Modelling and estimation of nonlinear quantile regression with clustered data.

ABSTRACT: In regression applications, the presence of nonlinearity and correlation among observations offer computational challenges not only in traditional settings such as least squares regression, but also (and especially) when the objective function is nonsmooth as in the case of quantile regression. Methods are developed for the modelling and estimation of nonlinear conditional quantile functions when data are clustered within two-level nested designs. The proposed estimation algorithm is a blend of a smoothing algorithm for quantile regression and a second order Laplacian approximation for nonlinear mixed models. This optimization approach has the appealing advantage of reducing the original nonsmooth problem to an approximated L 2 problem. While the estimation algorithm is iterative, the objective function to be optimized has a simple analytic form. The proposed methods are assessed through a simulation study and two applications, one in pharmacokinetics and one related to growth curve modelling in agriculture.

Project description:Additive models are flexible regression tools that handle linear as well as non-linear terms. The latter are typically modelled via smoothing splines. Additive mixed models extend additive models to include random terms when the data are sampled according to cluster designs (e.g. longitudinal).These models find applications in the study of phenomena like growth, certain disease mechanisms and energy expenditure in humans, when repeated measurements are available. We propose a novel additive mixed model for quantile regression. Our methods are motivated by an application to physical activity based on a data set with more than half a million accelerometer measurements in children of the UK Millennium Cohort Study. In a simulation study, we assess the proposed methods against existing alternatives.

Estimation and Inference of Quantile Regression for Survival Data Under Biased Sampling.

Project description:Biased sampling occurs frequently in economics, epidemiology, and medical studies either by design or due to data collecting mechanism. Failing to take into account the sampling bias usually leads to incorrect inference. We propose a unified estimation procedure and a computationally fast resampling method to make statistical inference for quantile regression with survival data under general biased sampling schemes, including but not limited to the length-biased sampling, the case-cohort design, and variants thereof. We establish the uniform consistency and weak convergence of the proposed estimator as a process of the quantile level. We also investigate more efficient estimation using the generalized method of moments and derive the asymptotic normality. We further propose a new resampling method for inference, which differs from alternative procedures in that it does not require to repeatedly solve estimating equations. It is proved that the resampling method consistently estimates the asymptotic covariance matrix. The unified framework proposed in this article provides researchers and practitioners a convenient tool for analyzing data collected from various designs. Simulation studies and applications to real datasets are presented for illustration. Supplementary materials for this article are available online.

Semiparametrically efficient estimation in quantile regression of secondary analysis.

Project description:Analysing secondary outcomes is a common practice for case-control studies. Traditional secondary analysis employs either completely parametric models or conditional mean regression models to link the secondary outcome to covariates. In many situations, quantile regression models complement mean-based analyses and provide alternative new insights on the associations of interest. For example, biomedical outcomes are often highly asymmetric, and median regression is more useful in describing the 'central' behaviour than mean regressions. There are also cases where the research interest is to study the high or low quantiles of a population, as they are more likely to be at risk. We approach the secondary quantile regression problem from a semiparametric perspective, allowing the covariate distribution to be completely unspecified. We derive a class of consistent semiparametric estimators and identify the efficient member. The asymptotic properties of the resulting estimators are established. Simulation results and a real data analysis are provided to demonstrate the superior performance of our approach with a comparison with the only existing approach so far in the literature.

Quantile regression for doubly censored data.

Project description:Double censoring often occurs in registry studies when left censoring is present in addition to right censoring. In this work, we propose a new analysis strategy for such doubly censored data by adopting a quantile regression model. We develop computationally simple estimation and inference procedures by appropriately using the embedded martingale structure. Asymptotic properties, including the uniform consistency and weak convergence, are established for the resulting estimators. Moreover, we propose conditional inference to address the special identifiability issues attached to the double censoring setting. We further show that the proposed method can be readily adapted to handle left truncation. Simulation studies demonstrate good finite-sample performance of the new inferential procedures. The practical utility of our method is illustrated by an analysis of the onset of the most commonly investigated respiratory infection, Pseudomonas aeruginosa, in children with cystic fibrosis through the use of the U.S. Cystic Fibrosis Registry.

Modelling of South African Hypertension: Application of Panel Quantile Regression.

Project description:Hypertension is one of the crucial risk factors for morbidity and mortality around the world, and South Africa has a significant unmet need for hypertension care. This study aims to establish the potential risk factors of hypertension amongst adults in South Africa attributable to high systolic and diastolic blood pressure over time by fitting panel quantile regression models. Data obtained from the South African National Income Dynamics Study (NIDS) Household Surveys carried out from 2008 to 2018 (Wave 1 to Wave 5) was employed to develop both the fixed effects and random effects panel quantile regression models. Age, BMI, gender (males), race, exercises, cigarette consumption, and employment status were significantly associated with either one of the BP measures across all the upper quantiles or at the 75th quantile only. Suggesting that these risk factors have contributed to the exacerbation of uncontrolled hypertension prevalence over time in South Africa.

Quantile regression for recurrent gap time data.

Project description:Evaluating covariate effects on gap times between successive recurrent events is of interest in many medical and public health studies. While most existing methods for recurrent gap time analysis focus on modeling the hazard function of gap times, a direct interpretation of the covariate effects on the gap times is not available through these methods. In this article, we consider quantile regression that can provide direct assessment of covariate effects on the quantiles of the gap time distribution. Following the spirit of the weighted risk-set method by Luo and Huang (2011, Statistics in Medicine 30, 301-311), we extend the martingale-based estimating equation method considered by Peng and Huang (2008, Journal of the American Statistical Association 103, 637-649) for univariate survival data to analyze recurrent gap time data. The proposed estimation procedure can be easily implemented in existing software for univariate censored quantile regression. Uniform consistency and weak convergence of the proposed estimators are established. Monte Carlo studies demonstrate the effectiveness of the proposed method. An application to data from the Danish Psychiatric Central Register is presented to illustrate the methods developed in this article.

Quantile association regression on bivariate survival data.

Project description:The association between two event times is of scientific importance in various fields. Due to population heterogeneity, it is desirable to examine the degree to which local association depends on different characteristics of the population. Here we adopt a novel quantile-based local association measure and propose a conditional quantile association regression model to allow covariate effects on local association of two survival times. Estimating equations for the quantile association coefficients are constructed based on the relationship between this quantile association measure and the conditional copula. Asymptotic properties for the resulting estimators are rigorously derived, and induced smoothing is used to obtain the covariance matrix. Through simulations we demonstrate the good practical performance of the proposed inference procedures. An application to age-related macular degeneration (AMD) data reals interesting varying effects of the baseline AMD severity score on the local association between two AMD progression times.

Tensor response quantile regression with neuroimaging data.

Project description:Collecting neuroimaging data in the form of tensors (i.e. multidimensional arrays) has become more common in mental health studies, driven by an increasing interest in studying the associations between neuroimaging phenotypes and clinical disease manifestation. Motivated by a neuroimaging study of post-traumatic stress disorder (PTSD) from the Grady Trauma Project, we study a tensor response quantile regression framework, which enables novel analyses that confer a detailed view of the potentially heterogeneous association between a neuroimaging phenotype and relevant clinical predictors. We adopt a sensible low-rank structure to represent the association of interest, and propose a simple two-step estimation procedure which is easy to implement with existing software. We provide rigorous theoretical justifications for the intuitive two-step procedure. Simulation studies demonstrate good performance of the proposed method with realistic sample sizes in neuroimaging studies. We conduct the proposed tensor response quantile regression analysis of the motivating PTSD study to investigate the association between fMRI resting-state functional connectivity and PTSD symptom severity. Our results uncover non-homogeneous effects of PTSD symptoms on brain functional connectivity, which cannot be captured by existing tensor response methods.

Competing risks regression for clustered data.

Project description:A population average regression model is proposed to assess the marginal effects of covariates on the cumulative incidence function when there is dependence across individuals within a cluster in the competing risks setting. This method extends the Fine-Gray proportional hazards model for the subdistribution to situations, where individuals within a cluster may be correlated due to unobserved shared factors. Estimators of the regression parameters in the marginal model are developed under an independence working assumption where the correlation across individuals within a cluster is completely unspecified. The estimators are consistent and asymptotically normal, and variance estimation may be achieved without specifying the form of the dependence across individuals. A simulation study evidences that the inferential procedures perform well with realistic sample sizes. The practical utility of the methods is illustrated with data from the European Bone Marrow Transplant Registry.

DGQR estimation for interval censored quantile regression with varying-coefficient models.

Project description:This paper propose a direct generalization quantile regression estimation method (DGQR estimation) for quantile regression with varying-coefficient models with interval censored data, which is a direct generalization for complete observed data. The consistency and asymptotic normality properties of the estimators are obtained. The proposed method has the advantage that does not require the censoring vectors to be identically distributed. The effectiveness of the method is verified by some simulation studies and a real data example.
