Project description:IntroductionBiological soil crusts (BSCs) constitute a substantial portion of primary production in dryland ecosystems. They successionally mature to deliver a series of ecosystem services. Bacteria, as an important community in BSCs, play critical roles in maintaining the structure and functions of BSCs. However, the process by which bacterial diversity and community are altered with BSC development is not fully understood.MethodsIn this study, amplicons sequencing was used to investigate bacterial diversity and community compositions across five developmental stages of BSCs (bare sand, microbial crusts, algae crusts, lichen crusts, and moss crusts) and their relationship with environmental variables in the Gonghe basin sandy land in Qinghai-Tibet Plateau, northwestern China.ResultsThe results showed that Proteobacteria, Actinobacteria, Cyanobacteria, Acidobacteria, Bacteroidetes, and Firmicutes were predominant in different developmental stages of BSCs, accounting for more than 77% of the total relative abundance. The phyla of Acidobacteria and Bacteroidetes were abundant in this region. With BSC development, bacterial diversity significantly increased, and the taxonomic community composition significantly altered. The relative abundance of copiotrophic bacteria, such as Actinobacteria, Acidobacteria, Bacteroidetes, Verrucomicrobia, Planctomycetes, and Gemmatimonadetes significantly increased, whereas the relative abundance of oligotrophic bacteria, such as Proteobacteria and Firmicutes significantly decreased. The relative abundance of Cyanobacteria in the algae crusts was significantly higher than that in the other developmental stages (p < 0.05).ConclusionVariations in bacterial composition suggested that the potential ecological functions of the bacterial community were altered with BSC development. The functions varied from enhancing soil surface stability by promoting soil particle cementation in the early stages to promoting material circulation of the ecosystem by fixing carbon and nitrogen and decomposing litter in the later stages of BSC development. Bacterial community is a sensitive index of water and nutrient alterations during BSC development. SWC, pH value, TC, TOC, TN, NO3 -, TP and soil texture were the primary environmental variables that promoted changes in the bacterial community composition of BSCs.

Project description:Caragana korshinskii, a leguminous shrub, a common specie, is widely planted to prevent soil erosion on the Loess Plateau. The objective of this study was to determine how the plantation ages affected soil, leaf and root nutrients and ecological stoichiometry. The chronosequence ages of C. korshinskii plantations selected for this study were 10, 20 and 30 years. Soil organic carbon (SOC) and soil total nitrogen (STN) of C. korshinskii plantations significantly increased with increase in the chronosequence age. However, soil total phosphorous (STP) was not affected by the chronosequence age. The soil C: N ratio decreased and the soil C: P and N: P ratios increased with increasing plantation age. The leaf and root concentrations of C, N, and P increased and the ratios C: N, C: P, and N: P decreased with age increase. Leaf N: P ratios were >20, indicating that P was the main factor limiting the growth of C. korshinskii. This study also demonstrated that the regeneration of natural grassland (NG) effectively preserved and enhanced soil nutrient contents. Compared with NG, shrub lands (C. korshinskii) had much lower soil nutrient concentrations, especially for long (>20 years) chronosequence age. Thus, the regeneration of natural grassland is an ecologically beneficial practice for the recovery of degraded soils in this area.

Project description:This study represents the first characterization of sand microbiota in migrating barchan sand dunes. Bacterial communities were studied through direct counts and cultivation, as well as 16S rRNA gene and metagenomic sequence analysis to gain an understanding of microbial abundance, diversity, and potential metabolic capabilities. Direct on-grain cell counts gave an average of 5.3 ± 0.4 x 105 cells g-1 of sand. Cultured isolates (N = 64) selected for 16S rRNA gene sequencing belonged to the phyla Actinobacteria (58%), Firmicutes (27%) and Proteobacteria (15%). Deep-sequencing of 16S rRNA gene amplicons from 18 dunes demonstrated a high relative abundance of Proteobacteria, particularly enteric bacteria, and a dune-specific-pattern of bacterial community composition that correlated with dune size. Shotgun metagenome sequences of two representative dunes were analyzed and found to have similar relative bacterial abundance, though the relative abundances of eukaryotic, viral and enterobacterial sequences were greater in sand from the dune closer to a camel-pen. Functional analysis revealed patterns similar to those observed in desert soils; however, the increased relative abundance of genes encoding sporulation and dormancy are consistent with the dune microbiome being well-adapted to the exceptionally hyper-arid Qatari desert.

Project description:Water is a limiting factor for plant growth and vegetation dynamics in alpine sandy land of the Tibetan Plateau, especially with the increasing frequency of extreme precipitation events and drought caused by climate change. Therefore, a relatively stable water source from either deeper soil profiles or ground water is necessary for plant growth. Understanding the water use strategy of dominant species in the alpine sandy land ecosystem is important for vegetative rehabilitation and ecological restoration. The stable isotope methodology of ?D, ?18O, and ?13C was used to determine main water source and long-term water use efficiency of Salix psammophila and S. cheilophila, two dominant shrubs on interdune of alpine sandy land in northeastern Tibetan Plateau. The root systems of two Salix shrubs were investigated to determine their distribution pattern. The results showed that S. psammophila and S. cheilophila absorbed soil water at different soil depths or ground water in different seasons, depending on water availability and water use strategy. Salix psammophila used ground water during the growing season and relied on shallow soil water recharged by rain in summer. Salix cheilophila used ground water in spring and summer, but relied on shallow soil water recharged by rain in spring and deep soil water recharged by ground water in fall. The two shrubs had dimorphic root systems, which is coincident with their water use strategy. Higher biomass of fine roots in S. psammophila and longer fine roots in S. cheilophila facilitated to absorb water in deeper soil layers. The long-term water use efficiency of two Salix shrubs increased during the dry season in spring. The long-term water use efficiency was higher in S. psammophila than in S. cheilophila, as the former species is better adapted to semiarid climate of alpine sandy land.

Project description:The vegetation dynamic (e.g., community productivity) is an important index used to evaluate the ecosystem function of grassland ecosystem. However, the critical factors that affect vegetation biomass are disputed continuously, and most of the debates focus on mean annual precipitation (MAP) or temperature (MAT). This article integrated these two factors, used the aridity index (AI) to describe the dynamics of MAP and MAT, and tested the hypothesis that vegetation traits are influenced primarily by the AI. We sampled 275 plots at 55 sites (five plots at each site, including alpine steppe and meadow) across an alpine grassland of the northern Tibet Plateau, used correlation analysis and redundancy analysis (RDA) to explore which key factors determine the biomass dynamic, and explained the mechanism by which they affect the vegetation biomass in different vegetation types via structural equation modelling (SEM). The results supported our hypothesis, in all of the environmental factors collected, the AI made the greatest contribution to biomass variations in RDA , and the correlation between the AI and biomass was the largest (R = 0.85, p < 0.05). The final SEM also validated our hypothesis that the AI explained 79.3% and 84.4% of the biomass variations in the alpine steppe and the meadow, respectively. Furthermore, we found that the soils with higher carbon to nitrogen ratio and soil total nitrogen had larger biomass, whereas soil organic carbon had a negative effect on biomass in alpine steppe; however, opposite effects of soil factors on biomass were observed in an alpine meadow. The findings demonstrated that the AI was the most critical factor affecting biomass in the alpine grasslands, and different reaction mechanisms of biomass response to the AI existed in the alpine steppe and alpine meadow.

Project description:Vegetation degradation, due to climate change and human activities, changes the biomass, vegetation species composition, and soil nutrient input sources and thus affects soil nutrient cycling and enzyme activities. However, few studies have focused on the responses of soil nutrients and enzymes to vegetation degradation in high-altitude wet meadows. In this study, we examined the effects of vegetation degradation on soil nutrients (soil organic carbon, SOC; total nitrogen, TN; total phosphorus, TP) and enzyme activities (i.e., urease, catalase, amylase) in an alpine meadow in the eastern margin of the Qinghai-Tibet Plateau. Four different levels of degradation were defined in terms of vegetation density and composition: primary wet meadow (CK), lightly degraded (LD), moderately degraded (MD), and heavily degraded (HD). Soil samples were collected at depth intervals of 0-10, 10-20, 20-40, 40-60, 60-80, and 80-100 cm to determine soil nutrient levels and enzyme activities. The results showed that SOC, TN, catalase and amylase significantly decreased with degradation level, while TP and urease increased with degradation level (P < 0.05). Soil nutrient and enzyme activity significantly decreased with soil depth (P < 0.05), and the soil nutrient and enzyme activity exhibited obvious "surface aggregation". The activities of soil urease and catalase were strongest in spring and weakest in winter. The content of TN in spring, summer, and autumn was significantly higher than observed in winter (P < 0.05). The soil TP content increased in winter. Soil amylase activity was significantly higher in summerm than in spring, autumn, and winter (P < 0.05). TP was the main limiting factor for plant growth in the Gahai wet meadow. Values of SOC and TN were positively and significantly correlated with amylase and catalase (P < 0.05), but negatively correlated with urease (P < 0.05). These results suggest the significant role that vegetation degradation and seasonal freeze-thaw cycle play in regulating enzyme activities and nutrient availability in wet meadow soil.

Project description:Global surface temperature is predicted to increase by at least 1.5°C by the end of this century. However, the response of soil microbial communities to global warming is still poorly understood, especially in high-elevation grasslands. We therefore conducted an experiment on three types of alpine grasslands on the Qinghai-Tibet Plateau to study the effect of experimental warming on abundance and composition of soil microbial communities at 0-10 and 10-20 cm depths. Plots were passively warmed for 3 years using open-top chambers and compared to adjacent control plots at ambient temperature. Soil microbial communities were assessed using phospholipid fatty acid (PLFA) analysis. We found that 3 years of experimental warming consistently and significantly increased microbial biomass at the 0-10 cm soil depth of alpine swamp meadow (ASM) and alpine steppe (AS) grasslands, and at both the 0-10 and 10-20 cm soil depths of alpine meadow (AM) grasslands, due primarily to the changes in soil temperature, moisture, and plant coverage. Soil microbial community composition was also significantly affected by warming at the 0-10 cm soil depth of ASM and AM and at the 10-20 cm soil depth of AM. Warming significantly decreased the ratio of fungi to bacteria and thus induced a community shift towards bacteria at the 0-10 cm soil depth of ASM and AM. While the ratio of arbuscular mycorrhizal fungi to saprotrophic fungi (AMF/SF) was significantly decreased by warming at the 0-10 cm soil depth of ASM, it was increased at the 0-10 cm soil depth of AM. These results indicate that warming had a strong influence on soil microbial communities in the studied high-elevation grasslands and that the effect was dependent on grassland type.

Project description:Effects of global changes on biodiversity have been paid more and more attention world widely, and the open top chambers (OTCs) are the most common tools to study the effects of climatic warming on plant diversity. However, it remains unclear how flowers evolve under environmental changes, which could help us to understand the changes of plant diversity in the OTCs. We compared the insect diversity and pollen:ovule (P/O) ratio of eight outcrossing species with different life histories inside and outside the OTCs on the Qinghai-Tibet Plateau, to examine the effects induced by OTCs on the evolution of floral traits. In the OTCs, P/O ratio decreased in annuals, but increased in perennials, indicating an overall trend toward selfing in annuals. We found that the insect diversity differed significantly inside and outside the OTCS, with decreases of dipteran insects and bees. We concluded that changes of P/O ratio in the studied plant species might result from pollination failure, which might be the results of mismatch between flowering time and pollinator activities. We also suggested annuals might be in a more extinction risk than perennials in OTCs, if strong inbreeding depression occurs in these annual outcrossing plants.

Project description:Understanding the interaction between large herbivores and pasture production, especially with respect to the grazing optimization hypothesis, is critical for pasture management and generating theoretical and testable predictions. However, the optimization hypothesis remains contradictory in alpine meadows on the Qinghai-Tibet Plateau (QTP). In this study, we tested the grazing optimization hypothesis using four yak-grazing intensities (no grazing, light grazing, moderate grazing and heavy grazing) in alpine meadow habitats from 2015 to 2017. The results indicated that species diversity did not differ significantly among grazing regimes during the experimental period. However, the aboveground net primary production (ANPP) under moderate grazing consistently significantly exceeded that in control enclosures over 3 years, confirming the grazing optimization hypothesis. Levels of overcompensation varied among grazing intensities and years, and grazing-induced plant compensation may only occur in the short term. The enhanced regrowth of Poaceae and Cyperaceae induced by yak grazing might contribute to the overall level of overcompensation by plant community. Our results strongly support the grazing optimization hypothesis in the context of alpine meadows grazed by yaks, emphasizing the complex interactions between ANPP, herbivores and other ecological factors in alpine meadows on the QTP. These findings provide new insights for the development of an ecological conservation strategy that will help restore this fragile ecosystem and balance the seemingly incompatible requirements of animal husbandry.

Project description:IntroductionIn grassland ecosystems dominated by asexual plants, the maintenance, renewal, and resistance of plant populations to disturbance are more dependent on the belowground bud bank (BBB). However, the response of the BBB to environmental factors in the alpine grassland of the Qinghai-Tibet Plateau (QTP) is still unknown.MethodsTherefore, a transect survey was conducted to measure the size and scale of BBB and 21 factors in the alpine grassland of the QTP. In addition, the critical driving factors of BBB were screened by boost regression tree analysis, and a structural equation model (SEM) was employed to express the path coefficients of the key factors on the BBB size.ResultsThe results showed that BBB size had no significant geographical pattern in the QTP, and the BBB size was mainly accounted for by soil leucine aminopeptidase (LAP, 17.32%), followed by Margalef and Shannon -Wiener indices of plants (12.63% and 9.24%, respectively), and precipitation (9.23%). SEM further indicated significant positive effects of plant diversity (scored at 0.296) and precipitation (scored at 0.180) on BBB size, and a significant negative effect of LAP (scored at 0.280) on BBB size.DiscussionGenerally, the findings allow for better understanding of the regulated mechanisms of BBB size and the importance of the role of bud bank in the restoration of the grassland ecosystem.
