: Sf21 Cell lines sequences, Sf2M: Xenobiotic Induced Midgut and Sf9L: Sf9 cell lines sequences. All assembled contigs and singlets were kindly sent by the website maintainers. The BLAST2GO software v.2.3.1 (http://www.blast2go.org) was used to annotate the EST database. 60-mer probes were designed for all 7,552 assembled contigs and 5,519 annotated singlets (BlastX), totaling 13,071 sequences. For contigs encoding detoxification enzymes (P450s, GSTs and CEs) three probes were designed. Additional probe groups for 15 control genes were also included. Project description:A new Anagnorisma species, Anagnorisma chamrani sp. n., is described from Binaloud Mountains of Khorasan-e-Razavi province in north-eastern Iran, and compared with its sister species, Anagnorisma eucratides (Boursin, 1960). The adults, and male and female genitalia of both species are illustrated in 11 figures. The genus Anagnorisma is recorded for the first time for the fauna of Iran.

Project description:The corn earworm, Helicoverpa zea, is a major target pest of the insecticidal Vip3Aa protein used in pyramided transgenic Bt corn and cotton with Cry1 and Cry2 proteins in the U.S. The widespread resistance to Cry1 and Cry2 proteins in H. zea will challenge the long-term efficacy of Vip3Aa technology. Determining the frequency of resistant alleles to Vip3Aa in field populations of H. zea is critically important for resistance management. Here, we provided the first F2 screen study to estimate the resistance allele frequency for Vip3Aa in H. zea populations in Texas, U.S. In 2019, 128 H. zea neonates per isofamily for a total of 114 F2 families were screened with a diagnostic concentration of 3.0 μg/cm2 of Vip3Aa39 protein in diet-overlay bioassays. The F2 screen detected two families carrying a major Vip3Aa resistance allele. The estimated frequency of major resistance alleles against Vip3Aa39 in H. zea in Texas from this study was 0.0065 with a 95% CI of 0.0014-0.0157. A Vip3Aa-resistant strain (RR) derived from the F2 screen showed a high level of resistance to Vip3Aa39 protein, with a resistance ratio of >588.0-fold relative to a susceptible population (SS) based on diet-overlay bioassays. We provide the first documentation of a major resistance allele conferring high levels of Vip3Aa resistance in a field-derived strain of H. zea in the U.S. Data generated from this study contribute to development of management strategies for the sustainable use of the Vip3Aa technology to control H. zea in the U.S.
