Project description:Insects are well known for their ability to keep track of their heading direction based on a combination of skylight cues and visual landmarks. This allows them to navigate back to their nest, disperse throughout unfamiliar environments, as well as migrate over large distances between their breeding and non-breeding habitats. The monarch butterfly (Danaus plexippus), for instance, is known for its annual southward migration from North America to certain trees in Central Mexico. To maintain a constant flight route, these butterflies use a time-compensated sun compass, which is processed in a region in the brain, termed the central complex. However, to successfully complete their journey, the butterflies' brain must generate a multitude of orientation strategies, allowing them to dynamically switch from sun-compass orientation to a tactic behavior toward a certain target. To study whether monarch butterflies exhibit different orientation modes and if they can switch between them, we observed the orientation behavior of tethered flying butterflies in a flight simulator while presenting different visual cues to them. We found that the butterflies' behavior depended on the presented visual stimulus. Thus, while a dark stripe was used for flight stabilization, a bright stripe was fixated by the butterflies in their frontal visual field. If we replaced a bright stripe with a simulated sun stimulus, the butterflies switched their behavior and exhibited compass orientation. Taken together, our data show that monarch butterflies rely on and switch between different orientation modes, allowing the animal to adjust orientation to its actual behavioral demands.

Project description:This study investigated the colourful secondary particles formed by controlling the aggregation states of colloidal silica particles and the enhancement of the structural colouration of the secondary particles caused by adding black particles. We obtained glossy, partially structurally coloured secondary particles in the absence of NaCl, but matte, whitish secondary particles were obtained in the presence of NaCl. When a small amount of carbon black was incorporated into both types of secondary particles, the incoherent multiple scattering of light from the amorphous region was considerably reduced. However, the peak intensities in the reflection spectra, caused by Bragg reflection and by coherent single wavelength scattering, were only slightly decreased. Consequently, a brighter structural colour of these secondary particles was observed with the naked eye. Furthermore, when magnetite was added as a black particle, the coloured secondary particles could be moved and collected by applying an external magnetic field.

Project description:The current study seeks confirmation for the hypothesis that 2D:4D (positively) predicts prosociality when people are more likely to rely on intuition than deliberation. We assess intuition and deliberation using the Cognitive Reflection Test (CRT) and measure prosociality via the validated Social Value Orientation (SVO) slider measure. Although our results do not provide collective evidence for our main proposition, we observe in the data that for low (right) 2D:4D men, the more intuitive they are, the less prosocial they become, whereas for high (right) 2D:4D men the thinking style does not affect their prosociality. Importantly, we find that two alternative measures of cognitive reflection, CRT and CRT-2, differently relate to prosocial decision making such that only CRT-2 (but not the classic CRT) positively predicts prosociality. Given that previous research on the role of cognitive reflection and 2D:4D in prosocial decision making provided inconsistent results, the present study findings are highly valuable to get a better understanding in this domain of study. Furthermore, some of our findings invite further confirmatory tests, thereby opening up multiple avenues for further research.

Project description:Chlorophylless flower petals are known to be composed of non-photosynthetic tissues. Here, we show that the light energy storage that can be photoacoustically measured in flower petals of Petunia hybrida is approximately 10-12%. We found that the supposed chlorophylless photosynthesis is an anoxygenic, anthocyanin-dependent process occurring in blue flower petals (ADAPFP), accompanied by non-respiratory light-dependent oxygen uptake and a 1.5-fold photoinduced increase in ATP levels. Using a simple, adhesive tape stripping technique, we have obtained a backside image of an intact flower petal epidermis, revealing sword-shaped ingrowths connecting the cell wall and vacuole, which is of interest for the further study of possible vacuole-related photosynthesis. Approaches to the interpretations of ADAPFP are discussed, and we conclude that these results are not impossible in terms of the known photochemistry of anthocyanins.

Project description:Every autumn, monarch butterflies migrate from North America to their overwintering sites in Central Mexico. To maintain their southward direction, these butterflies rely on celestial cues as orientation references. The position of the sun combined with additional skylight cues are integrated in the central complex, a region in the butterfly's brain that acts as an internal compass. However, the central complex does not solely guide the butterflies on their migration but also helps monarchs in their non-migratory form manoeuvre on foraging trips through their habitat. By comparing the activity of input neurons of the central complex between migratory and non-migratory butterflies, we investigated how a different lifestyle affects the coding of orientation information in the brain. During recording, we presented the animals with different simulated celestial cues and found that the encoding of the sun was narrower in migratory compared to non-migratory butterflies. This feature might reflect the need of the migratory monarchs to rely on a precise sun compass to keep their direction during their journey. Taken together, our study sheds light on the neural coding of celestial cues and provides insights into how a compass is adapted in migratory animals to successfully steer them to their destination.

Project description:During their fall migration, Eastern North American monarch butterflies (Danaus plexippus) use a time-compensated Sun compass to aid navigation to their overwintering grounds in central Mexico. It has been assumed that the circadian clock that provides time compensation resides in the brain, although this assumption has never been examined directly. Here, we show that the antennae are necessary for proper time-compensated Sun compass orientation in migratory monarch butterflies, that antennal clocks exist in monarchs, and that they likely provide the primary timing mechanism for Sun compass orientation. These unexpected findings pose a novel function for the antennae and open a new line of investigation into clock-compass connections that may extend widely to other insects that use this orientation mechanism.

Project description:A newly developed flight simulator allows monarch butterflies to fly actively for up to several hours in any horizontal direction while their fall migratory flight direction can be continuously recorded. From these data, long segments of virtual flight paths of tethered, flying, migratory monarch butterflies were reconstructed, and by advancing or retarding the butterflies' circadian clocks, we have shown that they possess a time-compensated sun compass. Control monarchs on local time fly approximately southwest, those 6-h time-advanced fly southeast, and 6-h time-delayed butterflies fly in northwesterly directions. Moreover, butterflies flown in the same apparatus under simulated overcast in natural magnetic fields were randomly oriented and did not change direction when magnetic fields were rotated. Therefore, these experiments do not provide any evidence that monarch butterflies use a magnetic compass during migration.

Project description:A new approach is presented for measuring the three-dimensional orientation of individual macromolecules using single molecule fluorescence polarization (SMFP) microscopy. The technique uses the unique polarizations of evanescent waves generated by total internal reflection to excite the dipole moment of individual fluorophores. To evaluate the new SMFP technique, single molecule orientation measurements from sparsely labeled F-actin are compared to ensemble-averaged orientation data from similarly prepared densely labeled F-actin. Standard deviations of the SMFP measurements taken at 40 ms time intervals indicate that the uncertainty for individual measurements of axial and azimuthal angles is approximately 10 degrees at 40 ms time resolution. Comparison with ensemble data shows there are no substantial systematic errors associated with the single molecule measurements. In addition to evaluating the technique, the data also provide a new measurement of the torsional rigidity of F-actin. These measurements support the smaller of two values of the torsional rigidity of F-actin previously reported.

Project description:Eastern North American migratory monarch butterflies (Danaus plexippus) have faced sharp declines over the past two decades. Captive rearing of monarch butterflies is a popular and widely used approach for both public education and conservation. However, recent evidence suggests that captive-reared monarchs may lose their capacity to orient southward during fall migration to their Mexican overwintering sites, raising questions about the value and ethics of this activity undertaken by tens of thousands of North American citizens, educators, volunteers and conservationists each year. We raised offspring of wild-caught monarchs on swamp milkweed (Asclepias incarnata) indoors at 29°C during the day and 23°C at night (~77% RH, 18L:6D), and after eclosion, individuals were either tested in a flight simulator or radio tracked in the wild using an array of automated telemetry towers. While 26% (10/39) of monarchs tested in the flight simulator showed a weakly concentrated southward orientation, 97% (28/29) of the radio-tracked individuals that could be reliably detected by automated towers flew in a south to southeast direction from the release site and were detected at distances of up to 200 km away. Our results suggest that, although captive rearing of monarch butterflies may cause temporary disorientation, proper orientation is likely established after exposure to natural skylight cues.

Project description:The molecular orientation of organic molecules of zinc phthalocyanine (ZnPc) in single-component films on copper iodide (CuI) substrates can be controlled to achieve a molecular orientation lying flat on the substrate (flat-on) owing to π-d orbital interactions between the ZnPc molecules and the CuI. A 3-fold enhancement in the performance of organic photovoltaic cells has been reported by introducing a CuI interlayer between a ZnPc:fullerene (C60) bulk heterojunction (BHJ) film and the substrate. However, the mechanism underpinning the resultant solar cell performance enhancement was unclear. Herein, we report on the results of using in situ reflection absorption spectroscopy measurements during the vacuum deposition of coevaporated ZnPc:C60 BHJ films on various substrates to investigate the ZnPc molecular orientation. Our results revealed that the flat-on molecular orientation of ZnPc molecules in ZnPc:C60 BHJ films on CuI interlayers and flat-on ZnPc substrates can be successfully identified via the strong π-π interactions between the BHJ film and the substrate. The π-π interactions between individual ZnPc molecules are stronger than the π-d interactions between ZnPc molecules and CuI in coevaporated ZnPc:C60 films, as is evident from the molecular orientation of ZnPc, as determined by in situ reflection absorption spectroscopy. Our findings demonstrate that precisely controlling the molecular orientations of the films could enhance organic photovoltaic (OPV) performance. The present work provides important insights that will enable the design of higher performance OPV cells.
