Project description:Paurodontella parapitica n. sp., collected from the rhizosphere of an apple tree in Kermanshah province, western Iran, is described. The new species is characterized by a body length of 505 to 723 µm (females) and 480 to 600 µm (males), lip region continuous by depression; 7 to 8 ?m broad, 3 to 4 µm high, stylet length 7 to 9 µm or 1 to 1.3 times the lip region diameter, short postuterine sac of 4 to 6 ?m long, lateral fields with five to six incisures; outer incisures crenated and inner incisures weakly crenated, excretory pore situated 90 to 100 µm from anterior end; functional males common in the population, with spicules 24 to 26 ?m long. Tail of both sexes similar, almost straight and elongate-conoid. The new species resembles in morphology and morphometrics to four known species of the genus, namely P. apitica, P. minuta, P. myceliophaga, and P. sohailai. The results of phylogenetic analyses based on sequences of D2/D3 expansion region of 28S rRNA gene revealed this genus is polyphyletic in four different clades in Tylenchid.

Project description:Recently, much attention has been focused on a group of rhabditid nematodes called Phasmarhabditis, a junior synonym of Pellioditis, as a promising source of biocontrol agents for invasive slugs. Pellioditis pelhamensis n. sp. was first isolated from earthworms near Pelham Bay Park in Bronx, New York, USA, in 1990 and has been found to be pathogenic to slugs as well as some earthworms. It has also been used in several comparative developmental studies. Here, we provide a description of this species, as well as a redescription of a similar earthworm-associated nematode, Pellioditis pellio Schneider, 1866, re-isolated from the type locality. Although P. pelhamensis n. sp. and P. pellio are morphologically similar, they are reproductively isolated. Molecular phylogenetic analysis places both species in a clade that includes all species previously described as Phasmarhabditis which are associated with gastropods. Phasmarhabditis Andrássy, 1976 is therefore a junior synonym of Pellioditis Dougherty, 1953. Also, Pellioditis bohemica Nermut', Půža, Mekete & Mráček, 2017, described to be a facultative parasite of slugs, is found to be a junior synonym of Pellioditis pellio (Schneider, 1866), adding to evidence that P. pellio is associated with both slugs and earthworms. The earthworm-associated species P. pelhamensis n. sp. and P. pellio represent different subclades within Pellioditis, suggesting that Pellioditis species in general have a broader host range than just slugs. Because of this, caution is warranted in using these species as biological control agents until more is understood about their ecology.

Project description:Aphelenchoides macrospica n. sp. is described and illustrated from the West Azerbayjan province, northwestern Iran. The new species is characterized by its body length of 807 to 963 μm (males) and 792 to 1,094 μm (females), offset cephalic region, lateral fields with four incisures, long stylet with 15 to 16 μm length, and excretory pore situated opposite or behind the nerve ring. Pharyngeal glands overlapping intestine dorsally and extending for 90 to 121 μm, tail terminus mucronate in both sexes. Vagina directed anteriad, and spicules are relatively large (27-32 μm) with well-developed broadly rounded apex and condylus. The new species comes close to seven known species of the genus namely A. arcticus, A. blastophthorus, A. haguei, A. huntensis, A. lucknowensis, A. parasaprophilus, and A. xui, but it differs from them by the body size, stylet length, size of spicules, and length of postvulval uterine sac. The results of phylogenetic analyses based on sequences of D2-D3 expansion region of 28S and 18S rDNA, confirmed its status as a new species.

Project description:Hemicycliophora ahvasiensis n. sp., recovered from the rhizospheric soil of date palm in Khuzestan province, southwest Iran, is described and illustrated based upon morphological, morphometric and molecular data. The new species is characterized by its sheath, closely fitting most of the body, cuticle with or without numerous irregular lines, sometimes appearing as blocks in distal body region. Lateral field without discrete longitudinal lines, but often with continuous broken striae or anastomoses. Continuous lip region with single annulus, slightly elevated labial disc, stylet with posteriorly sloping knobs. Vulva with or without slightly modified lips, spermatheca with sperm and tail conoid, symmetrically narrowing at distal region to form a narrow conical region. Morphologically, the new species looks similar to H. indica, H. labiata, H. siddiqii, H. tenuistriata and H. typica. The latter species appears more similar to the new species under light microscopy, but could be separated using the scanning electron microscopy and molecular data. The new species was also compared with H. epicharoides and H. dulli, two species with close phylogenetic affinities to it. The phylogenetic relationships of the new species were reconstructed and discussed using partial sequences of the D2-D3 expansion segments of large subunit, and internal transcribed spacer regions (LSU D2-D3 and ITS rDNA). Hemicycliophora conida, the second studied species, was recovered from north Iran and characterized by morphological and molecular data.

Project description:During a survey in Golestan province, north Iran, two populations belonging to the family Tylenchidae were recovered in association with Quercus sp., and a rotten wood sample of an unidentified forest tree. The first recovered species was mainly characterized by having a disc-like differentiation in the frontal end under the light microscope (LM), proposing it as a tentative member of the genus Discotylenchus. Detailed morphological studies using scanning electron microscope (SEM), however, did not reveal a true disc, but showing the smooth cephalic region, and a narrow annulus behind the cephalic plate. Based upon the cephalic region structure, and by lacking a true disc, the species was identified as a member of the genus Filenchus. This population was further characterized by 555 to 618 μm long females, lateral fields with four incisures, 9 to 10 μm long stylet, spermatheca large, including spheroid sperm, post-vulval uterine sac (PUS) 8 to 12 µm long and gradually tapering to an elongate conoid tail with pointed tip. It was compared with relevant species of Filenchus having four incisures in the lateral fields and similar general morphology. By having a disc-like differentiation in the frontal end under the LM, it was further compared with three similar known species of Discotylenchus. The morphological comparisons with species under two aforementioned genera showed the recovered population belongs to an unknown species, described herein as Filenchus pseudodiscus n. sp. The molecular phylogenetic relationships of the new species using partial small and large subunit ribosomal RNA gene (SSU and LSU D2-D3 rDNA) sequences were reconstructed and discussed. Malenchus gilanensis, the second recovered and studied species was originally established based upon traditional criteria. An updated LSU phylogeny of the genus Malenchus by including M. gilanensis was also presented and its results were discussed.

Project description:Aphelenchoides fuchsi sp. n. is described and illustrated from bark and wood samples of a weakened Mondell pine in Kermanshah Province, western Iran. The new species has body length of 332 to 400 µm (females) and 365 to 395 µm (males). Lip region set off from body contour. The cuticle is weakly annulated, and there are four lines in the lateral field. The stylet is 8 to 10 ?m long and has small basal swellings. The excretory pore is located ca one body diam. posterior to metacorpus valve or 51 to 62 ?m from the head. The postuterine sac well developed (60-90 µm). Spicules are relatively short (15-16 ?m in dorsal limb) with apex and rostrum rounded, well developed, and the end of the dorsal limb clearly curved ventrad like a hook. The male tail has usual three pairs of caudal papillae (2+2+2) and a well-developed mucro. The female tail is conical, terminating in a complicated step-like projection, usually with many tiny nodular protuberances. The new species belongs to the Group 2 sensu Shahina, category of Aphelenchoides species. Phylogenetic analysis based on small subunit (SSU) and partial large subunit (LSU) sequences of rRNA supported the morphological results.

Project description:Two new species, Neotobrilus nicsmolae n. sp. and Chronogaster carolinensis n. sp. are described from a small, acidic, temperate, natural lake in North Carolina. N. nicsmolae n. sp. comes close to three members of the genus reported from North America, N. filipjevi, N. longus, and N. hopei. However, N. nicsmolae is unique with in the genus in having a combination of characters: size smaller than 1,700 μm, shorter outer labial and cephalic setae, tail shorter than 250 μm, last ventromedian supplement close (about 5 μm) to cloacal opening, spicule length of 61 to 85 μm, flagelloid sperm, and possession of subterminal setae. Assessment of relationships among clades within the Triplonchida using DNA sequences of the D2D3 expansion segment of the LSU rDNA showed that the family Trichodoridae and the genus Tripyla were recovered as monophyletic. The genus Tobrilus was recovered as monophyletic in the neighbor-joining and maximum likelihood trees, but that was not so in the maximum-parsimony tree. The separation among genera of the Trichodoridae, i.e., Trichodorus and Paratrichodorus, was not clear-cut in all phylograms. Chronogaster carolinensis n. sp. in having one ventral mucro with no spine and vacuolated lateral glandular bodies comes close to C. typica and C. ethiopica but differs from all hitherto known species in a combination of characteristics: in having long cephalic setae, long stoma, crystalloid bodies, vacuolated lateral glandular bodies, and a tail terminus with blunt ventral mucro, and its lack of lateral line.
