Capture and light-induced release of antibiotics by an azo dye polymer.

ABSTRACT: The isomerisation of azo dyes can induce conformational changes which have potential applications in medicine and environmental protection. We developed an agar diffusion assay to test the capture and release of biologically active molecules from an azo electro-optic polymer, Poly (Disperse Red 1 methacrylate) (DR1/PMMA). The assay monitors the growth of bacteria placed in soft agar under a glass coverslip. Antibiotics can then be applied on the coverslip resulting in the clearance of the area under the coverslip due to growth inhibition. This assay demonstrates that DR1/PMMA is able to capture either tetracycline or ampicillin and the relative amount of DR1/PMMA required for capture was determined. Finally, the active antibiotics can be released from DR1/PMMA by exposure to green laser light. Exposure to white light from a torch or to heat does not release the antibiotic.

Light induced conch-shaped relief in an azo-polymer film.

Project description:We have discovered that a novel chiral structured surface relief (termed 'conch'-shaped surface relief) with a height of over 1 μm can be formed in an azo-polymer film merely by employing circularly polarized optical vortex irradiation with a total angular momentum of j = ±2. The temporal evolution of the conch-shaped surface relief in the azo-polymer film was also observed. The results provide physical insight into how the angular momentum of light is transferred to a material through mass transport by cis-trans photo-isomerization. Such conch-shaped surface reliefs with chirality, in which functional chemical composites can be doped, enable new applications, such as planar chiral metamaterials, plasmonic holograms, and identification of chiral chemical composites.

Glow discharge plasma stabilization of azo dye on PMMA polymer.

Project description:The effects of argon gas glow discharge plasma on the surface of DR1 dye-loaded PMMA polymer films are examined in this work. Plasma immobilizes the dye on the surface of polymer without using stabilizers. Argon plasma activates the surface through breaking some bonds and generation of radical sites. It affects the acrylate groups of PMMA leading to covalent bonds between dye and surface of polymer. In addition, plasma treatment and contact with ambient air may result in the creation of new polar components, such as carbonyl and carboxyl compounds and links that enhance the dye attachment to the polymer matrix. Besides, the dye adsorption on the polymer film is impacted by changes in surface topography. Furthermore, plasma modifies the dye conformation, which affects the adherence of the dye to the polymer surface through bringing the dye to the higher energy state. The chemical and topographical modification of dye-loaded PMMA films by plasma are investigated by spectroscopic and AFM methods. Furthermore, aging process was used to confirm dye retention on the polymer film after plasma modification as opposed to dye-loaded polymer film that was left untreated as a reference sample. Finally, investigated method suggests a novel and very affordable technique for fabrication of poly(MMA-co-DR1) copolymer in the form of a homogeneous surface layer.

Light-induced spiral mass transport in azo-polymer films under vortex-beam illumination.

Project description:When an azobenzene-containing polymer film is exposed to non-uniform illumination, a light-induced mass migration process may be induced, leading to the formation of relief patterns on the polymer-free surface. Despite many years of research effort, several aspects of this phenomenon remain poorly understood. Here we report the appearance of spiral-shaped relief patterns on the polymer film under the illumination of focused Laguerre-Gauss beams with helical wavefronts and an optical vortex at their axis. The induced spiral reliefs are sensitive to the vortex topological charge and to the wavefront handedness. These findings are unexpected because the doughnut-shaped intensity profile of Laguerre-Gauss beams contains no information about the wavefront handedness. We propose a model that explains the main features of this phenomenon through the surface-mediated interference of the longitudinal and transverse components of the optical field. These results may find applications in optical nanolithography and optical-field nanoimaging.

Effects of azo dye on simultaneous biological removal of azo dye and nutrients in wastewater.

Project description:The potential disrupting effects of Azo dye on wastewater nutrients removal deserved more analysis. In this study, 15 days exposure experiments were conducted with alizarin yellow R (AYR) as a model dye to determine whether the dye caused adverse effects on biological removal of both the dye and nutrients in acclimated anaerobic-aerobic-anoxic sequencing batch reactors. The results showed that the AYR removal efficiency was, respectively, 85.7% and 66.8% at AYR concentrations of 50 and 200 mg l-1, while higher AYR inlet (400 mg l-1) might inactivate sludge. Lower removal of AYR at 200 mg l-1 of AYR was due to the insufficient support of electron donors in the anaerobic process. However, the decolorized by-products p-phenylenediamine and 5-aminosalicylic were completely decomposed in the following aerobic stage at both 50 and 200 mg l-1 of AYR concentrations. Compared with the absence of AYR, the presence of 200 mg l-1 of AYR decreased the total nitrogen removal efficiency from 82.4 to 41.1%, and chemical oxygen demand (COD) removal efficiency initially decreased to 68.1% and then returned to around 83.4% in the long-term exposure time. It was also found that the inhibition of AYR, nitrogen and COD removal induced by a higher concentration of AYR was due to the increased intracellular reactive oxygen species production, which caused the rise of oxidation-reduction potential value and decreased ammonia monooxygenase and nitrite oxidoreductase activities.

Chitosan-azo dye bioplastics that are reversibly resoluble and recoverable under visible light irradiation.

Project description:Biopolymer composite materials were prepared by combining bio-sourced cationic water-soluble chitosan with bi-functional water-soluble anionic azo food dyes amaranth (AMA) or allura red (ALR) as ionic cross-linkers, mixing well in water, and then slow-drying in air. The electrostatically-assembled ionically-paired films showed good long-term stability to dissolution, with no re-solubility in water, and competitive mechanical properties as plastic materials. However, upon exposure of the bioplastics to low power light at sunlight wavelengths and intensities stirring in water, the stable materials photo-disassembled back to their water-soluble and low-toxicity (edible) constituent components, via structural photo-isomerization of the azo ionic crosslinkers. XRD, UV-vis, and IR spectroscopy confirmed that these assemblies are reversibly recoverable and so can in principle represent fully recyclable, environmentally degradable materials triggered by exposure to sunlight and water after use, with full recovery of starting components ready for re-use. A density functional theory treatment of the amaranth azo dye identified a tautomeric equilibrium favouring the hydrazone form and rationalized geometrical isomerization as a mechanism for photo-disassembly. The proof-of-principle suitability of films of these biomaterial composites as food industry packaging was assessed via measurement of mechanical, water and vapour barrier properties, and stability to solvent tests. Tensile strength of the composite materials was found to be 25-30 MPa, with elongation at break 3-5%, in a range acceptable as competitive for some applications to replace oil-based permanently insoluble non-recyclable artificial plastics, as fully recyclable, recoverable, and reusable low-toxicity green biomaterials in natural environmental conditions.

Solvent- and Light-Sensitive AIEE-Active Azo Dye: From Spherical to 1D and 2D Assemblies.

Project description:Fluorescent molecular assembly systems provide an exciting platform for creating stimuli-responsive nano- and microstructured materials with optical, electronic, and sensing functions. To understand the relationship between (i) the plausible molecular structures preferentially adopted depending on the solvent polarity (such as N,N-dimethylformamide [DMF], tetrahydrofuran [THF], and toluene), (ii) the resulting spectroscopic features, and (iii) self-assembled nano-, micro-, and macrostructures, we chose a sterically crowded triangular azo dye (3Bu) composed of a polar molecular core and three peripheral biphenyl wings. The chromophore changed the solution color from yellow to pink-red depending on the solvent polarity. In a yellow DMF solution, a considerable amount of the twisted azo form could be kept stable with the help of favorable intermolecular interactions with the solvent molecules. By varying the concentration of the DMF solution, the morphology of self-assembled structures was transformed from nanoparticles to micrometer-sized one-dimensional (1D) structures such as sticks and fibers. In a pink-red toluene solution, the periphery of the central ring became more planar. The resulting significant amount of the keto-hydrazone tautomer grew into micro- and millimeter-sized 1D structures. Interestingly, when THF-H2O (1:1) mixtures were stored at a low temperature, elongated fibers were stacked sideways and eventually developed into anisotropic two-dimensional (2D) sheets. Notably, subsequent exposure of visible-light-irradiated sphere samples to solvent vapor resulted in reversible fluorescence off↔on switching accompanied by morphological restoration. These findings suggest that rational selection of organic dyes, solvents, and light is important for developing reusable fluorescent materials.

Sulfoxide hemithioindigo tweezers - visible light addressable capture and release.

Project description:Introducing responsive elements into supramolecular recognition systems offers great advantages for the control of intermolecular interactions and represents an important stepping stone towards multi-purpose and reprogrammable synthetic systems. Of particular interest is implementation of light-responsiveness because of the unique ease and precision of this signal. Here we present visible light responsive hemithioindigo-based molecular tweezers that bear a highly polar sulfoxide function as an additional recognition unit inside their binding site. Sulfur oxidation allows to simultaneously enhance all crucial properties of this receptor type i.e. photoswitching capability, thermal stability of individual switching states, binding affinity, and binding modulation upon switching. With a novel titration method the thermodynamic binding parameters were determined using reduced sample amounts. Employing these strongly enhanced molecular tweezers allowed to demonstrate photocontrol of intermolecular charge transfer in a reversible manner.

Azo-Dye-Functionalized Polycarbonate Membranes for Textile Dye and Nitrate Ion Removal.

Project description:Challenges exist in the wastewater treatment of dyes produced by the world's growing textiles industry. Common problems facing traditional wastewater treatments include low retention values and breaking the chemical bonds of some dye molecules, which in some cases can release byproducts that can be more harmful than the original dye. This research illustrates that track-etched polycarbonate filtration membranes with 100-nanometer diameter holes can be functionalized with azo dye direct red 80 at 1000 µM, creating a filter that can then be used to remove the entire negatively charged azo dye molecule for a 50 µM solution of the same dye, with a rejection value of 96.4 ± 1.4%, at a stable flow rate of 114 ± 5 µL/min post-functionalization. Post-functionalization, Na+ and NO3- ions had on average 17.9%, 26.0%, and 31.1% rejection for 750, 500, and 250 µM sodium nitrate solutions, respectively, at an average flow rate of 177 ± 5 µL/min. Post-functionalization, similar 50 µM azo dyes had increases in rejection from 26.3% to 53.2%. Rejection measurements were made using ultraviolet visible-light spectroscopy for dyes, and concentration meters using ion selective electrodes for Na+ and NO3- ions.

Photocage-Selective Capture and Light-Controlled Release of Target Proteins.

Project description:Photochemical transformations enable exquisite spatiotemporal control over biochemical processes; however, methods for reliable manipulations of biomolecules tagged with biocompatible photo-sensitive reporters are lacking. Here we created a high-affinity binder specific to a photolytically removable caging group. We utilized chemical modification or genetically encoded incorporation of noncanonical amino acids to produce proteins with photocaged cysteine or selenocysteine residues, which were used for raising a high-affinity monoclonal antibody against a small photoremovable tag, 4,5-dimethoxy-2-nitrobenzyl (DMNB) group. Employing the produced photocage-selective binder, we demonstrate selective detection and immunoprecipitation of a variety of DMNB-caged target proteins in complex biological mixtures. This combined orthogonal strategy permits photocage-selective capture and light-controlled traceless release of target proteins for a myriad of applications in nanoscale assays.
