Irlactane and Tremulane Sesquiterpenes from the Cultures of the Medicinal Fungus Irpex lacteus HFG1102.

ABSTRACT: A new irlactane-type, namely irlactin K (1), and 22 tremulane-type sesquiterpenes including fourteen previously undescribed ones, namely irpexolactins A-N (2-15), and a known irlactane-type sesquiterpenoid, were isolated from the fermentation broth of the medicinal fungus Irpex lacteus HFG1102. The structures of all the isolates were characterized by extensive spectroscopic methods, including 1D and 2D NMR and MS spectroscopic analysis. The absolute configurations of irlactin K and the known compound conocenol B (20) were established by single-crystal X-ray diffraction analysis. The vasorelaxant effects of irlactin K (1), irpexolactins A (2), C (4), K (12), and irlactam (22) were evaluated.

Project description:Eight new furan derivatives, irpexins A‒H (1‒8), two new polyketides, irpexins I and J (9 and 10), together with nine known compounds were isolated from the fermentation of Irpex lacteus. All compounds shows no cytotoxicity to human MCF-7 and Hela cancer cell lines at the concentration of 10 μM.

Irpexoates A-D, Four Triterpenoids with Malonyl Modifications from the Fruiting Bodies of the Medicinal Fungus Irpex lacteus.

Project description:Four eburicane-type triterpenoids with malonyl modifications, namely irpexoates A-D (1-4), were isolated from the fruiting bodies of the medicinal fungus Irpex lacteus. The structures of the new compounds were established by extensive spectroscopic methods, including 1D and 2D NMR, HRESIMS spectroscopic analysis. Irpexoate B (2) displayed weak cytotoxicity against four human cancer cell lines (A-549, SMMC-7721, MCF-7, SW480) with IC50 values varying from 22.9 to 34.0 ?M, and irpexoate D (4) showed weak cytotoxicity against the human cancer cell line SW480 with an IC50 value of 35.2 ?M.

Characterization of Acetylxylan Esterase from White-Rot Fungus Irpex lacteus.

Project description:The carbohydrate esterase family 1 (CE1) in CAZy contains acetylxylan esterases (AXEs) and feruloyl esterases (FAEs). Here we cloned a gene coding for an AXE belonging to CE1 from Irpex lacteus (IlAXE1). IlAXE1 was heterologously expressed in Pichia pastoris, and the recombinant enzyme was purified and characterized. IlAXE1 hydrolyzed p-nitrophenyl acetate, α-naphthyl acetate and 4-methylumbelliferyl acetate, however, it did not show any activity on ethyl ferulate and methyl p-coumarate. We also examined the activity on partially acetylated and feruloylated xylan extracted from corncob by hydrothermal reaction. Similarly, ferulic and p-coumaric acids were not liberated, and acetic acid was only detected in the reaction mixture. The results indicated that IlAXE1 is an acetylxylan esterase actually reacted to acetyl xylan. However, since IlAXE1 was unable to completely release acetic acid esterifying xylopyranosyl residues, it is assumed that acetyl groups exhibiting resistance to deacetylation by IlAXE1 are present in corn cob xylan.

Genome sequence of the white-rot fungus Irpex lacteus F17, a type strain of lignin degrader fungus.

Project description:Irpex lacteus, a cosmopolitan white-rot fungus, degrades lignin and lignin-derived aromatic compounds. In this study, we report the high-quality draft genome sequence of I. lacteus F17, isolated from a decaying hardwood tree in the vicinity of Hefei, China. The genome is 44,362,654 bp, with a GC content of 49.64% and a total of 10,391 predicted protein-coding genes. In addition, a total of 18 snRNA, 842 tRNA, 15 rRNA operons and 11,710 repetitive sequences were also identified. The genomic data provides insights into the mechanisms of the efficient lignin decomposition of this strain.

Deciphering lignocellulose deconstruction by the white rot fungus Irpex lacteus based on genomic and transcriptomic analyses.

Project description:BackgroundIrpex lacteus is one of the most potent white rot fungi for biological pretreatment of lignocellulose for second biofuel production. To elucidate the underlying molecular mechanism involved in lignocellulose deconstruction, genomic and transcriptomic analyses were carried out for I. lacteus CD2 grown in submerged fermentation using ball-milled corn stover as the carbon source.ResultsIrpex lacteus CD2 efficiently decomposed 74.9% lignin, 86.3% cellulose, and 83.5% hemicellulose in corn stover within 9 days. Manganese peroxidases were rapidly induced, followed by accumulation of cellulase and hemicellulase. Genomic analysis revealed that I. lacteus CD2 possessed a complete set of lignocellulose-degrading enzyme system composed mainly of class II peroxidases, dye-decolorizing peroxidases, auxiliary enzymes, and 182 glycoside hydrolases. Comparative transcriptomic analysis substantiated the notion of a selection mode of degradation. These analyses also suggested that free radicals, derived either from MnP-organic acid interplay or from Fenton reaction involving Fe2+ and H2O2, could play an important role in lignocellulose degradation.ConclusionsThe selective strategy employed by I. lacteus CD2, in combination with low extracellular glycosidases cleaving plant cell wall polysaccharides into fermentable sugars, may account for high pretreatment efficiency of I. lacteus. Our study also hints the importance of free radicals for future designing of novel, robust lignocellulose-degrading enzyme cocktails.

New Sesquiterpenoids From Plant-Associated Irpex lacteus.

Project description:Bacteria produce a large number of virulence factors through the quorum sensing (QS) mechanism. Inhibiting such QS system of the pathogens without disturbing their growth is a potential strategy to control multi-drug-resistant pathogens. To accomplish this, two new tremulane-type sesquiterpenoids, irpexolaceus H (1) and I (2), along with two known furan compounds, irpexlacte B (3) and C (4), were isolated from Orychophragmus violaceus (L.) OE Schulz endophytic fungus Irpex lacteus (Fr.) Fr. Their structures were elucidated by detailed spectroscopic data (NMR, HRESIMS, IR, and UV), single-crystal X-ray diffraction, and electronic circular dichroism (ECD) analysis. Furthermore, those compounds were evaluated for anti-quorum sensing (anti-QS) activity, and compound 3 was found contributing to the potential QS inhibitory activity.

Steroids and Sesquiterpenes From Cultures of the Fungus Phellinus igniarius.

Project description:Two new steroids, 3α,17α,19,20-tetrahydroxy-4α-methylpregn-8-ene (1) and 3α,12α,17α,20-tetrahydroxy-4α-methylpregn-8-ene (2) and three new sesquiterpenoids, 12-hydroxy-α-cadinol (3), 3α,12-dihydroxy-δ-cadinol (4), and 3α,6α-dihydroxyspiroax-4-ene (5), have been isolated from cultures of the fungus Phellinus igniarius. Their structures were characterized based on extensive spectroscopic data. In preliminary in vitro assays, compounds 3 and 4 exhibited the vascular-activities against phenylephrine-induced vasoconstriction with the relaxing rates of 11.0 % and 7.0 % at 3 × 10-4 M, respectively.
