Se Nanoparticles Induce Changes in the Growth, Antioxidant Responses, and Fruit Quality of Tomato Developed under NaCl Stress.

ABSTRACT: Nanotechnology represents an opportunity to improve the use of elements in agriculture. Selenium is an element that is beneficial to plants and essential to the human diet. The size of nanoparticles gives them characteristics that can enhance the benefits that selenium provides to plants. The objective of the present study was to determine the effects of selenium nanoparticles on the growth, antioxidant responses, and fruit quality of tomato developed under NaCl stress. Four doses of selenium nanoparticles (1, 5, 10, and 20 mg L-1) under NaCl stress, only NaCl, and a control were evaluated. The results showed that the impact of salinity on the growth of the tomato crop can be reduced with the application of selenium nanoparticles. However, the amount of both enzymatic and non-enzymatic compounds significantly increased in the leaves and fruits of tomato. The results suggest that the application of selenium nanoparticles generated a positive effect against salinity in the tomato crop; moreover, it had a positive impact on the content of beneficial biocompounds for human health in tomato fruits.

Project description:Identification of tomato varieties able to exhibit higher accumulation of primary and secondary metabolites in their fruits is currently a main objective in tomato breeding. One tool to improve fruit quality is to cultivate the plants under salt stress, although improvement of fruit quality is generally accompanied by productivity losses. However, it is very interesting to implement strategies aiming at enhancing fruit quality of tomato by means of growing plants in moderate salt stress that allows for a sustainable fruit yield. The traditional tomato varieties adapted to the Mediterranean environmental constraints may be very attractive plant materials to achieve this goal, given the wide range of fruit quality traits because of their genetic diversity. Here, agronomic responses and fruit quality traits, including primary and secondary metabolites, were analyzed in fruits of two Mediterranean traditional tomato varieties named "Tomate Pimiento" ("TP") and "Muchamiel Aperado" ("MA") because of the pepper and pear shape of their fruits, using as reference the commercial cultivar "Moneymaker" ("MM"). Plants were grown without salt (control) and with moderate salt stress (50 mM NaCl), which did not affect fruit yield in any variety. "TP" is of great interest because of its high soluble solids content (SSC) in control, which is even higher in salt, whereas "MA" is very attractive because of its high Brix yield index (SSC × fruit yield), used as overall fruit quality measure. Similitude between both traditional varieties were found for primary metabolism, as they significantly increased sucrose contents compared with "MM" in red ripe fruits from plants in control and, especially, salt stress conditions. The most remarkable difference was the high constitutive levels of total amino acids in "TP" fruits, including the three major free amino acids found in tomato fruit, GABA, glutamate, and glutamine, which even increased under salinity. Regarding secondary metabolites, the most interesting change induced by salinity was the increase in α-tocopherol found in red ripe fruits of both "TP" and "MA." These results reveal the interest of traditional varieties as sources of genetic variation in breeding because of their improvement of tomato fruit quality without production losses under moderate salt stress.

Project description:In regions with intensive agricultural production, large amounts of organic waste are produced by livestock animals. Liquid digestate from manure-based biogas production could potentially serve as fertilizer if integrated with closed horticultural irrigation systems. The aim of this experiment was to investigate how fertilizer based on liquid biogas by-products of pig manure digestion can affect the growth and production of tomato plants. Integration of a nitrification bioreactor presumes a significantly lower concentration of nutrient solutions and a higher level of oxygenation than classical mineral cultivation. Therefore, additional controls were included. We compared plant growth and fruit quality traits of tomato plants grown in a hydroponic solution with organic fertilizer with two levels of mineral fertilizer. The tomatoes grown with organic waste-based liquid fertilizer showed reduced growth rates but increased mean fruit size, resulting in no significant change in total yield compared with high-mineral cultivation. The growth rate was similarly reduced in plants cultivated with low-mineral fertilizer. Plants cultivated with organic waste-based fertilizer had high Cl- concentration in xylem sap, leaves, and, ultimately, fruits. The leaves of plants cultivated with organic waste-based fertilizer contained higher concentrations of starch and soluble carbohydrate and low concentrations of phosphorous (P) and sulfur (S). The plants grown with organic waste-based or low-mineral medium showed significantly poorer fruit quality than the plants cultivated with the high-mineral solution. The low-mineral treatment increased xylem sap contribution to fruit weight because of higher root power. The organic waste-based fertilization did not change the root power but increased fruit size. In conclusion, organic waste-based cultivation is a possible solution for sustainable plant production in greenhouses. However, additional adjustment of nutrient supply is required to improve fruit quality.

Project description:Genome Wide Association Studies (GWAS) have been recently used to dissect complex quantitative traits and identify candidate genes affecting phenotype variation of polygenic traits. In order to map loci controlling variation in tomato marketable and nutritional fruit traits, we used a collection of 96 cultivated genotypes, including Italian, Latin American, and other worldwide-spread landraces and varieties. Phenotyping was carried out by measuring ten quality traits and metabolites in red ripe fruits. In parallel, genotyping was carried out by using the Illumina Infinium SolCAP array, which allows data to be collected from 7,720 single nucleotide polymorphism (SNP) markers.The Mixed Linear Model used to detect associations between markers and traits allowed population structure and relatedness to be evidenced within our collection, which have been taken into consideration for association analysis. GWAS identified 20 SNPs that were significantly associated with seven out of ten traits considered. In particular, our analysis revealed two markers associated with phenolic compounds, three with ascorbic acid, β-carotene and trans-lycopene, six with titratable acidity, and only one with pH and fresh weight. Co-localization of a group of associated loci with candidate genes/QTLs previously reported in other studies validated the approach. Moreover, 19 putative genes in linkage disequilibrium with markers were found. These genes might be involved in the biosynthetic pathways of the traits analyzed or might be implied in their transcriptional regulation. Finally, favourable allelic combinations between associated loci were identified that could be pyramided to obtain new improved genotypes.Our results led to the identification of promising candidate loci controlling fruit quality that, in the future, might be transferred into tomato genotypes by Marker Assisted Selection or genetic engineering, and highlighted that intraspecific variability might be still exploited for enhancing tomato fruit quality.
