Targeted Cortical Manipulation of Auditory Perception.

ABSTRACT: Driving perception by direct activation of neural ensembles in cortex is a necessary step for achieving a causal understanding of the neural code for auditory perception and developing central sensory rehabilitation methods. Here, using optogenetic manipulations during an auditory discrimination task in mice, we show that auditory cortex can be short-circuited by coarser pathways for simple sound identification. Yet when the sensory decision becomes more complex, involving temporal integration of information, auditory cortex activity is required for sound discrimination and targeted activation of specific cortical ensembles changes perceptual decisions, as predicted by our readout of the cortical code. Hence, auditory cortex representations contribute to sound discriminations by refining decisions from parallel routes.

Dynamics of auditory cortical activity during behavioural engagement and auditory perception.

Project description:Behavioural engagement can enhance sensory perception. However, the neuronal mechanisms by which behavioural states affect stimulus perception remain poorly understood. Here we record from single units in auditory cortex of rats performing a self-initiated go/no-go auditory task. Self-initiation transforms cortical tuning curves and bidirectionally modulates stimulus-evoked activity patterns and improves auditory detection and recognition. Trial self-initiation decreases the rate of spontaneous activity in the majority of recorded cells. Optogenetic disruption of cortical activity before and during tone presentation shows that these changes in evoked and spontaneous activity are important for sound perception. Thus, behavioural engagement can prepare cortical circuits for sensory processing by dynamically changing sound representation and by controlling the pattern of spontaneous activity.

Cortical auditory evoked potential in cochlear implant users: An objective method to improve speech perception.

Project description:ObjectiveTo investigate if cortical auditory evoked potential (CAEP) measures can be used to verify the cochlear implant (CI) map and consequently improve CI outcomes in adults with bilateral hearing loss.DesignCAEPs were measured in CI recipients using the speech tokens /m/, /g/, /t/ and /s/. If CAEP responses were present for all speech tokens, the participant's map was considered "satisfactory". If CAEP responses were absent, the CI map was considered "unsatisfactory" and therefore adjusted and CAEP measures repeated. This was repeated until auditory potentials were seen in response to all four speech tokens. Speech testing was conducted pre-CI, as well as before and after CAEP-guided map adjustments.Results108 adult unilateral CI users participated, whose sound processors were previously programmed using subjective methods. 42 CI users elicited a CAEP response to all four speech tokens and therefore no further mapping adjustments were made. 66 subjected lacked a CAEP response to at least one speech token and had their CI map adjusted accordingly. Of those, 31 showed a CAEP response to all four speech tokens, and the average speech score significantly improved after CI map adjustments based on CAEP responses.ConclusionCAEP's are an objective tool that can be used to guide and verify CI mapping in adults CI users. Significant improvement in speech scores was observed in participants who had their CI map adjusted based on CAEP responses.

Adaptive auditory brightness perception.

Project description:Perception adapts to the properties of prior stimulation, as illustrated by phenomena such as visual color constancy or speech context effects. In the auditory domain, only little is known about adaptive processes when it comes to the attribute of auditory brightness. Here, we report an experiment that tests whether listeners adapt to spectral colorations imposed on naturalistic music and speech excerpts. Our results indicate consistent contrastive adaptation of auditory brightness judgments on a trial-by-trial basis. The pattern of results suggests that these effects tend to grow with an increase in the duration of the adaptor context but level off after around 8 trials of 2 s duration. A simple model of the response criterion yields a correlation of r = .97 with the measured data and corroborates the notion that brightness perception adapts on timescales that fall in the range of auditory short-term memory. Effects turn out to be similar for spectral filtering based on linear spectral filter slopes and filtering based on a measured transfer function from a commercially available hearing device. Overall, our findings demonstrate the adaptivity of auditory brightness perception under realistic acoustical conditions.

Basal Forebrain Cholinergic-Auditory Cortical Network: Primary Versus Nonprimary Auditory Cortical Areas.

Project description:Acetylcholine (ACh) release in the cortex is critical for learning, memory, attention, and plasticity. Here, we explore the cholinergic and noncholinergic projections from the basal forebrain (BF) to the auditory cortex using classical retrograde and monosynaptic viral tracers deposited in electrophysiologically identified regions of the auditory cortex. Cholinergic input to both primary (A1) and nonprimary auditory cortical (belt) areas originates in a restricted area in the caudal BF within the globus pallidus (GP) and in the dorsal part of the substantia innominata (SId). On the other hand, we found significant differences in the proportions of cholinergic and noncholinergic projection neurons to primary and nonprimary auditory areas. Inputs to A1 projecting cholinergic neurons were restricted to the GP, caudate-putamen, and the medial part of the medial geniculate body, including the posterior intralaminar thalamic group. In addition to these areas, afferents to belt-projecting cholinergic neurons originated from broader areas, including the ventral secondary auditory cortex, insular cortex, secondary somatosensory cortex, and the central amygdaloid nucleus. These findings support a specific BF projection pattern to auditory cortical areas. Additionally, these findings point to potential functional differences in how ACh release may be regulated in the A1 and auditory belt areas.

Summary statistics in auditory perception.

Project description:Sensory signals are transduced at high resolution, but their structure must be stored in a more compact format. Here we provide evidence that the auditory system summarizes the temporal details of sounds using time-averaged statistics. We measured discrimination of 'sound textures' that were characterized by particular statistical properties, as normally result from the superposition of many acoustic features in auditory scenes. When listeners discriminated examples of different textures, performance improved with excerpt duration. In contrast, when listeners discriminated different examples of the same texture, performance declined with duration, a paradoxical result given that the information available for discrimination grows with duration. These results indicate that once these sounds are of moderate length, the brain's representation is limited to time-averaged statistics, which, for different examples of the same texture, converge to the same values with increasing duration. Such statistical representations produce good categorical discrimination, but limit the ability to discern temporal detail.

Decoding contextual influences on auditory perception from primary auditory cortex.

Project description:Perception can be highly dependent on stimulus context, but whether and how sensory areas encode the context remains uncertain. We used an ambiguous auditory stimulus - a tritone pair - to investigate the neural activity associated with a preceding contextual stimulus that strongly influenced the tritone pair's perception: either as an ascending or a descending step in pitch. We recorded single-unit responses from a population of auditory cortical cells in awake ferrets listening to the tritone pairs preceded by the contextual stimulus. We find that the responses adapt locally to the contextual stimulus, consistent with human MEG recordings from the auditory cortex under the same conditions. Decoding the population responses demonstrates that pitch-change selective cells are able to predict well the context-sensitive percept of the tritone pairs. Conversely, decoding the distances between the pitch representations predicts the opposite of the percept. The various percepts can be readily captured and explained by a neural model of cortical activity based on populations of adapting, pitch and pitch-direction selective cells, aligned with the neurophysiological responses. Together, these decoding and model results suggest that contextual influences on perception may well be already encoded at the level of the primary sensory cortices, reflecting basic neural response properties commonly found in these areas.

Auditory cortical connectivity in humans.

Project description:To understand auditory cortical processing, the effective connectivity between 15 auditory cortical regions and 360 cortical regions was measured in 171 Human Connectome Project participants, and complemented with functional connectivity and diffusion tractography. 1. A hierarchy of auditory cortical processing was identified from Core regions (including A1) to Belt regions LBelt, MBelt, and 52; then to PBelt; and then to HCP A4. 2. A4 has connectivity to anterior temporal lobe TA2, and to HCP A5, which connects to dorsal-bank superior temporal sulcus (STS) regions STGa, STSda, and STSdp. These STS regions also receive visual inputs about moving faces and objects, which are combined with auditory information to help implement multimodal object identification, such as who is speaking, and what is being said. Consistent with this being a "what" ventral auditory stream, these STS regions then have effective connectivity to TPOJ1, STV, PSL, TGv, TGd, and PGi, which are language-related semantic regions connecting to Broca's area, especially BA45. 3. A4 and A5 also have effective connectivity to MT and MST, which connect to superior parietal regions forming a dorsal auditory "where" stream involved in actions in space. Connections of PBelt, A4, and A5 with BA44 may form a language-related dorsal stream.

Sensorimotor Integration Can Enhance Auditory Perception.

Project description:Whenever we move, speak, or play musical instruments, our actions generate auditory sensory input. The sensory consequences of our actions are thought to be predicted via sensorimotor integration, which involves anatomical and functional links between auditory and motor brain regions. The physiological connections are relatively well established, but less is known about how sensorimotor integration affects auditory perception. The sensory attenuation hypothesis suggests that the perceived loudness of self-generated sounds is attenuated to help distinguish self-generated sounds from ambient sounds. Sensory attenuation would work for louder ambient sounds, but could lead to less accurate perception if the ambient sounds were quieter. We hypothesize that a key function of sensorimotor integration is the facilitated processing of self-generated sounds, leading to more accurate perception under most conditions. The sensory attenuation hypothesis predicts better performance for higher but not lower intensity comparisons, whereas sensory facilitation predicts improved perception regardless of comparison sound intensity. A series of experiments tested these hypotheses, with results supporting the enhancement hypothesis. Overall, people were more accurate at comparing the loudness of two sounds when making one of the sounds themselves. We propose that the brain selectively modulates the perception of self-generated sounds to enhance representations of action consequences.

McGurk illusion recalibrates subsequent auditory perception.

Project description:Visual information can alter auditory perception. This is clearly illustrated by the well-known McGurk illusion, where an auditory/aba/ and a visual /aga/ are merged to the percept of 'ada'. It is less clear however whether such a change in perception may recalibrate subsequent perception. Here we asked whether the altered auditory perception due to the McGurk illusion affects subsequent auditory perception, i.e. whether this process of fusion may cause a recalibration of the auditory boundaries between phonemes. Participants categorized auditory and audiovisual speech stimuli as /aba/, /ada/ or /aga/ while activity patterns in their auditory cortices were recorded using fMRI. Interestingly, following a McGurk illusion, an auditory /aba/ was more often misperceived as 'ada'. Furthermore, we observed a neural counterpart of this recalibration in the early auditory cortex. When the auditory input /aba/ was perceived as 'ada', activity patterns bore stronger resemblance to activity patterns elicited by /ada/ sounds than when they were correctly perceived as /aba/. Our results suggest that upon experiencing the McGurk illusion, the brain shifts the neural representation of an /aba/ sound towards /ada/, culminating in a recalibration in perception of subsequent auditory input.
