Two Species of Fireworms (Annelida: Amphinomidae: Chloeia) from Hong Kong.

ABSTRACT: A new fireworm species and a poorly described species of the genus Chloeia are described on the basis of specimens collected from Hong Kong. The new species, C. bimaculata sp. nov., is characterized by having two distinct mid-dorsal dark spots one behind the other on each segment. It can be distinguished from its sympatric species C. parva, which is characterized by having a mid-dorsal "Y"-shaped dark pigment on each segment. Chloeia bimaculata sp. nov. diverges from C. parva by 16.8% for the COI gene, 4.6% for the 16S rRNA gene and 0.6% for the 28S rRNA gene. Other Chloeia species with the corresponding gene sequences available in the GenBank show greater divergences.

Project description:Zhi Wang, Yanjie Zhang, and Jian-Wen Qiu (2018) Marphysa hongkongensa n. sp. (Annelida: Eunicidae) is described based on samples from the intertidal zone of Tolo Harbour, Hong Kong. This new species belongs to the Marphysa sanguinea species complex. It is characterized by a subacicular hook from chaetiger 26-58 to the posterior end, branchiae with up to 5-10 filaments from chaetiger 14-35 to the posterior end, and four types of pectinate chaetae. Molecular analyses indicated that the cytochrome oxidase c subunit I gene and 16S RNA gene of Marphysa hongkongensa diverged from the corresponding sequence of the closest related species of Marphysa in GenBank by 19.5% and 12.1%, respectively. An identification key is provided for species in the Marphysa sanguinea complex along the Chinese coast.

A new species of Linopherus (Annelida, Amphinomidae) from Beibu Gulf, South China Sea.

Project description:Linopherus beibuwanensissp. n. is described based on six specimens deposited in the Marine Biological Museum of the Chinese Academy of Sciences, Qingdao, collected from the Beibu Gulf (Tokin Gulf), South China Sea. The new species differs from all other Linopherus species by the shape of prostomium and caruncle. The posterior margin of prostomium is bilobed; the caruncle arises medially and confluently from the posterior margin of the prostomium, joining together with prostomium. A key to distinguish the known species of the genus is provided.

A new genus and species of bristle worm from Beibu Gulf, South China Sea (Annelida, Polychaeta, Amphinomidae).

Project description:Alleurythoe, a new genus with type species Alleurythoe tenuichaetasp. n., is described and illustrated based on material from Beibu Gulf, northwestern South China Sea. The new genus is distinguished from all genera within Amphinomidae by a combination of characters: caruncle trilobed, conspicuous, attached to and confluent with the posterior prostomial lobe, which is free from the body wall and has 6-7 folds on each of the lateral lobes; both noto- and neuropodial aciculae are spinose, extending beyond the chaetal lobe. Alleurythoe tenuichaetasp. n. is characterized by having branchiae present from chaetiger 4 and a bifurcate neurochaetae capillary. A key distinguishing the genera of Amphinominae is provided.

Transcriptome sequencing and annotation of the polychaete Hermodice carunculata (Annelida, Amphinomidae).

Project description:BackgroundThe amphinomid polychaete Hermodice carunculata is a cosmopolitan and ecologically important omnivore in coral reef ecosystems, preying on a diverse suite of reef organisms and potentially acting as a vector for coral disease. While amphinomids are a key group for determining the root of the Annelida, their phylogenetic position has been difficult to resolve, and their publically available genomic data was scarce.ResultsWe performed deep transcriptome sequencing (Illumina HiSeq) and profiling on Hermodice carunculata collected in the Western Atlantic Ocean. We focused this study on 58,454 predicted Open Reading Frames (ORFs) of genes longer than 200 amino acids for our homology search, and Gene Ontology (GO) terms and InterPro IDs were assigned to 32,500 of these ORFs. We used this de novo assembled transcriptome to recover major signaling pathways and housekeeping genes. We also identify a suite of H. carunculata genes related to reproduction and immune response.ConclusionsWe provide a comprehensive catalogue of annotated genes for Hermodice carunculata and expand the knowledge of reproduction and immune response genes in annelids, in general. Overall, this study vastly expands the available genomic data for H. carunculata, of which previously consisted of only 279 nucleotide sequences in NCBI. This underscores the utility of Illumina sequencing for de novo transcriptome assembly in non-model organisms as a cost-effective and efficient tool for gene discovery and downstream applications, such as phylogenetic analysis and gene expression profiling.

Are Fireworms Venomous? Evidence for the Convergent Evolution of Toxin Homologs in Three Species of Fireworms (Annelida, Amphinomidae).

Project description:Amphinomids, more commonly known as fireworms, are a basal lineage of marine annelids characterized by the presence of defensive dorsal calcareous chaetae, which break off upon contact. It has long been hypothesized that amphinomids are venomous and use the chaetae to inject a toxic substance. However, studies investigating fireworm venom from a morphological or molecular perspective are scarce and no venom gland has been identified to date, nor any toxin characterized at the molecular level. To investigate this question, we analyzed the transcriptomes of three species of fireworms-Eurythoe complanata, Hermodice carunculata, and Paramphinome jeffreysii-following a venomics approach to identify putative venom compounds. Our venomics pipeline involved de novo transcriptome assembly, open reading frame, and signal sequence prediction, followed by three different homology search strategies: BLAST, HMMER sequence, and HMMER domain. Following this pipeline, we identified 34 clusters of orthologous genes, representing 13 known toxin classes that have been repeatedly recruited into animal venoms. Specifically, the three species share a similar toxin profile with C-type lectins, peptidases, metalloproteinases, spider toxins, and CAP proteins found among the most highly expressed toxin homologs. Despite their great diversity, the putative toxins identified are predominantly involved in three major biological processes: hemostasis, inflammatory response, and allergic reactions, all of which are commonly disrupted after fireworm stings. Although the putative fireworm toxins identified here need to be further validated, our results strongly suggest that fireworms are venomous animals that use a complex mixture of toxins for defense against predators.

A new species of the rare, deep-sea polychaete genus Benthoscolex from the Sea of Kumano, Japan (Annelida, Amphinomidae).

Project description:A new species of amphinomid polychaete, Benthoscolex seisuiaesp. n., is described from the Sea of Kumano, Japan, from depths of 487-596 m. The species is distinguishable from its congeners by the following features: i) palps 1.8 times as long as lateral antennae; ii) branchiae do not reach to the tip of the notochaetae. This is the first record of Benthoscolex from Japan. A partial mitochondrial cytochrome c oxidase subunit I gene sequence from the holotype of B. seisuiaesp. n. is provided for reliable species identification in the future.

A novel Dirofilaria species causing human and canine infections in Hong Kong.

Project description:Dirofilariasis is globally the commonest manifestation of zoonotic filariasis. We report the detection of a novel canine species causing human and canine dirofilariasis in Hong Kong. Three human cases occurring over 10 months were identified, one presenting with cervical lymphadenopathy, one with an abdominal subcutaneous mass, and one with a subconjunctival nodule. Transected worms recovered from the resected abdominal subcutaneous mass were morphologically compatible with Dirofilaria. The cox1 gene sequences of the three human isolates were identical; however, they were only 96.2% and 89.3% identical to the cox1 gene of Dirofilaria repens and Dirofilaria immitis, respectively. Sequencing of the 18S-ITS1-5.8S gene cluster was successful in the intact worm, and the nucleotide sequences were 94.0% and 94.9% identical to those of D. repens and D. immitis, respectively. Screening of the blood samples from 200 dogs and 100 cats showed the presence of the novel Dirofilaria species in 3% (6/200) of the dogs' but none of the cats' blood samples. Nucleotide sequences of the cox1 gene and 18S-ITS1-5.8S gene clusters of the dogs' samples were identical to those in the human samples. The sera of canines infected by this novel Dirofilaria species were negative when tested with the SNAP 4Dx D. immitis detection kit, except in the case of dogs with a mixed infection with D. immitis as detected by PCR. The results from this study suggest that this novel Dirofilaria species is a cause of filarial infection in humans and dogs in Hong Kong. We propose to name this Dirofilaria species "Candidatus Dirofilaria hongkongensis."
