Seed Pretreatment and Foliar Application of Proline Regulate Morphological, Physio-Biochemical Processes and Activity of Antioxidant Enzymes in Plants of Two Cultivars of Quinoa (Chenopodium quinoa Willd.).

ABSTRACT: In the current study, the effects of exogenously applied proline (25 and 50 mM) and low-temperature treatment were examined on the physiochemical parameters in the plants of two cultivars (V1 and V2) of quinoa (Chenopodium quinoa Willd.). The seeds were also exposed to chilling stress at 4 °C before sowing. Plants raised from the seeds treated with low temperature showed reduced plant growth and contents of chlorophyll and carotenoids, but they had significantly increased contents of malondialdehyde, proline, ascorbic acid, total free amino acids, total soluble sugars, and total phenolics, as well as the activity of the peroxidase (POD) enzyme. Cold stress applied to seeds remained almost ineffective in terms of bringing about changes in plant root, hydrogen peroxide, glycine betaine and activities of superoxide dismutase (SOD), and catalase (CAT) enzymes. The exogenous application of proline significantly increased plant growth, the contents of chlorophyll, carotenoids, proline, ascorbic acid, total free amino acids, phenolics, and total soluble sugars, as well as the activities of SOD, POD, and CAT, but it decreased malondialdehyde content. Overall, foliar application of proline was better than the seed treatment in improving root dry weight, root length, chlorophyll a, carotenoids, glycine betaine, ascorbic acid and superoxide dismutase activity, whereas seed pre-treatment with proline was effective in improving shoot dry weight, shoot length, hydrogen peroxide, malondialdehyde, and peroxidase activity in both quinoa cultivars.

Project description:Quinoa (Chenopodium quinoa Willd.), an Andean native crop, is increasingly popular around the world due to its high nutritional content and stress tolerance. The production and the popularity of this strategic global food are greatly restricted by many limiting factors, such as seed pre-harvest sprouting, bitter saponin, etc. To solve these problems, the underlying mechanism of seed maturation in quinoa needs to be investigated. In this study, based on the investigation of morphological characteristics, a quantitative analysis of its global proteome was conducted using the combinational proteomics of tandem mass tag (TMT) labeling and parallel reaction monitoring (PRM). The proteome changes related to quinoa seed maturation conversion were monitored to aid its genetic improvement. Typical changes of morphological characteristics were discovered during seed maturation, including mean grain diameter, mean grain thickness, mean hundred-grain weight, palea, episperm color, etc. With TMT proteomics analysis, 581 differentially accumulated proteins (DAPs) were identified. Functional classification analysis and Gene Ontology enrichment analysis showed that most DAPs involved in photosynthesis were downregulated, indicating low levels of photosynthesis. DAPs that participated in glycolysis, such as glyceraldehyde-3-phosphate dehydrogenase, pyruvate decarboxylase, and alcohol dehydrogenase, were upregulated to fulfill the increasing requirement of energy consumption during maturation conversion. The storage proteins, such as globulins, legumins, vicilins, and oleosin, were also increased significantly during maturation conversion. Protein-protein interaction analysis and function annotation revealed that the upregulation of oleosin, oil body-associated proteins, and acyl-coenzyme A oxidase 2 resulted in the accumulation of oil in quinoa seeds. The downregulation of β-amyrin 28-oxidase was observed, indicating the decreasing saponin content, during maturation, which makes the quinoa "sweet". By the PRM and qRT-PCR analysis, the expression patterns of most selected DAPs were consistent with the result of TMT proteomics. Our study enhanced the understanding of the maturation conversion in quinoa. This might be the first and most important step toward the genetic improvement of quinoa.

Project description:The total area under quinoa (Chenopodium quinoa Willd.) cultivation and the consumption of its grain have increased in recent years because of its nutritional properties and ability to grow under adverse conditions, such as drought. Climate change scenarios predict extended periods of drought and this has emphasized the need for new crops that are tolerant to these conditions. The main goal of this work was to evaluate crop yield and quality parameters and to characterize the physiology of two varieties of quinoa grown under water deficit in greenhouse conditions. Two varieties of quinoa from the Chilean coast (Rainbow) and altiplano (Illpa) were used, grown under full irrigation or two different levels of water deficit applied during the grain filling period. There were no marked differences in yield and quality parameters between treatments, but the root biomass was higher in plants grown under severe water deficit conditions compared to control. Photosynthesis, transpiration and stomatal conductance decreased with increased water stress in both cultivars, but the coastal variety showed higher water use efficiency and less discrimination of 13C under water deficit. This response was associated with greater root development and a better stomatal opening adjustment, especially in the case of Rainbow. The capacity of Rainbow to increase its osmoregulant content (compounds such as proline, glutamine, glutamate, K and Na) could enable a potential osmotic adjustment in this variety. Moreover, the lower stomatal opening and transpiration rates were also associated with higher leaf ABA concentration values detected in Rainbow. We found negative logarithmic relationships between stomatal conductance and leaf ABA concentration in both varieties, with significant R2 values of 0.50 and 0.22 in Rainbow and Illpa, respectively. These moderate-to-medium values suggest that, in addition to ABA signaling, other causes for stomatal closure in quinoa under drought such as hydraulic regulation may play a role. In conclusion, this work showed that two quinoa cultivars use different strategies in the face of water deficit stress, and these prevent decreases in grain yield and quality under drought conditions.
