Optimisation by Design of Experiment of Benzimidazol-2-One Synthesis under Flow Conditions.

ABSTRACT: A novel flow-based approach for the preparation of benzimidazol-2-one (1) scaffold by the 1,1'-carbonyldiimidazole (CDI)-promoted cyclocarbonylation of o-phenylenediamine (2) is reported. Starting from a preliminary batch screening, the model reaction was successfully translated under flow conditions and optimised by means of design of experiment (DoE). The method allowed the efficient preparation of this privileged scaffold and to set up a general protocol for the multigram-scale preparation in high yield, purity, and productivity, and was successfully applied for the multigram flow synthesis of N-(2-chlorobenzyl)-5-cyano-benzimidazol-2-one, which is a key synthon for hit-to-lead explorations in our anti-inflammatory drug discovery program.

Project description:Photocatalyzed reactions of 2-(alkoxycarbonyl)benzenediazonium tetrafluoroborates with various alkenes afforded isochromanones in good yields, according to a mechanism that was investigated. Three-step-in-one synthesis of supercapacitor MWCNT superparamagnetic magnetite composite material under flow.

Project description:Composites of multi-walled carbon nanotubes (MWCNTs) and superparamagnetic magnetite nanoparticles, Fe3O4@MWCNT, were synthesized in DMF in a vortex fluidic device (VFD). This involved in situ generation of the iron oxide nanoparticles by laser ablation of bulk iron metal at 1064 nm using a pulsed laser, over the dynamic thin film in the microfluidic platform. The overall processing is a three-step in one operation: (i) slicing MWCNTs, (ii) generating the superparamagnetic nanoparticles and (iii) decorating them on the surface of the MWCNTs. The Fe3O4@MWCNT composites were characterized by transmission electron microscopy, scanning transmission electron microscope, TG analysis, X-ray diffraction and X-ray photoelectron spectroscopy. They were used as an active electrode for supercapacitor measurements, establishing high gravimetric and areal capacitances of 834 F g-1 and 1317.7 mF cm-2 at a scan rate of 10 mV s-1, respectively, which are higher values than those reported using similar materials. In addition, the designer material has a significantly higher specific energy of 115.84 W h kg-1 at a specific power of 2085 W kg-1, thereby showing promise for the material in next-generation energy storage devices.

Preparation of Aminals under Continuous Flow Conditions.

Project description:A laboratory experiment was developed for the continuous flow preparation of an aminal derived from the condensation of furfural with morpholine. The experiment introduces the students to concepts of green chemistry, heterogeneous catalysis, and continuous flow. A cheap and easy setup allows the heterogeneous catalyst reactor to be built in class. The use of furfural reinforces the importance of green chemistry by using one of the key synthons obtained from biomass.

Efficient Microwave-assisted One-pot Three-component Synthesis of 2,3-Disubstituted Benzofurans under Sonogashira Conditions.

Project description:An efficient one-pot method for the synthesis of 2,3-disubstituted benzo[b]furans from commercially available 2-iodophenols, terminal acetylenes and aryl iodides has been developed utilizing Sonogashira reaction conditions. After an initial Sonogashira coupling of the 2-iodophenol with the terminal alkyne, cyclization involving the aryl iodide provides the 2,3-disubstituted benzo[b]furan in good to excellent yields. The use of microwave irradiation shortens the reaction times and minimizes the side products. This methodology is especially useful for the construction of libraries of highly substituted benzo[b]furans and their analogues.

The Eschenmoser coupling reaction under continuous-flow conditions.

Project description:The Eschenmoser coupling is a useful carbon-carbon bond forming reaction which has been used in various different synthesis strategies. The reaction proceeds smoothly if S-alkylated ternary thioamides or thiolactames are used. In the case of S-alkylated secondary thioamides or thiolactames, the Eschenmoser coupling needs prolonged reaction times and elevated temperatures to deliver valuable yields. We have used a flow chemistry system to promote the Eschenmoser coupling under enhanced reaction conditions in order to convert the demanding precursors such as S-alkylated secondary thioamides and thiolactames in an efficient way. Under pressurized reaction conditions at about 220 °C, the desired Eschenmoser coupling products were obtained within 70 s residence time. The reaction kinetics was investigated and 15 examples of different building block combinations are given.

Formation of surface nanodroplets under controlled flow conditions.

Project description:Nanodroplets on a solid surface (i.e., surface nanodroplets) have practical implications for high-throughput chemical and biological analysis, lubrications, laboratory-on-chip devices, and near-field imaging techniques. Oil nanodroplets can be produced on a solid-liquid interface in a simple step of solvent exchange in which a good solvent of oil is displaced by a poor solvent. In this work, we experimentally and theoretically investigate the formation of nanodroplets by the solvent exchange process under well-controlled flow conditions. We find significant effects from the flow rate and the flow geometry on the droplet size. We develop a theoretical framework to account for these effects. The main idea is that the droplet nuclei are exposed to an oil oversaturation pulse during the exchange process. The analysis shows that the volume of the nanodroplets increases with the Peclet number Pe of the flow as ∝ Pe(3/4), which is in good agreement with our experimental results. In addition, at fixed flow rate and thus fixed Peclet number, larger and less homogeneously distributed droplets formed at less-narrow channels, due to convection effects originating from the density difference between the two solutions of the solvent exchange. The understanding from this work provides valuable guidelines for producing surface nanodroplets with desired sizes by controlling the flow conditions.

Coordinating Chiral Ionic Liquids: Design, Synthesis, and Application in Asymmetric Transfer Hydrogenation under Aqueous Conditions.

Project description:Hydrophilic coordinating chiral ionic liquids with an amino alcohol substructure were developed and efficiently applied to the asymmetric reduction of ketones. Their careful design and adaptability to the desired reaction conditions allow for these chiral ionic liquids to be used as the sole source of chirality in a ruthenium-catalyzed transfer hydrogenation reaction of aromatic ketones. When used in this reaction system, these chiral ionic liquids afforded excellent yields and high enantioselectivities.
