ABSTRACT: The African montane bamboo Yushania alpina provides both habitat and food for many species in the Albertine Rift region. In Volcanoes National Park (VNP), Rwanda, it is especially important as a key food resource for the Endangered mountain gorilla Gorilla beringei beringei and Endangered golden guenon Cercopithecus mitis kandti. We examined temporal and spatial variation in bamboo shoots regeneration and consumption by primates, monitored between 2013 and 2018 in 82 16-m² plots located along transects in VNP. Our analyses revealed a decline in vegetative regeneration of bamboo in recent years, which is mirrored by a decline in bamboo shoot consumption by primates; but an increase in proportional intake. Local declines in regeneration are potentially due to high intensities of herbivory, decreased amounts of rainfall during growing seasons, and natural processes that form part of the life cycle of bamboo. Moreover, spatial variation in bamboo regeneration can be explained by elevation, as well as by stand-level variation in soil acidity, vegetation density, and the density of dead bamboo culms. We discuss the potential mechanisms underlying observed temporal and spatial variations and outline possible effects of a decline in bamboo regeneration for primates and other aspects of biodiversity in VNP.

Project description:The spatial distribution of species has long sparked interest among ecologists and biogeographers, increasingly so in studies of species responses to climate change. However, field studies on spatial patterns of distribution, useful to inform conservation actions at local scales, are still lacking for many regions, especially the tropics. We studied elevational trends and species-area relationships among anurans in wetland habitats within Volcanoes National Park (VNP) in Rwanda, part of the biodiverse Albertine Rift region. In VNP, wetlands are key sites for anuran reproduction, and anurans are likely threatened by wetland desiccation which has occurred for the last few decades. Between 2012 and 2017, we sampled anuran communities in ten VNP wetlands located along an elevational gradient of c. 600 m (from 2,546 to 3,188 m a.s.l.) and found at least eight species, including at least two Albertine Rift Endemics. We show that species richness, diversity, and abundance likely decline with a decrease in wetland size and with an increase in elevation, though additional sampling (e.g., at night) might be needed to derive definite conclusions. Larger wetlands at lower elevations contained most species and individuals, which indicates the potential threat of wetland size reduction (through desiccation) for anuran conservation. However, we also found that wetlands differed in species composition and that some species (e.g., Sclerophrys kisoloensis) were likely restricted in distribution to only a few of the smaller wetlands-suggesting that the conservation of each individual wetland should be prioritized, regardless of size. We propose that all wetlands in VNP require additional conservation measures, which should be based on knowledge gathered through long-term monitoring of anuran communities and research on drivers of wetland decline. Only such extended research will allow us to understand the response of anurans in VNP to threats such as climate change and wetland desiccation.

Project description:IntroductionIn recent years, Volcanoes National Park has seen a rise in its wildlife population, primarily due to the diligent efforts of the Rwandan government in safeguarding endangered species, notably the mountain gorillas (Gorilla beringei spp. beringei). This population growth has led to a pressing need for more expansive habitats, ensuring these creatures have ample space, sustenance, and shelter for their wellbeing. Consequently, there are planned park expansion activities on the horizon. However, before initiating this expansion, a critical prelude involves identifying potential threats, particularly toxic substances stemming from agricultural activities in the surrounding environment of Volcanoes National Park.MethodsTo address this concern, a comprehensive study was conducted, aimed at pinpointing potential toxic hazards and assessing the awareness of the local population regarding the harm these hazards pose to wildlife species. Data was collected from individuals with no prior knowledge of the study using a pre-tested questionnaire. The questionnaire was divided into three sections: socio-demographic issues, potential toxic hazards assessment, and a section to determine awareness and risk of potential toxic hazards to humans, animals, and the environment. Respondents were selected based on specific criteria, which included being 18 years or older and residing within the National Volcano Park (NVP) area.ResultsThe study's findings revealed four main categories of potential toxic hazards, which include household chemicals, pharmaceutical products, agricultural pesticides, and poisonous plants. These hazards could jeopardize the health and survival of wildlife species if they consume or come into contact with them. Furthermore, the study exposed an inadequacy in the knowledge and skills of the local community in preventing these toxic hazards, which can result in death of wildlife species and ecosystem contamination and degradation.ConclusionStudy results also underscored the significance of education and training in enhancing the awareness of local communities concerning these toxic threats. Therefore, it is imperative to implement immediate measures to mitigate the adverse effects of these toxic hazards on wildlife species, especially in light of the planned park expansion.

Project description:BackgroundMoso bamboo (Phyllostachys edulis) is a typical East Asian bamboo that does not flower for > 60 years and propagates without seed reproduction. Thus, Moso bamboo can be propagated vegetatively, possibly resulting in highly heterozygous genetic inheritance. Recently, a draft genome of Moso bamboo was reported, followed by whole genome single nucleotide polymorphisms (SNP) analysis, which showed that the genome of Moso bamboo in China has regional characteristics. Moso bamboo in Japan is thought to have been introduced from China over the sea in 1736. However, it is unclear where and how Moso bamboo was introduced in Japan from China. Here, based on detailed analysis of heterozygosity in genome diversity, we estimate the spread of genome diversity and its pedigree of Moso bamboo.ResultsWe sequenced the whole genome of Moso bamboo in Japan and compared them with data reported previously from 15 regions of China. Only 4.1 million loci (0.37% of the analyzed genomic region) were identified as polymorphic loci. We next narrowed down the number of polymorphic loci using several filters and extracted more reliable SNPs. Among the 414,952 high-quality SNPs, 319,431 (77%) loci were identified as heterozygous common to all tested samples. The result suggested that all tested samples were clones via vegetative reproduction. Somatic mutations may accumulate in a heterozygous manner within a single clone. We examined common heterozygous loci between samples from Japan and elsewhere, from which we inferred that an individual closely related to the sample from Fujian, China, was introduced to Japan across the sea without seed reproduction. In addition, we collected 16 samples from four nearby bamboo forests in Japan and performed SNP and insertion/deletion analyses using a genotyping by sequencing (GBS) method. The results suggested that a small number of somatic mutations would spread within and between bamboo groves.ConclusionsHigh heterozygosity in the genome-wide diversity of Moso bamboo implies the vegetative propagation of Moso bamboo from China to Japan, the pedigree of Moso bamboo in Japan, and becomes a useful marker to approach the spread of genome diversity in clonal plants.

Project description:Oesophagostomum sp. is a parasitic nematode that frequently infects wild chimpanzees. Although nodular lesions are commonly associated with infection, some wild chimpanzee populations seem to tolerate Oesophagostomum nodular lesions while those at Gombe and other sites suffer from associated morbidity and mortality. From August 2004 to December 2013, we examined demographic (i.e., age, sex) and individual correlates (i.e., fecal consistency, Oesophagostomum egg production) to Oesophagostomum-associated pathology in 14 individually recognized chimpanzees at Gombe Stream National Park, Tanzania. In addition, we characterized Oesophagostomum-associated pathology in 14 individual sympatric primates including baboons, colobus, and cercopithecid monkeys. In five chimpanzees, there was no evidence of any significant underlying disease aside from oesophagostomiasis to explain the thin condition or diarrhea. All five of these chimpanzees had moderate to numerous parasitic nodules. In general, nodules were more numerous in older chimpanzees. Three of four chimpanzees with the highest average Oesophagostomum egg counts in feces collected during the year prior to their death had numerous parasitic nodules at necropsy. In contrast, the four chimpanzees with the lowest egg counts had only moderate numbers of nodules. No association (P = 0.74) was noted between frequency of diarrhea in the year prior to death and the number of nodules noted at necropsy. Nodules were also present in all baboons examined documenting pathology associated with Oesophagostomum infection in wild baboons. In contrast, no lesions were noted in colobus or cercopithecid monkeys, although it is uncertain if they are infected as no fecal studies have been completed in these species to date at Gombe. Sequence of DNA isolated from nodules in chimpanzees matched (99%) Oesophagostomum stephanostomum. Further research is needed to identify the types of Oesophagostomum causing lesions in baboons and to determine if baboons suffer from these infections. Am. J. Project description:BackgroundAfrican Trypanosomiases threaten the life of both humans and animals. Trypanosomes are transmitted by tsetse and other biting flies. In Rwanda, the African Animal Trypanosomiasis (AAT) endemic area is mainly around the tsetse-infested Akagera National Park (NP). The study aimed to identify Trypanosoma species circulating in cattle, their genetic diversity and distribution around the Akagera NP.MethodologyA cross-sectional study was carried out in four districts, where 1,037 cattle blood samples were collected. The presence of trypanosomes was determined by microscopy, immunological rapid test VerY Diag and PCR coupled with High-Resolution Melt (HRM) analysis. A parametric test (ANOVA) was used to compare the mean Packed cell Volume (PCV) and trypanosomes occurrence. The Cohen Kappa test was used to compare the level of agreement between the diagnostic methods.FindingsThe overall prevalence of trypanosome infections was 5.6%, 7.1% and 18.7% by thin smear, Buffy coat technique and PCR/HRM respectively. Microscopy showed a low sensitivity while a low specificity was shown by the rapid test (VerY Diag). Trypanosoma (T.) congolense was found at a prevalence of 10.7%, T. vivax 5.2%, T. brucei brucei 2% and T. evansi 0.7% by PCR/HRM. This is the first report of T.evansi in cattle in Rwanda. The non-pathogenic T. theileri was also detected. Lower trypanosome infections were observed in Ankole x Friesian breeds than indigenous Ankole. No human-infective T. brucei rhodesiense was detected. There was no significant difference between the mean PCV of infected and non-infected animals (p>0.162).ConclusionsOur study sheds light on the species of animal infective trypanosomes around the Akagera NP, including both pathogenic and non-pathogenic trypanosomes. The PCV estimation is not always an indication of trypanosome infection and the mechanical transmission should not be overlooked. The study confirms that the area around the Akagera NP is affected by AAT, and should, therefore, be targeted by the control activities. AAT impact assessment on cattle production and information on the use of trypanocides are needed to help policymakers prioritise target areas and optimize intervention strategies. Ultimately, these studies will allow Rwanda to advance in the Progressive Control Pathway (PCP) to reduce or eliminate the burden of AAT.
