Project description:Targeted memory reactivation (TMR) is a methodology employed to manipulate memory processing during sleep. TMR studies have great potential to advance understanding of sleep-based memory consolidation and corresponding neural mechanisms. Research making use of TMR has developed rapidly, with over 70 articles published in the last decade, yet no quantitative analysis exists to evaluate the overall effects. Here we present the first meta-analysis of sleep TMR, compiled from 91 experiments with 212 effect sizes (N = 2,004). Based on multilevel modeling, overall sleep TMR was highly effective (Hedges' g = 0.29, 95% CI [0.21, 0.38]), with a significant effect for two stages of non-rapid-eye-movement (NREM) sleep (Stage NREM 2: Hedges' g = 0.32, 95% CI [0.04, 0.60]; and slow-wave sleep: Hedges' g = 0.27, 95% CI [0.20, 0.35]). In contrast, TMR was not effective during REM sleep nor during wakefulness in the present analyses. Several analysis strategies were used to address the potential relevance of publication bias. Additional analyses showed that TMR improved memory across multiple domains, including declarative memory and skill acquisition. Given that TMR can reinforce many types of memory, it could be useful for various educational and clinical applications. Overall, the present meta-analysis provides substantial support for the notion that TMR can influence memory storage during NREM sleep, and that this method can be useful for understanding neurocognitive mechanisms of memory consolidation. Project description:BackgroundThe ability to encode and consolidate motor memories is essential for persons with Parkinson's disease (PD), who usually experience a progressive loss of motor function. Deficits in memory encoding, usually expressed as poorer rates of skill improvement during motor practice, have been reported in these patients. Whether motor memory consolidation (i.e., motor skill retention) is also impaired is unknown.ObjectiveTo determine whether motor memory consolidation is impaired in PD compared to neurologically intact individuals.MethodsWe conducted a pre-registered systematic review (PROSPERO: CRD42020222433) following PRISMA guidelines that included 46 studies.ResultsMeta-analyses revealed that persons with PD have deficits in retaining motor skills (SMD = -0.17; 95% CI = -0.32, -0.02; p = 0.0225). However, these deficits are task-specific, affecting sensory motor (SMD = -0.31; 95% CI -0.47, -0.15; p = 0.0002) and visuomotor adaptation (SMD = -1.55; 95% CI = -2.32, -0.79; p = 0.0001) tasks, but not sequential fine motor (SMD = 0.17; 95% CI = -0.05, 0.39; p = 0.1292) and gross motor tasks (SMD = 0.04; 95% CI = -0.25, 0.33; p = 0.7771). Importantly, deficits became non-significant when augmented feedback during practice was provided, and additional motor practice sessions reduced deficits in sensory motor tasks. Meta-regression analyses confirmed that deficits were independent of performance during encoding, as well as disease duration and severity.ConclusionOur results align with the neurodegenerative models of PD progression and motor learning frameworks and emphasize the importance of developing targeted interventions to enhance motor memory consolidation in PD.

Project description:Autobiographical memory retrieval is associated with activity of a distributed network that is similar to the default-mode network (DMN) identified via activity correlations measured during rest. We tested whether activity correlations could be used to identify the autobiographical network during extended bouts of retrieval. Global-correlativity analysis identified regions with activity correlation differences between autobiographical-retrieval and resting states. Increased correlations were identified for retrieval versus resting states within a distributed network that included regions prototypical for autobiographical memory. This network segregated into two subnetworks comprised of regions related to memory versus cognitive control, suggesting greater functional segregation during autobiographical retrieval than rest. DMN regions were important drivers of these effects, with increased correlations between DMN and non-DMN regions and segregation of the DMN into distinct subnetworks during retrieval. Thus, the autobiographical network can be robustly identified via activity correlations and retrieval is associated with network functional organization distinct from rest.

Project description:The present study concerns a preregistered replication of the study conducted by Johnson et al. (Johnson et al. 2013 Psychol. Sci. 24, 1104-1112 (doi:10.1177/0956797612466414)), in which they showed an inhibition-of-return-like effect in working memory. Inhibition of return is a well-known phenomenon observed in the field of perception and refers to the observation that it takes longer to look back at a location which has recently been explored than to look at an unexplored location. Working memory is a central concept in the field of cognitive psychology and refers to the capacity to process and maintain information simultaneously over short periods of time. Johnson's study applied the inhibition of return paradigm to the concept of working memory. Their results showed that it is harder to access a working memory representation that had just been thought of, i.e. refreshed, in comparison to an unrefreshed working memory representation. Contrary to this study of Johnson et al., who observed refreshing to result in inhibitory processes, most studies on refreshing have described its effect as increasing/prolonging the level of activation of the memory representations. In an attempt to integrate these opposite patterns produced by 'refreshing', we started by replicating one of the studies on the inhibition of return in working memory reported by Johnson et al.

Project description:Chronic noncancer pain (CNCP) is a significant health burden among adults. Standard behavioral therapies typically focus on targeting negative affect (NA) and yield only modest treatment effects. The aims of this study were to systematically review and investigate the association between positive affect (PA) and pain severity among adults with CNCP. Databases that were searched included MEDLINE (PubMed), PsycINFO, CINAHL, ProQuest Dissertations and Theses, OLASTER, Open Grey, and PsyArXiv (inception to July 23, 2019). We analyzed studies that: (1) used observational, experimental, or intervention study designs; (2) enrolled individuals with CNCP (pain ≥ 12 weeks); and (3) reported full quantitative results on outcomes. Two researchers independently screened articles, extracted data, and assessed the risk of bias. The main meta-analysis was followed by subgroup analyses. All analyses were performed using random-effects models. Formal tests for heterogeneity (Q-statistic; I) and publication bias (p-curve and p-uniform*) were performed. We meta-analyzed 29 studies with 3521 participants. Results demonstrated that PA inversely impacts pain severity in people with CNCP (r = -0.23). Subgroup analyses showed a significant effect for gender and marginally significant effects for age in studies that adjusted for NA. On average, effect sizes for observational studies were larger in studies with a higher proportion of female respondents and in studies that did not adjust for NA. Finally, larger effect sizes were found in intervention studies with older compared with younger samples.

Project description:We present a fully preregistered, high-powered conceptual replication of Experiment 1 by Smith, Tracy, and Murray (1993). They observed a cognitive deficit in people with elevated depressive symptoms in a task requiring flexible analytic processing and deliberate hypothesis testing, but no deficit in a task assumed to require more automatic, holistic processing. Specifically, they found that individuals with depressive symptoms showed impaired performance on a criterial-attribute classification task, requiring flexible analysis of the attributes and deliberate hypothesis testing, but not on a family-resemblance classification task, assumed to rely on holistic processing. While deficits in tasks requiring flexible hypothesis testing are commonly observed in people diagnosed with a major depressive disorder, these deficits are much less commonly observed in people with merely elevated depressive symptoms, and therefore Smith et al.'s (1993) finding deserves further scrutiny. We observed no deficit in performance on the criterial-attribute task in people with above average depressive symptoms. Rather, we found a similar difference in performance on the criterial-attribute versus family-resemblance task between people with high and low depressive symptoms. The absence of a deficit in people with elevated depressive symptoms is consistent with previous findings focusing on different tasks.

Project description:BackgroundTill et al. (2016) reported that in an Austrian sample approximately one in ten respondents incorrectly believed that Austria still practices, or recently practiced, the death penalty, and that there is a positive association between the amount of weekly television viewing and this gross misperception of the Austrian justice system.MethodsAn endorsed, prereviewed, preregistered close (N = 597) served to test the veracity of these reported effects. This was coupled with the conceptual extension part, which (a) investigated the potential influence of watching American crime series, (b) accounted for further possible confounds, and (c) tested the generalizability of the effect of television viewing to online streaming.ResultsOnline survey data (N = 597) replicated the one-in-ten prevalence of incorrect answers with the 5-item death penalty questionnaire used in the original study, but not, when asking directly about Austria's death penalty practices (prevalence: 0.3%). Younger age, but not the amount of television viewing or online streaming, suggestibility, or preferred TV genre consistently predicted incorrect answers in the death penalty questionnaire. Incorrect answers were Mokken-scalable (i.e., formed a common scale, complying with a non-parametric item response model) and were highly consistent. In contrast to the replication study results, a small meta-analysis of all available evidence (three studies, including the present replication) suggested that the aggregate effect of television viewing nominally was significant, albeit small.ConclusionThe replication study yielded mixed results, which indicate the perception of a high prevalence of beliefs that there is capital punishment in a country without death penalty probably is due to a faultily designed questionnaire and thus a research artifact. Also, positive associations of television viewing with such beliefs likely are only small at best.

Project description:We used microarrays to assess gene expression differences in the hippocampus between FoxO6 mutant and wild-type siblings before (basal) or after novel object learning. The cohort of basal mice were housed individually for at least 3 days prior to tissue harvesting. The mice used to collect hippocampal samples after the object learning task were handled daily in the procedure room, prior to the object learning task. 24 hours before the object learning task, each mouse was individually habituated to the empty novel object arena for 10 min. On the days of the novel object learning task and the empty arena habituation, the mice were transferred to the procedure room and acclimatized for 60 min. For the novel object learning task, the mice were allowed to explore two identical and novel objects for 10 min in a 70 x 70 x 70 cm black plastic arena with a white PVC vinyl material on the base. After the object exploration, mice were transferred to an empty cage, and were euthanized after 60 min. The brains were dissected and incubated in ice-cold RNAlater (Ambion) for 24 hours at 4M-BM-:C. The brains were then rapidly frozen in O.C.T. Tissue-Tek (Sakura) at -80M-BM-:C. Coronal brain sections (300 M-BM-5m) were made using a microtome (Microm), the hippocampus was finely dissected and collected into M-bM-^@M-^XRNA lysis bufferM-bM-^@M-^Y (Ambion), and homogenized for 30 sec using a rotar-stat homogenizer. Total RNA was extracted using the RNAqueous column (Ambion) following the manufacturerM-bM-^@M-^Ys protocol.

The brains of Home Cage controls and Fear Conditioned animals (n = 4/group) were then punched to collect amygdala tissue. miRNA were extracted using the Qiagen miRNeasy Kit, and then shipped to Exiqon. Project description:Research into strategies that can combat episodic memory decline in healthy older adults has gained widespread attention over the years. Evidence suggests that a short period of rest immediately after learning can enhance memory consolidation, as compared to engaging in cognitive tasks. However, a recent study in younger adults has shown that post-encoding engagement in a working memory task leads to the same degree of memory consolidation as from post-encoding rest. Here, we tested whether this finding can be extended to older adults. Using a delayed recognition test, we compared the memory consolidation of word-picture pairs learned prior to 9 min of rest or a 2-Back working memory task, and examined its relationship with executive functioning and mindwandering propensity. Our results show that (1) similar to younger adults, memory for the word-picture associations did not differ when encoding was followed by post-encoding rest or 2-Back task and (2) older adults with higher mindwandering propensity retained more word-picture associations encoded prior to rest relative to those encoded prior to the 2-Back task, whereas participants with lower mindwandering propensity had better memory performance for the pairs encoded prior to the 2-Back task. Overall, our results indicate that the degree of episodic memory consolidation during both active and passive post-encoding periods depends on individual mindwandering tendency.
