Prerequisites for an Artificial Self.

ABSTRACT: Traditionally investigated in philosophy, body ownership and agency-two main components of the minimal self-have recently gained attention from other disciplines, such as brain, cognitive and behavioral sciences, and even robotics and artificial intelligence. In robotics, intuitive human interaction in natural and dynamic environments becomes more and more important, and requires skills such as self-other distinction and an understanding of agency effects. In a previous review article, we investigated studies on mechanisms for the development of motor and cognitive skills in robots (Schillaci et al., 2016). In this review article, we argue that these mechanisms also build the foundation for an understanding of an artificial self. In particular, we look at developmental processes of the minimal self in biological systems, transfer principles of those to the development of an artificial self, and suggest metrics for agency and body ownership in an artificial self.

Self-assembled artificial cilia.

Project description:Due to their small dimensions, microfluidic devices operate in the low Reynolds number regime. In this case, the hydrodynamics is governed by the viscosity rather than inertia and special elements have to be introduced into the system for mixing and pumping of fluids. Here we report on the realization of an effective pumping device that mimics a ciliated surface and imitates its motion to generate fluid flow. The artificial biomimetic cilia are constructed as long chains of spherical superparamagnetic particles, which self-assemble in an external magnetic field. Magnetic field is also used to actuate the cilia in a simple nonreciprocal manner, resulting in a fluid flow. We prove the concept by measuring the velocity of a cilia-pumped fluid as a function of height above the ciliated surface and investigate the influence of the beating asymmetry on the pumping performance. A numerical simulation was carried out that successfully reproduced the experimentally obtained data.

Cultural prerequisites of socioeconomic development.

Project description:In the centuries since the enlightenment, the world has seen an increase in socioeconomic development, measured as increased life expectancy, education, economic development and democracy. While the co-occurrence of these features among nations is well documented, little is known about their origins or co-evolution. Here, we compare this growth of prosperity in nations to the historical record of cultural values in the twentieth century, derived from global survey data. We find that two cultural factors, secular-rationality and cosmopolitanism, predict future increases in GDP per capita, democratization and secondary education enrollment. The converse is not true, however, which indicates that secular-rationality and cosmopolitanism are among the preconditions for socioeconomic development to emerge.

DNA Self-assembly Catalyzed by Artificial Agents.

Project description:Nucleic acids have been shown to be versatile molecules and engineered to produce various nanostructures. However, the poor rate of these uncatalyzed nucleic acid reactions has restricted the development and applications. Herein, we reported a novel finding that DNA self-assembly could be nonenzymatically catalyzed by artificial agents with an increasing dissociation rate constant K2. The catalytic role of several artificial agents in DNA self-assembly was verified by real-time fluorescent detection or agarose gel electrophoresis. We found that 20% PEG 200 could significantly catalyze DNA self-assembly and increase the reaction efficiency, such as linear hybridization chain reaction (HCR) and exponential hairpin assembly (EHA). Therefore, we foresee that a fast and efficient DNA self-assembly in structural DNA nanotechnology will be desirable.

Stretchable and self-healable hydrogel artificial skin.

Project description:Hydrogels have emerged as promising materials for the construction of skin-like mechanical sensors. The common design of hydrogel-based artificial skin requires a dielectric sandwiched between two hydrogel layers for capacitive sensing. However, such a planar configuration limits the sensitivity, stretchability and self-healing properties. Here, we report the design of single-layer composite hydrogels with bulk capacitive junctions as mechanical sensors. We engineer dielectric peptide-coated graphene (PCG) to serve as homogenously dispersed electric double layers in hydrogels. Any mechanical motions that alter the microscopic distributions of PCG in the hydrogels can significantly change the overall capacitance. We use peptide self-assembly to render strong yet dynamic interfacial interactions between the hydrogel network and graphene. The resulting hydrogels can be stretched up to 77 times their original length and self-heal in a few minutes. The devices can effectively sense strain and pressure in both air and aqueous environments, providing tremendous opportunities for next-generation iontronics.

Decent Living Standards: Material Prerequisites for Human Wellbeing.

Project description:We define a set of universal, irreducible and essential set of material conditions for achieving basic human wellbeing, along with indicators and quantitative thresholds, which can be operationalized for societies based on local customs and preferences. We draw support for this decent living standard (DLS) from different accounts of basic justice, including the capability approach and basic needs. The DLS goes beyond existing multidimensional poverty indicators by comprehensively addressing living conditions and the means of social participation. The DLS offers a normative basis to develop minimum wage and reference budgets, and to assess the environmental impacts, such as climate change, of eradicating poverty.

DNA cross-triggered cascading self-amplification artificial biochemical circuit.

Project description:The construction of compact and robust artificial biochemical circuits based on nucleic acids can help researchers to understand the essential mechanisms of complex biological systems, and design sophisticated strategies for various requirements. In this study, a novel DNA cross-triggered cascading self-amplification artificial biochemical circuit was developed. Once triggered by trace amounts (as low as 2 amol) of either of two fully independent oligonucleotide factors under homogeneous isothermal conditions, the circuit simultaneously amplified both factors by 105-107 fold, which was proved using mass spectrometry. The compact and robust circuit was successfully used to construct a multi-input Boolean logic operation and a sensitive DNA biosensor based on the dual-amplification of both the target and reporter. The circuit showed great potential for signal gain in complicated molecular programming, and flexible control of nucleic acid nanomachines in biochemical network systems and nanotechnology.

Structural prerequisites for G-protein activation by the neurotensin receptor.

Project description:We previously determined the structure of neurotensin receptor NTSR1 in an active-like conformation with six thermostabilizing mutations bound to the peptide agonist neurotensin. This receptor was unable to activate G proteins, indicating that the mutations restricted NTSR1 to relate agonist binding to G-protein activation. Here we analyse the effect of three of those mutations (E166A(3.49), L310A(6.37), F358A(7.42)) and present two structures of NTSR1 able to catalyse nucleotide exchange at Gα. The presence of F358(7.42) causes the conserved W321(6.48) to adopt a side chain orientation parallel to the lipid bilayer sealing the collapsed Na(+) ion pocket and linking the agonist with residues in the lower receptor part implicated in GPCR activation. In the intracellular receptor half, the bulkier L310(6.37) side chain dictates the position of R167(3.50) of the highly conserved D/ERY motif. These residues, together with the presence of E166(3.49) provide determinants for G-protein activation by NTSR1.

People's self-reported encounters of Perceiving Mind in Artificial Intelligence.

Project description:This article presents the data from two surveys that asked about everyday encounters with artificial intelligence (AI) systems that are perceived to have attributes of mind. In response to specific attribute prompts about an AI, the participants qualitatively described a personally-known encounter with an AI. In survey 1 the prompts asked about an AI planning, having memory, controlling resources, or doing something surprising. In survey 2 the prompts asked about an AI experiencing emotion, expressing desires or beliefs, having human-like physical features, or being mistaken for a human. The original responses were culled based on the ratings of multiple coders to eliminate responses that did not adhere to the prompts. This article includes the qualitative responses, coded categories of those qualitative responses, quantitative measures of mind perception and demographics. Artificial Self-assembling Nanocompartment for Organizing Metabolic Pathways in Yeast.

Project description:Metabolic pathways are commonly organized by sequestration into discrete cellular compartments. Compartments prevent unfavorable interactions with other pathways and provide local environments conducive to the activity of encapsulated enzymes. Such compartments are also useful synthetic biology tools for examining enzyme/pathway behavior and for metabolic engineering. Here, we expand the intracellular compartmentalization toolbox for budding yeast (Saccharomyces cerevisiae) with Murine polyomavirus virus-like particles (MPyV VLPs). The MPyV system has two components: VP1 which self-assembles into the compartment shell and a short anchor, VP2C, which mediates cargo protein encapsulation via binding to the inner surface of the VP1 shell. Destabilized green fluorescent protein (GFP) fused to VP2C was specifically sorted into VLPs and thereby protected from host-mediated degradation. An engineered VP1 variant displayed improved cargo capture properties and differential subcellular localization compared to wild-type VP1. To demonstrate their ability to function as a metabolic compartment, MPyV VLPs were used to encapsulate myo-inositol oxygenase (MIOX), an unstable and rate-limiting enzyme in d-glucaric acid biosynthesis. Strains with encapsulated MIOX produced ∼20% more d-glucaric acid compared to controls expressing "free" MIOX─despite accumulating dramatically less expressed protein─and also grew to higher cell densities. This is the first demonstration in yeast of an artificial biocatalytic compartment that can participate in a metabolic pathway and establishes the MPyV platform as a promising synthetic biology tool for yeast engineering.

Confidence and self-attribution bias in an artificial stock market.

Project description:Using an agent-based model we examine the dynamics of stock price fluctuations and their rates of return in an artificial financial market composed of fundamentalist and chartist agents with and without confidence. We find that chartist agents who are confident generate higher price and rate of return volatilities than those who are not. We also find that kurtosis and skewness are lower in our simulation study of agents who are not confident. We show that the stock price and confidence index-both generated by our model-are cointegrated and that stock price affects confidence index but confidence index does not affect stock price. We next compare the results of our model with the S&P 500 index and its respective stock market confidence index using cointegration and Granger tests. As in our model, we find that stock prices drive their respective confidence indices, but that the opposite relationship, i.e., the assumption that confidence indices drive stock prices, is not significant.
