Air purifiers with high-efficiency particulate air (HEPA) filters remove not only particulate matter but also airborne microorganisms in indoor environments. We investigated the bacterial community in HEPA filters (used for 1 year) and that in the floor dust of 12 office rooms in Beijing. We found that the viable bacteria proportion in the filter was significantly higher than that in the floor dust (p < 0.001). Project description:The present study deals with an evaluation of the air purifier's effectiveness in reducing the concentration of different sized particulate matter (PM) and ions in the real-world indoor environment. Two types of air purifiers (API and APII) mainly equipped with High-Efficiency Particulate Air (HEPA) filters that differed in other specifications were employed in general indoor air and the presence of an external source (candles and incense). The gravimetric sampling of PM was carried out by SKC Cascade Impactor and further samples were analyzed for determining ions' concentration while real-time monitoring of different sized PM was done through Grimm Aerosol Spectrometer (1.109). The result showed that API reduced PM levels of different sizes ranged from 12-52% and 29-53% in general indoor air and presence of external source respectively. Concerning the APII, a higher decrease percent in PM level was explored in presence of an external source (52-68%) as compared to scenarios of general indoor air (37-64%). The concentrations of the ions were noticed to be decreased in all three size fractions but surprisingly some ions' (not specific) concentrations increased on the operation of both types of air purifiers. Overall, the study recommends the use of air purifiers with mechanical filters (HEPA) instead of those which release ions for air purification.

Project description:Our aim was to assess whether airborne particle counting is an immediate indicator of biocontamination in controlled environment rooms with HEPA filters in a hospital. A prospective study was carried out in a tertiary care hospital between 2016 and 2018. The study was divided in two periods and the measurements were performed in different controlled environment rooms with HEPA filters. The Environmental Biosafety Criterion (EBC) was defined as the absence of fungal and bacterial contamination. In the training period, the area under the ROC curve (aROC) of airborne particle counting and EBC was calculated for each particle size as well as the cut-off points that optimize the combination of sensitivity and specificity in the association between them. aROC is created by plotting sensitivity against 1-specificity. In the testing period, the cut-off points previously selected were validated. 328 measurements were carried out in the training period and 301 in the testing period. In the training period, an association was found between airborne particle counting and EBC. An aROC?=?0.760, 95% Confidence Interval (95% CI) 0.695-0.825 was observed for 0.3?µm particles; an aROC?=?0.797 (95% CI 0.734-0.860) for 0.5?µm particles; and an aROC?=?0.751 (95% CI 0.673-0.829) for 5?µm particles. The cut-off points that optimized the combination of sensitivity and specificity were 9.0?×?103 for 0.3?µm particles, 3.6?×?103 particles for 0.5?µm, and 3.2?×?102 particles for 5?µm. In the testing period, the previous cut-off points were validated. We conclude that airborne particle counting is a useful, immediate, and preliminary measure to identify the presence of biocontamination in controlled environment rooms with HEPA filters.

Project description:UNLABELLED:Air travel can rapidly transport infectious diseases globally. To facilitate the design of biosensors for infectious organisms in commercial aircraft, we characterized bacterial diversity in aircraft air. Samples from 61 aircraft high-efficiency particulate air (HEPA) filters were analyzed with a custom microarray of 16S rRNA gene sequences (PhyloChip), representing bacterial lineages. A total of 606 subfamilies from 41 phyla were detected. The most abundant bacterial subfamilies included bacteria associated with humans, especially skin, gastrointestinal and respiratory tracts, and with water and soil habitats. Operational taxonomic units that contain important human pathogens as well as their close, more benign relatives were detected. When compared to 43 samples of urban outdoor air, aircraft samples differed in composition, with higher relative abundance of Firmicutes and Gammaproteobacteria lineages in aircraft samples, and higher relative abundance of Actinobacteria and Betaproteobacteria lineages in outdoor air samples. In addition, aircraft and outdoor air samples differed in the incidence of taxa containing human pathogens. Overall, these results demonstrate that HEPA filter samples can be used to deeply characterize bacterial diversity in aircraft air and suggest that the presence of close relatives of certain pathogens must be taken into account in probe design for aircraft biosensors. PRACTICAL IMPLICATIONS:A biosensor that could be deployed in commercial aircraft would be required to function at an extremely low false alarm rate, making an understanding of microbial background important. This study reveals a diverse bacterial background present on aircraft, including bacteria closely related to pathogens of public health concern. Furthermore, this aircraft background is different from outdoor air, suggesting different probes may be needed to detect airborne contaminants to achieve minimal false alarm rates. This study also indicates that aircraft HEPA filters could be used with other molecular techniques to further characterize background bacteria and in investigations in the wake of a disease outbreak.

Project description:PurposeTo evaluate the effects of air purifiers on the concentrations of indoor air pollutants and on asthma control in children.Materials and methodsIn this randomized crossover trial, daily use of an air purifier filter was compared with a matched placebo with the filter off. Thirty elementary school students who had asthma were enrolled and randomly allocated to one of two groups. The primary endpoints were changes in indoor air quality, asthma severity, lung function, airway inflammatory, urine microbiome, and phthalate after the installation of air purifiers. PM2.5 and CO? were measured as indoor air pollutants. Asthma severity was assessed in terms of both symptom and medication scores acquired using a daily questionnaire. The higher the score, the better the symptom or the less frequent the use of medication. Peak expiratory flow rate and fractional exhaled nitric oxide were also measured.ResultsThe mean age of the enrolled patients was 9.2±1.98 years. The mean concentration of PM2.5 was 17.0 ?g/m³ in the filter-off condition, but significantly lower at 9.26 ?g/m³ in the filter-on condition. Medication scores were 6.9 for the filter-off and 7.12 for the filter-on conditions, reflecting a statistically significant decrease in the frequency of medications used during air purifier operation. Bacterial richness, as determined using the Chao 1 index, was markedly lower in the filter-on than the filter-off condition.ConclusionThis study suggests that air purifiers benefit medication burden in children with asthma by reducing PM2.5 levels.

Project description:BackgroundPolychlorinated biphenyls (PCBs) are persistent pollutants identified worldwide as human blood and breast milk contaminants. Because they bioaccumulate, consumption of meat, fish, and dairy products predicts human blood concentrations. PCBs were also used widely in building materials, including caulks and paints, but few studies have evaluated the contribution of these exposures to body burden.MethodsIn an earlier study, we detected PCBs in indoor air in 31% of 120 homes on Cape Cod, MA. Two of the homes had much higher concentrations than the rest, so we retested to verify the initial finding, evaluate blood PCB concentrations of residents, and identify the PCB source.ResultsAir and dust concentrations remained elevated over 5 years between initial and follow-up sampling. Blood serum concentrations of PCBs in residents of the homes were generally elevated above the 95th percentile of a representative sample of the US population. Serum concentrations in residents and air and dust concentrations were especially high in a home where a resident reported use of PCB-containing floor finish in the past, and where the floor of one room was sanded and refinished just prior to sample collection.ConclusionThis case-study suggests that PCB residues in homes may be more significant contributors to overall exposure than diet for some people, and that use of a commercially-available PCB-containing wood floor finish in residences during the 1950s and 1960s is an overlooked but potentially important source of current PCB exposure in the general population.

Project description:Viral aerosols can lead to respiratory viral infections with high infectivity. About 90% of people's time is spent in closed environments. A few studies have pointed out that the ventilation systems in air handling units (AHUs) that treat and transmit a new synthetic clean and conditioned environment can also spread and transport viral particles in buildings. The aim of this work is to study the characterization of adenovirus, a DNA non-enveloped respiratory virus, on the F7 fiberglass filter used in AHUs. In this study, an experimental setup simulating an AHU was used. The SYBR® QPCR, Electrical Low-Pressure Impactor (ELPI™) and Scanning Mobility Particle Sizer (SMPS™) were used to detect, measure and characterize the aerosolized adenovirus solution. The characterization results showed that the nebulized adenovirus could be aerosolized in different forms associated or not with cell debris and proteins. The quantification and level of infectivity of adenovirus demonstrated that viruses passed through filters and remained infectious up- and downstream of the system during the 25 min of aerosolization. This study showed that AHUs should be considered an indoor source of viral contamination.

Project description:Controlling bioaerosols has become more important with increasing participation in indoor activities. Treatments using natural-product nanomaterials are a promising technique because of their relatively low toxicity compared to inorganic nanomaterials such as silver nanoparticles or carbon nanotubes. In this study, antimicrobial filters were fabricated from natural Euscaphis japonica nanoparticles, which were produced by nebulizing E. japonica extract. The coated filters were assessed in terms of pressure drop, antimicrobial activity, filtration efficiency, major chemical components, and cytotoxicity. Pressure drop and antimicrobial activity increased as a function of nanoparticle deposition time (590, 855, and 1150 µg/cm2(filter) at 3-, 6-, and 9-min depositions, respectively). In filter tests, the antimicrobial efficacy was greater against Staphylococcus epidermidis than Micrococcus luteus; ~61, ~73, and ~82% of M. luteus cells were inactivated on filters that had been coated for 3, 6, and 9 min, respectively, while the corresponding values were ~78, ~88, and ~94% with S. epidermidis. Although statistically significant differences in filtration performance were not observed between samples as a function of deposition time, the average filtration efficacy was slightly higher for S. epidermidis aerosols (~97%) than for M. luteus aerosols (~95%). High-performance liquid chromatography (HPLC) and electrospray ionization-tandem mass spectrometry (ESI/MS) analyses confirmed that the major chemical compounds in the E. japonica extract were 1(ß)-O-galloyl pedunculagin, quercetin-3-O-glucuronide, and kaempferol-3-O-glucoside. In vitro cytotoxicity and disk diffusion tests showed that E. japonica nanoparticles were less toxic and exhibited stronger antimicrobial activity toward some bacterial strains than a reference soluble nickel compound, which is classified as a human carcinogen. This study provides valuable information for the development of a bioaerosol control system that is environmental friendly and suitable for use in indoor environments.

Project description:There has been discrepancies between the daily air quality reports of the Beijing municipal government, observations recorded at the U.S. Embassy in Beijing, and Beijing residents' perceptions of air quality. This study estimates Beijing's daily area PM(2.5) mass concentration by means of a novel technique SPA (Single Point Areal Estimation) that uses data from the single PM(2.5) observation station of the U.S Embassy and the 18 PM(10) observation stations of the Beijing Municipal Environmental Protection Bureau. The proposed technique accounts for empirical relationships between different types of observations, and generates best linear unbiased pollution estimates (in a statistical sense). The technique extends the daily PM(2.5) mass concentrations obtained at a single station (U.S. Embassy) to a citywide scale using physical relations between pollutant concentrations at the embassy PM(2.5) monitoring station and at the 18 official PM(10) stations that are evenly distributed across the city. Insight about the technique's spatial estimation accuracy (uncertainty) is gained by means of theoretical considerations and numerical validations involving real data. The technique was used to study citywide PM(2.5) pollution during the 423-day period of interest (May 10, 2010 to December 6, 2011). Finally, a freely downloadable software library is provided that performs all relevant calculations of pollution estimation.
