Dye Clicked Thermoplastic Polyurethane as a Generic Platform toward Chromic-Polymer Applications.

ABSTRACT: Chromic dyes responding against external stimuli are useful in various field of applications especially to colorimetric sensors. However, there have been several limitations in generic application because of its cost, stability and reliability. Here, we introduced highly functionalizable polymeric materials as a supporter covalently modified with controlled amount of chromic dyes. The photochromic organic dye (spiropyran) and highly functional thermoplastic polyurethanes (TPU) have been adopted as a representative example. Conventional polymeric solution processes such as film processing, wet-spinning, electrospinning and ink-writing are readily applicable because dye-TPU maintains its own solubility in various organic solvents. Additionally, since the concentration of dye on TPU are precisely controllable, these dye-TPU solution can be adopted in broad range of specific applications, such as secret coding, smart fabric, and chromic polymeric film layer.

Project description:The fabrication of polymeric nanofibers and its potential versatility instigated to foster smart hybrid nanomaterials for the removal of environmental pollutants. In this pursuit, in this research work, polyacrylonitrile (PAN)-based two-dimensional (2D) nanofibrous mats with polyethyleneimine (PEI)/Fe and quaternary ammonium (QA)/Fe as hybrid fillers were prepared by the electrospinning process for the effective dye removal and bacterial disinfection. The characteristics of the fabricated nanomaterials were extensively explored by several analytical techniques such as field emission-scanning electron microscopy, transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, and Brunauer-Emmett-Teller analysis. Magnetic and thermal properties were investigated by superconducting quantum interference device and thermogravimetric measurements. The kinetic and isothermal models affirmed the adsorption behavior of the PAN-PEI/Fe nanofibers, and further regenerative studies substantiated the sustainability of the mats for the removal of industrial dye effluents. Subsequently, the magnetic-QA-loaded PAN nanofiber mats exhibited bactericidal killing efficacy of 99 and 89.5% in both Staphylococcus aureus and green fluorescence protein expressing Escherichia coli bacterial models evaluated from the conventional quantitative bacterial colony-counting assay. Disk diffusion method and microscopic investigations corroborated the disinfection efficacy with zone of inhibitions of ∼23 and 33 mm, respectively. Interestingly, in vitro cell culture studies conducted in BHK-21 and NIH 3T3 cell lines demonstrated the cytocompatibility, and the in vivo toxicity investigations using the zebrafish models necessitated the real-time application of these nanofibrous mats. Therefore, the comprehensive study of the fabricated PAN-templated functionalized 2D nanofibrous mats affirmed to be competent for the remediation of industrial dye effluents and bacteria in water bodies.

Project description:An optical molecular imaging contrast agent that is tailored toward lymphatic mapping techniques implementing near-infrared (NIR) fluorescence image-guided navigation in the planning and surgical treatment of cancers would significantly aid in enabling the real-time visualization of the potential metastatic tumor-draining lymph node(s) for their needed surgical biopsy and/or removal, thereby ensuring unmissed disease to prevent recurrence and improve patient survival rates. Here, the development of the first NIR fluorescent rosol dye (THQ-Rosol) tailored to overcome the limitations arising from the suboptimal properties of the generic molecular fluorescent dyes commonly used for such applications is described. In developing THQ-Rosol, we prepared a progressive series of torsionally restrictive N-substituted non-NIR fluorescent rosol dyes based on density functional theory (DFT) calculations, wherein we discerned high correlations amongst their calculated energetics, modeled N-C3' torsion angles, and evaluated properties. We leveraged these strong relationships to rationally design THQ-Rosol, wherein DFT calculations inspired an innovative approach and synthetic strategy to afford an uncharged xanthene core-based scaffold/molecular platform with an aptly elevated p Ka value alongside NIR fluorescence emission (ca.700-900 nm). THQ-Rosol exhibited 710 nm NIR fluorescence emission, a 160 nm Stokes shift, robust photostability, and an aptly elevated p Ka value (5.85) for affording pH-insensitivity and optimal contrast upon designed use. We demonstrated the efficacy of THQ-Rosol for lymphatic mapping with in vitro and in vivo studies, wherein it revealed timely tumor drainage and afforded definitive lymph node visualization upon its administration and accumulation. THQ-Rosol serves as a proof-of-concept for the effective tailoring of an uncharged xanthene core-based scaffold/molecular platform toward a specific imaging application using rational design.
