First report of the root parasite Cansjera rheedei (Santalales: Opiliaceae) in Taiwan.

ABSTRACT:

Background

The family Opiliaceae in Santalales comprises approximately 38 species within 12 genera distributed worldwide. In Taiwan, only one species of the tribe Champereieae, Champereia manillana, has been recorded. Here we report the first record of a second member of Opiliaceae, Cansjera in tribe Opilieae, for Taiwan.

New information

The newly-found species, Cansjera rheedei J.F. Gmelin (Opiliaceae), is a liana distributed from India and Nepal to southern China and western Malaysia. This is the first record of both the genus Cansjera and the tribe Opilieae of Opiliaceae in Taiwan. In this report, we provide a taxonomic description for the species and colour photographs to facilitate identification in the field.

First report of root-lesion nematode, Pratylenchus oleae from pistachio in Iran.

Project description:Pistachio, Pistacia vera is one of the most important cash crops in Iran that is scattered in arid and semi-arid regions. During a survey of plant-parasitic nematodes of pistachio in Ardakan city in Yazd Province, a species of root-lesion nematode was isolated and identified by morphological, morphometrical, and molecular methods as Pratylenchus oleae Palomares-Rius et al., 2014. This species was isolated from several pistachio trees rhizosphere regarding to molecular analysis, D2-D3 expansion segments of 28S rRNA was amplified by PCR and sequenced. The sequence was deposited in GenBank (Accession No. MW338666). Along with the related phylogenetic analysis, placed this species in a monophyletic clade with other Pratylenchus oleae isolates, based on Bayesian inference (BI) phylogeny. A PCR reaction with the P. oleae specific primer set produced a 547 bp fragment. This is the first report of P. oleae infecting Pistachio tree in the world.

First report of leopard fossils from a limestone cave in Kenting area, southern Taiwan.

Project description:Longshia-dong Cave, a limestone cave located in the Kenting area within the Kenting National Park of southern Taiwan, yields numerous terrestrial mammalian fossils. Many of them were not reported in historical literature and are neither present in Taiwan. For instance, no historical literature mentioned leopards inhabited in Taiwan, and thus their existence remained unknown. This study describes three fossil leopard (Panthera pardus) teeth uncovered from Longshia-dong Cave. Two isolated lower premolars and one lower molar, respectively p3, p4 and m1, were discovered in a very small area (11 × 6 cm) and show a series of progressive increase in size. Thus, the three teeth should have been belonging to the same individual from the subfamily of Pantherinae. Traditional linear measurements and two-dimensional geometric morphometric analysis for the occlusal surface outlines were conducted on the fossil teeth and extant pantherines inhabited in Asia such as clouded leopards (Neofelis nebulosa), leopards (Panthera pardus), and tigers (Panthera tigiris). Results show that the fossil teeth are similar both in size and morphology to the teeth of extant leopards, suggesting the assignment of the fossil teeth to leopards. This study, for the first time, reported the presence of leopards in the Late Pleistocene of Taiwan. In addition, the smaller size of the fossil teeth in comparison with Chinese fossil leopards is putatively attributed to insular dwarfism or individual size variability, yet more studies are required.

First Report of Root Rot Caused by Plectosphaerella cucumerina on Cabbage in China.

Project description:Severe root rot was observed in fields of cabbages (Brassica oleracea L.) in 2015 in China. Cardinal symptoms of this disease included root rot and wilting leaves. A fungus was isolated from diseased tissues consistently. Based on the morphological features and molecular analysis of the ITS-5.8S rDNA and D1/D2 domain of the 28S rRNA gene, it was identified as Plectosphaerella cucumerina. This is the first report of P. cucumerina causing cabbage root rot in China and the world.

First Report of Root Rot Caused by Phytophthora bilorbang on Olea europaea in Italy.

Project description:Leaf chlorosis, severe defoliation and wilt associated with root rot were observed on mature olive trees cv. Nera di Gonnos in an experimental orchard at Mirto Crosia (Calabria, southern Italy). An oomycete was consistently isolated from rotten roots of symptomatic olive trees. It was identified as Phytophthora bilorbang by morphological characters and sequencing of Internal Transcribed Spacer (ITS) regions of ribosomal DNA (rDNA). Pathogenicity was verified by inoculating potted two-month-old rooted cuttings of Olea europaea var. Nera di Gonnos in a soil infestation trial. P. bilorbang was re-isolated from roots of symptomatic, artificially inoculated olive cuttings to fulfill Koch's postulates. This is the first report of P. bilorbang on O. europaea L. and on a species of the Oleaceae family worldwide.

First report of diazotrophic Brevundimonas spp. as growth enhancer and root colonizer of potato.

Project description:Rhizobacteria contain various plant-beneficial traits and their inoculation can sustainably increase crop yield and productivity. The present study describes the growth-promoting potential of Brevundimonas spp. isolated from rhizospheric soil of potato from Sahiwal, Pakistan. Four different putative strains TN37, TN39, TN40, and TN44 were isolated by enrichment on nitrogen-free malate medium and identified as Brevundimonas spp. based on their morphology, 16S rRNA gene sequence, and phylogenetic analyses. All strains contained nifH gene except TN39 and exhibited nitrogen fixation potential through acetylene reduction assay (ARA) except TN40. Among all, the Brevundimonas sp. TN37 showed maximum ARA and phosphate solubilization potential but none of them exhibited the ability to produce indole acetic acid. Root colonization studies using transmission electron microscopy and confocal laser scanning microscopy showed that Brevundimonas sp. TN37 was resident over the root surface of potato; forming sheets in the grooves in the rhizoplane. TN37, being the best among all was further evaluated in pot experiment using potato cultivar Kuroda in sterilized sand. Results showed that Brevundimonas sp. TN37 increased growth parameters and nitrogen uptake as compared to non-inoculated controls. Based on the results obtained in this study, it can be suggested that Brevundimonas spp. (especially TN37) possess the potential to improve potato growth and stimulate nitrogen uptake. This study is the first report of Brevundimonas spp. as an effective PGPR in potato.

First report of northern root-knot nematode, Meloidogyne hapla (Chitwood, 1949) on strawberry in Turkey.

Project description:Strawberry is one of the most economically important crops worldwide. Several species of plant-parasitic nematodes have been reported to be pathogenic on strawberries, among them the northern root-knot nematode (Meloidogyne hapla), which considered to be strawberry most important nematode pest worldwide. In August 2019, strawberry growers at Silifke (Mersin, Turkey) identified nematode-like symptoms on strawberry roots and infected seedlings were brought to the nematology laboratory at of Mersin University for diagnostics. Roots were separated into small pieces and nematode extraction was performed by a modified Baermann funnel method and identified under the microscope. DNA was extracted from individual nematodes using Worm Lysis Buffer(WLB (+)). The species-specific SCAR markers (JMV1, JMV2, and JMVhapla) yielded a 440 bp band specific to M. hapla. The 28S rRNA gene region, obtained using the general primers D2\D3, sequence was analysed from. The analyzed sequence was 100% identicle to M. hapla. The gene sequences were deposited into GenBank database with accession numbers MN897751 and MN895037. Both morphological and molecular diagnostic methods confirmed that the strawberry plants collected in Silifke were infested with M. hapla. To our best knowledge this is the first report of plant-parasitic nematode species M. hapla infecting strawberry in Turkey. Currently, the adverse effect of RKN on strawberry production in the region is unknown to strawberry growers.

Comparative Plastome Analysis of Root- and Stem-Feeding Parasites of Santalales Untangle the Footprints of Feeding Mode and Lifestyle Transitions.

Project description:In plants, parasitism triggers the reductive evolution of plastid genomes (plastomes). To disentangle the molecular evolutionary associations between feeding on other plants below- or aboveground and general transitions from facultative to obligate parasitism, we analyzed 34 complete plastomes of autotrophic, root- and stem-feeding hemiparasitic, and holoparasitic Santalales. We observed inexplicable losses of housekeeping genes and tRNAs in hemiparasites and dramatic genomic reconfiguration in holoparasitic Balanophoraceae, whose plastomes have exceptionally low GC contents. Genomic changes are related primarily to the evolution of hemi- or holoparasitism, whereas the transition from a root- to a stem-feeding mode plays no major role. In contrast, the rate of molecular evolution accelerates in a stepwise manner from autotrophs to root- and then stem-feeding parasites. Already the ancestral transition to root-parasitism coincides with a relaxation of selection in plastomes. Another significant selectional shift in plastid genes occurs as stem-feeders evolve, suggesting that this derived form coincides with trophic specialization despite the retention of photosynthetic capacity. Parasitic Santalales fill a gap in our understanding of parasitism-associated plastome degeneration. We reveal that lifestyle-genome associations unfold interdependently over trophic specialization and feeding mode transitions, where holoparasitic Balanophoraceae provide a system for exploring the functional realms of plastomes.

The first archaic Homo from Taiwan.

Project description:Recent studies of an increasing number of hominin fossils highlight regional and chronological diversities of archaic Homo in the Pleistocene of eastern Asia. However, such a realization is still based on limited geographical occurrences mainly from Indonesia, China and Russian Altai. Here we describe a newly discovered archaic Homo mandible from Taiwan (Penghu 1), which further increases the diversity of Pleistocene Asian hominins. Penghu 1 revealed an unexpectedly late survival (younger than 450 but most likely 190-10 thousand years ago) of robust, apparently primitive dentognathic morphology in the periphery of the continent, which is unknown among the penecontemporaneous fossil records from other regions of Asia except for the mid-Middle Pleistocene Homo from Hexian, Eastern China. Such patterns of geographic trait distribution cannot be simply explained by clinal geographic variation of Homo erectus between northern China and Java, and suggests survival of multiple evolutionary lineages among archaic hominins before the arrival of modern humans in the region.

Nematode species were identified using both molecular analysis and morphology of perineal patterns. Meloidogyne enterolobii and M. javanica were identified from U. parvifolia cv. Allee. Meloidogyne arenaria and M. javanica were identified from U. parvifolia cv. Drake.
