Establishment of cumulative assessment groups of pesticides for their effects on the thyroid.

ABSTRACT: Cumulative assessment groups of pesticides have been established for two specific effects on the thyroid: firstly hypothyroidism, and secondly parafollicular cell (C-cell) hypertrophy, hyperplasia and neoplasia. Sources of uncertainties resulting from the methodological approach and from the limitations in available data and scientific knowledge have been identified and considered. This report supports the publication of a scientific report on cumulative risk assessment to pesticides affecting the thyroid, in which all uncertainties identified for either the exposure assessment or the establishment of the cumulative assessment groups are incorporated into a consolidated risk characterisation.

Sources of uncertainties resulting from the methodological approach and from the limitations in available data and scientific knowledge have been identified and considered. Cumulative dietary exposure assessment of pesticides that have chronic effects on the thyroid using SAS® software.

Project description:Retrospective dietary exposure assessments were conducted for two groups of pesticides that have chronic effects on the thyroid: hypertrophy, hyperplasia and neoplasia of C-cells, and hypothyroidism. The pesticides considered in this assessment were identified and characterised in the scientific report on the establishment of cumulative assessment groups of pesticides for their effects on the thyroid. The exposure assessments used monitoring data collected by Member States under their official pesticide monitoring programmes in 2014, 2015 and 2016, and individual food consumption data from 10 populations of consumers from different countries and from different age groups. Exposure estimates were obtained for each group of pesticides by means of a 2-dimensional probabilistic model, which was implemented in SAS ® software. Results were validated against exposure estimates obtained by the Dutch National Institute for Public Health and the Environment (RIVM) using the Monte Carlo Risk Assessment (MCRA) software. Both tools produced nearly identical results and minor differences were mainly attributed to the random effect of probabilistic modelling. The exposure estimates obtained in this report are used in the final scientific report on the cumulative dietary risk characterisation of pesticides that have chronic effects on the thyroid. The latter combines the hazard assessment and exposure assessment into a consolidated risk characterisation including all uncertainties.

Project description:A retrospective cumulative risk assessment of dietary exposure to pesticide residues was conducted for chronic inhibition of acetylcholinesterase. The pesticides considered in this assessment were identified and characterised in a previous scientific report on the establishment of cumulative assessment groups of pesticides for their effects on the nervous system. The exposure assessments used monitoring data collected by Member States under their official pesticide monitoring programmes in 2016, 2017 and 2018, and individual food consumption data from 10 populations of consumers from different countries and from different age groups. Exposure estimates were obtained by means of a two-dimensional probabilistic model, which was implemented in SAS ® software. The characterisation of cumulative risk was supported by an uncertainty analysis based on expert knowledge elicitation. For each of the 10 populations, it is concluded with varying degrees of certainty that cumulative exposure to pesticides contributing to the chronic inhibition of acetylcholinesterase does not exceed the threshold for regulatory consideration established by risk managers.

Pesticides With Potential Thyroid Hormone-Disrupting Effects: A Review of Recent Data.

Project description:Plant Protection Products, more commonly referred to as pesticides and biocides, are used to control a wide range of yield-reducing pests including insects, fungi, nematodes, and weeds. Concern has been raised that some pesticides may act as endocrine disrupting chemicals (EDCs) with the potential to interfere with the hormone systems of non-target invertebrates and vertebrates, including humans. EDCs act at low doses and particularly vulnerable periods of exposure include pre- and perinatal development. Of critical concern is the number of pesticides with the potential to interfere with the developing nervous system and brain, notably with thyroid hormone signaling. Across vertebrates, thyroid hormone orchestrates metamorphosis, brain development, and metabolism. Pesticide action on thyroid homeostasis can involve interference with TH production and its control, displacement from distributor proteins and liver metabolism. Here we focused on thyroid endpoints for each of the different classes of pesticides reviewing epidemiological and experimental studies carried out both in in vivo and in vitro. We conclude first, that many pesticides were placed on the market with insufficient testing, other than acute or chronic toxicity, and second, that thyroid-specific endpoints for neurodevelopmental effects and mixture assessment are largely absent from regulatory directives.

Novel genetic variants in differentiated thyroid cancer and assessment of the cumulative risk.

Project description:A genome-wide association study (GWAS) performed on a high-incidence Italian population followed by replications on low-incidence cohorts suggested a strong association of differentiated thyroid cancer (DTC) with single nucleotide polymorphisms (SNPs) at 9q22.33, 2q35, 20q11.22-q12 and 14q24.3. Moreover, six additional susceptibility loci were associated with the disease only among Italians. The present study had two aims, first to identify loci involved in DTC risk and then to assess the cumulative effect of the SNPs identified so far in the Italian population. The combined analysis of the previous GWAS and the present Italian study provided evidence of association with rs7935113 (GALNTL4, OR = 1.36, 95%CI 1.20-1.53, p-value = 7.41 × 10(-7)) and rs1203952 (FOXA2, OR = 1.29, 95%CI 1.16-1.44, p-value = 4.42 × 10(-6)). Experimental ENCODE and eQTL data suggested that both SNPs may influence the closest genes expression through a differential recruitment of transcription factors. The assessment of the cumulative risk of eleven SNPs showed that DTC risk increases with an increasing number of risk alleles (p-trend = 3.13 × 10(-47)). Nonetheless, only a small fraction (about 4% on the disease liability scale) of DTC is explained by these SNPs. These data are consistent with a polygenic model of DTC predisposition and highlight the importance of association studies in the discovery of the disease hereditability.

Competitive androgen receptor antagonism as a factor determining the predictability of cumulative antiandrogenic effects of widely used pesticides.

Project description:BackgroundMany pesticides in current use have recently been revealed as in vitro androgen receptor (AR) antagonists, but information about their combined effects is lacking.ObjectiveWe investigated the combined effects and the competitive AR antagonism of pesticide mixtures.MethodsWe used the MDA-kb2 assay to test a combination of eight AR antagonists that did not also possess AR agonist properties ("pure" antagonists; 8 mix: fludioxonil, fenhexamid, ortho-phenylphenol, imazalil, tebuconazole, dimethomorph, methiocarb, pirimiphos-methyl), a combination of five AR antagonists that also showed agonist activity (5 mix: cyprodinil, pyrimethanil, vinclozolin, chlorpropham, linuron), and all pesticides combined (13 mix). We used concentration addition (CA) and independent action (IA) to formulate additivity expectations, and Schild plot analyses to investigate competitive AR antagonism.ResultsA good agreement between the effects of the mixture of eight "pure" AR antagonists and the responses predicted by CA was observed. Schild plot analysis revealed that the 8 mix acted by competitive AR antagonism. However, the observed responses of the 5 mix and the 13 mix fell within the "prediction window" boundaries defined by the predicted regression curves of CA and IA. Schild plot analysis with these mixtures yielded anomalous responses incompatible with competitive receptor antagonism.ConclusionsA mixture of widely used pesticides can, in a predictable manner, produce combined AR antagonist effects that exceed the responses elicited by the most potent component alone. Inasmuch as large populations are regularly exposed to mixtures of antiandrogenic pesticides, our results underline the need for considering combination effects for these substances in regulatory practice.

Relationship between cumulative exposure to pesticides and sleep disorders among greenhouse vegetable farmers.

Project description:BACKGROUND:In the northern region of China, many greenhouse vegetable farmers are exposed to high cumulative levels of pesticides due to long-term work in greenhouses that impacts their health. The aim of the current study was to identify the relationship between cumulative pesticide exposure and sleep disorders among farmers in Yinchuan, Northwest China. METHODS:A cross-sectional study was conducted for 3 consecutive years in 2015, 2016 and 2017. Using a random sampling to select the resident teams, 1366 participants were enrolled, and information was collected via face-to-face interviews by trained investigators. Ordinal logistic, multinomial logistic and poisson logistic regression models were used to identify the associations between cumulative exposure intensity (CEI) and sleep disorders. RESULTS:High CEI (OR = 1.56, 95% CI: 1.02-3.38) was associated with short sleep duration when compared with low CEI in the Full Model. CEI was not associated with long sleep duration. Self-rated sleep quality was associated with medium (OR = 1.46, 95% CI: 1.10-2.00) and high (OR = 2.50, 95% CI: 1.83-3.40) CEI. Similarly, having difficulty sleeping was associated with medium (OR = 1.52, 95% CI: 1.02-2.24) and high (OR = 1.74, 95% CI: 1.16-2.62) CEI. Differences in the associations by gender were also noted. CONCLUSION:CEI was associated with sleep disorders, and gender differences were observed. Efforts should be made by local governments to address sleep problems that result from cumulative pesticide exposure in farmers, and gender differences should be considered.

<i>In vitro</i> assessment of pesticides capacity to act as agonists/antagonists of the thyroid hormone nuclear receptors.

Project description:Chemicals acting as thyroid hormone disruptors (THDs) are of a particular concern for public health, considering the importance of this hormone in neurodevelopment and metabolic processes. They might either alter the circulating level of thyroid hormone (TH) or interfere with the cellular response to the hormonal stimulation. In order to assess this later possibility we selected 39 pesticides and combined several in vitro tests. Reporter assays respectively addressed the transactivation capacity of the full-length TH nuclear receptor TRα1, the transactivation capacity of its C-terminal ligand binding domain, or the ability of the hormone to destabilize the interaction between TRα1 and the transcriptional corepressor NcoR. Although some pesticides elicit a cellular response, which sometimes interferes with TH signaling, RNA-seq analysis provided no evidence that they can act as TRα1 agonists or antagonists. Their neurodevelopmental toxicity in mammals cannot be explained by an alteration of the response to TH.

Assessment of early exaggerated treatment effects in orthodontic interventions using cumulative meta-analysis.

Project description:BackgroundThe reported initial strong treatment effects reported in early trials that are refuted in subsequent future studies assessing the same interventions have been attributed to novelty bias. The aim of this study was to determine whether there is any evidence of novelty bias in the reported treatment effects of orthodontics interventions.Materials and methodsRelevant orthodontic systematic review (SRs) topics containing at least one meta-analysis on either binary or continuous outcomes with a minimum of three trials considered important areas in the field of orthodontic practice were identified. SR, meta-analysis, and primary study-level characteristics were extracted. Descriptive statistics were calculated at the SRs, meta-analysis, and at the individual study level. All SR and trial-level data were imported into the statistical software and all meta-analyses were replicated using the cumulative random-effects meta-analysis approach. Changes in the size and direction of the estimates between the first trial and the cumulative effect over time were recorded.ResultsForty-seven meta-analyses were included. The total number of primary studies included within these meta-analyses was 408 (N = 408). Overall, the final effect size estimate decreased in 29 (61.7%, N = 29/47) cumulative meta-analyses whilst it increased in the remaining 18 (38.3%, N = 18/47). No association between the level of risk of bias and the cumulative absolute effect size was evident (OR 1.00; 95% CI: 0.98, 1.03; P = 0.717) after adjusting for year of the primary study (P = 0.22).ConclusionsClinicians should be wary of the results of trials reporting the effectiveness of new interventions as there is a possibility that the reported effect size will be often exaggerated.
