The PIWI-interacting RNA (piRNA) pathway is a small RNA-based immune system that controls the expression of transposons and maintains genome integrity in animal gonads. In <i>Drosophila</i>, piRNA-guided silencing is achieved, in part, via co-transcriptional repression of transposons by Piwi.
