The taxonomic identities of Pholidota wenshanica and P. subcalceata (Orchidaceae, Coelogyninae).

ABSTRACT: P. wenshanica S.C.Chen & Z.H.Tsi and P. subcalceata Gagnep. have long been recognized as synonyms of P. leveilleana Schltr. In the present study, detailed morphological comparisons suggest that specimens referred to as P. wenshanica and P. subcalceata differ significantly in both vegetative and floral characters from those of P. leveilleana. Here we resurrect P. wenshanica and P. subcalceata as independent species. Key diagnostic characters essential for delineating identities of these species are presented.

The complete chloroplast genome of Pholidota imbricata (Orchidaceae).

Project description:Pholidota imbricata belongs to tribe Coelogninae in Orchidaceae distributed in Sichuan, Xizang, and Yunnan. Here, we report the first complete chloroplast (cp) genome and the cp genome features of P. imbricata. The complete cp genome sequence of P. imbricata is 159,292?bp in length and presented a typical quadripartite structure including one large single-copy region (LSC, 87,515?bp), one small single-copy region (SSC, 20,999?bp), and two inverted repeat regions (IRs, 25,389?bp each). The cp genome encoded 141 genes, of which 108 were unique genes (80 protein-coding genes, 24 tRNAs, and 4 rRNAs). The phylogenetic relationships show that P. imbricata is sister to the species of the genus Pleione in tribe Coelogninae.

The complete chloroplast genome of Pholidota yunnanensis Rolfe (Orchidaceae: Coelogyninae).

Project description:In this study, the complete chloroplast genome of Pholidota yunnanensis is presented, which represents first complete plastid genome of the genus Pholidota in the subtribe Coelogyninae. The chloroplast genome size is 159,729 bp, including a GC content of 37.3% and 135 genes (89 protein-coding genes, 38 tRNA genes, 8 rRNA genes). The genome structure is typical quadripartite, consisting of a pair of inverted repeat regions (26,638 bp) separated by a large single-copy region (LSC, 87,610 bp) and a small single-copy region (SSC, 18,843 bp). Phylogenetic analysis among 15 species based on cp genomes recovered a well-supported phylogenetic tree and indicated a close relationship between Pholidota yunnanensis and Pleione bulbocodioides.

The complete chloroplast genome sequence of Pholidota articulata (Orchidaceae), a rarely medicinal orchid.

Project description:Pholidota Lindl. ex Hook. was placed in tribe Arethuseae Lindl. (Epidendroideae, Orchidaceae), while its generic relationship has been unclear. Since the plastid genome could play a key role in plant systematics, the complete chloroplast (cp) genome of P. articulata was reported in this paper. The cp genome was 160,114 bp in length with four typical quadripartite structures, which was consisted of a large single copy (LSC) region of 87,756 bp, a small single copy (SSC) region of 18,872 bp, and two inverted repeats (IR) of 26,734 bp. In addition, the cp genome encoded 132 genes in total, of which were 113 unique genes, including 79 protein-coding genes, 30 tRNAs, and 4 rRNAs. The phylogenetic analysis indicated that P. articulata was closely clustered with other two species of Pholidota and that they appeared to be related to Pleione in Arethuseae Lindl.

A taxonomic revision of Herminium L. (Orchidoideae, Orchidaceae).

Project description:Herminium (Orchidaceae, Orchidoideae) is a medium-sized genus widespread in the northern hemisphere, with a clear centre of diversity in the Himalayas. We present a comprehensive taxonomic revision of Herminium based on field observations and morphological studies, for which we examined about 2500 specimens. We recognize 49 species grouped into six formal sections, including one new species, Herminium tibeticum, from Tibet. We provide an identification key to the species, descriptions of the species, notes on ecology and distribution, and complete nomenclature for each species, including typifications. We here designate lectotypes for five species and reduce four taxa to synonymy.

Taxonomic revision of the genus Cranichis (Orchidaceae, Cranichideae) in Colombia.

Project description:The geographical range of the orchid genus Cranichis extends from USA, Central America, and the Caribbean to Bolivia and Argentina-with the greatest diversity observed in the Andean region. This taxon embraces herbs with the scape and basal leaves developing from a single, terminal bud and having the scape enclosed in several sheaths. The small, non-resupinate, flowers are arranged in a racemose inflorescence. The petals and lip are free from the column part and their gynostemium is short and massive. In this paper, the synopsis of Cranichis in Colombia is presented. Each species occurring in the country is characterized and illustrated. The information about habitat and distribution of national genus representatives are provided. Several groups of species that are very similar in their morphology are delineated to facilitate process of species identification. A total of 10 species are described here as new. Lectotypes for the six species Cranichis ciliata, C. fendleri, C. mandonii, C. tenuis, C. viereckii, and C. wageneri have been selected. Additionally, two new nomenclatorial combinations within Ocampoa are proposed.

The taxonomic identity of three varieties of Lecanorchis nigricans (Vanilleae, Vanilloideae, Orchidaceae) in Japan.

Project description:To elucidate the taxonomy of the Lecanorchis nigricans Honda, 1931 species complex, the present study investigated the detailed morphology of three L. nigricans varieties in Japan. While L. nigricans var. patipetala Y.Sawa, 1980 and L. nigricans var. yakusimensis T.Hashim., 1990 have often been treated as synonyms of L. nigricans var. nigricans, the present study demonstrates that the three varieties are morphologically distinct. More specifically, L. nigricans var. nigricans only produces complete cleistogamous flowers and is distinct from the plants currently called "L. nigricans", which are identical to the chasmogamous variety L. nigricans var. patipetala. The other chasmogamous variety L. nigricans var. yakusimensis can be easily distinguished from L. nigricans var. patipetala by its more spatulate tepals and higher cucullate lip. Therefore, the present study provides emended description of the three L. nigricans varieties based on type specimens and specimens collected from type localities. In addition, the isotype specimen of L. nigricans var. patipetala is designated as the lectotype because the holotype has been lost.

Molecular phylogenetic study of the tribe Tropidieae (Orchidaceae, Epidendroideae) with taxonomic and evolutionary implications.

Project description:The orchid tribe Tropidieae comprises three genera, Tropidia, Corymborkis and Kalimantanorchis. There are three fully mycoheterotrophic species within Tropidieae: Tropidia saprophytica, T. connata and Kalimantanorchis nagamasui. A previous phylogenetic study of K. nagamasui, based only on plastid matK data, placed K. nagamasui outside the clade of Tropidia and Corymborkis without support. In this study, we performed phylogenetic analyses using a nuclear ribosomal DNA spacer (ITS1-5.8S-ITS2), a low-copy nuclear coding gene (Xdh) and a mitochondrial intron (nad1b-c intron) to study the phylogenetic relationships within Tropidieae. We included six photosynthetic and all three fully mycoheterotrophic Tropidieae species. The resulting phylogenetic trees placed these fully mycoheterotrophic species inside the Tropidia clade with high support. In our trees, these three species do not form a monophyletic group together, because the photosynthetic T. graminea is nested amongst them. Our results also suggest that the loss of photosynthetic ability occurred at least twice in Tropidia.

Taxonomic placement of Paphiopedilum rungsuriyanum (Cypripedioideae; Orchidaceae) based on morphological, cytological and molecular analyses.

Project description:BackgroundPaphiopedilum rungsuriyanum from Northern Laos was discovered and described in 2014. It is characterized by having miniature tessellated leaves, a flower having a helmet shaped lip with a V-shaped neckline, and a semi-lunate, 3-dentate staminode with an umbo. These morphological features distinguish P. rungsuriyanum from the other known sections/subgenera of Paphiopedilum, making it difficult to group with existing infrageneric units.ResultsPaphiopedilum rungsuriyanum has chromosome number of 2n = 26. Fluorescence in situ hybridization study demonstrates that there are two 45S rDNA signals in the telomeric region of chromosomes, and more than 20 5S rDNA signals dispersed signals in the pericentromeric and centromeric regions. Phylogenetic analyses based on four nuclear (i.e. ITS, ACO, DEF4 and RAD51) and four plastid (i.e. atpI-atpH, matK, trnS-trnfM and ycf1) gene regions indicate that P. rungsuriyanum is nested in subgenus Paphiopedilum and is a sister to section Paphiopedilum.ConclusionsThe results in combination with karyomorphological, rDNA FISH patterns, morphological and phylogenetic analyses suggest a new section Laosianum to accommodate this species in the current sectional circumscription of subgenus Paphiopedilum.
