Molecular Identification of Endophytic Fungi and Their Pathogenicity Evaluation Against Dendrobium nobile and Dendrobium officinale.

ABSTRACT: : Dendrobium are tropical orchid plants that host diverse endophytic fungi. The role of these fungi is not currently well understood in Dendrobium plants. We morphologically and molecularly identified these fungal endophytes, and created an efficient system for evaluating the pathogenicity and symptoms of endophytic fungi on Dendrobium nobile and Dendrobium officinale though in vitro co-culturing. ReThe colony morphological traits of Dendrobium myco-endophytes (DMEs) were recorded for their identification. Molecular identification revealed the presence of Colletotrichum tropicicola, Fusarium keratoplasticum, Fusarium oxysporum, Fusarium solani, and Trichoderma longibrachiatum. The pathogenicity results revealed that T. longibrachiatum produced the least pathogenic effects against D. nobile protocorms. In seedlings, T. longibrachiatum showed the least pathogenic effects against D. officinale seedlings after seven days. C. tropicicola produced highly pathogenic effects against both Dendrobium seedlings. The results of histological examination of infected tissues revealed that F. keratoplasticum and T. longibrachiatum fulfill Koch's postulates for the existence of endophytes inside the living tissues. The DMEs are cross-transmitted inside the host plant cells, playing an important role in plant host development, resistance, and alkaloids stimulation.

Project description:Endophytic fungi are microorganisms that, exhibiting within the plant tissues without causing any apparent harm to the host, establish a symbiotic relationship with plants. Host plants provide endophytic fungi with essential nutrients and a protected environment. In exchange, the fungi can enhance the plant's ability to acquire nutrients. They can also play a crucial role in increasing the host plant's tolerance to various abiotic and biotic stresses. Endophytic fungi can produce a wide range of bioactive compounds, some similar to those found in the host plant. In Iraq's Duhok province of the Kurdistan region, the plant species Vitis vinifera has been explored as a habitat for diverse endophytic microorganisms across various ecological environments. During the period from 2021 to 2022, a total of 600 samples were collected from four distinct locations: Bagera, Besfke, Barebhar, and Atrush. From these samples, twelve endophytic fungal species were isolated, including Aspergillus flavipes, Botryosphaeria dothidea, Fusarium oxysporum, Fusarium ruscicol, Fusarium venenatum, Chaetomium globosum, Clonostachys rosea, Mucor racemosus, Penicillium glabrum, Aspergillus terreus, Aspergillus nidulans, and Aspergillus niger, Alternaria alternata, Paecilomyces maximus, Curvularia buchloes. These fungi were introduced for their potential as biocontrol agents against grapevine trunk diseases and grape rotting fungi, which pose significant risks to grapevine health and productivity. Penicilium radiatolobatum, Botrysphaeria dothidea, Fusarium ruscicola, Fusarium venenatum, and Paecilomyces maximus represented the first record as endophytes on grapevine in Iraq. Based on ITS and SSU sequencing, molecular identification confirmed these fungi's presence with sequence identities ranging from 99% to 100%. Phylogenetic analysis revealed that these endophytes could be categorized into five main clusters (A, B, C, D, and E), showing high intra-group similarity. Utilizing the Dual Culture method, the endophyte Paecilomyces maximus demonstrated a 70.83% inhibition rate against Ilyonectria destructans. In the Food Poisoning method, A. flavipes and P. maximus emerged as the most effective inhibitors of Ilyonectria destructans, whereas A. terreus, M. racemosus, and P. maximus achieved complete inhibition (100%) of Botrytis cinerea. Additionally, M. racemosus was identified as the most effective biocontrol agent against Neoscytalidium dimidiatum. In conclusion, the study emphasizes the potential of endophytic fungi from Vitis vinifera as effective biocontrol agents against grapevine diseases, highlighting their role in sustainable vineyard management. These findings lead to further exploration and implementation of these fungi-inserted pest management strategies.

Project description:IntroductionDendrobium nobile (D. nobile), a valued Chinese herb known for its diverse pharmacological effects, owes much of its potency to the bioactive compound dendrobine. However, dendrobine content varies significantly with plant age, and the mechanisms governing this variation remain unclear. This study delves into the potential role of endophytic fungi in shaping host-microbe interactions and influencing plant metabolism.MethodsUsing RNA-seq, we examined the transcriptomes of 1-year-old, 2-year-old, and 3-year-old D. nobile samples and through a comprehensive analysis of endophytic fungal communities and host gene expression in D. nobile stems of varying ages, we aim to identify associations between specific fungal taxa and host genes.ResultsThe results revealing 192 differentially expressed host genes. These genes exhibited a gradual decrease in expression levels as the plants aged, mirroring dendrobine content changes. They were enriched in 32 biological pathways, including phagosome, fatty acid degradation, alpha-linolenic acid metabolism, and plant hormone signal transduction. Furthermore, a significant shift in the composition of the fungal community within D. nobile stems was observed along the age gradient. Olipidium, Hannaella, and Plectospherella dominated in 1-year-old plants, while Strelitziana and Trichomerium prevailed in 2-year-old plants. Conversely, 3-year-old plants exhibited additional enrichment of endophytic fungi, including the genus Rhizopus. Two gene expression modules (mediumpurple3 and darkorange) correlated significantly with dominant endophytic fungi abundance and dendrobine accumulation. Key genes involved in dendrobine synthesis were found associated with plant hormone synthesis.DiscussionThis study suggests that the interplay between different endophytic fungi and the hormone signaling system in D. nobile likely regulates dendrobine biosynthesis, with specific endophytes potentially triggering hormone signaling cascades that ultimately influence dendrobine synthesis.

Project description:IntroductionDendrobine, a valuable alkaloid found in Dendrobium nobile, possesses significant pharmaceutical potential.MethodsIn this study, we explored innovative approaches to enhance dendrobine production by utilizing endophytic fungi in a Temporary Immersion Bioreactor System (TIBS, Nanjing BioFunction Co. Ltd., China) and traditional test bottles. Dendrobine was unequivocally identified and characterised in D. nobile co-culture seedlings through UHPLC analysis and LC-MS qTOF analysis, supported by reference standards.ResultsThe CGTB (control group) and EGTB (experimental group) 12-month-old D. nobile seedlings exhibited similar peak retention times at 7.6±0.1 minutes, with dendrobine identified as C16H25NO2 (molecular weight 264.195). The EGTB, co-cultured with Trichoderma longibrachiatum (MD33), displayed a 2.6-fold dendrobine increase (1804.23 ng/ml) compared to the CGTB (685.95 ng/ml). Furthermore, a bioanalytical approach was applied to investigate the mono-culture of T. longibrachiatum MD33 with or without D. nobile seedlings in test bottles. The newly developed UHPLC-MS method allowed for dendrobine identification at a retention time of 7.6±0.1 minutes for control and 7.6±0.1 minutes for co-culture. Additionally, we explored TIBS to enhance dendrobine production. Co-culturing D. nobile seedlings with Trichoderma longibrachiatum (MD33) in the TIBS system led to a substantial 9.7-fold dendrobine increase (4415.77 ng/ml) compared to the control (454.01 ng/ml) after just 7 days. The comparative analysis of dendrobine concentration between EGTB and EGTIBS highlighted the remarkable potential of TIBS for optimizing dendrobine production. Future research may focus on scaling up the TIBS approach for commercial dendrobine production and investigating the underlying mechanisms for enhanced dendrobine biosynthesis in D. nobile. The structural elucidation of dendrobine was achieved through 1H and 13C NMR spectroscopy, revealing a complex array of proton environments and distinct carbon environments, providing essential insights for the comprehensive characterization of the compound.DiscussionThese findings hold promise for pharmaceutical and industrial applications of dendrobine and underline the role of endophytic fungi in enhancing secondary metabolite production in medicinal plants.
