Versatile Surface Modification of Hydrogels by Surface-Initiated, Cu0-Mediated Controlled Radical Polymerization.

ABSTRACT: Surface-initiated controlled radical polymerization mediated by a Cu0 plate (SI-Cu0 CRP) emerges as a versatile and efficient method for the functionalization of the exposed surfaces of hydrogels with a wide variety of polymer brushes. When a Cu0 plate is placed in contact with initiator-bearing hydrogel surfaces in the presence of ligand and monomer and under ambient conditions, it rapidly consumes dissolved oxygen from the reaction mixture, further acting as a source of catalyst and leading to the rapid growth of hydrogel-bound polymer chains. Three types of functional surfaces have been prepared as examples of the wide range of potential materials that can be synthesized in this way, including a hydrogel with a protective, hydrophobic surface, a lubricious hydrogel, as well as a hydrogel with thermally switchable frictional properties.

Modular and Versatile Spatial Functionalization of Tissue Engineering Scaffolds through Fiber-Initiated Controlled Radical Polymerization.

Project description:Native tissues are typically heterogeneous and hierarchically organized, and generating scaffolds that can mimic these properties is critical for tissue engineering applications. By uniquely combining controlled radical polymerization (CRP), end-functionalization of polymers, and advanced electrospinning techniques, a modular and versatile approach is introduced to generate scaffolds with spatially organized functionality. Poly-ε-caprolactone is end functionalized with either a polymerization-initiating group or a cell-binding peptide motif cyclic Arg-Gly-Asp-Ser (cRGDS), and are each sequentially electrospun to produce zonally discrete bilayers within a continuous fiber scaffold. The polymerization-initiating group is then used to graft an antifouling polymer bottlebrush based on poly(ethylene glycol) from the fiber surface using CRP exclusively within one bilayer of the scaffold. The ability to include additional multifunctionality during CRP is showcased by integrating a biotinylated monomer unit into the polymerization step allowing postmodification of the scaffold with streptavidin-coupled moieties. These combined processing techniques result in an effective bilayered and dual-functionality scaffold with a cell-adhesive surface and an opposing antifouling non-cell-adhesive surface in zonally specific regions across the thickness of the scaffold, demonstrated through fluorescent labelling and cell adhesion studies. This modular and versatile approach combines strategies to produce scaffolds with tailorable properties for many applications in tissue engineering and regenerative medicine.

A well-defined, versatile photoinitiator (salen)Co-CO2CH3 for visible light-initiated living/controlled radical polymerization.

Project description:The control of the polymerization of a wide range of monomers under mild conditions by a single catalyst remains a major challenge in polymer science. We report a versatile, well-defined organocobalt salen complex to control living radical polymerization of different categories of monomers, including acrylates, acrylamides and vinyl acetate, under visible light irradiation at ambient temperature. Both household light and sunlight were effectively applied in the synthesis of polymers with controlled molecular weights and narrow polydispersities. Narrowly dispersed block copolymers (Mw/Mn < 1.2) were obtained under various conditions. The structures of the polymers were analyzed by 1H NMR, 2D NMR, 13C NMR, GPC, MALDI-TOF-MS and isotopic labeling experiments, which showed that the ? and ? ends of the polymer chains were capped with (salen)Co and -CO2CH3 segments, respectively, from the photoinitiator (salen)Co-CO2CH3. The ? end was easily functionalized through oxygen insertion followed by hydrolysis from 18O2 to -18OH. This robust system can proceed without any additives, and offers a versatile and green way to produce well-defined homo and block copolymers.

Protein-polymer bioconjugates via a versatile oxygen tolerant photoinduced controlled radical polymerization approach.

Project description:The immense application potential of amphiphilic protein-polymer conjugates remains largely unexplored, as established "grafting from" synthetic protocols involve time-consuming, harsh and disruptive deoxygenation methods, while "grafting to" approaches result in low yields. Here we report an oxygen tolerant, photoinduced CRP approach which readily affords quantitative yields of protein-polymer conjugates within 2 h, avoiding damage to the secondary structure of the protein and providing easily accessible means to produce biomacromolecular assemblies. Importantly, our methodology is compatible with multiple proteins (e.g. BSA, HSA, GOx, beta-galactosidase) and monomer classes including acrylates, methacrylates, styrenics and acrylamides. The polymerizations are conveniently conducted in plastic syringes and in the absence of any additives or external deoxygenation procedures using low-organic content media and ppm levels of copper. The robustness of the protocol is further exemplified by its implementation under UV, blue light or even sunlight irradiation as well as in buffer, nanopure, tap or even sea water.

Visible Light-Induced Metal Free Surface Initiated Atom Transfer Radical Polymerization of Methyl Methacrylate on SBA-15.

Project description:Surface-initiated atom transfer radical polymerization (SI-ATRP) is one of the most versatile techniques to modify the surface properties of materials. Recent developed metal-free SI-ATRP makes such techniques more widely applicable. Herein photo-induced metal-free SI-ATRP of methacrylates, such as methyl methacrylate, N-isopropanyl acrylamide, and N,N-dimethylaminoethyl methacrylate, on the surface of SBA-15 was reported to fabricate organic-inorganic hybrid materials. A SBA-15-based polymeric composite with an adjustable graft ratio was obtained. The structure evolution during the SI-ATRP modification of SBA-15 was monitored and verified by FT-IR, XPS, TGA, BET, and TEM. The obtained polymeric composite showed enhanced adsorption ability for the model compound toluene in aqueous conditions. This procedure provides a low-cost, readily available, and easy modification method to synthesize polymeric composites without the contamination of metal.

Self-Cleaning Cotton Obtained after Grafting Thermoresponsive Poly(N-vinylcaprolactam) through Surface-Initiated Atom Transfer Radical Polymerization.

Project description:Although the performance of smart textiles would be enhanced if they could display self-cleaning ability toward various kinds of contamination, the procedures that have been used previously to impart the self-cleaning potential to these functional fabrics (solvent casting, dip coating, spin coating, surface crosslinking) have typically been expensive and/or limited by uncontrollable polymer thicknesses and morphologies. In this paper, we demonstrate the use of atomic transfer radical polymerization for the surface-initiated grafting of poly(N-vinylcaprolactam), a thermoresponsive polymer, onto cotton. We confirmed the thermoresponsiveness and reusability of the resulting fabric through water contact angle measurements and various surface characterization techniques (scanning electron microscopy, atomic force microscopy, Fourier transform infrared spectroscopy). Finally, we validated the self-cleaning performance of the fabric by washing away an immobilized fluorescent protein in deionized water under thermal stimulus. Fluorescence micrographs revealed that, after the fifth wash cycle, the fabric surface had undergone efficient self-cleaning of the stain, making it an effective self-cleaning material. This approach appears to have potential for application in the fields of smart textiles, responsive substrates, and functional fabrics.

Surface initiated atom transfer radical polymerization grafting of sodium styrene sulfonate from titanium and silicon substrates.

Project description:This study investigates the grafting of poly-sodium styrene sulfonate (pNaSS) from trichlorosilane/10-undecen-1-yl 2-bromo-2-methylpropionate functionalized Si and Ti substrates by atom transfer radical polymerization (ATRP). The composition, molecular structure, thickness, and topography of the grafted pNaSS films were characterized with x-ray photoelectron spectroscopy (XPS), time-of-flight secondary ion mass spectrometry (ToF-SIMS), variable angle spectroscopic ellipsometry (VASE), and atomic force microscopy (AFM), respectively. XPS and ToF-SIMS results were consistent with the successful grafting of a thick and uniform pNaSS film on both substrates. VASE and AFM scratch tests showed the films were between 25 and 49 nm thick on Si, and between 13 and 35 nm thick on Ti. AFM determined root-mean-square roughness values were ∼2 nm on both Si and Ti substrates. Therefore, ATRP grafting is capable of producing relatively smooth, thick, and chemically homogeneous pNaSS films on Si and Ti substrates. These films will be used in subsequent studies to test the hypothesis that pNaSS-grafted Ti implants preferentially adsorb certain plasma proteins in an orientation and conformation that modulates the foreign body response and promotes formation of new bone.

Near-infrared light-responsive hydrogels via peroxide-decorated MXene-initiated polymerization.

Project description:Two-dimensional MXene Ti3C2T x nanosheets with peroxide decoration (p-Ti3C2T x ) are synthesized by a sonication-assisted MILD etching method. The obtained MXenes can generate hydroxyl radical species and act as an initiator for free-radical polymerization of a series of acrylic monomers without the use of light illumination or co-initiators. The monomers analyzed include acrylamide, N-isopropylacrylamide (NIPAM), N,N-dimethylacrylamide, methyl methacrylate, and hydroxyethyl methacrylate. By simply mixing N-isopropylacrylamide monomers and p-Ti3C2T x nanosheets under deoxygenated conditions, PNIPAM-based nanocomposite hydrogels are synthesized using a high concentration of the monomer. The nanocomposite hydrogels have a photothermal conversion efficiency of 34.7% and photothermal stability superior to that of pristine Ti3C2T x . Taking advantage of the thermal responsive behavior of PNIPAM, the nanocomposite hydrogels are successfully exploited as remotely near-infrared light controlled "smart" windows, fluidic valves and photodetectors.

Synthesizing a Hybrid Nanocomposite as an Affinity Adsorbent through Surface-Initiated Atom Transfer Radical Polymerization Catalyzed by Myoglobin.

Project description:A hybrid bifunctional core-shell nanostructure was synthesized for the first time via surface-initiated atom transfer radical polymerization (SI-ATRP) using myoglobin as a biocatalyst (ATRPase) in an aqueous solution. N-Isopropyl acrylamide (NIPA) and N-(3-aminopropyl)methacrylamide (APMA) were applied to graft flexible polymer brushes onto initiator-functionalized silica nanoparticles. Two different approaches were implemented to form the core-shell nanocomposite: (a) random copolymerization, Si@p(NIPA-co-APMA) and (b) sequential block copolymerization, Si@pNIPA-b-pAPMA. These nanocomposites can be used as versatile intermediates, thereby leading to different types of materials for targeted applications. In this work, a phenylboronic acid ligand was immobilized on the side chain of the grafted brushes during a series of postmodification reactions to create a boronate affinity adsorbent. The ability to selectively bind glycoproteins (ovalbumin and glycated hemoglobin) via boronic acid was assessed at two different temperatures (20 and 40 °C), where Si@pNIPA-b-APMABA (163 mg OVA/g of particle) displayed an approximately 1.5-fold higher capacity than Si@p(NIPA-co-APMA)BA (107 mg OVA/g of particle). In addition to selective binding to glycoproteins, the nanocomposites exhibited selective binding for myoglobin due to the molecular imprinting effect during the postmodification process, that is, 72 and 111 mg Mb/g for Si@p(NIPA-co-APMA)BA and Si@pNIPA-b-pAPMABA, respectively.

A generalizable method for the construction of MOF@polymer functional composites through surface-initiated atom transfer radical polymerization.

Project description:We report a generalizable approach to construct MOF@polymer functional composites through surface-initiated atom transfer radical polymerization (SI-ATRP). Unlike conventional SI-ATRP that requires covalent pre-anchoring of the initiating group on substrate surfaces, in our approach, a rationally designed random copolymer (RCP) macroinitiator first self-assembles on MOF surfaces through inter-chain hydrogen bond crosslinking. Subsequent polymerization in the presence of a crosslinking monomer covalently threads these polymer chains into a robust network, physically confining the MOF particle inside the polymer shell. We demonstrated the universality of this approach by growing various polymers on five MOFs of different metals (Zr, Zn, Co, Al, and Cr) with complete control over shell thickness, functionality and layer sequence while still retaining the inherent porosity of the MOFs. Moreover, the wettability of UiO-66 can be continuously tuned from superhydrophilic to superhydrophobic simply through judicious monomer(s) selection. We also demonstrated that a 7 nm polystyrene shell can effectively shield UiO-66 particles against 1 M H2SO4 and 1 M NaOH at elevated temperature, enabling their potential application in demanding chemical environments.
