A new stonefly species (Plecoptera, Perlidae) from the Interior Highlands USA, with morphological and molecular comparison to other congeneric species.

ABSTRACT: Thirty-one species of Nearctic Perlesta Banks, 1906 (Plecoptera: Perlidae) are recognized. A new species is described from western Arkansas and eastern Oklahoma, USA, Perlestasublobata South & DeWalt, sp. nov., from the adult male, adult female, and egg. Perlestasublobata males are differentiated from other congeners by a combination of a prominent ventral caecum and a distinct dorsal extension of the lateral sclerites of the aedeagus. A preliminary molecular phylogenetic hypothesis is proposed for Perlesta based on 17 congeners and three outgroup taxa using partial mitochondrial cytochrome c oxidase subunit I sequence data. Illustrations, stereomicroscope images, and scanning electron micrographs support the description and comparison to other Perlesta.

Project description:A new Nearctic species of Perlidae (Insecta, Plecoptera), Perlesta ephelidasp. n., is described from the male, female, and egg stages. This species has been previously reported as, or confused with, Perlesta shubuta Stark from several central and eastern U.S. states. Perlesta ephelida is distinctive from Perlesta shubuta and other regional Nearctic congeners mainly according to male genitalic and egg characteristics. Perlesta ephelida is a widely-distributed eastern Nearctic species, whereas Perlesta shubuta appears to be restricted to a narrow latitudinal belt in the Gulf Coast region from Louisiana east conservatively to the Florida panhandle. The egg of Perlesta shubuta is depicted with scanning electron microscopy for the first time.

The mitochondrial genome from the stonefly: Claassenia sp. (Plecoptera: Perlidae).

Project description:Claasseniini is the apparent sister group to all the other groups of Perlidae, and the small tribe Claasseniini has only one genus includes 12 species in the world. Only five mitochondrial genomes in the Perlinae have previously been reported. In this study, we sequenced the mitochondrial genome of Claassenia sp. Wu in genus Claassenia for the first time. The total length of the mitochondrial genome is 15,774 bp, and the DNA base composition is 34.0% A, 27.4% T, 25.6% C and 13.0% G. The whole genome contains 37 genes and one control region, which is similar to other published stoneflies. All PCG started with ATN except CCG for COI gene, TTG for ND1 gene, GTG for ND4 and ND5 genes. The termination codon of 11 PCGs used the normal complete codon TAA or TAG, and the COII and ND5 genes were terminated by a single T. All tRNAs except tRNASer(AGN) fold into a classic clover structure. The phylogenetic tree by Maximum likelihood (ML) and Bayesian (BI) methods supported that Claassenia sp. was the basal taxa of the Perlidae clade. This study provides new data for further mitochondrial genome study on the Perlidae.

Mitochondrial genome of the stonefly Kamimuria wangi (Plecoptera: Perlidae) and phylogenetic position of plecoptera based on mitogenomes.

Project description:This study determined the mitochondrial genome sequence of the stonefly, Kamimuria wangi. In order to investigate the relatedness of stonefly to other members of Neoptera, a phylogenetic analysis was undertaken based on 13 protein-coding genes of mitochondrial genomes in 13 representative insects. The mitochondrial genome of the stonefly is a circular molecule consisting of 16,179 nucleotides and contains the 37 genes typically found in other insects. A 10-bp poly-T stretch was observed in the A+T-rich region of the K. wangi mitochondrial genome. Downstream of the poly-T stretch, two regions were located with potential ability to form stem-loop structures; these were designated stem-loop 1 (positions 15848-15651) and stem-loop 2 (15965-15998). The arrangement of genes and nucleotide composition of the K. wangi mitogenome are similar to those in Pteronarcys princeps, suggesting a conserved genome evolution within the Plecoptera. Phylogenetic analysis using maximum likelihood and Bayesian inference of 13 protein-coding genes supported a novel relationship between the Plecoptera and Ephemeroptera. The results contradict the existence of a monophyletic Plectoptera and Plecoptera as sister taxa to Embiidina, and thus requires further analyses with additional mitogenome sampling at the base of the Neoptera.

The mitochondrial genome of the stonefly Togoperla limbata Pictet (Plecoptera: Perlidae).

Project description:We sequenced the complete mitochondrial genomes of Togoperla limbata which is the unique species distributed in Japan to provide supplementary data for future study in phylogenetic studies of Perlidae. This complete mitogenome of T. limbata is 15,915 bp long, including 37 typical genes and a control region as other stoneflies. Through the data analysis, the A + T content of the whole mitogenome, PCGs, tRNAs, rRNAs, and control region was 63.7%, 61.3%, 68.7%, 66.8%, and 74.8%. Most PCGs used typical start or stop codon except COI, ND5, and ND1 used the exceptional start codon and the COII and ND5 used the single T as the stop codon. Phylogenetic tree was constructed by using Bayesian inference (BI) and maximum-likelihood (ML) analysis based on the sequences of the 13 PCGs and two rRNAs and the result showed that subfamily Perlinae was a monophyletic group and the clade Togoperla and Paragnetina had a closer relationship.

Project description:Two new Neoperla species (Neoperla mesospina, Neoperla latispina) are described from the adult male stage from the Jinhuacha Nature Reserve, Guangxi of China. The new species are compared with similar taxa. Taxonomic remarks are also provided for N. transversprojecta Du & Sivec and N. yao Stark. The latter species is newly recorded for Guangxi.

The complete mitochondrial genome analysis of the stonefly, Flavoperla sp. (Plecoptera: Perlidae).

Project description:Flavoperla is a small genus in Acroneuriinae, which was established by Chu but was treated as a synonym for a long time until Uchida and Harper recognized Flavoperla as a valid genus. This genus includes 15 species in the world now, including six species from China. Here, we sequenced and annotated the mitochondrial genome of Flavoperla sp. under the next-generation sequence technology, the first representative of Genus Flavoperla. The complete mitochondrial genome of Flavoperla sp. is 15,796 bp in length with an A + T content of 68.3% showed a positive AT-skew (0.037) and a negative GC-skew (-0.262). The gene order and organization of the mitochondrial genome is consistent with other stoneflies. The control region had the highest A + T content. Eleven PCGs started with the typical codon (ATN), the remaining PCGs started with TTG (ND1and ND5). All PCGs terminated with TAA/TAG, except COII and ND5 used single T. The phylogenetic tree by maximum likelihood (ML) and Bayesian (BI) methods supported that Flavoperla sp. was the sister group to Niponiella limbatella.

Characterization of the complete mitochondrial genome of a stonefly, Perlesta teaysia (Plecoptera: Perlidae).

Project description:In this study, we sequenced the mitochondrial genome of Perlesta teaysia. The complete mitochondrial genome was 16,023 bp in length, including 37 typical genes and a control region. The overall nucleotide composition was biased toward the A/T nucleotides. All the protein-coding genes (PCGs) started with typical ATN codon as the start codon except for ND1 and ND5, which started with TTG and GTG, respectively. In addition, 11 of 13 PCGs possessed the typical stop codon TAA/TAG, whereas ND4 and ND5 terminated with an incomplete stop codon. Phylogenetic analysis suggested that P. teaysia was a sister group to Calineuria stigmatica.
