Structural and Thermal Studies of New Triphenyltin Complexes.

ABSTRACT: Crystal and molecular structures of new triorganotin complexes have been determined via X-ray diffraction. These complexes include, among others, the second polymorph of Ph3Sn(thiocytosine), the double complex salt Ph3Sn(methimazole)2·Ph3SnCl2, and five-coordinated triphenyltin chloride with methimazole, tetrahydopyrimidine-2-thione, dimethylformamide, and dimethyl sulfoxide. Hirshfeld surface analysis allowed for better visualization and precise pinpointing of the differences between polymorphs as well as easier analysis of intermolecular interactions. All of the new structure of the Ph3Sn(L)Cl type displayed interesting thermal features and therefore were also analyzed via thermogravimetric analysis/simultaneous thermal analysis/differential scanning calorimetry methods. These analyses showed the different ways in which these complexes underwent thermal decomposition. In some cases, to solve the problems that arose, powder X-ray diffraction analyses have also been performed.

Project description:AbstractMany distinct amino acid and aromatic amine-derived transition metal complexes are used as physiologically active compounds. A few Cobalt (II) complexes have been synthesized by reacting cobalt (II) chloride with 1, 8-diaminonapthalene-based tetraamide macrocyclic ligands in an ethanolic media. These synthesized ligands (TAML1-3) and associated Co(II) complexes were fully characterized with various spectroscopic techniques, such as IR, NMR, CHN analysis, EPR, molar conductance, and magnetic susceptibility measurements, TGA, UV-visible spectra, powder X-ray diffraction and DFT analysis. The IR spectra reveal interactions between the core metal atom and ligands through N of 1, 8-diaminonapthalene. The distorted octahedral geometry of synthesized Co(II) macrocyclic complexes were confirmed by ESR, UV-Vis and DFT studies. The synthesized ligands (TAML1-TAML3) and their Co(II) complexes were tested for antimicrobial activity against A. niger, C. albicans, and F. oxysporum in addition to bacteria like S. aureus, B. subtilis, and Gram-negative bacteria like E. coli. The ligand TAML1 and complex [Co(TAML1)Cl2] showed an excellent antibacterial activity. The minimum inhibitory concentration of TAML1 and [Co(TAML1)Cl2] against S. aureus were found to be 7 mm and 10 mm zone of inhibition at 500 ppm, respectively, compared to drug ampicillin (3 mm). Additionally, each molecule exhibited notable antioxidant activity. The biological significance of the synthesized compounds was then evaluated by molecular docking experiments with the active site of the receptor protein such as Sars-Cov-2, C. Albicans, X. campestris and E. coli. Inorganic Phosphate and Arsenate within New Tetranuclear Copper and Zinc Complexes: Syntheses, Crystal Structures, Magnetic, Electrochemical, and Thermal Studies.

Project description:Three, PO4 3-/HPO4 2- and AsO4 3--incorporated, new tetranuclear complexes of copper(II) and zinc(II) ions have been synthesized and fully characterized. In methanol-water, reactions of H3cpdp (H3cpdp = N,N'-Bis[2-carboxybenzomethyl]-N,N'-Bis[2-pyridylmethyl]-1,3-diaminopropan-2-ol) with copper(II) chloride in the presence of either NaOH/Na2HPO4·2H2O or KOH/Na2HAsO4·7H2O lead to the isolation of the tetranuclear complexes Na3[Cu4(cpdp)2(μ4-PO4)](OH)2·14H2O (1) and K2[Cu4(cpdp)2(μ4-AsO4)](OH)·162/3H2O (2), respectively. Similarly, the reaction of H3cpdp with zinc(II) chloride in the presence of NaOH/Na2HPO4·2H2O yields a tetranuclear complex, Na(H3O)2[Zn4(cpdp)2(μ4-HPO4)]Cl3·121/2H2O (3). All complexes are characterized by single-crystal X-ray diffraction and other analytical techniques, such as Fourier transform infrared and UV-vis spectroscopy, thermogravimetric and electrochemical studies. The solid-state molecular framework of each complex contains two monocationic [M2(cpdp)]+ (M = Cu, Zn) units, which are exclusively coordinated to either phosphate/hydrogen phosphate or arsenate groups in a unique mode. All three complexes exhibit a μ4:η1:η1:η1:η1 bridging mode of the PO4 3-/HPO4 2-/AsO4 3- groups, with each bridging among four metal ions. The thermal properties of all three complexes have been investigated by thermogravimetric analysis. Low-temperature magnetic studies of complexes 1 and 2 disclose moderate antiferromagnetic interactions mediated among the copper centers through alkoxide and phosphate/arsenate bridges. Electrochemical studies of complexes 1 and 2 in dimethylformamide using cyclic voltammetry reveal the presence of a fairly assessable one-electron metal-based irreversible reduction and one quasireversible oxidation couple.

Spectral, thermal, antimicrobial studies for silver(I) complexes of pyrazolone derivatives.

Project description:BackgroundSynthesize new complexes of Ag(I) to enhance efficacy or stability and also, pharmacological activities on the operation of pyrazolone's biological properties.ResultsEfficient and high yielding pathways starting from the versatile and readily available 3-methyl-1-phenyl-5-pyrazolone by Knoevenagel condensation of a sequence of 4-arylidene-3-methyl-1-phenyl-5-pyrazolone derivatives (2a-c) have been formed by the reaction of various substituted aromatic aldehydes Used as ligands to synthesize Ag(I) chelates. Synthesized compounds and their complexes have been characterized by elemental analysis, magnetic and spectroscopic methods (IR, 13C, 1HNMR, mass) and thermal analysis. The spectrophotometric determinations suggest distorted octaedral geometry for all complexes. Both ligands and their metal complexes have also been tested for their antibacterial and antifungal efficacy.ConclusionsNewly synthesized compounds have shown potent antimicrobial activity. The results showed that the complex 's high activity was higher than its free ligands, and that Ag(I)-L3 had the highest activity.

Target selection of soluble protein complexes for structural proteomics studies.

Project description:BACKGROUND: Protein expression in E. coli is the most commonly used system to produce protein for structural studies, because it is fast and inexpensive and can produce large quantity of proteins. However, when proteins from other species such as mammalian are produced in this system, problems of protein expression and solubility arise 1. Structural genomics project are currently investigating proteomics pipelines that would produce sufficient quantities of recombinant proteins for structural studies of protein complexes. To investigate how the E. coli protein expression system could be used for this purpose, we purified apoptotic binary protein complexes formed between members of the Caspase Associated Recruitment Domain (CARD) family. RESULTS: A combinatorial approach to the generation of protein complexes was performed between members of the CARD domain protein family that have the ability to form hetero-dimers between each other. In our method, each gene coding for a specific protein partner is cloned in pET-28b (Novagen) and PGEX2T (Amersham) expression vectors. All combinations of protein complexes are then obtained by reconstituting complexes from purified components in native conditions, after denaturation-renaturation or co-expression. Our study applied to 14 soluble CARD domain proteins revealed that co-expression studies perform better than native and denaturation-renaturation methods. In this study, we confirm existing interactions obtained in vivoin mammalian cells and also predict new interactions. CONCLUSION: The simplicity of this screening method could be easily scaled up to identify soluble protein complexes for structural genomic projects. This study reports informative statistics on the solubility of human protein complexes expressed in E.coli belonging to the human CARD protein family.

Preparation of cooperative RNA recognition complexes for crystallographic structural studies.

Project description:It is essential that mRNA-binding proteins recognize specific motifs in target mRNAs to control their processing, localization, and expression. Although mRNAs are typically targets of many different regulatory factors, our understanding of how they work together is limited. In some cases, RNA-binding proteins work cooperatively to regulate an mRNA target. A classic example is Drosophila melanogaster Pumilio (Pum) and Nanos (Nos). Pum is a sequence-specific RNA-binding protein. Nos also binds RNA, but interaction with some targets requires Pum to bind first. We recently determined crystal structures of complexes of Pum and Nos with two different target RNA sequences. A crystal structure in complex with the hunchback mRNA element showed how Pum and Nos together can recognize an extended RNA sequence with Nos binding to an A/U-rich sequence 5' of the Pum sequence element. Nos also enables recognition of elements that contain an A/U-rich 5' sequence, but imperfectly match the Pum sequence element. We determined a crystal structure of Pum and Nos in complex with the Cyclin B mRNA element, which demonstrated how Nos clamps the Pum-RNA complex and enables recognition of the imperfect element. Here, we describe methods for expression and purification of stable Pum-Nos-RNA complexes for crystallization, details of the crystallization and structure determination, and guidance on how to analyze protein-RNA structures and evaluate structure-driven hypotheses. We aim to provide tips and guidance that can be applied to other protein-RNA complexes. With hundreds of mRNA-binding proteins identified, combinatorial control is likely to be common, and much work remains to understand them structurally.

Synthesis and Structural Investigation of New Bio-Relevant Complexes of Lanthanides with 5-Hydroxyflavone: DNA Binding and Protein Interaction Studies.

Project description:In the present work, we attempted to develop new metal coordination complexes of the natural flavonoid 5-hydroxyflavone with Sm(III), Eu(III), Gd(III), Tb(III). The resultant hydroxo complexes have been characterized by a variety of spectroscopic techniques, including fluorescence, FT-IR, UV-Vis, EPR and mass spectral studies. The general chemical formula of the complexes is [Ln(C15H?O?)?(OH)?(H?O)x]·nH?O, where Ln is the lanthanide cation and x = 0 for Sm(III), x = 1 for Eu(III), Gd(III), Tb(III) and n = 0 for Sm(III), Gd(III), Tb(III), n = 1 for Eu(III), respectively. The proposed structures of the complexes were optimized by DFT calculations. Theoretical calculations and experimental determinations sustain the proposed structures of the hydroxo complexes, with two molecules of 5-hydroxyflavone acting as monoanionic bidentate chelate ligands. The interaction of the complexes with calf thymus DNA has been explored by fluorescence titration and UV-Vis absorption binding studies, and revealed that the synthesized complexes interact with DNA with binding constants (Kb) ~ 10?. Human serum albumin (HSA) and transferrin (Tf) binding studies have also been performed by fluorescence titration techniques (fluorescence quenching studies, synchronous fluorescence spectra). The apparent association constants (Ka) and thermodynamic parameters have been calculated from the fluorescence quenching experiment at 299 K, 308 K, and 318 K. The quenching curves indicate that the complexes bind to HSA with smaller affinity than the ligand, but to Tf with higher binding affinities than the ligand.

Triphenyltin recognition by primary structures of effector proteins and the protein network of Bacillus thuringiensis during the triphenyltin degradation process.

Project description:Herein, triphenyltin (TPT) biodegradation efficiency and its transformation pathway have been elucidated. To better understand the molecular mechanism of TPT degradation, the interactions between amino acids, primary structures, and quaternary conformations of effector proteins and TPT were studied. The results verified that TPT recognition and binding depended on amino acid sequences but not on secondary, tertiary or quaternary protein structure. During this process, TPT could change the molecular weight and isoelectric point of effector proteins, induce their methylation or demethylation, and alter their conformation. The effector proteins, alkyl hydroperoxide reductase and acetyl-CoA acetyltransferase, recognizing TPT were crucial to TPT degradation. Electron transfer flavoprotein subunit alpha, phosphoenolpyruvate carboxykinase, aconitate hydratase, branched-chain alpha-keto acid dehydrogenase E1 component, biotin carboxylase and superoxide dismutase were related to energy and carbon metabolism, which was consistent with the results in vivo. The current findings develop a new approach for investigating the interactions between proteins and target compounds.

Diverse Coordinative Zinc Complexes Containing Amido-Pyridinate Ligands: Structural and Catalytic Studies.

Project description:In this work, zinc complexes containing amidopyridinate ligands substituted with different pendant arms have been described. Treatment of ligand precursors with ZnEt2 at a 1:1 ratio in THF yields zinc ethyl complexes (PyN C1Py )2(ZnEt)2 (1) and (PyN C2NMe2 )2(ZnEt)2 (2), respectively. Complexes 1 and 2 show the same geometry as a distorted tetrahedron, but adopt different coordination behaviors supported by the ligands. Complex 1 represents a rare and a non-centrosymmetric mode, which the amido group bridges two zinc centers to form a six-membered ring. However, complex 2 shows a centrosymmetric mode, which the pyridine group links to the zinc centers to form an eight-membered ring. Recrystallization of complex 2 gives an additional complex (PyN C2NMe2 )4Zn3(?3-O) (3). We attempted to prepare zinc benzyl oxide complexes but afforded only a self-assembly cubane complex Zn7Et6(OBn)8 (4). All molecular structures 1-4 are characterized depending on both single-crystal X-ray and spectroscopic data. Furthermore, their catalytic properties toward the ring opening polymerization of ?-caprolactone and L-lactide, using benzyl alcohol as the initiation reagent, are under investigation.
