Identification and characterization of cresol degrading Pseudomonas monteilii strain SHY from Soil samples.

ABSTRACT: Cresol is an organic pollutant discharged by pharmaceutical, pesticide, coal and gasification industries causing severe organ failure in humans. Therefore, it is of interest to isolate microbes from contaminated site for degrading cresol. We isolated a strain (CR-13) that survives at 5000ppm cresol with about 80 percent cresol degradation ability. Immobilized cells showed >99 percent degradation at high concentration of cresol. The 16S rRNA sequence (accession number: MF278026) deposited in GenBank was used for phylogenetic tree analysis and the strain was grouped under Pseudomonas monteilii. The isolated cresol degrading strain was subsequently named as Pseudomonas monteilii SHY.

Characterization and application of a newly isolated pyrene-degrading bacterium, Pseudomonas monteilii.

Project description:Pseudomonas monteilii PL5 (PL5) was newly isolated from soil sample and was identified by 16S rDNA sequence analysis. The strain PL5 had a high potential to degrade pyrene (PYR) in both liquid solution and soil and was able to degrade 51.8% of PYR at 25 °C and pH 7.0 condition within 10 days. At 25 °C, the ability of strain PL5 to degrade PYR at different pH values followed the following order pH 6.0 > pH 7.0 > pH 8.0 > pH 9.0. Degradation of total PYR was 56.5 and 51.8% after 10 days at pH 6.0 and 7.0 with PYR half-lives of 8.8 and 9.2 days, respectively. The ability of strain PL5 degraded PYR under different temperatures was 35 > 25 > 15 °C at pH 6.0. Among the tested soils contaminated by PYR, the best degradation of PYR by strain PL5 occurred in paddy soil where the degradation was 57.5% after 10 days, and the half-life of PYR was reduced 19-fold in the presence of strain PL5. This study suggested that P. monteilii PL5 could be used for the bioremediation of the contaminated soil and water through the degradation of PYR.

Here we present a 5.76-Mb assembly of the P. monteilii genome for the first time. It may provide abundant molecular information for the transformation of aromatics.

Isolation and characterization of an estrogen-degrading Pseudomonas putida strain SJTE-1.

Project description:In this report, Pseudomonas putida SJTE1 isolated from an enrichment culture of sludge was confirmed to degrade natural estrogens (17?-estradiol, estrone, estriol), estrogenic chemicals (naphthalene and phenanthrene) and testosterone. The strain completely degraded 1 mg/L 17?-estradiol in 24 h and transformed it into estrone; 90% and 75% of 50 mg/L and 100 mg/L 17?-estradiol were utilized in 7 days, respectively. The transformation efficiency of this strain against natural estrogens was much higher than that against other estrogenic chemicals. Organic carbon sources, lipopolysaccharide and surfactants could enhance the degradation efficiency of strain SJTE-1 against 17?-estradiol. The adsorption of 17?-estradiol onto the biomass was the premise for transmembrane and cellular utilization of this chemical. This work has the potential to bioremediate the environmental estrogens.

Complete Genome Sequences of Pseudomonas monteilii SB3078 and SB3101, Two Benzene-, Toluene-, and Ethylbenzene-Degrading Bacteria Used for Bioaugmentation.

Project description:Pseudomonas monteilii SB3078 and SB3101 are benzene-, toluene-, and ethylbenzene-degrading strains used for bioaugmentation in relation to treatment of wastewater contaminated with petrochemical hydrocarbons. Complete genome sequencing of the bioaugmentation strains confirms that they are very closely related (100.0% average nucleotide identity). Both strains contain extensive integration of phage elements, with the main difference being insertion of additional phage elements in the SB3078 genome.

Characterization of a Pyrethroid-Degrading Pseudomonas fulva Strain P31 and Biochemical Degradation Pathway of D-Phenothrin.

Project description:D-phenothrin is one of the most popular pyrethroid insecticides for its broad spectrum and high insecticidal activity. However, continuous use of D-phenothrin has resulted in serious environmental contamination and raised public concern about its impact on human health. Biodegradation of D-phenothrin has never been investigated and its metabolic behaviors remain unknown. Here, a novel bacterial strain P31 was isolated from active sludge, which completely degraded (100%) D-phenothrin at 50 mg?L-1 in 72 h. Based on the morphology, 16S rRNA gene and Biolog tests, the strain was identified as Pseudomonas fulva. Biodegradation conditions were optimized as 29.5°C and pH 7.3 by utilizing response surface methodology. Strain P31 depicted high tolerance and strong D-phenothrin degradation ability through hydrolysis pathway. Strain P31 degraded D-phenothrin at inhibition constant (Ki) of 482.1673 mg?L-1 and maximum specific degradation constant (qmax) of 0.0455 h-1 whereas critical inhibitor concentration remained as 41.1189 mg?L-1. The 3-Phenoxybenzaldehyde and 1,2-benzenedicarboxylic butyl dacyl ester were identified as the major intermediate metabolites of D-phenothrin degradation pathway through high-performance liquid chromatography and gas chromatography-mass spectrometry. Bioaugmentation of D-phenothrin-contaminated soils with strain P31 dramatically enhanced its degradation, and over 75% of D-phenothrin was removed from soils within 10 days. Moreover, the strain illustrated a remarkable capacity to degrade other synthetic pyrethroids, including permethrin, cyhalothrin, ?-cypermethrin, deltamethrin, fenpropathrin, and bifenthrin, exhibiting great potential in bioremediation of pyrethroid-contaminated environment.

Isolation and characterization of a novel 2-sec-butylphenol-degrading bacterium Pseudomonas sp. strain MS-1.

Project description:A novel bacterium capable of utilizing 2-sec-butylphenol as the sole carbon and energy source, Pseudomonas sp. strain MS-1, was isolated from freshwater sediment. Within 30 h, strain MS-1 completely degraded 1.5 mM 2-sec-butylphenol in basal salt medium, with concomitant cell growth. A pathway for the metabolism of 2-sec-butylphenol by strain MS-1 was proposed on the basis of the identification of 3 internal metabolites-3-sec-butylcatechol, 2-hydroxy-6-oxo-7-methylnona-2,4-dienoic acid, and 2-methylbutyric acid-by gas chromatography-mass spectrometry analysis. Strain MS-1 degraded 2-sec-butylphenol through 3-sec-butylcatechol along a meta-cleavage pathway. Degradation experiments with various alkylphenols showed that the degradability of alkylphenols by strain MS-1 depended strongly on the position (ortho >> meta = para) of the alkyl substitute, and that strain MS-1 could degrade 2-alkylphenols with various sized and branched alkyl chain (o-cresol, 2-ethylphenol, 2-n-propylphenol, 2-isopropylphenol, 2-sec-butylphenol, and 2-tert-butylphenol), as well as a dialkylphenol (namely, 6-tert-butyl-m-cresol).

We present the draft genome sequence, which is 6.4 Mb in size; analysis suggests the chromosomal localization of genes coding for naphthalene utilization. The operons coding for glucose and other aromatic compounds might also be annotated in another study.

Identification and Characterization of HD1, a Novel Ofloxacin-Degrading Bacillus Strain.

Project description:In recent years, an increasing number of lakes and soils around the world have been polluted by antibiotics, seriously threatening the ecological balance and human health. Currently, there is a lack of understanding of the biodegradation mechanism of typical antibiotics by microorganisms. In this study HD1, a novel Bacillus sp. strain called capable of effectively degrading ofloxacin (OFL), a typical antibiotic with a high detection rate in the environment, was isolated from soil contaminated by OFL. The results of single-factor experiments showed that the optimal conditions for OFL degradation included 30°C, pH 7.0, and 10 g L-1 NaCl. After 7 days of incubation under aerobic conditions, the degradation efficiency of OFL (5 mg L-1) was about 66.2%. Five degradation products were detected by LC-MS analysis, and it was deduced that the possible degradation pathways of OFL included the oxidation of the piperazine ring, demethylation, hydroxylation, and methoxy cleavage. Metabolomics analysis indicated that key pathways with the highest difference with HD1 metabolites included the phenylalanine, arginine, and proline metabolism pathways. By regulating energy, amino acid metabolism, and carbohydrate metabolism, HD1 could alleviate OFL stress to degrade better. This study explored the degradation mechanism of OFL by HD1 and provides a theoretical basis and technical support for the remediation of OFL-contaminated environments by functional microorganisms.

Exacerbation of bronchiectasis by Pseudomonas monteilii: a case report.

Project description:BackgroundPseudomonas spp are important opportunistic and nosocomial pathogens. One such species is Pseudomonas monteilii (P. monteilii). It has been described as an environmental contaminant and potential pathogen. We identified this organism as the causative agent of an exacerbation of bronchiectasis and an environmental contaminant in our hospital on two separate occasions.Case presentationP. monteilii was the cause of an exacerbation of bronchiectasis in a 30-year-old HIV negative male. Patient presented with cough with sputum production and exertional dyspnea. The isolate was recovered from a sputum sample in significant counts and definitively identified by Matrix-Assisted Laser Desorption/Ionisation- Time of Flight Mass Spectrometry (MALDI-TOF MS). He was treated with piperacillin-tazobactam and recovered clinically and microbiologically. Another two isolates of the organism were contaminants from the hospital environment. The three isolates were susceptible to all tested antibiotics. Typing by Random amplification of polymorphic DNA (RAPD) found no clonal relationship between them.ConclusionsLess common species of Pseudomonas need to be identified accurately. This organism is identified by commonly used phenotypic systems as P. putida which may have contributed to a lower reported prevalence. P. monteilii is a known environmental contaminant and must also be considered as a potential pathogen, particularly in patients with chronic lung disease.
